| 1 | Cordycepin protects renal ischemia/reperfusion injury through regulating inflammation,apoptosis,and oxidative stress显示文摘Cordycepin(3-deoxyadenosine)is a naturally occurring adenosine analog and one of the bioactive constituents isolated from Cordyceps sinensis,species of the fungal genus Cordyceps.It has traditionally been a prized Chinese folk medicine for the human well-being.However,the actions of cordycepin against renal ischemia/reperfusion injury(/R)are still unknown.In the present study,rats were subject to VR and cordycepin was intragastrically administered for seven consecutive days before surgery to investigate the effects and mechanisms of cordycepin against renal/R injury.The test results of kidney and peripheral blood samples of experimental animals showed that cordycepin significantly decreased serum blood urea nitrogen and creatinine levels and markedly attenuated cell injury.Mechanistic studies showed that cordycepin significantly regulated inflammation,apoptosis,and oxidative stress.These data provide new insights for investigating the natural product with the nephroprotective effect against I/R,which should be developed as a new therapeutic agent for the treatment of I/R in the future. | Feng Han Meng Dou Yuxiang Wang Cuixiang Xu Yang Li Xiaoming Ding Wujun Xue Jin Zheng Puxun Tian Chenguang Ding | 2020 | Acta Biochimica et Biophysica Sinica2020,52,2: | 9 |

| 3 | miR-124/IRE-1α affects renal ischemia/reperfusion injury by regulating endoplasmic reticulum stress in renal tubular epithelial cells显示文摘Acute kidney injury(AKI)refers to a clinical syndrome that occurs as a result of a rapid decline in renal function caused by multiple factors.Renal ischemia/reperfusion(/R)injury is one of the main causes of AKI and has a high incidence and mortality,However,the specific pathogenesis of renal I/R injury is still unclear.In recent years,a major breakthrough has been made in the study of endoplasmic reticulum stress(ERS)-mediated apoptosis in I/R injury.It has been reported that miRNAs play protective roles in ischemic/reperfused organs,but the molecular mechanisms have not been investigated deeply.In this study,the renal VR mouse model was used to explore the roles of miR-124 in ERS and in renal I/R injury.The western blot results showed that the expression levels of ERS-related proteins IRE-1α,XBP-1,and glucose-regulated protein 78(GRP78)were significantly increased in the IVR model group when compared with those in the control group.Meanwhile,qPCR results showed that miR-124 expression was decreased in the I/R injury model,and overexpression of miR-124 using miR-124 mimics effectively reduced the expression of ERS-related proteins and alleviated renal LR injury.In addition,luciferase reporter assay was performed,and the results showed that IRE 1αand miR-124 may have direct interaction.In conclusion,our data indicated that miR-124 was a negative regulator of ERS via binding to IRE-1α,ultimately conferring its protective effect on the kidney,which demonstrates the regulatory mechanism of miR-124 in renal I/R injury and provides new ideas and methods for the prevention and treatment of renal I/R injury. | Chenguang Ding Meng Dou Yuxiang Wang Yang Li Ying Wang Jin Zheng Xiao Li Wujun Xue Xiaoming Ding Puxun Tian | 2020 | Acta Biochimica et Biophysica Sinica2020,52,2: | 4 |
| 4 | The effects of diltiazem in renal transplantation patients treated with cyclosporine A显示文摘Objective:To investigate the effects of diltiazem and cyclosporine A(CsA)combination therapy on protecting the kidney,promoting graft functioning and improving post-transplanted kidney recovery.Methods:The blood concentrations of CsA,the condition of the post-transplant kidney,the rate of acute rejection(AR),as well as hepatic and renal toxicity in 636 cases of renal transplant recipients were determined after being treated by CsA,with or without diltiazem.Results:Compared with the control group which received CsA,mycophenolate mofetil(MMF) and prednisolone(Pred)but lacked diltiazem,the group receiving these agents together with diltiazem required reduced dosage of CsA(P<0.01),while blood concentrations of CsA were significantly increased(P<0.01);the recovery time of graft function was reduced from(6.2±1.5)d to(3.9±1.4)d(P<0.01),and the rate of AR was decreased from 13.2%to 7.9%(P<0.01).Conclusion:In renal transplantation patients treated with CsA and diltiazem,blood concentrations of CsA were increased while the dosage was decreased.This efficient combination therapy reduced patients'economic burden,at the same time retained kidney function,promoted graft function recovery and decreased hepatic and renal toxicity and the rate of AR. | Wujun Xue Yong Song Puxun Tian Xiaoming Ding Xiaoming Pan Hang Yan Jun Hou Xinshun Feng Heli Xiang Xiaohui Tian | 2010 | The Journal of Biomedical Research2010,24,4: | 4 |

| 8 | Predictive value of hypothermic machine perfusion parameters combined perfusate biomarkers in deceased donor kidney transplantation显示文摘Background:Delayed graft function(DGF)is the main cause of renal function failure after kidney transplantation.This study aims at investigating the value of hypothermic machine perfusion(HMP)parameters combined with perfusate biomarkers on predicting DGF and the time of renal function recovery after deceased donor(DD)kidney transplantation.Methods:HMP parameters,perfusate biomarkers and baseline characteristics of 113 DD kidney transplantations from January 1,2019 to August 31,2019 in the First Affiliated Hospital of Xi’an Jiaotong University were retrospectively analyzed using univariate and multivariate logistic regression analysis.Results:In this study,the DGF incidence was 17.7%(20/113);The multivariate logistic regression results showed that terminal resistance(OR:1.879,95%CI 1.145-3.56)and glutathione S-transferase(GST)(OR=1.62,95%CI 1.23-2.46)were risk factors for DGF;The Cox model analysis indicated that terminal resistance was an independent hazard factor for renal function recovery time(HR=0.823,95%CI 0.735-0.981).The model combining terminal resistance and GST(AUC=0.888,95%CI:0.842-0.933)significantly improved the DGF predictability compared with the use of terminal resistance(AUC=0.756,95%CI 0.693-0.818)or GST alone(AUC=0.729,95%CI 0.591-0.806).Conclusion:According to the factors analyzed in this study,the combination of HMP parameters and perfusate biomarkers displays a potent DGF predictive value. | Yuxi Qiao Chenguang Ding Yang Li Xiaohui Tian Puxun Tian Xiaoming Ding Heli Xiang Jin Zheng Wujun Xue | 2022 | Chinese Medical Journal2022,,2: | 1 |
| 9 | Daclizumab prevents acute renal allograft rejection: 1 year analysis显示文摘Objective: To investigate the clinical effect of Daclizumab on preventing acute rejection in renal transplant recipients.Methods: 71 patients were randomly divided into two groups: Daclizumab group (n =26) and control group (n = 45). Baselineregimen of mycophenolate mofetil(MMF), cyclosporin(CsA), methylprednisolone(MPD) and prednisone(Pred) were adminis-tered to all patients. The treatment of Daclizumab was based on baseline regimen. The Daclizumab group received Daclizumabtwice before and after renal transplant. The occurrence of post-transplantation acute rejection, renal function and T lymphocytesubtypes were sequentially monitored; meanwhile adverse events, infection episode, and patient and graft survival were observed.All of patients received a follow-up of 12 months at least. Results: The occurrence of acute rejection in Daclizumab group in 1,3, 6 and 12 months after renal transplantation was 7.7%, 19.2%, 23.1% and 30.8%, respectively, while it was 15.6%, 28.9%,35.6% and 46.7% in the control group. There was significant difference between the two group(P < 0.05). There was no differencein infection episodes and adverse events between the Daclizumab group and control group. One year patient survival was 92.3% inDaclizumab group, 91.1 % in control group (P > 0.05), compared with graft survival of 96.2 % and 93.3 % for Daclizumab andcontrol group, respectively (P > 0. 05). The renal function in Daclizumab group in 1, 6 and 12 months after renal transplantationwas better than that in control group (P < 0.05). The CD3+ and CD4+ subtypes decreased in both two groups after operation butno significant difference(P > 0.05). | 14 | Effects of mature Sertoli cells on allogeneic islets cocultured in vitro显示文摘Objective: To set up a method for isolation and culture of mature Sertoli cells and to estimate their effects on allogeneic islets cocultured in vitro. Methods: Adult SD rat testicular Sertoli cells were prepared successfully by three-step enzyme digestion. Then they were cocultured respectively with allogeneic islets and activated Wistar rat splenocytes. 24-hour cumulative insulin release and glucose-stimulated insulin secretion test were performed to detect islet function between pure islets culture group and coculture group. Splenocyte proliferation activity was determined by MTT colorimetry assay to observe the inhibition effect of Sertoli cells in different densities. Result: Firstly, in pure islet culture group, the 24-hour cumulative insulin release was gradually decreased in 21-day culture time. Compared to day 3, this change was significant on day 7 (P < 0.05) and on day 10,14,21(P < 0.01). In contrast, in coculture group, compared to day 3, the 24-hour cumulative insulin release was increased significantly on day 7 (P < 0.01), and then gradually decreased on day 10 and 14, but still higher than that of day 3. It was on day 21 that it began to decrease compared to day 3(P < 0.05). During the culture time in vitro, the 24-hour cumulative insulin release of islet coculture group was significantly higher than that of pure islets culture group(P < 0.01). In the case of stimulation index(SI), there was a similar tendency as insulin release in the two groups. Secondly, mature Sertoli cells(1×106/mL) pretreated by 15 grays irradiation could decrease proliferation activity of activated splenocytes compared to that of control group(P < 0.01). This inhibition effect was dose-dependent. Conclusion: Mature Sertoli cells can improve the function and prolong the survival of islet cells cultured in vitro. They can also provide an immune protection to islet cells.
