1Three-dimensional (3D) hydrogel serves as a platform to identify potential markers of chondrocyte dedifferentiation by combining RNA sequencing显示文摘Dedifferentiation of chondrocyte greatly restricts its function and application,however,it is poorly understood except a small number of canonical markers.The non-cell-adhesive property endows polysaccharide hydrogel with the ability to maintain chondrocyte phenotype,which can serve as a platform to identify new molecular markers and therapeutic targets of chondrocyte dedifferentiation.In this study,the high-throughput RNA sequencing(RNA-seq)was first performed on articular chondrocytes at primary(P0)and passage 1(P1)stages to explore the global alteration of gene expression along with chondrocyte dedifferentiation.Significantly,several potential marker genes,such as PFKFB3,KDM6B,had been identified via comparatively analyzing their expression in P0 and P1 chondrocytes as well as in 3D constructs(i.e.chondrocyte-laden alginate hydrogel and HA-MA hydrogel)at both mRNA and protein level.Besides,the changes in cellular morphology and enriched pathway of differentially expressed genes during chondrocyte dedifferentiation was studied in detail.This study developed the use of hydrogel as a platform to investigate chondrocyte dedifferentiation;the results provided new molecular markers and potential therapeutic targets of chondrocyte dedifferentiation.Yang Ling Weiyuan Zhang Peiyan Wang Wanhua Xie Wei Yang Dong-An Wang Changjiang Fan 2021Bioactive Materials2021,6,9:3

3Scriptaid affects histone acetylation and the expression of development-related genes at different stages of porcine somatic cell nuclear transfer embryo during early development显示文摘Although the somatic cell nuclear transfer(SCNT) technique has been used extensively for cloning and generating transgenic pigs,the cloning efficiency is still very low.It has been proposed that the low efficiency of this technique is the result of incomplete epigenetic reprogramming and abnormal gene expression during early embryonic development.In this study,we investigate the effect of Scriptaid,a low-toxicity histone deacetylase inhibitor,on the developmental competence of porcine SCNT embryos.We found that treating SCNT embryos with 500 nmol/L Scriptaid for 15 h after activation significantly enhanced the blastocyst formation rate(27.7%) compared with the untreated group(control)(12.2%,P<0.05).Using an immunofluorescence technique to measure the average fluorescence intensity,we also found that treating SCNT embryos with Scriptaid increased the level of histone acetylation on histone H3 at lysine 14(acH3K14).Furthermore,treating embryos with Scriptaid increased the expression level of three genes that play important roles during embryonic development(Oct4,Klf4 at the blastocyst stage and Nanog at the 4-cell stage).Moreover,the expression level of the apoptosis-related gene Caspase-3 was significantly lower in the Scriptaid-treated SCNT embryos compared with the control SCNT embryos at the 4-cell and blastocyst stages.In conclusion,these results indicate that Scriptaid treatment improves the development and nuclear reprogramming of porcine SCNT embryos.ZHOU Yan HUANG YongYe XIE WanHua SONG Qi JI Yuan ZHANG YanPing OUYANG HongSheng LAI LiangXue PANG DaXin TANG XiaoChun 2013Chinese Science Bulletin2013,58,18:1
4Photochemical graft of γ-cyclodextrin's interior leading to in-situ charge-transfer complexes with unusual regioselectivity and its application in 3D photo-printing显示文摘Photoirradiation of anthraquinone(AQ)derivatives withγ-cyclodextrin(γ-CD)in aqueous solution led to aγ-CD derivative with a 9,10-dihydroxylanthracene grafting unprecedentedly at the 5′-position of one glucoside ofγ-CD in high efficiency.Meanwhile,another AQ enters into the remaining space of the 9,10-dihydroxylanthracene-occupiedγ-CD cavity to in-situ form a selfincluded quinhydrone charge transfer(CT)complex.The structure of the supramolecular CT complex was unambiguously demonstrated by 1 D and 2 D NMR(^(1)H NMR,^(13)C NMR,DEPT,^(1)H-^(1)H COSY,HSQC,HMBC,TOCSY,and ROESY)spectroscopies,which revealed a highly stereoselective stacking of the quinhydrone pairs in theγ-CD cavity.The supramolecular CT complex showed an unusually high binding affinity and improved oxidation resistance.Polyacrylamides bearingγ-CD or anthraquinone were respectively synthesized by co-polymerization of acrylamide with the corresponding monomer.Photoirradiation of aqueous mixtures containing these two types of polymers led to gelation,which was applied to 3 D photo-printing of soft materials.Dongjing Zhang Wenting Liang Jigao Yi Jinlei Chen Yongjun Lv Ting Zhao Chao Xiao Xiangyu Xie Wanhua Wu Cheng Yang 2022Science China Chemistry2022,65,6:0
