1Gastric cancer: Epidemiology, risk factors and prevention strategies显示文摘Gastric cancer(GC) is a global health problem, with more than 1 million people newly diagnosed with GC worldwide each year. GC is more prevalent in less developed countries than in more developed countries. About half of all GC cases worldwide occur in East Asia, notably China. Globally, overall incidence rates of GC are declining, which is potentially attributed to a decrease in Helicobacter pylori(H. pylori) infection and the use of refrigeration to preserve foods rather than salt. GC is a multifactorial disease, and its occurrence and development were impacted by environmental and genetic factors. H. pylori infection is the primary risk factor for GC, especially for non-cardia. The prognosis of GC is poor due to stages at the first diagnosis. The 5-year survival rate is less than 10% when patients are diagnosed at an advanced stage, but the rate is as high as 85% if patients are detected at an earlier stage. Endoscopic screening can potentially prevent GC by early diagnosis and early treatment and has been widely adopted in screening programs in East Asian countries, such as Japan and Korea. 2The TanSat mission: preliminary global observations显示文摘The Chinese global carbon dioxide monitoring satellite (Tan Sat) was launched successfully in December2016 and has completed its on-orbit tests and calibration. Tan Sat aims to measure the atmospheric column-averaged dry air mole fractions of carbon dioxide (XCO_2) with a precision of 4 ppm at the regional scale, and in addition, to derive global and regional CO2fluxes. Progress towards these objectives is reviewed and the first scientific results from Tan Sat measurements are presented. Tan Sat on-orbit tests indicate that the Atmospheric Carbon dioxide Grating Spectrometer is in normal working status and is beginning to produce L1B products. The preliminary Tan Sat XCO_2products have been retrieved by an algorithm and compared to NASA Orbiting Carbon Observatory-2 (OCO-2) measurements during an overlapping observation period. 4Zika virus infection induces RNAi-mediated antiviral immunity in human neural progenitors and brain organoids显示文摘The re-emergence of Zika virus(ZIKV)in the Western Hemisphere has resulted in global public health crisis since 2015.ZIKV preferentially infects and targets human neural progenitor cells(hNPCs)and causes fetal microcephaly upon maternal infection.hNPCs not only play critical roles during fetal brain development,but also persist in adult brain throughout life.Yet the mechanism of innate antiviral immunity in hNPCs remains largely unknown.Here,we show that ZIKV infection triggers the abundant production of virus-derived small interfering RNAs in hNPCs,but not in the more differentiated progenies or somatic cells.Ablation of key RNAi machinery components significantly enhances ZIKV replication in hNPCs.Furthermore,enoxacin,a broad-spectrum antibiotic that is known as an RNAi enhancer,exerts potent anti-ZIKV activity in hNPCs and other RNAi-competent cells.Strikingly,enoxacin treatment completely prevents ZIKV infection and circumvents ZIKV-induced microcephalic phenotypes in brain organoid models that recapitulate human fetal brain development.Our findings highlight the physiological importance of RNAi-mediated antiviral immunity during the early stage of human brain development,uncovering a novel strategy to combat human congenital viral infections through enhancing RNAi.Yan-Peng Xu Yang Qiu Boya Zhang Guilai Chen Qi Chen Miao Wang Fan Mo Jiuyue Xu Jin Wu Rong-Rong Zhang Meng-Li Cheng Na-Na Zhang Bao Lyu Wen-Liang Zhu Meng-Hua Wu Qing Ye Da Zhang Jiang-Hong Man Xiao-Feng Li Jie Cui Zhiheng Xu Baoyang Hu Xi Zhou Cheng-Feng Qin 2019Cell Research2019,29,4:8

6Spatial Variation of Dissolved Organic Carbon in Soils of Riparian Wetlands and Responses to Hydro-geomorphologic Changes in Sanjiang Plain, China显示文摘Spatial variation of dissolved organic carbon(DOC) in soils of riparian wetlands and responses to hydro-geomorphologic changes in the Sanjiang Plain were analyzed through in situ collecting soil samples in the Naoli River and the Bielahong River. The results showed that the average contents of DOC for soil layer of 0–100 cm were 730.6 mg/kg, 250.9 mg/kg, 423.0 mg/kg and 333.1 mg/kg respectively from riverbed to river terrace along the transverse directions of the Naoli watershed. The content of the soil DOC was the highest in the riverbed, lower in the high floodplain and much lower in the river terrace, and it was the lowest in the low floodplain. The difference in the content and vertical distribution of DOC between the riverbed and the three riparian wetlands was significant, while it was not significant among the low floodplain, the high floodplain and the river terrace. The variability of soil DOC was related to the hydrological connectivity between different landscape position of the riparian wetlands and the adjacent stream. Extremely significant correlations were observed between DOC and total organic carbon(TOC), total iron(TFe), ferrous iron(Fe(II)) whose correlation coefficients were 0.819, –0.544 and –0.709 in riparian wetlands of the Naoli River. With the increase of wetland destruction, soil p H increased and soil DOC content changed. The correlation coefficients between soil DOC and TOC, TFe, Fe(II) also changed into 0.759, –0.686 and –0.575 respectively in the Bielahong River. Under the impact of drainage ditches, the correlations between soil DOC and TFe, Fe(II) were not obvious, while the soil p H was weakly alkaline and was negatively correlated with soil DOC in the previous high floodplain. 8Source Contributions to PM2.5 under Unfavorable Weather Conditions in Guangzhou City,China显示文摘Historical haze episodes(2013–16) in Guangzhou were examined and classified according to synoptic weather systems.Four types of weather systems were found to be unfavorable, among which 'foreside of a cold front'(FC) and 'sea high pressure'(SP) were the most frequent(> 75% of the total). Targeted case studies were conducted based on an FC-affected event and an SP-affected event with the aim of understanding the characteristics of the contributions of source regions to fine particulate matter(PM_(2.5)) in Guangzhou. Four kinds of contributions—namely, emissions outside Guangdong Province(super-region), emissions from the Pearl River Delta region(PRD region), emissions from Guangzhou–Foshan–Shenzhen(GFS region), and emissions from Guangzhou(local)—were investigated using the Weather Research and Forecasting–Community Multiscale Air Quality model. The results showed that the source region contribution differed with different weather systems. SP was a stagnant weather condition, and the source region contribution ratio showed that the local region was a major contributor(37%), while the PRD region, GFS region and the super-region only contributed 8%, 2.8% and 7%, respectively, to PM_(2.5) concentrations. By contrast, FC favored regional transport. The super-region became noticeable,contributing 34.8%, while the local region decreased to 12%. A simple method was proposed to quantify the relative impact of meteorology and emissions. Meteorology had a 35% impact, compared with an impact of-18% for emissions, when comparing the FC-affected event with that of the SP. 9Association between PTCH1 gene polymorphisms and chronic obstructive pulmonary disease susceptibility in a Chinese Han population:a case-control study显示文摘Background:Chronic obstructive pulmonary disease(COPD)is a leading cause of morbidity and mortality worldwide.Genome-wide association studies in non-Asian population revealed a link between COPD and mutations in the PTCH1 gene encoding Patched1,a receptor in the Hedgehog signaling pathway important for lung morphogenesis and pulmonary function.The aim of this study was to investigate the association between PTCH1 polymorphisms and the COPD risk in the Chinese Han population.Methods:We performed a case-control study including 296 patients with COPD and 300 healthy individuals.Single-nucleotide polymorphisms in the PTCH1 gene were identified and genotyped based on the linkage disequilibrium analysis in all participants.Odds ratios(ORs)and 95%confidence intervals(95%CIs)were estimated using logistic regression analysis after adjustment for age,gender,and smoking.Results:In total,28 single-nucleotide polymorphisms were identified in patients with COPD.Among them,'A'allele of rs28491365(OR:1.388,95%CI:1.055-1.827,P=0.018),and'G'alleles of rs10512248(OR:1.299,95%CI:1.021-1.653,P=0.033)and rs28705285(OR:1.359,95%CI:1.024-1.803,P=0.033;respectively)were significantly associated with an increased COPD risk.Genetic model analysis revealed that the'T/T'genotype of rs34695652 was associated with a decreased COPD risk under the recessive model(OR:0.490,95%CI:0.270-0.880,P=0.010),whereas rs28504650/rs10512248 haplotype CG was significantly associated with an increased COPD risk after adjustment for age,gender,and smoking status(OR:6.364,95%CI:1.220-33.292,P=0.028).Conclusions:The study provides a new insight into the role of PTCH1 polymorphisms in the susceptibility to COPD in the Chinese Han population.Xi Kang Ting Guo Lyu Liu Shui-Zi Ding Cheng Lei Hong Luo 2020Chinese Medical Journal2020,,17:3
10Refined spatial temporal epigenomic profiling reveals intrinsic connection between PRDM9-mediated H3K4me3 and the fate of double-stranded breaks显示文摘Meiotic recombination is initiated by the formation of double-strand breaks(DSBs),which are repaired as either crossovers(COs)or noncrossovers(NCOs).In most mammals,PRDM9-mediated H3K4me3 controls the nonrandom distribution of DSBs;however,both the timing and mechanism of DSB fate control remain largely undetermined.Here,we generated comprehensive epigenomic profiles of synchronized mouse spermatogenic cells during meiotic prophase I,revealing spatiotemporal and functional relationships between epigenetic factors and meiotic recombination.We find that PRDM9-mediated H3K4me3 at DSB hotspots,coinciding with H3K27ac and H3K36me3,is intimately connected with the fate of the DSB.Our data suggest that the fate decision is likely made at the time of DSB formation:earlier formed DSBs occupy more open chromatins and are much more competent to proceed to a CO fate.Our work highlights an intrinsic connection between PRDM9-mediated H3K4me3 and the fate decision of DSBs,and provides new insight into the control of CO homeostasis.Yao Chen Ruitu Lyu Bowen Rong Yuxuan Zheng Zhen Lin Ruofei Dai Xi Zhang Nannan Xie Siqing Wang Fuchou Tang Fei Lan Ming-Han Tong 2020Cell Research2020,30,3:3
11Mechanism of Shengmai Injection on Anti-Sepsis and Protective Activities of Intestinal Mucosal Barrier in Mice显示文摘Objective:To study the mechanism of Shengmai Injection(SMI)on anti-sepsis and protective activities of intestinal mucosal barrier.Methods:The contents of 11 active components of SMI including ginsenoside Rb1,Rb2,Rb3,Rd,Re,Rf,Rg1,Rg2,ophioposide D,schisandrol A and schisantherin A were determined using ultra-performance liquid chromatography.Fifty mice were randomly divided into the blank,the model,the low-,medium-and high-dose SMI groups(0.375,0.75,1.5 mL/kg,respectively)by random number table,10 mice in each group.On SMI group,SMI was administrated to mice daily via tail vein injection for 3 consecutive days,while the mice in the blank and model groups were given 0.1 mL of normal saline.One hour after the last SMI administration,except the blank group,the mice in other groups were intraperitoneally injected with lipopolysaccharide(LPS)saline solution(2 mL/kg)at a dosage of 5 mL/kg for development of endotoxemia mice model.The mice in the blank group were given the same volume of normal saline.Inflammatory factors including interferon-γ(INF-γ),tumor necrosis factor-α(TNF-α),interleukin(IL)-2 and IL-10 were measured by flow cytometry.Myosin light-chain kinase(MLCK),nuclear factorκB(NF-κB)levels,and change of Occludin proteins in jejunum samples were analyzed by Western blot.Results:The decreasing trends of INF-γ,TNF-α and IL-2 were found in serum of SMI treatment groups.In SMI-treated mice,the content of Occludin increased and MLCK protein decreased compared with the model group(P<0.05 or P<0.01).The content of cellular and nuclear NF-κB did not change significantly(P>0.05).Conclusion:SMI may exert its anti-sepsis activity mainly through NF-κB-pro-inflammatory factor-MLCK-TJ cascade.LU Juan YU Yue WANG Xiao-jing CHAI Rui-ping LYU Xin-kai DENG Ming-hui HU Mei-geng QI Yun CHEN Xi 2022Chinese Journal of Integrative Medicine2022,28,9:2

13Progress and Knowledge Transfer from Science to Technology in the Research Frontier of CRISPR Based on the LDA Model显示文摘Purpose:This study explores the underlying research topics regarding CRISPR based on the LDA model and figures out trends in knowledge transfer from science to technology in this area over the latest 10 years.Design/methodology/approach:We collected publications on CRISPR between 2011 and2020 from the Web of Science,and traced all the patents citing them from lens.org.15,904 articles and 18,985 patents in total are downloaded and analyzed.The LDA model was applied to identify underlying research topics in related research.In addition,some indicators were introduced to measure the knowledge transfer from research topics of scientific publications to IPC-4 classes of patents.Findings:The emerging research topics on CRISPR were identified and their evolution over time displayed.Furthermore,a big picture of knowledge transition from research topics to technological classes of patents was presented.We found that for all topics on CRISPR,the average first transition year,the ratio of articles cited by patents,the NPR transition rate are respectively 1.08,15.57%,and 1.19,extremely shorter and more intensive than those of general fields.Moreover,the transition patterns are different among research topics.Research limitations:Our research is limited to publications retrieved from the Web of Science and their citing patents indexed in lens.org.A limitation inherent with LDA analysis is in the manual interpretation and labeling of'topics'.Practical implications:Our study provides good references for policy-makers on allocating scientific resources and regulating financial budgets to face challenges related to the transformative technology of CRISPR.Originality/value:The LDA model here is applied to topic identification in the area of transformative researches for the first time,as exemplified on CRISPR.Additionally,the dataset of all citing patents in this area helps to provide a full picture to detect the knowledge transition between S&T.Yushuang Lyu Muqi Yin Fangjie Xi Xiaojun Hu 2022Journal of Data and Information Science2022,7,1:2

15Inhibitory effects of petasin on human colon carcinoma cells mediated by inactivation of Akt/mTOR pathway显示文摘Background:Colorectal cancer is the third most common cancer worldwide and still lack of effective therapy so far.Petasin,a natural product found in plants of the genus Petasites,has been reported to possess anticancer activity.The present study aimed to investigate the anticolon cancer activity of petasin both in vitro and in vivo.The molecular mechanism of petasin was also further explored.Methods:Caco-2,LoVo,SW-620,and HT-29 cell lines were used to detect the inhibitory effect of petasin on colon cancer proliferation.Cell viability was determined using the MTT (3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromide) assay.Cell apoptosis was analyzed by flow cytometry.Hoechst 33258 staining was used to visualize morphological changes.Cell migration was assessed using a wound-healing migration assay,and cell invasion was investigated using Transwell chambers.Western blotting assays were employed to evaluate the expression levels of proteins in the protein kinase B/mammalian target of rapamycin (Akt/mTOR) signaling pathway.Finally,in vivo activity of petasin was evaluated using the SW-620 subcutaneous tumor model established in Balb/c nude mice.Twelve rats were randomly divided into control group and 10 mg/kg petasin group.The tumor volume was calculated every 7 days for 28 days.Terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling (TUNEL) assay was performed to assess the apoptotic effect of petasin.Differences between two groups were assessed by analysis of independent-sample t tests.Results:Petasin significantly inhibited the proliferation of human colon carcinoma cell lines,induced apoptosis,and suppressed migration and invasion in SW-620 cells.Western blotting results showed that petasin decreased the phosphorylation of Akt (1.01 ± 0.16 vs.0.74 ± 0.06,P = 0.042),mTOR (0.71 ± 0.12 vs.0.32 ± 0.11,P = 0.013),and P70S6K (1.23 ± 0.21 vs.0.85 ± 0.14,P = 0.008),elevated the expression of caspase-3 (0.41 ± 0.09 vs.0.74 ± 0.12,P = 0.018) and caspase-9 (1.10 ± 0.27 vs.1.98 ± 0.22,P = 0.009),decreased the Bcl-2 protein (2.75 ± 0.47 vs.1.51 ± 0.36,P = 0.008),downregulated the expression of matrix metalloproteinase (MMP)-3 (1.51 ± 0.31 vs.0.82 ± 0.11,P = 0.021) and MMP-9 (1.56 ± 0.32 vs.0.94 ± 0.15,P = 0.039) in SW-620 cell.In vivo,10 mg/kg petasin inhibited tumor growth in Balb/c nude mice (924.18 ± 101.23 vs.577.67 ± 75.12 mm3 at day 28,P = 0.001) and induced apoptosis (3.6 ± 0.7% vs.36.0 ± 4.9%,P = 0.001) in tumor tissues.Conclusions:Petasin inhibits the proliferation of colon cancer SW-620 cells via inactivating the Akt/mTOR pathway.Our findings suggest petasin as a potential candidate for colon cancer therapy.Xi Lyu Ai-Lin Song Yin-Liang Bai Xiao-Dong Xu Dong-Qiang He You-Cheng Zhang 2019Chinese Medical Journal2019,,9:2

17Integrated strategies for chemotherapy cycles in nasopharyngeal carcinoma patients: Real-world data from two epidemic centers guiding decision-making显示文摘objective:Two cycles of induction chemotherapy(IC)followed by 2 cycles of platinum-based concurrent chemoradiotherapy(CCRT)(2IC+2CCRT)for locoregionally advanced nasopharyngeal carcinoma(LA-NPC)is widely adopted but not evidence-confirmed.This study aimed to determine the clinical value of 2IC+2CCRT regarding efficacy,toxicity and cost-effectiveness.Methods:This real-world study from two epidemic centers used propensity score matching(PSM)and inverse probability of treatment weighting(IPTW)analyses.The enrolled patients were divided into three groups based on treatment modality:Group A(2IC+2CCRT),Group B(3IC+2CCRT or 2IC+3CCRT)and Group C(3IC+3CCRT).Long-term survival,acute toxicities and cost-effectiveness were compared among the groups.We developed a prognostic model dividing the population into high-and low-risk cohorts,and survivals including overall survival(OS),progression-free survival(PFS),distant metastasis-free survival(DMFS)and locoregional relapse-free survival(LRRFS)were compared among the three groups according to certain risk stratifications.Results:Of 4,042 patients,1,175 were enrolled,with 660,419,and 96 included in Groups A,B and C,respectively.Five-year survivals were similar among the three groups after PSM and confirmed by IPTW.Grade 3-4 neutropenia and leukocytopenia were significantly higher in Groups C and B than in Group A(52.1%vs.41.5%vs.25.2%;41.7%vs.32.7%vs.25.0%)as were grade 3-4 nausea/vomiting and oral mucositis(29.2%vs.15.0%vs.6.1%;32.3%vs.25.3%vs.18.0%).Cost-effective analysis suggested that 2IC+2CCRT was the least expensive,while the health benefits were similar to those of the other groups.Further exploration showed that 2IC+2CCRT tended to be associated with a shorter PFS in high-risk patients,while 3IC+3CCRT potentially contributed to poor PFS in low-risk individuals,mainly reflected by LRRFS.Conclusions:In LA-NPC patients,2IC+2CCRT was the optimal choice regarding efficacy,toxicity and costeffectiveness;however,2IC+2CCRT and 3IC+3CCRT probably shortened LRRFS in high-and low-risk populations,respectively.Zejiang Zhan Yingying Huang Jiayu Zhou Zhuochen Cai Haoyang Huang Ying Deng Wenze Qiu Xun Cao Xi Chen Chixiong Liang Lulu Zhang Xiang Guo Taize Yuan Xing Lyu 2023Chinese Journal of Cancer Research2023,35,2:1
18Analysis of retinal vasculature changes in indirect traumatic optic neuropathy using optic coherence tomography angiography显示文摘AIM:To assess the retinal vasculature alterations in indirect traumatic optic neuropathy(ITON)patients following craniofacial trauma by optic coherence tomography angiography(OCTA).METHODS:Patients diagnosed of monocular ITON were recruited from August 2016 to May 2020.OCTA was performed using the Angio Vue OCT-A system for two cube scans centered at the optic nerve head and fovea.OCTA data included thicknesses of peripapillary retinal nerve fiber layer(RNFL)and macular ganglion cell complex(GCC),as well as proportion of capillary perfusion and data were analyzed for correlation with post-injury timepoints:within 7,8-30,31-90,and 91-365d.RESULTS:A total of 73 ITON patients were studied.Significant thinning of RNFL and GCC layers and attenuation of microvascular perfusion were observed in ITON eyes as compared to contralateral unaffected eyes(for most of the analyzed sectors and quadrants,P<0.05).Without respect to surgical intervention and vision recovery,the decrease in retinal layer thicknesses and microvascular perfusion was time-dependent,and most significant within three months(P<0.001).CONCLUSION:ITON presents with time-dependent thinning of retinal layers and attenuation of microvasculature,indicating possible degeneration of retinal ganglion cells due to reduced retinal blood supply.Huan Ma Yang Gao Jin-Miao Li Yue-Kun Bao Cong Nie Pan Yin Xi Lyu Xiao-Yan Ding Rong Lu 2022International Journal of Ophthalmology(English edition)2022,15,8:1
19Enhanced catalytic oxidation of 2,4-dichlorophenol via singlet oxygen dominated peroxymonosulfate activation on CoOOH@Bi_(2)O_(3)composite显示文摘Cobalt oxyhydroxide(CoOOH)has been turned out to be a high-efficiency catalyst for peroxymonosulfate(PMS)activation.In this study,CoOOH was loaded on bismuth oxide(Bi_(2)O_(3))using a facile chemical precipitation process to improve its catalytic activity and stability.The result showed that the catalytic performance on the 2,4-dichlorophenol(2,4-DCP)degradation was significantly enhanced with only 11 wt%Bi_(2)O_(3)loading.The degradation rate in the CoOOH@Bi_(2)O_(3)/PMS system(0.2011 min−1)was nearly 6.0 times higher than that in the CoOOH/PMS system(0.0337 min−1).Furthermore,CoOOH@Bi_(2)O_(3)displayed better stability with less Co ions leaching(16.4%lower than CoOOH)in the PMS system.These phenomena were attributed to the Bi_(2)O_(3)loading which significantly increased the conductivity and specific surface area of the CoOOH@Bi_(2)O_(3)composite.Faster electron transfer facilitated the redox reaction of Co(III)/Co(II)and thus was more favorable for reactive oxygen species(ROS)generation.Meanwhile,larger specific surface area furnished more active sites for PMS activation.More importantly,there were both non-radical(^(1)O_(2))and radicals(SO_(4)^(−)•,O_(2)^(−)•,and OH•)in the CoOOH@Bi_(2)O_(3)/PMS system and^(1)O_(2)was the dominant one.In general,this study provided a simple and practical strategy to enhance the catalytic activity and stability of cobalt oxyhydroxide in the PMS system.Tianhao Xi Xiaodan Li Qihui Zhang Ning Liu Shu Niu Zhaojun Dong Cong Lyu 2021Frontiers of Environmental Science & Engineering2021,15,4:1
20Opposite effects of Drosophila C3PO on gene silencing mediated by esi-2.1 and miRNA-bantam显示文摘Translin/TRAX complex,also named as C3PO,is evolutionarily conserved and participates in diverse cellular processes in different organisms from yeast to human.C3PO plays a critical role in the activation of RNA-induced silencing complexes by promoting the unwinding and degradation of passenger strand of exogenous siRNAs(exo-siRNAs)in Drosophila and human.Moreover,human C3PO(hC3PO)has been found to broadly repress miRNAs by degrading miRNA precursors.However,the effect of Drosophila melanogaster C3PO(dmC3PO)on endogenous siRNA(endo-siRNA)and miRNA pathways remains unknown.Here,we found that the loss of dmC3PO promoted the accumulation of the passenger strand of esi-2.1(hp-CG4068B),and resulted in the de-repression of the DNA-damage-response gene mutagensensitive 308(mus308),which is an endogenous slicer target of esi-2.1 in Drosophila.Moreover,we also found that depletion of dmC3PO increased the accumulation of miR-bantam.Taken together,our findings indicated that dmC3PO not only involves in siRNA pathway triggered by dsRNA,but also regulates the abundance of certain endogenous small RNAs in Drosophila.Yujie Liu Yuan Fang Yongxiang Liu Zhaowei Wang Bao Lyu Yuanyang Hu Xi Zhou 2019Acta Biochimica et Biophysica Sinica2019,51,2:1
