| 1 | A transient expression system in soybean mesophyll protoplasts reveals the formation of cytoplasmic GmCRY1 photobody-like structures显示文摘Soybean(Glycine max(L.) Merr.), grown for its plant oils and proteins, is one of the most important crops throughout the world.Generating stable and heritable transgenic soybeans is relatively inefficient;therefore, there is an urgent need for a simple and high-efficient transient transformation method by which to enable the investigation of gene functions in soybeans, which will facilitate the elucidation and improvement of the molecular mechanisms regulating the associated agronomic traits. We established a system of transient expression in soybean mesophyll protoplasts and obtained a high level of protoplast transfection efficiency(up to 83.5%). | 2 | Utility of Droplet Digital PCR Assay for Quantitative Detection of Norovirus in Shellfish, from Production to Consumption in Guangxi, China显示文摘Objective Shellfish are recognized as important vehicles of norovirus-associated gastroenteritis. The present study aimed to monitor norovirus contamination in oysters along the farm-to-fork continuum in Guangxi, a major oyster production area in Southwestern China. Methods Oyster samples were collected monthly from farms, markets, and restaurants, from January to December 2016. Norovirus was detected and quantified by one-step reverse transcription-droplet digital polymerase chain reaction(RT-ddPCR). Results A total of 480 oyster samples were collected and tested for norovirus genogroups I and II. Norovirus was detected in 20.7% of samples, with genogroup II predominating. No significant difference was observed in norovirus prevalence among different sampling sites. The norovirus levels varied widely, with a geometric mean of 19,300 copies/g in digestive glands. Both norovirus prevalence and viral loads showed obvious seasonality, with a strong winter bias. Conclusion This study provides a systematic analysis of norovirus contamination ‘from the farm to the fork' in Guangxi. RT-ddPCR can be a useful tool for detection and quantification of low amounts of norovirus in the presence of inhibitors found particularly in foodstuffs. | 5 | Effects of processing technology on essential oil and pharmacological action of frankincense显示文摘Background:According to the traditional Chinese medicine theory,the common processing methods for Ruxiang(frankincense)include stir-frying processing and vinegar processing.Methods:With network pharmacology and thermal analysis methods,we selected and identified the main and irritating ingredients,established the pyrolysis characteristic parameters for identifying frankincense quality,quantified the starting and optimum temperature limits for processing frankincense,and analyzed the influences of processing adjuvants(vinegar)on combustion pyrolysis characteristics of frankincense.By applying Fourier transformation infrared spectrometry,high performance liquid chromatography,gas chromatography and UV-visible spectrometry,we evaluated the processing technology developed in our study of frankincense and its processed products.Results:Based on network pharmacology,we can find that the classical compounds of essential oil are the main pharmacodynamics components of frankincense possessing the anti-inflammatory,analgesia,anti-tumor,anti-ulcer and ant-inflammatory bowel disease effects.The pyrolysis combustion rate peaks of frankincense at 285.27±7.05℃and 476.99±13.46℃were the characteristic peaks used to identify frankincense quality.The specific temperature of slow-fire processing,including the vinegar processing and stir-frying processing,was 148.72±5.09℃.Heating for 5.67±0.98 min after reaching processing temperature was needed for processing frankincense with vinegar.The kinetics analysis showed that both frankincense and its vinegar mixture exhibited good linear relationships.The feasibility and practicability for our processing technology were verified through the determination of medicinal compounds by Fourier transformation infrared spectrometry,high performance liquid chromatography,gas chromatography and UV.Conclusion:Frankincense essential oils were the main active components of frankincense,which also can be taken as the internal chemical control indexes to judge the quality control and to optimize the processing technology of frankincense.The pyrolysis combustion rate peaks of frankincense at 285.27±7.05℃and 476.99±13.46℃were the characteristic peaks that can be used to identify frankincense quality.The optimum processing conditions of vinegar-processed frankincense with were as follows:with vinegar,20%;heating temperature,148.72±5.09℃;and heating time,5.67±0.98 min.Under these conditions,frankincense essential oil could cause a moderate effect on combustion pyrolysis characteristics to exert its corresponding pharmacological effects. | Xiang-Long Meng Yong-Lin Liang Chen-Zi Lyu Xiao-Juan Su Cong Hu Chen-Xu Ning Shuo-Sheng Zhang | 2021 | Traditional Medicine Research2021,6,3: | 2 |

| 9 | Enhanced catalytic oxidation of 2,4-dichlorophenol via singlet oxygen dominated peroxymonosulfate activation on CoOOH@Bi_(2)O_(3)composite显示文摘Cobalt oxyhydroxide(CoOOH)has been turned out to be a high-efficiency catalyst for peroxymonosulfate(PMS)activation.In this study,CoOOH was loaded on bismuth oxide(Bi_(2)O_(3))using a facile chemical precipitation process to improve its catalytic activity and stability.The result showed that the catalytic performance on the 2,4-dichlorophenol(2,4-DCP)degradation was significantly enhanced with only 11 wt%Bi_(2)O_(3)loading.The degradation rate in the CoOOH@Bi_(2)O_(3)/PMS system(0.2011 min−1)was nearly 6.0 times higher than that in the CoOOH/PMS system(0.0337 min−1).Furthermore,CoOOH@Bi_(2)O_(3)displayed better stability with less Co ions leaching(16.4%lower than CoOOH)in the PMS system.These phenomena were attributed to the Bi_(2)O_(3)loading which significantly increased the conductivity and specific surface area of the CoOOH@Bi_(2)O_(3)composite.Faster electron transfer facilitated the redox reaction of Co(III)/Co(II)and thus was more favorable for reactive oxygen species(ROS)generation.Meanwhile,larger specific surface area furnished more active sites for PMS activation.More importantly,there were both non-radical(^(1)O_(2))and radicals(SO_(4)^(−)•,O_(2)^(−)•,and OH•)in the CoOOH@Bi_(2)O_(3)/PMS system and^(1)O_(2)was the dominant one.In general,this study provided a simple and practical strategy to enhance the catalytic activity and stability of cobalt oxyhydroxide in the PMS system. | Tianhao Xi Xiaodan Li Qihui Zhang Ning Liu Shu Niu Zhaojun Dong Cong Lyu | 2021 | Frontiers of Environmental Science & Engineering2021,15,4: | 1 |
| 10 | Molecular cloning,characterization and expression analysis of three key starch synthesis-related genes from the bulb of a rare lily germplasm,Lilium brownii var.giganteum显示文摘Starch is the predominant compound in bulb scales,and previous studies have shown that bulblet development is closely associated with starch enrichment.However,how starch synthesis affects bulbification at the molecular level is unclear.In this study,we demonstrate that Lilium brownii var.giganteum,a wild lily with a giant bulb in nature,and L.brownii,the native species,have different starch levels and characteristics according to cytological and ultra-structural observations.We cloned the complete sequence of three key gene-encoding enzymes(LbgAGPS,LbgGBSS,and LbgSSⅢ)during starch synthesis by rapid amplification of 5’and 3’complementary DNA(cDNA)ends(RACE)technology.Bioinformatics analysis revealed that the proteins deduced by these genes contain the canonical conserved domains.Constructed phylogenetic trees confirmed the evolutionary relationships with proteins from other species,including monocotyledons and dicotyledons.The transcript levels of various tissues and time course samples obtained during bulblet development uncovered relatively high expression levels in bulblets and gradual increase expression accompanying bulblet growth.Moreover,a set of single nucleotide polymorphisms(SNPs)was discovered in the AGPS genes of four lily genotypes,and a purifying selection fashion was predicted according to the non-synonymous/synonymous(Ka/Ks)values.Taken together,our results suggested that key starch-synthesizing genes might play important roles in bulblet development and lead to distinctive phenotypes in bulblet size. | Yun WU Minyi SUN Shiqi LI Ruihan MIN Cong GAO Qundan LYU Ziming REN Yiping XIA | 2021 | Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2021,22,6: | 1 |
| 11 | Enhanced Peroxymonosulfate Activation by NixCo1-xOOH for Efficient Catalytic Oxidation of Organic Pollutants显示文摘Peroxymonosulfate(PMS) has received increasing attention as viable technology for recalcitrant organics removal from polluted waters. Although promising, alternative heterogeneous catalysts with stable structure, strong hydrophilicity, environmental friendliness and excellent catalytic performance are highly desirable to facilitate the wide industrialization of PMS. In this work, Ni doped CoOOH catalyst was employed as PMS activator. Ni dopant had a significant influence on the morphology, structure and catalytic performance of CoOOH. NiojCoo.gOOH exhibited the best catalytic performance. Reaction rate ofNio.2Coo.8OOH was 2, 4, and 4.4 times that of CoOOH, CoFe2O4 and CO3O4, respectively. Moreover, Ni。2C00.8OOH/PMS system had potential application to organic pollutants and displayed a great catalytic activity over a broader pH value(e.g., 4-10). More importantly, Ni doping accelerated the transfonnation of Co(Ⅲ) and Co(Ⅱ) and formed active species CoOH^+ and NiOH^+ which were responsible to the enhancement of PMS activation.OH, SO4^-:O2^- and 1^O2 were detected, indicating both non-radical and radical processes in the Nio.2Coo.8OOH/PMS system. | 14 | Micrometer-sized NiOOH hierarchical spheres for enhanced degradation of sulfadiazine via synergistic adsorption and catalytic oxidation in peroxymonosulfate system显示文摘As an antibiotic,sulfadiazine has posed a serious threat to humans and ecosystems due to its chronic toxicity.The advanced oxidation processes (AOPs) via heterogeneous catalytic activation of peroxymonosulfate (PMS) have significant potential for the degradation of antibiotics.However,there are multiple restrictions including non-specifically binding to target contaminants,which would deplete oxidation capacity,and lacking energy effectiveness due to inefficient utilization of reactive oxygen species (ROS).To overcome these obstacles,we adopted the“bait-hook&destroy”strategy in this study.Herein,we synthesized a novel micrometer-sized Ni OOH hierarchical spheres assembled from nanosheets,which have relatively large specific surface areas and yield specified cavities to“bait-hook”sulfadiazine and PMS onto the surface cavities.This process was further conductive to effective generation of ROS and subsequently“destruction”of sulfadiazine with elevated mass transformation rate.20.4%of sulfadiazine can adsorb to Ni OOH surface in less than 30 min (0.0051 min^(-1)),and then sulfadiazine was completely degraded in 90min intervals in the Ni OOH/PMS system.The degradation rate constant (k=0.0537 min^(-1)) was about5.3,2.5 and 2.2 times higher than that in Ni_(2)O_(3)/PMS,NiO/PMS and Ni(OH)_(2)/PMS system,respectively.This was ascribed to the synergistic catalytic oxidation and adsorption process occurred on the surface of Ni OOH.Appreciably,there were both non-radicals (^(1)O_(2)) and radicals (O_(2)^(·-)and SO_(4)^(·-)) involved in the Ni OOH/PMS system,and^(1)O_(2)was distinguished as the dominated ROS for degradation of sulfadiazine.This study provides a novel strategy via synergistic adsorption and catalytic oxidation,and indicates that the micrometer-sized Ni OOH hierarchical sphere as heterogeneous catalyst is an attractive candidate for potential application of the SR-AOPs technology in water treatment. | Cong Lyu Lu Zhang Dan He Boyuan Su Ying Lyu | 2022 | Chinese Chemical Letters2022,33,2: | 0 |

| 19 | Expansion and adaptive evolution of the WRKY transcription factor family in Avicennia mangrove trees显示文摘Mangroves are adapted to intertidal zones,which present extreme environmental conditions.WRKYs are among the most prominent transcription factors(TFs)in higher plants and act through various interconnected networks to regulate responses to multiple abiotic stressors.Here,based on omic data,we investigated the landscape and evolutionary patterns of WRKYs in the main mangrove genus Avicennia.We found that both the number and the proportion of TFs and WRKYs in Avicennia species exceeded their inland relatives,indicating a signifcant expansion of WRKYs in Avicennia.We identifed 109 WRKY genes in the representative species Avicennia marina.Comparative genomic analysis showed that two recent wholegenome duplication(WGD)events played a critical role in the expansion of WRKYs,and 88%of Avicennia marina WRKYs(AmWRKYs)have been retained following these WGDs.Applying comparative transcriptomics on roots under experimental salt gradients,we inferred that there is high divergence in the expression of WGD-retained AmWRKYs.Moreover,we found that the expression of 16 AmWRKYs was stable between freshwater and moderately saline water but increased when the trees were exposed to high salinity.In particular,14 duplicates were retained following the two recent WGD events,indicating potential neo-and sub-functionalization.We also found that WRKYs could interact with other upregulated genes involved in signalling pathways and natural antioxidant biosynthesis to enhance salt tolerance,contributing to the adaptation to intertidal zones.Our omic data of the WRKY family in A.marina broadens the understanding of how a TF family relates to the adaptive evolution of mangroves. | Xiao Feng Guohong Li Weihong Wu Haomin Lyu Jiexin Wang Cong Liu Cairong Zhong Suhua Shi Ziwen He | 2023 | Marine Life Science & Technology2023,5,2: | 0 |
