2Advanced smart biomaterials and constructs for hard tissue engineering and regeneration显示文摘Hard tissue repair and regeneration cost hundreds of billions of dollars annually worldwide, and the need has substantially increased as the population has aged. Hard tissues include bone and tooth structures that contain calcium phosphate minerals.Smart biomaterial-based tissue engineering and regenerative medicine methods have the exciting potential to meet this urgent need. Smart biomaterials and constructs refer to biomaterials and constructs that possess instructive/inductive or triggering/stimulating effects on cells and tissues by engineering the material's responsiveness to internal or external stimuli or have intelligently tailored properties and functions that can promote tissue repair and regeneration. The smart material-based approaches include smart scaffolds and stem cell constructs for bone tissue engineering; smart drug delivery systems to enhance bone regeneration; smart dental resins that respond to pH to protect tooth structures; smart pH-sensitive dental materials to selectively inhibit acid-producing bacteria; smart polymers to modulate biofilm species away from a pathogenic composition and shift towards a healthy composition; and smart materials to suppress biofilms and avoid drug resistance. These smart biomaterials can not only deliver and guide stem cells to improve tissue regeneration and deliver drugs and bioactive agents with spatially and temporarily controlled releases but can also modulate/suppress biofilms and combat infections in wound sites. 3Characteristics and Preliminary Causes of Tropical Cyclone Extreme Rainfall Events over Hainan Island显示文摘The characteristics of tropical cyclone(TC) extreme rainfall events over Hainan Island from 1969 to 2014 are analyzed from the viewpoint of the TC maximum daily rainfall(TMDR) using daily station precipitation data from the Meteorological Information Center of the China Meteorological Administration, TC best-track data from the Shanghai Typhoon Institute,and NCEP/NCAR reanalysis data. The frequencies of the TMDR reaching 50, 100 and 250 mm show a decreasing trend[-0.7(10 yr)^(-1)], a weak decreasing trend [-0.2(10 yr)^(-1)] and a weak increasing trend [0.1(10 yr)^(-1)], respectively. For seasonal variations, the TMDR of all intensity grades mainly occurs from July to October, with the frequencies of TMDR 50 mm and 100 mm peaking in September and the frequency of TMDR 250 mm [TC extreme rainstorm(TCER) events]peaking in August and September. The western region(Changjiang) of the Island is always the rainfall center, independent of the intensity or frequencies of different intensity grades. The causes of TCERs are also explored and the results show that topography plays a key role in the characteristics of the rainfall events. TCERs are easily induced on the windward slopes of Wuzhi Mountain, with the coordination of TC tracks and TC wind structure. A slower speed of movement, a stronger TC intensity and a farther westward track are all conducive to extreme rainfall events. A weaker northwestern Pacific subtropical high is likely to make the 500-h Pa steering flow weaker and results in slower TC movement, whereas a stronger South China Sea summer monsoon can carry a higher moisture flux. 4Personalized neoantigen pulsed dendritic cell vaccine for advanced lung cancer显示文摘Neoantigens are considered to be ultimate target of tumor immunotherapy due to their high tumor specificity and immunogenicity.Dendritic cell(DCs)vaccines based on neoantigens have exciting effects in treatment of some malignant tumors and are a promising therapeutic modality.Lung cancer is a lethal disease with the highest morbidity and mortality rate in the world.Despite the rapid development of targeted therapy and immune checkpoint inhibitors for lung cancer in recent years,their efficacy is still unsatisfactory overall.Therefore,there is an urgent unmet clinical need for lung cancer treatment.Here,we attempted to treat lung cancer using a personalized neoantigen peptide-pulsed autologous DC vaccine and conducted a single-arm,2 medical centers,pilot study initiated by the investigator(ChiCTR-ONC 16009100,NCT02956551).The patients enrolled were patients with heavily treated metastatic lung cancer.Candidate neoantigens were derived from whole-exome sequencing and RNA sequencing of fresh biopsy tissues as well as bioinformatics analysis.A total of 12 patients were enrolled in this study.A total of 85 vaccine treatments were administered with a median value of 5 doses/person(range:3-14 doses/person).In total,12-30 peptide-based neoantigens were selected for each patient.AIl treatment-related adverse events were grade 1-2 and there were no delays in dosing due to toxic effects.The objective effectiveness rate was 25%;the disease control rate was 75%;the median progression-free survival was 5.5 months and the median overall survival was 7.9 months.This study provides new evidence for neoantigen vaccine therapy and new therapeutic opportunities for lung cancer treatment.Zhenyu Ding Qing Li Rui Zhang Li Xie Yang shu Song Gao Peipei Wang Xiaoqing Su Yun Qin Yuelan Wang Juemin Fang Zhongzheng Zhu Xuyang Xia Guochao Wei Hui Wang Hong Qian Xianling Guo Zhibo Gao Yu Wang Yuquan Wei Qing Xu Heng Xu Li Yang 2021Signal Transduction and Targeted Therapy2021,6,2:3
5Fusobacterium nucleatum enhances the efficacy of PD-L1 blockade in colorectal cancer显示文摘Given that only a subset of patients with colorectal cancer(CRC)benefit from immune checkpoint therapy,efforts are ongoing to identify markers that predict immunotherapeutic response.Increasing evidence suggests that microbes influence the efficacy of cancer therapies.Fusobacterium nucleatum induces different immune responses in CRC with different microsatellite-instability(MSI)statuses.Here,we investigated the effect of F.nucleatum on anti-PD-L1 therapy in CRC.We found that high F.nucleatum levels correlate with improved therapeutic responses to PD-1 blockade in patients with CRC.Additionally,F.nucleatum enhanced the antitumor effects of PD-L1 blockade on CRC in mice and prolonged survival.Combining F.nucleatum supplementation with immunotherapy rescued the therapeutic effects of PD-L1 blockade.Furthermore,F.nucleatum induced PD-L1 expression by activating STING signaling and increased the accumulation of interferon-gamma(IFN-γ)^(+)CD8^(+)tumor-infiltrating lymphocytes(TILs)during treatment with PD-L1 blockade,thereby augmenting tumor sensitivity to PD-L1 blockade.Finally,patient-derived organoid models demonstrated that increased F.nucleatum levels correlated with an improved therapeutic response to PD-L1 blockade.These findings suggest that F.nucleatum may modulate immune checkpoint therapy for CRC.Yaohui Gao Dexi Bi Ruting Xie Man Li Jing Guo Hu Liu Xianling Guo Juemin Fang Tingting Ding Huiyuan Zhu Yuan Cao Meichun Xing Jiayi Zheng Qing Xu Qian Xu Qing Wei Huanlong Qin 2021Signal Transduction and Targeted Therapy2021,6,12:3
6Preparation and Arc Erosion Resistance of C_f/Cu Composite by Vacuum Melting Infiltration显示文摘Cf/Cu composite was prepared by vacuum melting infiltration. Tiand Cr were doped to the Cu alloy to improve the wettability between Cu and carbon. The microstructure was investigated by XRD, SEM and EDS. The arc erosion rate of Cf/Cu composite was investigated in vacuum. The results showed that the Tiand Cr could improve the wettability between Cu and C/C preform and the infiltration ability of Cu into C/C preform greatly. A TiC interface formed between the fibers and matrix. The good bonding between the fiber and matrix guaranteed that part of the Cu matrix can still be bonded on the fibers even when the material was exposed to the plasma. Consequently, the carbon fibers were protected from the erosion. In comparison, Cu was completely consumed by the arc erosion. Hence, the graphite was eroded and presented a caulifl ower-like morphology. 8A hydrophilic fluorescent polymer containing naphthalimide moiety as chemosensor for microbioreactors显示文摘A novel hydrophilic copolymer P(NI-HEMA) containing the pendant group of naphthalimide moiety has been prepared. In the aqueous buffer solution, its film on the glass substrate shows an obvious fluorescence change and excellent linear relevance in the pH range of 5.8-8.0. Specifically, using the isobestic point at 406 nm as excitation wavelength can avoid the UV irradiation and the tedious calibration process. The film stability and hydrophilicity of copolymer P(NI-HEMA) were studied. 10Construction of miRNA-mRNA network reveals crucial miRNAs and genes in acute myocardial infarction显示文摘Acute myocardial infarction(AMI)is a severe cardiovascular disease.This study aimed to identify crucial microRNAs(miRNAs)and mRNAs in AMI by establishing a miRNA-mRNA network.The microarray datasets GSE31568,GSE148153,and GSE66360 were downloaded from the Gene Expression Omnibus(GEO)database.We identified differentially expressed miRNAs(DE-miRNAs)and mRNAs(DE-mRNAs)in AMI samples compared with normal control samples.The consistently changing miRNAs in both GSE31568 and GSE148153 datasets were selected as candidate DE-miRNAs.The interactions between the candidate DE-miRNAs and DE-mRNAs were analyzed,and a miRNA-mRNA network and a protein-protein interaction network were constructed,along with functional enrichment and pathway analyses.A total of 209 DE-miRNAs in the GSE31568 dataset,857 DE-miRNAs in the GSE148153 dataset,and 351 DE-mRNAs in the GSE66360 dataset were identified.Eighteen candidate DE-miRNAs were selected from both the GSE31568 and GSE148153 datasets.Furthermore,miR-646,miR-127-5p,miR-509-5p,miR-509-3-5p,and miR-767-5p were shown to have a higher degree in the miRNA-mRNA network.THBS-1 as well as FOS was a hub gene in the miRNA-mRNA network and the protein-protein interaction(PPI)network,respectively.CDKN1A was important in both miRNA-mRNA network and PPI network.We established a miRNA-mRNA network in AMI and identified five miRNAs and three genes,which might be used as biomarkers and potential therapeutic targets for patients with AMI.Kai Wang Zhongming Li Wenjie Ma Yan Sun Xianling Liu Lijun Qian Jian Hong Dasheng Lu Jing Zhang Di Xu 2021The Journal of Biomedical Research2021,35,6:2

16Sequence analysis and typing of Saprolegnia strains isolated from freshwater fish from Southern Chinese regions显示文摘Saprolegniasis,caused by Saprolegnia infection,is one of the most common diseases in freshwater fish.Our study aimed to determine the epidemiological characteristics of saprolegniasis in Chinese regions of high incidence.Saprolegnia were isolated and identified by morphological and molecular methods targeting the internal transcribed spacer(ITS)ribosomal DNA(rDNA)and building neighbor-joining(NJ)and maximum parsimony(MP)phylogenetic trees.The ITS sequences of eight isolated strains were compared with GenBank sequences and all strains fell into three clades:CLADE1(02,LP,04 and 14),CLADE2(S1),and CLADE3(CP,S2,L5 and the reference ATCC200013).Isolates 02 and LP shared 80%sequence similarity with S.diclina,S.longicaulis,S.ferax,S.mixta,and S.anomalies.Further,isolates 04 and 14 shared 80%similarity with S.bulbosa and S.oliviae.Finally,extremely high ITS sequence similarities were identified between isolates S1 and S.australis(100%);CP and S.hypogyna(96%);and S2,L5,ATCC200013 and S.salmonis(98%).This research provides insights into the identification,prevention and control of saprolegniasis pathogens and the potential development of effective drugs.Siya Liu Pengpeng Song Renjian Ou Wenhong Fang Mao Lin Jiming Ruan Xianle Yang Kun Hu 2017Aquaculture and Fisheries2017,2,5:1
