1Regional Δ~14C patterns and fossil fuel derived CO_2 distribution in the Beijing area using annual plants显示文摘The level of atmosphericΔ14C and the fossil fuel derived CO 2 concentration in the Beijing area from May to September,2009, were systematically analyzed based on radiocarbon(14C)measurements of annual plants by accelerator mass spectrometry(AMS). The results show that the maximumΔ14C in Beijing was 29.6‰±2.2‰,and the minimum was–28.2‰±2.5‰,with a trend of decreasingΔ14C from the outer suburbs to inner suburbs to the urban center.This trend correlates well with increases in fossil fuel derived CO2 caused by human activities such as population density,industrial emissions and traffic,with lower values of atmosphericΔ14C associated with more intensive human activities.The fossil fuel derived CO 2 concentrations from May to September, 2009,ranged from 3.9±1.0 ppm to 25.4±1.0 ppm.It was calculated that each additional 1 ppm of CO2 from fossil fuels depleted the atmosphericΔ14C by approximately 2.70‰.This study suggests that 14C measurements of annual plants by AMS provide an effective method to rapidly trace fossil fuel derived CO2.XI XianTing DING XingFang FU DongPo ZHOU LiPing LIU KeXin 2011Chinese Science Bulletin2011,56,16:7
2Rarefaction and Temperature Gradient Effect on the Performance of the Knudsen Pump显示文摘The prediction of the multiscale flow in the Knudsen pump is important for understanding its pumping mechanism.However,there is little research on such interesting multiscale phenomenon in the Knudsen pumps.In this paper,a novel numerical analysis method combining the direct simulation Monte Carlo(DSMC) method with the smoothed particle hydrodynamics(SPH) method is presented for simulating the multiscale flow,which is often encountered in the application of the Knudsen pumps.Validity and accuracy of the new method are given by comparing its results with that of the previous research.Using the coupled multiscale approach,the rarefaction and the temperature drive are studied,which are two main factors on the performance of the Knudsen pumps.To investigate the effect of rarefaction on the performance of the Knudsen pump,various pump operation pressures are compared.The flow characteristics and pumping ability at different rarefaction are analyzed,and the phenomenon of the multiscale flow is also discussed.Several cases with different linear or nonlinear temperature gradients are set to investigate the effect of temperature gradient on the performance of the Knudsen pump.The flow characteristics of the Knudsen pump such as the velocity,pressure increase,and the mass flowrate are presented.A unique phenomenon,the reverse transpiration effect caused by the nonlinear temperature gradient is studied,and the reason of the significant pressure increase in the pump channel is also analyzed.Since the multiscale gas flow is widely encountered in the microflow systems,the above method and its results can also be greatly beneficial and provide significant insights for the design of the MEMS devices.YE Jianjun YANG Jian ZHENG Jinyang DING Xianting WONG Ieong LI Weizhong CHEN Cong 2012Chinese Journal of Mechanical Engineering2012,25,4:6
3DNA tetrahedron-mediated immune-sandwich assay for rapid and sensitive detection of PSA through a microfluidic electrochemical detection system显示文摘Prostate-specific antigen(PSA)is the most widely used biomarker for the early diagnosis of prostate cancer.Existing methods for PSA detection are burdened with some limitations and require improvement.Herein,we developed a novel microfluidic–electrochemical(μFEC)detection system for PSA detection.First,we constructed an electrochemical biosensor based on screen-printed electrodes(SPEs)with modification of gold nanoflowers(Au NFs)and DNA tetrahedron structural probes(TSPs),which showed great detection performance.Second,we fabricated microfluidic chips by DNA TSP-Au NF-modified SPEs and a PDMS layer with designed dense meandering microchannels.Finally,theμFEC detection system was achieved based on microfluidic chips integrated with the liquid automatic conveying unit and electrochemical detection platform.TheμFEC system we developed acquired great detection performance for PSA detection in PBS solution.For PSA assays in spiked serum samples of theμFEC system,we obtained a linear dynamic range of 1–100 ng/mL with a limit of detection of 0.2 ng/mL and a total reaction time<25 min.Real serum samples of prostate cancer patients presented a strong correlation between the“gold-standard”chemiluminescence assays and theμFEC system.In terms of operation procedure,cost,and reaction time,our method was superior to the current methods for PSA detection and shows great potential for practical clinical application in the future.Dezhi Feng Jing Su Yi Xu Guifang He Chenguang Wang Xiao Wang Tingrui Pan Xianting Ding Xianqiang Mi 2021Microsystems & Nanoengineering2021,7,2:1

5Integrated microfluidic single-cell immunoblotting chip enables high-throughput isolation,enrichment and direct protein analysis of circulating tumor cells显示文摘Effective capture and analysis of a single circulating tumor cell(CTC)is instrumental for early diagnosis and personalized therapy of tumors.However,due to their extremely low abundance and susceptibility to interference from other cells,high-throughput isolation,enrichment,and single-cell-level functional protein analysis of CTCs within one integrated system remains a major challenge.Herein,we present an integrated multifunctional microfluidic system for highly efficient and label-free CTC isolation,CTC enrichment,and single-cell immunoblotting(ieSCI).The ieSCI-chip is a multilayer microfluidic system that combines an inertia force-based cell sorter with a membrane filter for label-free CTC separation and enrichment and a thin layer of a photoactive polyacrylamide gel with microwell arrays at the bottom of the chamber for single-cell immunoblotting.The ieSCI-chip successfully identified a subgroup of apoptosis-negative(Bax-negative)cells,which traditional bulk analysis did not detect,from cisplatin-treated cells.Furthermore,we demonstrated the clinical application of the ieSCI-chip with blood samples from breast cancer patients for personalized CTC epithelial-to-mesenchymal transition(EMT)analysis.The expression level of a tumor cell marker(EpCAM)can be directly determined in isolated CTCs at the single-cell level,and the therapeutic response to anticancer drugs can be simultaneously monitored.Therefore,the ieSCI-chip provides a promising clinical translational tool for clinical drug response monitoring and personalized regimen development.Aynur Abdulla Ting Zhang Shanhe Li Wenke Guo Antony RWarden Yufang Xin Nokuzola Maboyi Jiatao Lou Haiyang Xie Xianting Ding 2022Microsystems & Nanoengineering2022,8,1:1
6Single-Cell Microwell Platform Reveals Circulating Neural Cells as a Clinical Indicator for Patients with Blood-Brain Barrier Breakdown显示文摘Central nervous system diseases commonly occur with the destruction of the blood-brain barrier.As a primary cause of morbidity and mortality,stroke remains unpredictable and lacks cellular biomarkers that accurately quantify its occurrence and development.Here,we identify NeuN^(+)/CD45^(−)/DAPI^(+)phenotype nonblood cells in the peripheral blood of mice subjected to middle cerebral artery occlusion(MCAO)and stroke patients.Since NeuN is a specific marker of neural cells,we term these newly identified cells as circulating neural cells(CNCs).We find that the enumeration of CNCs in the blood is significantly associated with the severity of brain damage in MCAO mice(p<0:05).Meanwhile,the number of CNCs is significantly higher in stroke patients than in negative subjects(p<0:0001).These findings suggest that the amount of CNCs in circulation may serve as a clinical indicator for the real-time prognosis and progression monitor of the occurrence and development of ischemic stroke and other nervous system disease.Yu Zhang Antony R.Warden Khan Zara Ahmad Yanlei Liu Xijun He Minqiao Zheng Xinlong Huo Xiao Zhi Yuqing Ke Hongxia Li Sijia Yan Wenqiong Su Deng Cai Xianting Ding 2021Research2021,,1:0
7Determination of Polynomial Degree in the Regression of Drug Combinations显示文摘Studies on drug combinations are becoming more and more popular in the past few decades, with the development of computer and algorithms. One of the most common methods in optimizing drug combinations is regression of a polynomial model based on certain number of experimental observations. In this paper, we study how to determine the degree of polynomials in different circumstances of drug combination optimization. Using cross-validation, we have found that in most cases, a high degree results in failures of accurate prediction, named overfitting. 9Exploring phytochemicals of Withania somnifera from different vicinity for functional foods显示文摘The well-known relation between food and bioactives explores the great prospective of innovative and novel food to maintain or improve health,increasing the demand investigating advance products with medicinal effect.Withania somnifera also known as‘Ashwagandha’in India,is one of the most utilized plants in different ancient medicine systems.The present work is an effort to develop a knowledge-based concept and a quality control approach for the development of functional foods.The study took into account the standardization criteria and examined how different geographies or different field sites(of India)would influence the phytoconstituents and their distribution in the different samples of W.somnifera.Phytoconstituents level were strongly influenced by soil quality,cultivar,and growing season.An increase in phenolic content,flavonoid content,antioxidants and the quality yield,were all correlated with increased phytoconstituent concentrations.The highest hydro-methanolic extract yield(by reflux(Soxhlet),sonication(20 min)and maceration(16 h)was from the sample collected from Rajasthan(18.61%,m/m)followed by West Bengal(11.21%)and Madhya Pradesh(12.67%)respectively.The methanolic extract of Rajasthan sample(produced by the reflux(Soxhlet)extraction method)had the highest concentrations of flavonoids(65.32 mg quercetin equivalent(QE)/g)and the total phenols(60.35 mg gallic acid equivalent(GAE)/g),while Madhya Pradesh sample(distilled water reflux(Soxhlet))extract had the highest antioxidant content(74.13%).High performance thin layer chromatography(HPTLC)method was also utilized for identification of withanolides A.Chromatographic separations were carried out using V(ethyl acetate):V(toluene):V(acetic acid)=9.1:1:0.6 as the mobile phase.The gas chromatograph-mass spectrometer(GC-MS)study of hydro-methanolic Soxhlet extract of W.somnifera from 3 different regions of India was done and the total phytochemical substances identified were 132 in sample from Rajasthan,107 in sample from West Bengal,and 164 in sample from Madhya Pradesh respectively.To conclude,the present study demonstrates how,within a framework of integrated agricultural practices,yield and phytochemicals merge into a new paradigm of functional foods.Pooja Dhama Xianting Ding Alok Sharma 2023Journal of Future Foods2023,3,3:0

11Bioinspired Rotation Microneedles for Accurate Transdermal Positioning and Ultraminimal-Invasive Biomarker Detection with Mechanical Robustness显示文摘Microneedle permits transdermal biosensing and drug delivery with minor pain.However,accurate microneedle transdermal positioning with minimal skin deformation remains a significant technical challenge due to inhomogeneous skin topology and discontinuous force applied to the microneedle.Here,we introduce bioinspired rotation microneedles for in vivo accurate microneedle positioning as inspired by honeybees’stingers.We demonstrate the benefits of rotation microneedles in alleviating skin resistance through finite element analysis,full-thickness porcine validations,and mathematical derivations of microneedleskin interaction stress fields.The max penetration force was mitigated by up to 45.7%and the force attenuation rate increased to 2.73 times in the holding stage after penetration.A decrease in max skin deflection and a faster deformation recovery introduced by rotation microneedles implied a more precise penetration depth.Furthermore,we applied the rotation microneedles in psoriasis mice,a monogenic disorder animal model,for minimally invasive biological sample extraction and proinflammatory cytokine monitoring.An ultrasensitive detection method is realized by using only one microneedle to achieve cytokine mRNA level determination compared to commonly required biopsies or blood collection.Thus,rotation microneedles permit a simple,rapid,and ultraminimal-invasive method for subcutaneous trace biological sample acquisition and subsequent point-of-care diagnostics with minimal damage to both microneedles and skins.Yilin Cai Shiyi Huang Zhinan Zhang Jiazheng Zhang Xingyue Zhu Xiaoxiang Chen Xianting Ding 2022Research2022,,3:0
12A Hashing-Based Framework for Enhancing Cluster Delineation of High-Dimensional Single-Cell Profiles显示文摘Although many methods have been developed to explore the function of cells by clustering high-dimensional(HD)single-cell omics data,the inconspicuously differential expressions of biomarkers of proteins or genes across all cells disturb the cell cluster delineation and downstream analysis.Here,we introduce a hashing-based framework to improve the delineation of cell clusters,which is based on the hypothesis that one variable with no significant differences can be decomposed into more diversely latent variables to distinguish cells.By projecting the original data into a sparse HD space,fly and densefly hashing preprocessing retain the local structure of data,and improve the cluster delineation of existing clustering methods,such as PhenoGraph.Moreover,the analyses on mass cytometry dataset show that our hashing-based framework manages to unveil new hidden heterogeneities in cell clusters.The proposed framework promotes the utilization of cell biomarkers and enriches the biological findings by introducing more latent variables.Xiao Liu Ting Zhang Ziyang Tan Antony RWarden Shanhe Li Edwin Cheung Xianting Ding 2022Phenomics2022,2,5:0
13Safety evaluation of nanodiamond-doxorubicin complexes in a Naïve Beagle canine model using hematologic,histological,and urine analysis显示文摘While doxorubicin(DOX)is one of the most common chemotherapeutic drugs for treating cancer,use of DOX must be managed carefully due to dose-related toxicity.Nanodiamond(ND)drug delivery system conjugated with DOX(NDX)has been reported to enhance treatment efficacy and attenuate toxicity in murine cancer models.In addition,extensive biocompatibility studies indicate that NDs seem to be well tolerated in non-human primates.Before the clinical translation of NDX,it is necessary to verify the safety of ND in large mammals.Studies of nanomedicine drug safety for large animal are not commonly reported,and this work represents a key milestone in bridging earlier advances towards clinical assessment.Herein,NDs’safety as a drug-delivery platform was evaluated in Naïve Beagle dogs.The study is performed with DOX,ND,and NDX in a dual-gender animal model using intravenous(IV)injection and hepatic portal vein(HPV)injection methods.The dogs are monitored for their health phenotype changes in continuous 5 days.Blood and urine obtained are for clinical pathology research.The results indicate that ND drug delivery platform significantly relieves DOX toxicity for Naïve Beagle dog model.This study provides guidance for the pre-clinical safety assessment of NDX therapy at large animal level.Liping Wang Wenqiong Su Khan Zara Ahmad Xin Wang Ting Zhang Youyi Yu Edward Kai-Hua Chow Dean Ho Xianting Ding 2022Nano Research2022,15,4:0
