2Development of a Haploid-Inducer Mediated Genome Editing System for Accelerating Maize Breeding显示文摘Crop breeding aims to generate pure in bred lines with multiple desired traits. Doubled haploid (DH) and genome editing using CRISPR/Cas9 are two powerful game-changing technologies in crop breeding. However, both of them still fall short for rapid generation of pure elite lines with integrated favorable traits. Here, we report the development of a Haploid-Inducer Mediated Genome Editing (IMGE) approach, which utilizes a maize haploid inducer line carrying a CRISPR/Cas9 cassette targeting for a desired agronomic trait to pollinate an elite maize in bred line and to generate genome-edited haploids in the elite maize background. 3Spurious Thermoluminescence Characteristics of the Ediacaran Doushantuo Formation(ca. 635–551 Ma) and Its Implications for Marine Dissolved Organic Carbon Reservoir显示文摘The Ediacaran Doushantuo Formation(ca. 635–551 Ma) deposited immediately after the last Neoproterozoic glaciations and recorded the most prominent negative excursions of carbonate carbon isotopic composition(δ^(13)C_(carb)). These excursions have been interpreted as a result of widespread remineralization of a large dissolved organic carbon(DOC) reservoir in the Ediacaran deep oceans. However, there is no direct evidence so far found in rocks for the proposed DOC reservoir, which devalues such an interpretation. Here, we conducted a detailed study on the glow-curves characteristics and signal origins of spurious thermoluminescence(TL) of the Doushantuo Formation at Jiulongwan in Yangtze Gorges area, South China, through sequential tests under CO_2, N_2 and air. Spurious TL intensities for test samples before and after removing soluble organic matter via accelerated solvent extraction(ASE) are nearly identical. Further, significant positive correlation between the spurious TL intensity and total inorganic carbon(TIC) content(R^2=0.7) indicate that the Doushantuo spurious TL with the characteristic peak at 393.5 °C from the sequential test is chemiluminescence(CL) which is derived from the oxidation of a type of non-volatile organic matter strongly associated with carbonate mineral lattice(termed as 'X-OM'). A most likely explanation is that the X-OM is a type of dissolved organic matter which co-precipitated with carbonate minerals into sediments in the Ediacaran Doushantuo Ocean. Furthermore, a significant exponential negative correlation(R^2=0.55) is observed between the CL data and the isotopic difference between carbonate and coexisting bulk organic matter(i.e., Δ^(13)C_(carb-org), a proxy for remineralization degree of DOC reservoir in proposed DOC hypothesis), suggesting that the X-OM was derived from the oxidation of the DOC reservoir in the Ediacaran Ocean. We thus propose that the X-OM and its CL detected in our study may have recorded the evolution of the possible DOC reservoir in the Ediacaran Doushantuo Ocean. If this is correct, the stratigraphic variations of the CL intensity in the Doushantuo Formation at Jiulongwan support the pulsed oxidation of the DOC reservoir in the Ediacaran Ocean. Our findings indicate that the CL derived from the oxidation of non-volatile organic matter which is strongly associated with carbonate mineral lattices in rocks may provide a feasible approach for probing the evolution of DOC reservoir in the ancient oceans, thus likely provide direct geological evidence for the development of oceanic DOC reservoir in geological times.Haiyang Wang Chao Li Chaoyong Hu Shucheng Xie 2015Journal of Earth Science2015,26,6:12
4FHY3 and FAR1 Integrate Light Signals with the miR156-SPL Module-Mediated Aging Pathway to Regulate Arabidopsis Flowering显示文摘In response to competition for light from their neighbors,shade-intolerant plants flower precociously to ensure reproductive success and survival.However,the molecular mechanisms underlying this key developmental switch are not well understood.Here,we show that a pair of Arabidopsis transcription factors essential for phytochrome A signaling,FAR-RED ELONGATED HYPOCOTYL3(FHY3)and FAR-RED IMPAIRED RESPONSE1(FAR1),regulate flowering time by integrating environmental light signals with the miR156-SPL module-mediated aging pathway.We found that FHY3 and FAR1 directly interact with three flowering-promoting SQUAMOSA-PROMOTER BINDING PROTEIN-LIKE(SPL)transcription factors,SPL3,SPL4,and SPL5,and inhibit their binding to the promoters of several key flowering regulatory genes,including FRUITFUL(FUL),LEAFY(LFY),APETALA1(AP1),and MIR172C,thus downregulating their transcript levels and delaying flowering.Under simulated shade conditions,levels of SPL3/4/5 proteins increase,whereas levels of FHY3 and FAR1 proteins decline,thus releasing SPL3/4/5 from FHY3/FAR1 inhibition to allow activation of FUL,LFY,AP1,and MIR172C and,consequently,early flowering.Taken together,these results unravel a novel mechanism whereby plants regulate flowering time by integrating environmental cues(such as light conditions)and an internal developmental program(the miR156-SPL module-mediated aging pathway).Yurong Xie Qin Zhou Yongping Zhao Quanquan Li Yang Liu Mengdi Ma Baobao Wang Rongxin Shen Zhigang Zheng Haiyang Wang 2020Molecular Plant2020,13,3:8
5Stereotactic body radiation therapy versus radiofrequency ablation in patients with small hepatocellular carcinoma:a systematic review and meta-analysis显示文摘Background:This study aimed to compare the clinical outcomes and toxicity between small hepatocellular carcinoma(HCC)patients treated with stereotactic body radiation therapy(SBRT)and those treated with radiofrequency ablation(RFA).Methods:We searched databases for relevant clinical studies.The primary outcomes of interest were overall survival(OS)at 1 and 2 years,freedom from local progression(FFLP)rate at 2 years,and complications.Results:Five cohorts from 5 retrospective studies and 4,814 patients with HCC were included.Pooled OS at 2 years was significantly lower for SBRT than for RFA[odds ratio(OR):0.63;95%confidence interval(CI):0.51-0.79;P<0.0001],but the pooled FFLP rate at 2 years was higher for SBRT than for RFA(OR:1.66;95%CI:1.05-2.61;P=0.03).In addition,there was no significant difference in the local and liver toxicities of the two treatments.The contradictory conclusion between the OS and FFLP outcome may be attributed to the difference in radiological dose and location,but there were no uniform criteria to illustrate the radiological dose and location in the included studies.Conclusions:SBRT had a higher local control ratio but poorer prognosis than RFA in patients with small HCC.The local toxicity was comparable in both treatments.Further trials should be designed with uniform standards for SBRT and RFA treatments.Jiawei Hong Linping Cao Haiyang Xie Yuanxing Liu Jun Yu Shusen Zheng 2021Hepatobiliary Surgery and Nutrition2021,10,5:5
6Structural shifts in the intestinal microbiota of rats treated with cyclosporine A after orthotropic liver transplantation显示文摘Understanding the effect of immunosuppressive agents on intestinal microbiota is important to reduce the mortality and morbidity from orthotopic liver transplantation (OLT). We investigated the relationship between the commonly used immunosuppressive agent cyclosporine A (CSA) and the intestinal microbial variation in an OLT model. The rat samples were divided as follows:(1) N group (normal control);(2) I group (isograft LT, Brown Norway [BN] rat to BN);(3) R group (allograft LT, Lewis to BN rat);and (4) CSA group (R group treated with CSA). The intestinal microbiota was assayed by denaturing gradient gel electrophoresis profiles and by using real-time polymerase chain reaction. The liver histopathology and the alanine/aspartate aminotransferase ratio after LT were both ameliorated by CSA. In the CSA group, the numbers of rDNA gene copies of Clostridium cluster I, Clostridium cluster XIV, and Enterobacteriaceae decreased, whereas those of Faecalibacterium prausnitzii increased compared with the R group. Cluster analysis indicated that the samples from the N, I, and CSA groups were clustered, whereas the other clusters contained the samples from the R group. 7Genome-wide profiling of long noncoding RNA expression patterns and CeRNA analysis in mouse cortical neurons infected with different strains of borna disease virus显示文摘Borna disease virus 1(BoDV-1)is neurotropic prototype of Bornaviruses causing neurological diseases and maintaining persistent infection in brain cells of mammalian species.Long non-coding RNA(lncRNA)is transcript of more than 200 nucleotides without proteincoding function regulating various biological processes as proliferation,apoptosis,cell migration and viral infection.However,regulatory of lncRNAs in BoDV-1 infection remains unknown.To identify differential expression profiles and predict functions of lncRNA in BoDV-1 infection,microarray data showed that 3528 lncRNAs and 2661 lncRNAs were differentially expressed in Strain V and Hu-H1 BoDV-infected groups compared with control groups,respectively.Gene Ontology(GO)and pathway analysis suggested that differential lncRNAs may be involved in regulation of metabolic,biological regulation,cellular process,endocytosis,viral infections and cell adhesion processes,cancer in both BoDV-infected strains.ENSMUST00000128469 was found down-regulated in both BoDV-infected groups compared with control groups consistent with microarray(p<0.05).ceRNA analysis indicated possible interaction networks as ENSMUST00000128469/miR-22-5p,miR-206-3p,miR-302b-5p,miR-302c-3p,miR-1a-3p/Igf1.Igf1 was found up-regulated in both BoDV-infected groups compared with control groups(p<0.05).Possible functions of predicted target mRNAs and miRNAs of ENSMUST00000128469 were involved in cell proliferation,transcriptional misregulation and proteoglycan pathways enriched in cancer.lncRNA may be involved in regulation of Hu-H1 inhibited cell proliferation and promoted apoptosis through NF-kB,JNK/MAPK signaling,BCL2 and CDK6/E2F1 pathways different from Strain V.Possible interaction networks as ENSMUST00000128469/miR-22-5p,miR-206-3p,miR-302b-5p,miR-302c-3p,miR-1a-3p/Igf1 may involve in regulation of cell proliferation,apoptosis,and cancer.Lin Sun Yujie Guo Peng He Xiaoyan Xu Xiong Zhang Haiyang Wang Tian Tang Wei Zhou Ping Xu Peng Xie 2019Genes & Diseases2019,6,2:5
8Severity of early allograft dysfunction following donation after circulatory death liver transplantation:a multicentre study显示文摘Background:Early allograft dysfunction(EAD)is associated with decreased graft and patient survival rates.This study aimed to identify the severity of EAD and develop a predictive model for EAD after donation after circulatory death(DCD)liver transplantation(LT).Furthermore,the influence of operative time on EAD incidence was also evaluated.Methods:In this retrospective,multicentre cohort study,nomograms were established based on a single-centre training cohort(n=321)and validated in a 3-center validation cohort(n=501).Results:The incidence rate of EAD was 46.4%(149/321)in the training cohort and 40.5%(203/501)in the validation cohort.Of the 149 EAD patients in the training cohort,77 patients with either elevated alanine aminotransferase(ALT)or aspartate aminotransferase(AST)were classified as having EAD type A,and the rest of the EAD patients were classified as having EAD type B.Recipients with EAD type B had lower graft and patient survival rates than recipients with EAD type A(P=0.043 and 0.044,respectively).We further developed a nomogram to predict EAD(graft weight,cold ischemia time,donor age,model for end-stage liver disease(MELD)score)and another nomogram to predict EAD type B(graft weight,cold ischemia time,MELD score).The nomograms for the prediction of EAD and EAD type B had good discrimination[concordance index(C-index)=0.712(0.666-0.758),0.707(0.641-0.773)]and calibration[Hosmer-Lemeshow(HL)P=0.384,P=0.425]in the validation cohort.An increased operative time(>6 h)was associated with increased EAD and EAD type B incidence in the high-risk group(P=0.005,P=0.020,respectively).Conclusions:EAD type B was associated with decreased graft and patient survival rates.The novel nomograms effectively predicted the incidence of EAD and EAD type B in DCD LT patients.Kun Wang Di Lu Yuhui Liu Wangyao Li Li Zhuang Zhenyu Ma Qinfen Xie Binhua Pan Yichao Wu Junli Chen Lidan Lin Xiaowen Feng Qiang Wei Xuyong Wei Haiyang Xie Zhengxin Wang Shusen Zheng Xiao Xu 2021Hepatobiliary Surgery and Nutrition2021,10,1:4

10The circFASN/miR-33a pathway participates in tacrolimusinduced dysregulation of hepatic triglyceride homeostasis显示文摘Dyslipidemia exhibits a high incidence after liver transplantation,in which tacrolimus,a widely used immunosuppressant,plays a fundamental role.MicroRNAs and related circRNAs represent a class of noncoding RNAs that have been recognized as important regulators of genes associated with lipid metabolism.However,their transcriptional activities and functional mechanisms in tacrolimus-related dyslipidemia remain unclear.In this study,we observed that tacrolimus could induce triglyceride accumulation in hepatocytes by stimulating sterol response element-binding proteins(SREBPs)and miR-33a.Our in silico and experimental analyses identified miR-33a as a direct target of circFASN.Tacrolimus could downregulate circFASN and result in elevated miR-33a in vivo and in vitro.Overexpression of circFASN or silencing of miR-33a decreased the promoting effects of tacrolimus on triglyceride accumulation.Clinically,the incidence of dyslipidemia in liver transplant recipients with elevated serum miR-33a after liver transplantation was higher than that in patients without elevated serum miR-33a(46.3%vs.18.8%p=0.012,n=73).Our results showed that the circFASN/miR-33a regulatory system plays a distinct role in tacrolimus-induced disruption of lipid homeostasis.MiR-33a is likely a risk factor for tacrolimus-related dyslipidemia,providing a potential therapeutic target to combat tacrolimus-induced dyslipidemia after liver transplantation.Chenzhi Zhang Kangchen Chen Rongli Wei Guanghan Fan Xuechun Cai Li Xu Beini Cen Jianguo Wang Haiyang Xie Shusen Zheng Xiao Xu 2020Signal Transduction and Targeted Therapy2020,5,1:2
11Effects of surface roughness on interfacial dynamic recrystallization and mechanical properties of Ti-6Al-3Nb-2Zr-1Mo alloy joints produced by hot-compression bonding显示文摘The influence of surface roughness on the interfacial dynamic recrystallization kinetics and mechanical properties of Ti-6 Al-3 Nb-2 Zr-1 Mo hot-compression bonding joints was systematically investigated.It is found that for the bonding interface of rough surfaces,elongated fine grains are formed at the bonding interface due to shear deformation of the interfacial area.As the surface roughness increases,the proportion of elongated grains drastically decreases as they further reorient to form equiaxed grains along the bonding interface of rougher surfaces resulting from severe incompatible deformation of the interface area.Meanwhile,high-density geometrically necessary dislocations accumulate around the interfacial recrystallization area to accommodate the incompatible strain and lattice rotation.A rotational dynamic recrystallization mechanism is thereby proposed to rationalize the formation of fine interfacial recrystallization grains during bonding of rough surfaces.In contrast to that of rough surfaces,bonding interface of polished surfaces exists in the form of straight interface grain boundaries without fine grains under the same deformation conditions.While with the increase of deformation strain,small grain nuclei form along the bonding interface,which is associated with discontinuous dynamic recrystallization assisted by strain-induced boundary migration of interface grain boundaries.Moreover,the bonding joints of rough surfaces show lower elongation compared with that of polished surfaces.This is because the formation of heterogeneous fine grains with low Schmid factor along the bonding interface of rough surfaces,leading to worse compatible deformation capability and thereby poor ductility of bonding joints.Bijun Xie Zhenxiang Yu Haiyang Jiang Bin Xu Chunyang Wang Jianyang Zhang Mingyue Sun Dianzhong Li Yiyi Li 2022Journal of Materials Science & Technology2022,,1:2
12Mining a coal seam below a heating goaf with a force auxiliary ventilation system at Longhua underground coal mine, China显示文摘Extraction of a coal seam which lies not far below a heating goaf can be a major safety challenge. A force auxiliary ventilation system was adopted as a control method in successful extraction and recovery of the panel 30110 of the #3 1coal seam, which is about 30–40 m below the heating goaf of the #2 2seam at Longhua underground coal mine, Shanxi Province, China. Booster fans and ventilation control devices such as doors and regulators were used in the system. The results show that, provided that a force auxiliary ventilation system is properly designed to achieve a pressure balance between a panel and its overlying goaf, the system can be used to extract a coal seam overlain by a heating goaf. 13Room-temperature conversion of ethane and the mechanism understanding over single iron atoms confined in graphene显示文摘The catalytic conversion of ethane to high value-added chemicals is significantly important for utilization of hydrocarbon resources.However, it is a great challenge due to the typically required high temperature(> 400 ℃) conditions.Herein, a highly active catalytic conversion process of ethane at room temperature(25 ℃) is reported on single iron atoms confined in graphene via the porphyrin-like N4-coordination structures.Combining with the operando time of flight mass spectrometer and density functional theory calculations, the reaction is identified as a radical mechanism, in which the C–H bonds of the same C atom are preferentially and sequentially activated, generating the value-added C2 chemicals, simultaneously avoiding the over-oxidation of the products to CO2.The in-situ formed O–FeN4–O structure at the single iron atom serves as the active center for the reaction and facilitates the formation of ethyl radicals.This work deepens the understanding of alkane C–H activation on the FeN4 center and provides the reference in development of efficient catalyst for selective oxidation of light alkane.Suheng Wang Haobo Li Mengqi He Xiaoju Cui Lei Hua Haiyang Li Jianping Xiao Liang Yu N.Pethan Rajan Zhaoxiong Xie Dehui Deng 2019Journal of Energy Chemistry2019,28,9:1
