| 2 | LGR5 is a promising biomarker for patients with stage Ⅰ and Ⅱ gastric cancer显示文摘Objective: To investigate Leucine-rich repeat-containing G protein-coupled receptor 5 (LGR5) expressions in gastric cancer and to evaluate its clinical significance. Methods: LGR5 expression was assessed by immunohistochemistry in 257 gastric cancer patients after surgery. The relationships between LGR5 expression and clinicopathological features and patients prognosis were statistically analyzed. Results: The expression of LGR5 was significantly higher in gastric cancers as a cancer stem cell marker than in adjacent normal tissues (P<0.001), and more frequently in patients with intestinal type, well-moderate differentiation and stage Ⅰ and Ⅱ (P<0.05). Although we found gastric cancer patients with LGR5 positive expression had a poorer prognosis, it didn't meet statistical significance (P>0.05). LGR5 negative expression was significantly related to the favorable overall survival in stage Ⅰ and Ⅱ gastric cancer patients (P<0.05). Furthermore, patients with high LGR5 expression tended to be more likely to get progression and have poorer progress-free survival (P<0.05). Multivariate Cox regression analysis revealed that LGR5 expression was an independent factor of overall survival for the patients with stage Ⅰ and Ⅱ gastric cancer (P<0.05). | 3 | High salt diet accelerates the progression of murine lupus through dendritic cells via the p38 MAPK and STAT1 signaling pathways显示文摘The increased incidence of systemic lupus erythematosus(SLE)in recent decades might be related to changes in modern dietary habits.Since sodium chloride(NaCl)promotes pathogenic T cell responses,we hypothesize that excessive salt intake contributes to the increased incidence of autoimmune diseases,including SLE.Given the importance of dendritic cells(DCs)in the pathogenesis of SLE,we explored the influence of an excessive sodium chloride diet on DCs in a murine SLE model.We used an induced lupus model in which bone marrow-derived dendritic cells(BMDCs)were incubated with activated lymphocyte-derived DNA(ALD-DNA)and transferred into C57BL/6 recipient mice.We observed that a high-salt diet(HSD)markedly exacerbated lupus progression,which was accompanied by increased DC activation.NaCl treatment also stimulated the maturation,activation and antigenpresenting ability of DCs in vitro.Pretreatment of BMDCs with NaCl also exacerbated BMDC-ALD-DNA-induced lupus.These mice had increased production of autoantibodies and proinflammatory cytokines,more pronounced splenomegaly and lymphadenopathy,and enhanced pathological renal lesions.The p38 MAPK–STAT1 pathway played an important role in NaClinduced DC immune activities.Taken together,our results demonstrate that HSD intake promotes immune activation of DCs through the p38 MAPK–STAT1 signaling pathway and exacerbates the features of SLE.Thus,changes in diet may provide a novel strategy for the prevention or amelioration of lupus or other autoimmune diseases. | Ze Xiu Xiao Xiaojiang Hu Ximei Zhang Zhigang Chen Julie Wang Ke Jin Feng Lin Cao Baoqing Sun Joseph A.Bellanti Nancy Olsen Song Guo Zheng | 2020 | Signal Transduction and Targeted Therapy2020,5,1: | 6 |

Yunnanensis显示文摘The phenomena of anther opening and closing in Paris polyphylla var. yunnanensis (Franch) Hand.-Mazz were described in detail, and the effects of ecological factors on those phenomena related to anther opening, closing and the fly-pollination mechanism were discussed. Anthers of P. polyphylla var. yunnanensis open in the morning and close in the evening every day during an over 20 days’ period of anthesis. Light was detected as the main factor controlling this daily anther opening and closing. Anther opening can be reversed in response to rain and re-opens when rain stops. Relative humidity (RH) has a slight effect on anther opening and closing. Flower spans can be prolonged under laboratory conditions. | 5 | Autologous fibroblasts induce fibrosis of the nucleus pulposus to maintain the stability of degenerative intervertebral discs显示文摘Lumbar degenerative disc diseases cause low back pain(LBP). The maintenance of the height and stability of the intervertebral disc(IVD) space is an effective treatment for LBP. The following study evaluated the effects of fibroblast injection on intervertebral disc degeneration(IDD) in a preclinical setting. Compared with the IDD group, the fibroblast treatment group demonstrated effective maintenance of IVD height, reduced endplate degeneration, and improved nuclear magnetic resonance signals and overall histological structure. In doing so, fibrotic IVDs maintained the stability and biomechanics of the vertebra. This finding is in agreement with clinical findings that human nucleus pulposus(NP) fibrosis is essential for the maintenance of IVD height and mechanical properties in patients following percutaneous endoscopic lumbar discectomy(PELD). Mechanistically, we demonstrated that injected fibroblasts not only proliferated but also induced NP cells to adopt a fibrotic phenotype via the secretion of TGF-β.Finally, to better mimic human conditions, the efficacy of autologous fibroblast injection in the treatment of IDD was further examined in a nonhuman primate cynomolgus monkey model due to their capacity for upright posture. We showed that the injection of fibroblasts could maintain the IVD height and rescue IVD signals in cynomolgus monkeys. Taken together, the results of our study reveal that autologous fibroblast injection can enhance the natural process of fibrosis during acute and subacute stages of stress-induced IDD. | 6 | The role of autophagic and lysosomal pathways in ischemic brain injury显示文摘Autophagy is involved in neural cell death after cerebral ischemia. Our previous studies showed that rapamycin-induced autophagy decreased the rate of apoptosis, but the rate of apoptosis was in-creased after the autophagy inhibitor, 3-methyladenine, was used. In this study, a suture-occluded method was performed to generate a rat model of brain ischemia. Under a transmission electron microscope, autophagic bodies and autophagy lysosomes were markedly accumulated in neurons at 4 hours post brain ischemic injury, with their numbers gradually reducing over time. Western blotting demonstrated that protein levels of light chain 3-II and cathepsin B were significantly in-creased within 4 hours of ischemic injury, but these levels were not persistently upregulated over time. Confocal microscopy showed that autophagy was mainly found in neurons with positive light chain 3 signal. Injection of rapamycin via tail vein promoted the occurrence of autophagy in rat brain tissue after cerebral ischemia and elevated light chain 3 and cathepsin B expression. However, in-jection of 3-methyladenine significantly diminished light chain 3-II and cathepsin B expression. Results verified that autophagic and lysosomal activity is increased in ischemic neurons. Abnormal components in cells can be eliminated through upregulating cell autophagy or inhibiting autophagy after ischemic brain injury, resulting in a dynamic balance of substances in cells. | 12 | Integrated Equivalent Model of Permanent Magnet Synchronous Generator Based Wind Turbine for Large-scale Offshore Wind Farm Simulation显示文摘The high-speed simulation of large-scale offshore wind farms(OWFs) preserving the internal machine information has become a huge challenge due to the large wind turbine(WT) count and microsecond-range time step. Hence, it is undoable to investigate the internal node information of the OWF in the electro-magnetic transient(EMT) programs. To fill this gap,this paper presents an equivalent modeling method for largescale OWF, whose accuracy and efficiency are guaranteed by integrating the individual devices of permanent magnet synchronous generator(PMSG) based WT. The node-elimination algorithm is used while the internal machine information is recursively updated. Unlike the existing aggregation methods, the developed EMT model can reflect the characteristics of each WT under different wind speeds and WT parameters without modifying the codes. The access to each WT controller is preserved so that the time-varying dynamics of all the WTs could be simulated. Comparisons of the proposed model with the detailed model in PSCAD/EMTDC have shown very high precision and high efficiency. | 13 | Characteristics of shale gas reservoirs in Wufeng Formation and Longmaxi Formation in Well WX2,northeast Chongqing显示文摘To further understand shale reservoir characteristics of Wufeng Formation and Longmaxi Formation in the Wuxi area,northeast Chongqing,based on drilling data of Well WX2,and taking the graptolite biostratigraphy as the standard marker of stratigraphic division and comparison,the geochemistry,petrology,reservoir space and properties of organic-rich black shale were well investigated,and its gasbearing capacity and controlling factors were also analyzed.The result shows that in the Wufeng Formation and Longmaxi Formation of Well WX2,the organic-rich shale is 89.8 m thick and is characterized by good kerogen type,high organic abundance,moderate maturity and favorable hydrocarbongeneration condition,and the graptolite sequence is developed completely and continuously;the organic abundance is influenced by depositional rate,and the slow depositional rate is favorable for accumulation of organic matter in the black graptolite shale;from top to bottom,content of siliceous minerals increases and content of clay minerals decrease,therefore the brittleness increases;the organicrich siliceous shale and clay siliceous shale are favorable lithofacies for development of shale reservoirs;the nanopore is dominated by the parallel-plate pore with four open sides and has good connectivity;the pore size distribution curve has the multimodal characteristic,and the pore diameter mainly is in the range of 0.42e0.62 nm and the range of 3e5 nm;organic pores and interlayer pores of clay minerals make the greatest contribution to the total pore volume,while pores of brittle minerals have the least contribution;from top to bottom,organic pores gradually increase while interlayer pores of clay minerals gradually decrease;the on-site core gas content exceeds 8 m3/t,and the gas-bearing capacity is jointly controlled by hydrocarbon generation,reservoir and preservation conditions;and the WF2-LM6 biozone of the Katian to the early Aeronian is the high-quality shale reservoir,where the LM1 biozone of the Hirnantian was the best“sweet spot”which is the target of horizontal well drilling. | Jin Wu Feng Liang Wen Lin Hongyan Wang Wenhua Bai Chao Ma Shasha Sun Qun Zhao Xiaojiang Song Rongze Yu | 2017 | Petroleum Research2017,2,4: | 1 |

| 16 | Evaluation of suspected local recurrence in lung cancer and head & neck cancer:A comparison between ^(99m)Tc-HL91 SPECT and CT for biopsy proven lesions显示文摘CT has rather low accuracy for the follow-up of tumors after therapy. This study was to determine whether the diagnostic accuracy can be improved with 99mTc-HL91 SPECT in comparison with parallel results of CT imaging. Thirty patients of lung cancer or head & neck cancer, suspected of recurrences on clinical symptoms and CT during clinical follow-up after therapy, underwent 99mTc-HL91 SPECT. The radioactivity ratios of tumor to normal tissues (T/NT) were calculated using the region of interest technique. Results of 99mTc-HL91 SPECT were verified by histopathology. The 99mTc-HL91 average uptake ratios of T/NT in the group of recurrent lesions and non-recurrent lesions were 1.58±0.16 and 1.18±0.14, respectively. A significant difference was found between T/NT data of the two phases (t=4.87, P<0.001). The 99mTc-HL91 SPECT shows sensitivity of 72.73%, specificity of 89.47% and accuracy of 83.83% for differentiating recurrent lesion, while the CT shows sensitivity of 63.63%, specificity of 84.21% and accuracy of 76.67%. A combination of 99mTc-HL91 SPECT and CT for 21 patients with lung cancers or head & neck cancers with congruent results shows sensitivity of 100%, specificity of 94.12% and accuracy of 95.23%. It is concluded that 99mTc-HL91 SPECT may play a role in differentiating recurrent lesions in patients with lung cancer and head & neck cancer. | 17 | Three-dimensional structure of axonal mitochondria reflects the age of drosophila显示文摘This study aimed to reconstruct a three-dimensional map of axonal mitochondria using Fiji and Neurolucida software, and to observe directly the morphology and distribution of mitochondria in axons of motor neurons in dorsal longitudinal flight muscles of drosophila aged 5 days and 20 days, using electron microscopy. Results indicated that there was no difference in the total area and volume of mitochondria between 5-day-old drosophila and 20-day-old drosophila in all sections, but the ratio of mitochondrial total areas to axon total areas, as well as mitochondrial density of 20-day-old drosophila, was lower than that of 5-day-old drosophila. The number of mitochondria, whose volume was less than 1 000 000 μm 3 , and between 1 000 000 μm 3 and 10 000 000 μm 3 , was higher in 20-day-old drosophila than that in 5-day-old drosophila. The number of mitochondria with a volume between 1 000 000 μm 3 and 100 000 000 μm 3 was apparently higher than those with a volume less than 1 000 000 μm 3 or larger than 100 000 000 μm 3 . In addition, the number of mitochondria with a volume more than 100 000 000 μm 3 was small; however, the volume was nearly 70% of the total volume in both 5-day-old and 20-day-old drosophila. In contrast, the number of mitochondria with a volume between 1 000 000 μm 3 and 10 000 000 μm 3 was large, but the volume was less than 30% of the total volume.
