| 3 | Recent progress in mass spectrometry proteomics for biomedical research显示文摘Proteins are the key players in many cellular processes. Their composition, trafficking, and interactions underlie the dynamic processes of life. Furthermore, diseases are frequently accompanied by malfunction of proteins at multiple levels. Understanding how biological processes are regulated at the protein level is critically important to understanding the molecular basis for diseases and often shed light on disease prevention, diagnosis, and treatment. With rapid advances in mass spectrometry(MS)instruments and experimental methodologies, MS-based proteomics has become a reliable and essential tool for elucidating biological processes at the protein level. Over the past decade, we have witnessed great expansion of knowledge of human diseases with the application of MS-based proteomic technologies, which has led to many exciting discoveries. Herein we review the recent progress in MS-based proteomics in biomedical research, including that in establishing disease-related proteomes and interactomes. | 4 | CFD-DEM simulation of spouting of corn-shaped particles显示文摘Three dimensionally coupled computational fluid dynamics (CFD) and discrete element method (DEM) were used to investigate the flow of corn-shaped particles in a cylindrical spouted bed with a conical base. The particle motion was modeled by the DEM, and the gas motion by the k-ε two-equation turbulent model. A two-way coupling numerical iterative scheme was used to incorporate the effects of gas-particle interactions in terms of momentum exchange. The corn-shaped particles were constructed by a multisphere method. Drag force, contact force, Saffman lift force, Magnus lift force, and gravitational force acting on each individual particle were considered in establishing the mathematical modeling. Calculations were carried out in a cylindrical spouted bed with an inside diameter of 200 mm, a height of 700 mm, and a conical base of 60 . Comparison of simulations with experiments showed the availability of the multi-sphere method in simulating spouting action with corn-shaped particles, but it depended strongly on the number and the arrangement of the spherical elements. Gas-solid flow patterns, pressure drop, particle velocity and particle concentration at various spouting gas velocity were discussed. The results showed that particle velocity reaches a maximum at the axis and then decreases gradually along the radial direction in the whole bed. Particle concentration increases along the radial direction in the spout region but decreases in the fountain region, while it is nearly constant in the annulus region. | 5 | Pediatric reference intervals in China(PRINCE):design and rationale for a large,multicenter collaborative cross-sectional study显示文摘There is a lack of accurate pediatric reference intervals(RIs) in China, with most commonly used RIs established without consideration of the effect of age and gender. The Pediatric Reference Intervals in China(PRINCE) project aims to establish and verify pediatric RIs for 31 common laboratory measurands.The project will be a large, multicenter cross-sectional study:14,490 healthy children and adolescents aged up to 19 years will be surveyed by 10 children's hospitals and one pediatric department of a university hospital. To evaluate the feasibility and efficiency of the study methods, 602 children were surveyed in the pilot phase of the PRINCE study in April 2017: it found that some measurands were distinctly age dependent and that there were differences between values for males and females. The results of the pilot study affirmed the necessity of the PRINCE project for Chinese pediatrics. The pilot also indicated potential difficulties in the full survey, e.g., difficulties in recruiting children aged under 3 years and insufficient collection of blood samples from infants. | 6 | Prognostic value of programmed death.1, programmed death-ligand 1, programmed death-ligand 2 expression, and CD8(+) T cell density in primary tumors and metastatic lymph nodes from patients with stage T1.4N+M0 gastric adenocarcinoma显示文摘Background: Anti-programmed death-1/programmed death-ligand 1(PD-1/PD-L1) immunotherapy has been proved to be effective on gastric cancer in ongoing clinical trials. However, the value of PD-L1 in predicting responses of patients with gastric cancer to anti-PD-1/PD-L1 immunotherapy is controversial. Some studies suggested that intra-and inter-tumoral heterogeneity of PD-L1 expression might explain the controversy.This study aimed to analyze the expression of PD-L1, PD-L2, and PD-1 as well as CD8(+) T-cell density in primary tumors and lymph nodes from patients with stage T1-4 N+M0 gastric adenocarcinoma to explore the heterogeneity of PD-1 signaling pathway molecules.Methods: In primary tumors and metastatic as well as non-metastatic lymph nodes from patients with stage T1-4 N+M0 gastric adenocarcinoma, we detected PD-L1 and PD-L2 expression with immunohistochemistry. CD8(+)T-cell density in primary tumors and PD-1 expression on CD8(+)T cells were detected with immunofluorescence. Univariate analysis was used to determine the prognostic values of them. Cox proportional hazard regression model was used to identify independent risk factors that affect patients' overall survival and disease-free survival.Results: Among 119 eligible patients who had undergone surgical resection, the positive rate of PD-L1 was higher in metastatic lymph nodes than in primary tumors(45.4% vs. 38.7%, P = 0.005); the positive rate of PD-1 on CD8(+)T cells was significantly higher in primary tumors and metastatic lymph nodes than in tumor-free lymph nodes(both P < 0.001). The intensity of PD-1 expression on CD8(+) T cells in primary tumors and in metastatic lymph nodes were stronger than that in tumor-free lymph nodes from the same patient. Beside, the positive rate of PD-L2 did not show any differences between primary tumors and metastatic lymph nodes. | 8 | Elucidating Variations in Nitrogen Requirement According to Yield, Variety and Cropping System for Chinese Rice Production显示文摘Better understanding of the factors that influence crop nitrogen(N) requirement plays an important role in improving regional N recommendations for rice(Oryza sativa L.) production. We collected data from 1 280 plot-level measurements in different reaches of the Yangtze River, China to determine which factors contributed to variability in N requirement in rice. Yield, variety, and cropping system were significantly related to N requirement. The N requirement remained consistent at about 18.6 kg N Mg^(-1)grain as grain yield increased from 7 to 9 Mg ha^(-1), then decreased to 18.1, 16.9, and 15.9 kg N Mg^(-1)grain as yield increased to 9–10, 10–11, and > 11 Mg ha^(-1), respectively. The decreased requirement for N with increasing yield was attributable to declining N concentrations in grain and straw and increased harvest index. Super rice variety had lower N requirement(17.7 kg N Mg^(-1)grain) than ordinary inbred and hybrid varieties(18.5 and 18.3 kg N Mg^(-1)grain, respectively), which was a result of lower grain and straw N concentrations of super rice. The N requirements were 19.2, 17.8, and 17.5 kg N Mg^(-1)grain for early, middle, and late rice cropping systems, respectively. | 9 | Creating a novel herbicide-tolerance OsALS allele using CRISPR/Cas9-mediated gene editing显示文摘Weeds and weedy rice plague commercial rice fields in many countries. Developingherbicide-tolerance rice is the most efficient strategy to control weed proliferation. CRISPR/Cas9-mediated gene editing, which generates small InDels and nucleotide substitutions atand around target sites using error-prone non-homologous end joining DNA repairing, hasbeen widely adopted for generation of novel crop germplasm with a wide range of geneticvariation in important agronomic traits. We created a novel herbicide-tolerance allele inrice by targeting the acetolactate synthase (OsALS) gene using CRISPR/Cas9-mediated geneediting. The novel allele (G628W) arose from a G-to-T transversion at position 1882 of OsALSand conferred a high level of herbicide tolerance. Transgene-free progeny carryinghomozygous G628W allele were identified and showed agronomic performance similar tothat of wild-type plants, suggesting that the G628W allele is a valuable resource fordeveloping elite rice varieties with strong herbicide tolerance. To promote use of the G628Wallele and to accelerate introgression and/or pyramiding of the G628W allele with other elitealleles, we developed a DNA marker for the G628W allele that accurately and robustlydistinguished homozygous from heterozygous segregants. | 14 | Construction and analysis of a plant transformation binary vector pBDGG harboring a bi-directional promoter fusing dual visible reporter genes显示文摘The constitutive promoter of cauliflower mosaic virus 35S (CaMV 35S) is a polar unidirectional promoter and is widely used in plant genetic engineering. In the present study, the unidirectional CaMV 35S promoter has been modified to a bi-directional promoter by fusing its minimal promoter element to the 5′end of CaMV 35S promoter in the opposite orientation. To qualitatively and quantitatively esti-mate its bi-directional transcriptional function and activity, two visible reporter genes, gusA (β-glucuronidase, GUS) and gfp (green fluo-rescent protein, GFP), were fused to the two ends of the promoter in bi-orientations ending with NOS terminator sequences, respectively. Stable expression of gusA and gfp genes in transgenic tobacco (Nicotiana tabacum L.) was visulized by histochemically staining for GUS and fluorescence microscopic observation under UV for GFP in transgenic plants. The expression of two reporter genes showed that the constructed bi-directional promoter did have the bi-directional transcriptional function in both expected orientations. The quantitative estimation of GUS and GFP were determined on a HITACHI F1000 Fluorescence Spectrophotometer with various wavelengths of excita-tion and emission. The GUS activity varied from 8 to 250 pmol 4-MU / min / mg protein and the GFP content varied from 0.9 to 1.8 μg / mg protein in various lines of transgenic tobacco plants. | 15 | Identification of micro RNAs in six solanaceous plants and their potential link with phosphate and mycorrhizal signalings显示文摘To date, only a limited number of solanaceous miRNAs have been deposited in the miRNA database. Here,Rgenome-wide bioinformatic identification of miRNAs was performed in six solanaceous plants(potato, tomato, tobacco,eggplant, pepper, and petunia). A total of 2,239 miRNAs were identified following a range of criteria, of which 982 were from potato, 496 from tomato, 655 from tobacco, 46 from eggplant,45 were from pepper, and 15 from petunia. The sizes of miRNA families and miRNA precursor length differ in all the species.Accordingly, 620 targets were predicted, which could be functionally classified as transcription factors, metabolic enzymes, RNA and protein processing proteins, and other proteins for plant growth and development. We also showed evidence for miRNA clusters and sense and antisense miR NAs.Additionally, five Pi starvation- and one arbuscular mycorrhiza(AM)-related cis-elements were found widely distributed in the putative promoter regions of the miRNA genes. Selected miRNAs were classified into three groups based on the presence or absence of P1BS and MYCScis-elements, and their expression in response to Pi starvation and AM symbiosis was validated by quantitative reverse transcription-polymerase chain reaction(qRT-PCR). | 19 | Interfacial characteristics and mechanical properties of additive manufacturing martensite stainless steel on the Cu-Cr alloy substrate by directed energy deposition显示文摘Copper/steel is a typical bimetal functional material,combining the excellent electrical and thermal conductivity of copper alloy and the high strength and hardness of stainless steel.There has been recent interest in manufacturing copper/steel bimetal by directed energy deposition(DED)due to its layer-bylayer method.However,cracks tend to form on the copper/steel interface because of the great difference in thermal expansion coefficient and crystal structure between copper and steel.In this work,interfacial characteristics and mechanical properties of the copper/steel bimetal were studied from one layer to multilayers.The laser power has a great influence on the Cu element distribution of the molten pool,affecting the crack formation dramatically on the solidification stage.Cracks tend to form along columnar grain boundaries because of the Cu-rich liquid films and spherical particles in the cracks.Crack-free and good metallurgical bonding copper/steel interface is formed at a scanning velocity of 800 mm/min and the laser power of 3000 W.The ultimate tensile strength(UTS)and the break elongation(EL)of the vertically combined crack-free copper/steel bimetal are 238.2±4.4 MPa and 20.6±0.7%,respectively.The fracture occurs on the copper side instead of the copper/steel interface,indicating that the bonding strength is higher than that of the Cu-Cr alloy.The UTS of the horizontally combined crack-free copper/steel bimetal is 746.7±22.6 MPa,which is 200%higher than that of the Cu-Cr alloy substrate.The microhardness is 398.6±5.4 HV at the steel side and is 235.3±64.1 HV at the interface,which is400%higher than that of the Cu-Cr alloy substrate.This paper advances the understanding of the interfacial characteristics of heterogeneous materials and provides guidance and reference for the fabrication of multi-material components by DED. | Wenqi Zhang Hailong Liao Zhiheng Hu Shasha Zhang Baijin Chen Huanqing Yang Yun Wang Haihong Zhu | 2021 | Journal of Materials Science & Technology2021,,31: | 2 |
| 20 | Seroprevalence of SARS-CoV-2 infections among children visiting a hospital显示文摘Importance:In this study,we retrospectively investigated the seroprevalence of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)antibodies within serum samples from children in Beijing,China.These findings provide preliminary guidance regarding population susceptibility to SARS-CoV-2,which will aid in establishing policy toward coronavirus disease 2019(COVID-19)prevention and control.Objective:To understand the seropositivity of anti-SARS-CoV-2 IgM/IgG antibodies among children in Beijing,China,evaluate the susceptibility of children in Beijing to SARS-CoV-2,and provide prima facie evidence to guide SARS-CoV-2 prevention and control.Methods:IgM/IgG antibody kits(colloidal gold)were used to conduct preliminary screening of SARS-CoV-2 IgM/IgG antibodies in serum samples of children who presented to Beijing Children’s Hospital,Capital Medical University,having fever or requiring hospitalization,from March 2020 to August 2020.Statistical analysis of anti-SARS-CoV-2 antibody seropositivity was performed according to the children’s general demographic characteristics,timing of admission to hospital,presence of pneumonia,and viral nucleic acid test results.Results:The study included 19797 children with both IgM and IgG antibody results.Twenty-four children had anti-SARS-CoV-2 IgM-positive results(positive rate of 1.2‰),twelve children had anti-SARS-CoV-2 IgG-positive results(positive rate of 0.6‰).Viral nucleic acid test results were negative for the above-mentioned children with positive antibody findings;during the study,two children exhibited positive viral nucleic acid test results,but their anti-SARS-CoV-2 IgM/IgG antibody results were negative.Anti-SARS-CoV-2 IgM antibody seropositivity was higher in the<1-year-old group than in the≥6-year-old group.The rates of anti-SARS-CoV-2 IgM seropositivity was highest in August from March to August;IgG results did not significantly differ over time.The rates of anti-SARS-CoV-2 IgM or IgG seropositivity among children with and without suspected pneumonia did not significantly differ between groups.Interpretation:During the study period,the rates of anti-SARS-CoV-2 IgM/IgG antibody seropositivity were low among children who presented to Beijing Children’s Hospital,Capital Medical University.The findings suggest that children in Beijing are generally susceptible to SARS-CoV-2 infection;COVID-19 prevention and control measures should be strengthened to prevent disease in children. | Ran Wang Fang Jin Shuhui Cao Hong Yuan Jingchen Qu Jiaqi Zhang Yuxuan Li Xiangpeng Chen Wenqi Song Zhengde Xie | 2020 | Pediatric Investigation2020,4,4: | 2 |
