1Spatiotemporal Variations and Regional Transport of Air Pollutants in Two Urban Agglomerations in Northeast China Plain显示文摘Characteristics of air pollution in Northeast China(NEC) received less research attention in the past comparing to other heavily polluted regions in China.Spatiotemporal variations of six criteria air pollutants(PM10, PM2.5, SO2, NO2, O3 and CO) in Central Liaoning Urban Agglomeration(CLUA) and Harbin-Changchun Urban Agglomeration(HCUA) in NEC Plain were analyzed in this study based on three-year hourly observations of air pollutants and meteorological variables from 2015 to 2017.The results indicated that the annual mean concentrations of air pollutants are generally higher in the middle and southern regions in NEC Plain and lower in the northern region.Megacities such as Shenyang, Harbin and Changchun experience severe air pollution, with a three-year averaged air quality index(AQI) larger than 80, far exceeding the daily AQI standard at the first-level of 50 in China.The annual mean PM and SO2 concentrations decrease most significantly in NEC urban agglomerations from 2015 to 2017, followed by CO and NO2, while O3 shows a slight increasing trend.All the six pollutants exhibit obvious seasonal and diurnal variations, and these variations are dictated by local emission and meteorological conditions.PM2.5 and O3 concentrations in NEC urban agglomerations strongly depend on wind conditions.High O3 concentrations at different cities usually occur in presence of strong winds but are independent on wind direction(WD), while high PM2.5 is usually accompanied by weak winds and poor dispersion condition, and sometimes also occur when the northerly or southerly winds are strong.Regional transport of air pollutants between NEC urban agglomerations is common.A severe haze event on November 1–4, 2017 is examined to demonstrate the role of regional transport on pollution.LI Xiaolan HU Xiaoming SHI Shuaiyi SHEN Lidu LUAN Lan MA Yanjun 2019Chinese Geographical Science2019,29,6:9
2Si-substrate vertical-structure InGaN/GaN micro-LED-based photodetector for beyond 10 Gbps visible light communication显示文摘Visible light communication(VLC)has emerged as a promising communication method in 6G.However,the development of receiving devices is much slower than that of transmitting devices,limited by materials,structures,and fabrication.In this paper,we propose and fabricate an InGaN/GaN multiple-quantum-well-based vertical-structure micro-LED-based photodetector(μPD)on a Si substrate.A comprehensive comparison of the photoelectrical performance and communication performance of three sizes ofμPDs,10,50,and 100μm,is presented.The peak responsivity of all threeμPDs is achieved at 400 nm,while the passband full-widths at half maxima are 87,72,and 78 nm for 10,50,and 100μmμPDs,respectively.The−20 dB cutoff bandwidth is up to 822 MHz for 50μmμPD.A data rate of 10.14 Gbps is experimentally demonstrated by bit and power loading discrete multitone modulation and the proposed digital pre-equalizer algorithm over 1 m free space utilizing the self-designed 4×450μmμPD array as a receiver and a 450 nm laser diode as a transmitter.This is the first time a more than 10 Gbps VLC system has been achieved utilizing a GaN-based micro-PD,to the best of our knowledge.The investigation fully demonstrates the superiority of Si substrates and vertical structures in InGaN/GaNμPDs and shows its great potential for high-speed VLC links beyond 10 Gbps.JIANYANG SHI ZENGYI XU WENQING NIU DONG LI XIAOMING WU ZIWEI LI JUNWEN ZHANG CHAO SHEN GUANGXU WANG XIAOLAN WANG JIANLI ZHANG FENGYI JIANG SHAOHUA YU NAN CHI 2022Photonics Research2022,10,10:5
3Two-dimensional Pd-based nanomaterials for bioapplications显示文摘Noble metal nanomaterials have been extensively explored in cancer diagnostic and therapeutic applications owing to their unique physical and chemical properties, such as facile synthesis, straightforward surface functionalization, strong photothermal effect, and excellent biocompatibility. Herein, we summarize the recent development of two-dimensional(2D) Pd-based nanomaterials and their applications in cancer diagnosis and therapy. Different synthetic strategies for Pd nanosheets and the related nanostructures, including Pd@Au, Pd@Ag nanoplates and mesocrystalline Pd nanocorolla, are first discussed.Together with their unique properties, the potential bioapplications of these 2D Pd nanomaterials are then demonstrated. With strong absorption in near-infrared(NIR) region, these nanomaterials have great potentials in cancer photothermal therapy(PTT). They also readily act as contrast agents in photoacoustic(PA) imaging or X-ray computed tomography(CT) to achieve image-guided cancer therapy. Moreover,significant efforts have been devoted to studying the combination of PTT and other treatment modalities(e.g., chemotherapy or photodynamic therapy) based on Pd nanomaterials. The remarkable synergistic or collaborative effects to achieve better therapeutic efficacy are discussed as well. Additionally, the biosafety of 2D Pd-based nanomaterials in vitro and in vivo was evaluated. 4Antiviral activity of Jinchai capsule against influenza virus显示文摘OBJECTIVE:To evaluate the effect on influenza virus of Jinchai,a capsule made of Traditional Chinese Medicine.METHODS:Madin-darby canine kidney(MDCK) cells were infected with the FM1 strain of influenza virus A(subtype H1N1) in vitro.They were used to explore how Jinchai affected cell adsorption,cell membrane fusion,transcription and replication of the influenza virus.Hemagglutinin(HA) protein,intracellular pH,and influenza virus protein acid(PA) polymerase subunit were detected with confocal microscopy and real-time fluorescent quantitative polymerase chain reaction.RESULTS:Jinchai significantly reduced the expression of HA and PA polymerase subunit mRNA in infected MDCK cells.Jinchai also significantly decreased intracellular pH in infected cells.CONCLUSIONS:Jinchai had strong anti-influenza activity against the influenza virus.It weakened the ability of the influenza virus to adsorb to cell wall and fuse with cell membranes in the early infection stage,and inhibited the transcription and replication of the virus.Juying Zhong Xiaolan Cui Yujing Shi Yingjie Gao Hongxin Cao 2013Journal of Traditional Chinese Medicine2013,33,2:4

6Effect of glutathione on in vivo biodistribution and clearance of surface-modified small Pd nanosheets显示文摘Plasmonic Pd nanosheets have been emerging as promising materials for application in near-infrared(NIR) photothermal therapy(PTT) of cancer. However, animal in mice studies indicated that the original synthesized poly(vinylpyrrolidone)(PVP)-protected small Pd nanosheets(Pd-PVP) and some further surface-modified small Pd nanosheets such as Pd-PEG(SH) easily accumulated in reticuloendothelial system(RES) organs(liver, spleen, etc.) and were difficult to be cleared from these organs quickly. In the work, we surprisingly found that glutathione(GSH) could promote the clearance of surface-modified small Pd nanosheets(e.g. Pd-PVP, Pd-PEG(SH) and Pd-GSH) from the RES organs efficiently. The effects of GSH on the biodistribution and clearance of different surface-modified Pd nanosheets were investigated. Our results indicated that these surface-modified Pd nanosheets with or without GSH added caused no morbidity at target primary organs, and GSH can promote the clearance of different surface-modified Pd nanosheets in the order of Pd-PVP≈Pd-PEG(SH)>Pd-GSH. 7Concentration of trace elements and their sources in a snow pit from Yuzhu Peak, north-east Qinghai-Tibetan Plateau显示文摘In order to understand the potential influence of pollution from human activity on the natural cycle of trace elements in the at- mosphere over the eastern Kunlun mountains, north-eastern part of the Qinghai-Tibetan Plateau, concentrations of Al, Fe, Ba, Cd, Co, Cr, Cu, Li, Ni, Zn, Pb, Sb, Sr, U and V in a 75 cm-deep snow pit recovered from 5,800 m a.s.l. of the Yuzhu Peak Glacier on October 25, 2009, were determined by a sector field inductively coupled plasma mass spectrometry. The results indicate that con- centrations of the measured trace elements vary by orders of magnitude from one element to another ranging from the minimum value 1.2 pg/g (U) to the maximum value 293 ng/g (Fe); the value of Max/Min ranges from 7 pg/g (Sd) to 358 pg/g (Li). EFc val- ues of measured trace elements show that there are other important sources except dust for Cd, Sb, Zn, Pb and Cu. Comparison of the concentrations of the measured trace elements with those in other areas in the Tibetan Plateau indicates that trace element con- centrations for Yuzhu Peak are lower than those in east Tianshan and Muztagh Ata (Pamirs). Concentrations of some trace ele- ments are also lower than those in East Rongbuk Glacier. 8Effect of Chinese Herbal Medicines on Helicobacter pylori-associated gastroduodenal ulcers: a systematic review and Meta-analysis显示文摘OBJECTIVE: To evaluate the effect and safety of Chinese herbal medicines on Helicobacter pylori(HP)-associated gastroduodenal ulcers.METHODS: All randomized controlled trials(RCTs)listed in the PubMed, EMBASE, Cochrane Library,China National Knowledge Infrastructure Database,WanFang, China Science and Technology Journal Database and SinoMed databases that were published in English or Chinese were searched, and the retrieval time range was from database inception to December 31, 2018. A comprehensive Meta-analysis of all publications was performed with RevMan 5.3 software, and the quality of the evidence report-ed in the results of Meta-analysis was analyzed with GRADE profiler software(version 3.6.1). Dichotomous data were analyzed by calculating odds ratios and 95% confidence intervals(CIs). Outcome measures included the HP clearance rate and percentage of adverse effects.RESULTS: Eight trials with 919 participants were included in this Meta-analysis. Compared with the effects of single drug therapy on HP-associated gastroduodenal ulcers, according to the statistical analysis, odds ratios for the HP clearance rate and percentage of adverse effects of Chinese herbal medicines administered as complementary medicines combined with drugs were 3.10 [95% CI(2.21, 4.36),P < 0.01] and 0.28 [95% CI(0.15, 0.52), P < 0.01], respectively, and the differences were statistically significant. According to the GRADE analysis, the quality of evidence for the HP clearance rate and percentage of adverse effects were both very low.CONCLUSION: Compared with using the drug therapy only, the combination of Chinese herbal medicines with the drug therapy more effectively eliminates HP and alleviates adverse reactions. 9Genetic dissection of husk number and length across multiple environments and fine-mapping of a major-effect QTL for husk number in maize(Zea mays L.)显示文摘Husk number(HN)and husk length(HL)influence the mechanical harvesting of maize grain.We investigated the genetic basis of HN and HL using a population of 204 recombinant inbred lines phenotypically evaluated in five environments.The two husk traits showed broad phenotypic variation and high heritability.Nine stable quantitative trait loci(QTL)were identified by single-environment mapping,comprising four QTL for HN and five for HL,and three QTL explained>10%of the phenotypic variation.Joint mapping revealed 22 additive QTL and 46 epistatic QTL.Both additive and epistatic(additive×additive)effects as well as a few large-effect QTL and some minor-effect QTL appeared to contribute to the genetic architecture of HN and HL.The QTL for HN located on chromosome 7,q HN7,which accounted for^20%of phenotypic variation,was detected in all five environments.q HN7 was fine-mapped to a 721.1 kb physical region based on the maize B73 Ref Gen_v3 genome assembly.Within this interval,four genes associated with plant growth and development were selected as candidate genes.The results will be useful for improvement of maize husk traits by molecular breeding and provide a basis for the cloning of q HN7.Guangfei Zhou Yuxiang Mao Lin Xue Guoqing Chen Huhua Lu Mingliang Shi Zhenliang Zhang Xiaolan Huang Xudong Song Derong Hao 2020The Crop Journal2020,8,6:2
10Endogenous Fe^(2+)-activated ROS nanoamplifier for esterase-responsive and photoacoustic imaging-monitored therapeutic improvement显示文摘Chemodynamic therapy(CDT)is well acknowledged as potent reactive oxygen species(ROS)-mediated anticancer strategy.Especially,the study about labile iron pool(LIP)as endogenous ferrous catalyzer has paved the way for future CDT development.However,limited H_(2)O_(2) expression,mild acidity,reduced glutathione(GSH)ablation of ROS,etc.,all require employing alternate peroxo-complex to achieve enhanced CDT effect.As a non-Fenton-type substrate,artesunate(ART)has been utilized as a source of free radicals through decomposition of endoperoxide bridges catalyzed by ferrous ions,nonetheless,the non-tumor-specific delivery,inferior pharmacokinetics,and hydrophobic nature minimize the efficacy of ART in physiological systems.Herein,we devise a PPA nanoamplifier by conjugating ART with PEG-functionalized Pd@Pt nanoplates(PP NPs)to form ester linkage,ensuring specific intratumoral esterase-responsive ART release.Significantly,the PPA nanoamplifier combines the in situ decomposition of ARTs endoperoxide bridges by Fe^(2+) to superoxide anions(O_(2)^(·-))and peroxidase(POD)-like enzymatic catalysis of endogenous H_(2)O_(2) by PP to hydroxyl radicals(*OH),thus achieving amplified ROS-mediated tumor therapy.Besides,PPA displays GSH destruction potential,thereby protecting ROS from the cleavage by GSH oxidation.In addition,the strong absorption of PPA in near-infrared(NIR)region also endows PPA with photoacoustic property to realize imaging-guided CDT.In short,by taking advantages of the high enrichment and esterase-responsive drug release at tumor sites,PPA amplified ROS signals via dual pathways,killing tumor cells in vitro are inhibiting tumor growth in vivo,thereby realizing high-efficiency non-Fenton CDT.We believe our novel anti-tumor strategy based on PPA will broaden the future of ROS-mediated tumor-targeted therapy.Sijin Xiang Zhongxiong Fan Ziehen Ye Tianbao Zhu Dao Shi Shefang Ye Zhenqing Hou Xiaolan Chen 2022Nano Research2022,15,2:2
11Spatial heterogeneity and host repression in fig-fig wasp mutualism显示文摘It is generally believed that physical heterogeneity in common resource or evolutionary restraint can sufficiently prevent direct conflict between host and symbionts in mutualism systems. Our data on fig/fig wasp reciprocal mutualism(Ficus racemosa), however, show that structural barriers of female flowers or genetic constraints of pollinators previously hypothesized exist, but cannot sufficiently maintain the mutualism stability. The results show that a positive relationship between seed and wasp production could be maintained in warm season, which might be because of density dependence restraint among foundresses and their low oviposition and pollination efficiency, keeping common resource(female flowers) utilization unsaturated. Whilst, a negative correlation between wasp offspring and viable seed production was also observed in cold season, which might be that the increased oviposition and pollination efficiency maximized the common resource utilization. The fitness trade-off between fig and pollinator wasps is greatly affected by environmental or ecological variations. The local stability might result from temporal low exploitation efficiency of pollinators together with interference competition among pollinators. We suggest that host repression through the active regulation of bract closure, which can create interference competition among the foundresses and prevent extra more foundresses sequential entry in fruit cavities, would help the figs avoiding the cost of over-exploitation. 12Biomimetic Ti-6Al-4V alloy/gelatin methacrylate hybrid scaffold with enhanced osteogenic and angiogenic capabilities for large bone defect restoration显示文摘Titanium-based scaffolds are widely used implant materials for bone defect treatment.However,the unmatched biomechanics and poor bioactivities of conventional titanium-based implants usually lead to insufficient bone integration.To tackle these challenges,it is critical to develop novel titanium-based scaffolds that meet the bioadaptive requirements for load-bearing critical bone defects.Herein,inspired by the microstructure and mechanical properties of natural bone tissue,we developed a Ti-6Al-4V alloy(TC4)/gelatin methacrylate(GelMA)hybrid scaffold with dual bionic features(GMPT)for bone defect repair.GMPT is composed of a hard 3D-printed porous TC4 metal scaffold(PT)backbone,which mimics the microstructure and mechanical properties of natural cancellous bone,and a soft GelMA hydrogel matrix infiltrated into the pores of PT that mimics the microenvironment of the extracellular matrix.Ascribed to the unique dual bionic design,the resultant GMPT demonstrates better osteogenic and angiogenic capabilities than PT,as confirmed by the in vitro and rabbit radius bone defect experimental results.Moreover,controlling the concentration of GelMA(10%)in GMPT can further improve the osteogenesis and angiogenesis of GMPT.The fundamental mechanisms were revealed by RNA-Seq analysis,which showed that the concentration of GelMA significantly influenced the expression of osteogenesis-and angiogenesis-related genes via the Pi3K/Akt/mTOR pathway.The results of this work indicate that our dual bionic implant design represents a promising strategy for the restoration of large bone defects.Limin Ma Xiaolan Wang Ye Zhou Xiongfa Ji Shi Cheng Dong Bian Lei Fan Lei Zhou Chengyun Ning Yu Zhang 2021Bioactive Materials2021,6,10:1
13Electrochemical behaviors of silicon wafers in silica slurry显示文摘The electrochemical behaviors of n-type silicon wafers in silica-based slurry were investigated, and the influences of the pH value and solid content of the slurry on the corrosion of silicon wafers were studied by using electrochemical DC polarization and AC impedance techniques. The results revealed that these factors affected the corrosion behaviors of silicon wafers to different degrees and had their suitable parameters that made the maximum corrosion rate of the wafers. 14Reduced smooth muscle-fibroblasts transformation potentially decreases intestinal wound healing and colitis-associated cancer in ageing mice显示文摘Cancer and impaired tissue wound healing with ageing are closely related to the quality of life of the elderly population.Given the increased incidence of cancer and the population ageing trend globally,it is very important to explore how ageing impairs tissue wound healing and spontaneous cancer.In a murine model of DSS-induced acute colitis and AOM/DSS-induced colitis-associated cancer(CAC),we found ageing significantly decreases intestinal wound healing and simultaneous CAC initiation,although ageing does not affect the incidence of AOM-induced,sporadic non-inflammatory CRC.Mechanistically,reduced fibroblasts were observed in the colitis microenvironment of ageing mice.Through conditional lineage tracing,an important source of fibroblasts potentially derived from intestinal smooth muscle cells(ISMCs)was identified orchestrating intestinal wound healing and CAC initiation in young mice.However,the number of transformed fibroblasts from ISMCs significantly decreased in ageing mice,accompanied by decreased intestinal wound healing and decreased CAC initiation.ISMCs-fibroblasts transformation in young mice and reduction of this transformation in ageing mice were also confirmed by ex-vivo intestinal muscular layer culture experiments.We further found that activation of YAP/TAZ in ISMCs is required for the transformation of ISMCs into fibroblasts.Meanwhile,the reduction of YAP/TAZ activation in ISMCs during intestinal wound healing was observed in ageing mice.Conditional knockdown of YAP/TAZ in ISMCs of young mice results in reduced fibroblasts in the colitis microenvironment,decreased intestinal wound healing and decreased CAC initiation,similar to the phenotype of ageing mice.In addition,the data from intestine samples derived from inflammatory bowel disease(IBD)patients show that activation of YAP/TAZ also occurs in ISMCs from these patients.Collectively,our work reveals an important role of the ageing stromal microenvironment in intestinal wound healing and CAC initiation.Furthermore,our work also identified a potential source of fibroblasts involved in colitis and CAC.Yi Liu Yanhong Ji Ruiyi Jiang Chao Fang Gang Shi Lin Cheng Yinan Zuo Yixin Ye Xiaolan Su Junshu Li Huiling Wang Yuan Wang Yi Lin Lei Dai Shuang Zhang Hongxin Deng 2023Signal Transduction and Targeted Therapy2023,8,9:0
15Variation in Sunshine Duration and Related Aerosol Influences at Shangdianzi GAW Station, China: 1958–2021显示文摘Sunshine duration(SD) is adopted widely to study global dimming/brightening. However, long-term simultaneous measurements of SD and closely related impact factors require further analysis to elucidate how and why SD has varied during the past decades. In this study, a long-term(1958–2021) SD data series obtained from the Shangdianzi Global Atmosphere Watch(GAW) station in China was analyzed to detect linear trends, climatic jumps, and climatic periods in SD using linear fitting, the Mann–Kendall trend test, and the continuous wavelet transform method. Annual SD exhibited steady dimming(-67.3 h decade-1) before 2010, followed by a period of brightening(189.9 h decade-1)during 2011–2020. An abrupt jump in annual SD occurred in 1995, and the annual SD anomaly exhibited significant oscillation with ~3-yr periodicity during 1960–1978. Partial least squares analysis revealed that annual SD anomaly was associated with variations in relative humidity, gale days, cloud cover, and black carbon(BC). Further analysis of the clear-sky daily sunshine percentage(DSP) and simultaneous measurements of aerosol properties, including aerosol optical depth, aerosol extinction coefficient, single scattering albedo(SSA), BC, and total suspended particulates, suggested that variation in DSP was affected primarily by aerosol scattering and absorption. Furthermore, the hourly clear-sky SD at high aerosol loading was approximately 60% and 56% of that at middle and low aerosol loadings, respectively. 17Feedforward loop profile among transcription factor, miRNA and mRNA in influenza A virus-infected mouse lungs显示文摘Purpose:In the present study,we focused on the 46 microRNAs and 719 genes in the microRNA-gene network,reported by us,and aimed to build a research blueprint of feedforward loops and reveal the key TFs in H1N1-infected mouse lung.Method:Based on microRNAs and genes in the microRNA-gene network previously reported by us,we used Jemboss software to find relationships between TFs and microRNAs(or genes),and then built a TF-microRNA-gene network exploiting the interactions between TFs and microRNAs(or genes).Next,we searched the sequences of above genes or microRNAs near the transcription start site(TSS)area,and then used the MatchTM algorithm to predict relevant TFs,and built the TF-Gene-Network.Result:We built a TF-microRNAgene network and exploreed eight key TFs,namely NF-AT1,GKLF,SRY,SOX10,AML1,MZF1,CRX and myogenin,in the network,and then constructed subgraphs of these eight TFs.Simultaneously,we predicted the possible target genes of microRNAs and identified the feedforward regulation relationship of possible TFs,microRNAs and mRNAs.The results showed that all eight factors with a score greater than 100 were TFs,namely NF-AT1,GKLF,SRY,SOX10,AML1,CRX,myogenin and MZF1.We then constructed subtables of the above eight TFs.Conclusion:In this study,TFs including NF-AT1,GKLF,SRY,SOX10,AML1,MZF1,CRX and myogenin showed the highest score(>100)not only in the TF-microRNA-gene network but also in feedforward loops,indicating that these eight TFs play the most important roles in mouse H1N1 influenza virus infection biology.Yanyan Bao Yujing Shi Yingjie Gao Lirun Zhou Xiaolan Cui 2019Medical Data Mining2019,2,4:0
18MiR-93-5p in BM-MSCs-derived exosomes amelio­rates renal fibrosis by affecting macrophage polar­ization显示文摘MiRNAs and macrophages play important roles in renal fibrosis.The exosomes secreted by bone marrow mesenchymal stem cells(BM-MSCs)can alleviate renal fibrosis.What is not clear,however,is whether a type of miRNAs in the BM-MSCs exosomes can alleviate renal fibrosis by modulating macrophage polarization.First,we take a high-throughput sequencing of miRNAs in exo­somes of BM-MSCs from chronic kidney disease(CKD)and normal people.Then we used the UUO mouse model and injected exosomes into the tail vein.The macrophages were stimulated with lipopolysaccharide(LPS).MSC-Exo or exosomes from BM-MSCs transfected with miR-93-5p inhibitor(Inhi-Exo)were added to the culture medium.The macrophages were transfected with miR-93-5p inhibitor or miR-93-5p mimic alone.The expression of miR-93-5p in exosomes of CKD patients was significantly decreased compared with normal people and in the LPS-stimulated macrophages and UUO mice kidneys.After stimulation with LPS,the macrophages polarized toward M1 subtype.MSC-Exo or miR-93-5p mimic promoted macrophages from M1 to M2 sub­type.Inhi-Exo or miR-93-5p inhibitor blocked the differentiation from M1 to M2 subtype.Significant fibrotic changes occurred in the kidneys of UUO mice,and M1 macrophages were significantly increased.After injecting exosomes into the tail vein of UUO mice,the degree of renal fibrosis was alleviated,the expression of miR-93-5p in the kidney was significantly increased,and the renal macrophages differentiated from M1 to M2 subtype.These results demonstrated that miR-93-5p in the exosomes derived from BM-MSCs can improve renal fibrosis by inducing macrophage differentiation from M1 to M2 subtype.Anqi Nie Jiaqi Shi Xuerong Wang Yuqing Lu Wufei Dai Jianhua Wu Jing Liu Xiaolan Chen 2022Life Research2022,5,3:0
19The role of Smad6 in immunity of the pearl oyster Pinctada fucata martensii显示文摘Inhibitory Smads(I-Smads),which belong to the Smad family and inhibit bone morphogenic protein 2(BMP2)signaling by a variety of mechanisms,can suppress innate immunity responses in vertebrates.However,there are no reports for the role of Smad6 in immunity in mollusks.In this study,we showed that Smad6 of the pearl oyster Pinctada fucata martensii was located in the Smad6 cluster of the phylogenetic tree;mRNA expression of Smad6 and Smad3 was up-regulated after lipopolysaccharide and polyinosinic:polycytidylic challenge;and transcript levels of Smad6 and Smad3 showed opposite patterns during wound healing.Under salinity stress,water inflow and outflow in the gills appear to be regulated by BMP2-Smads signals,and BMP2-Smads signaling may be closely related to the immune response.Our results indicate that Smad6 is involved in immunity,that it plays a positive role in the response to immune challenge and an inhibitory role during wound healing,and that Smad6 and Smad3 may work against each other.Yu SHI Xiaolan PAN Meng XU Huiru LIU Hanzhi XU Maoxian HE 2022Journal of Oceanology and Limnology2022,40,3:0
20Clinical heterogeneity of NLRP12-associated autoinflammatory diseases显示文摘Nod-like receptor family pyrin domain-containing protein 12 (NLRP12) is one of the critical pattern recognition receptors which participates in the regulation of multiple inflammatory responses. Mutations in NLRP12 cause exceptionally rare NLRP12-associated autoinflammatory disease (NLRP12-AID). So far, very few patients with NLRP12-AID have been identified worldwide;therefore, data on the clinical phenotype and genetic profile are limited. In this study, we reported 10 patients who presented mainly with periodic fever syndrome or arthritis. Next-generation sequencing (NGS) identified 6 heterozygous mutations of NLRP12, including 2 novel null mutations. Of the patients, some with same mutations showed different clinical features. Compared to healthy controls, the increased levels of cytokines were revealed in the patients' plasmas, as well as in the supernatants of patients’ cells stimulated with lipopolysaccharide (LPS) or tumor necrosis factor-α (TNF-α). The missense mutations did not change the protein expression;but decreased level of NLRP12 protein was shown in the null mutations. And in vitro expression assay demonstrated a truncating protein induced by the frameshift mutation. Further functional studies revealed the deleterious effect of mutations on nuclear factor-kappa B (NF-κB) signaling. Both the null and missense mutations impaired their inhibition of NF-κB activation induced by p65. Collectively, this study reported a relatively large NLRP12-AID case series. Our findings expand the clinical spectrum, and reinforce the diversity of genetic mutations and clinical phenotypes.
