3Effects of different aperture-sized type I collagen/silk fibroin scaffolds on the proliferation and differentiation of human dental pulp cells显示文摘This study aimed at evaluate the effects of different aperture-sized type I collagen/silk fibroin(CSF)scaffolds on the proliferation and differentiation of human dental pulp cells(HDPCs).The CSF scaffolds were designed with 3D mapping software Solidworks.Three different aperture-sized scaffolds(CSF1-CSF3)were prepared by low-temperature deposition 3D printing technology.The morphology was observed by scanning electron microscope(SEM)and optical coherence tomography.The porosity,hydrophilicity and mechanical capacity of the scaffold were detected,respectively.HDPCs(third passage,1105 cells)were seeded into each scaffold and investigated by SEM,CCK-8,alkaline phosphatase(ALP)activity and HE staining.The CSF scaffolds had porous structures with macropores and micropores.The macropore size of CSF1 to CSF3 was 421627 lm,579636 lm and 707643 lm,respectively.The porosity was 69.862.2%,80.162.8%and 86.563.3%,respectively.All these scaffolds enhanced the adhesion and proliferation of HDPCs.The ALP activity in the CSF1 group was higher than that in the CSF3 groups(P<0.01).HE staining showed HDPCs grew in multilayer within the scaffolds.CSF scaffolds significantly improved the adhesion and ALP activity of HDPCs.CSF scaffolds were promising candidates in dentine-pulp complex regeneration.Shihui Jiang Zhaoxia Yu Lanrui Zhang Guanhua Wang Xiaohua Dai Xiaoli Lian Yan Yan Linpu Zhang Yue Wang Ruixin Li Huiru Zou 2021Regenerative Biomaterials2021,8,4:2

5Metagenomic insights into the effects of submerged plants on functional potential of microbial communities in wetland sediments显示文摘Submerged plants in wetlands play important roles as ecosystem engineers to improve self-purification and promote elemental cycling.However,their effects on the functional capacity of microbial communities in wetland sediments remain poorly understood.Here,we provide detailed metagenomic insights into the biogeochemical potential of microbial communities in wetland sediments with and without submerged plants(i.e.,Vallisneria natans).A large number of functional genes involved in carbon(C),nitrogen(N)and sulfur(S)cycling were detected in the wetland sediments.However,most functional genes showed higher abundance in sediments with submerged plants than in those without plants.Based on the comparison of annotated functional genes in the N and S cycling databases(i.e.,NCycDB and SCycDB),we found that genes involved in nitrogen fixation(e.g.,nifD/H/K/W),assimilatory nitrate reduction(e.g.,nasA and nirA),denitrification(e.g.,nirK/S and nosZ),assimilatory sulfate reduction(e.g.,cysD/H/J/N/Q and sir),and sulfur oxidation(e.g.,glpE,soeA,sqr and sseA)were significantly higher(correctedp<0.05)in vegetated vs.unvegetated sediments.This could be mainly driven by environmental factors including total phosphorus,total nitrogen,and C:N ratio.The binning of metagenomes further revealed that some archaeal taxa could have the potential of methane metabolism including hydrogenotrophic,acetoclastic,and methylotrophic methanogenesis,which are crucial to the wetland methane budget and carbon cycling.This study opens a new avenue for linking submerged plants with microbial functions,and has further implications for understanding global carbon,nitrogen and sulfur cycling in wetland ecosystems.Binhao Wang Xiafei Zheng Hangjun Zhang Xiaoli Yu Yingli Lian Xueqin Yang Huang Yu Ruiwen Hu Zhili He Fanshu Xiao Qingyun Yan 2021Marine Life Science & Technology2021,3,4:1

8Environmental selection and evolutionary process jointly shape genomic and functional profiles of mangrove rhizosphere microbiomes显示文摘Mangrove reforestation with introduced species has been an important strategy to restore mangrove ecosystem functioning.However,how such activities affect microbially driven methane(CH4),nitrogen(N),and sulfur(S)cycling of rhizosphere microbiomes remains unclear.To understand the effect of environmental selection and the evolutionary process on microbially driven biogeochemical cycles in native and introduced mangrove rhizospheres,we analyzed key genomic and functional profiles of rhizosphere microbiomes from native and introduced mangrove species by metagenome sequencing technologies.Compared with the native mangrove(Kandelia obovata,KO),the introduced mangrove(Sonneratia apetala,SA)rhizosphere microbiome had significantly(p<0.05)higher average genome size(AGS)(5.8 vs.5.5 Mb),average 16S ribosomal RNA gene copy number(3.5 vs.3.1),relative abundances of mobile genetic elements,and functional diversity in terms of the Shannon index(7.88 vs.7.84)but lower functional potentials involved in CH4 cycling(e.g.,mcrABCDG and pmoABC),N2 fixation(nifHDK),and inorganic S cycling(dsrAB,dsrC,dsrMKJOP,soxB,sqr,and fccAB).Similar results were also observed from the recovered Proteobacterial metagenome-assembled genomes with a higher AGS and distinct functions in the introduced mangrove rhizosphere.Additionally,salinity and ammonium were identified as the main environmental drivers of functional profiles of mangrove rhizosphere microbiomes through deterministic processes.This study advances our understanding of microbially mediated biogeochemical cycling of CH_(4),N,and S in the mangrove rhizosphere and provides novel insights into the influence of environmental selection and evolutionary processes on ecosystem functions,which has important implications for future mangrove reforestation.Xiaoli Yu Qichao Tu Jihua Liu Yisheng Peng Cheng Wang Fanshu Xiao Yingli Lian Xueqin Yang Ruiwen Hu Huang Yu Lu Qian Daoming Wu Ziying He Longfei Shu Qiang He Yun Tian Faming Wang Shanquan Wang Bo Wu Zhijian Huang Jianguo He Qingyun Yan Zhili He 2023mLife2023,2,3:0
9Verify the Function of a Potential Growth-Regulating Gene in Marine Bivalve Using a Candidate Model Organism Mulinia lateralis显示文摘A better understanding of genetic bases of growth regulation is essential for bivalve breeding,which is helpful to improve the yield of the commercially important bivalves.While previous studies have identified some candidate genes accounting for variation in growth-related traits through genotype-phenotype association analyses,seldom of them have verified the functions of these putative,growth-related genes beyond the genomic level due to the difficulty of culturing commercial bivalves under laboratory conditions.Fortunately,dwarf surf clam Mulinia lateralis can serve as a model organism for studying marine bivalves given its short generation time,the feasibility of being grown under experimental conditions and the availability of genetic and biological information.Using dwarf surf clam as a model bivalve,we characterize E2F3,a gene that has been found to account for variation in growth in scallops by a previous genome-wide association study,and verify its function in growth regulation through RNA interference(RNAi)experiments.For the first time,E2F3 in dwarf surf clam,which is termed as MulE2F3,is characterized.The results reveal that dwarf surf clams with MulE2F3 knocked down exhibit a reduction in both shell size and soft-tissue weight,indicating the functions of MulE2F3 in positively regulating bivalve growth.More importantly,we demonstrate how dwarf surf clam can be used as a model organism to investigate gene functions in commercial bivalves,shedding light on genetic causes for variation in growth to enhance the efficiency of bivalve farming.WANG Chen KONG Lingling LIAN Shanshan YANG Zujing MENG Deting LI Moli ZHANG Xiangchao BAO Zhenmin HU Xiaoli 2023Journal of Ocean University of China2023,22,4:0
10Multi-Objective Optimization for an Industrial Grinding and Classification Process Based on PBM and RSM显示文摘The grinding and classification process is one of the key sub-processes in mineral processing, which influences the final process indexes significantly and determines energy and ball consumption of the whole plant. Therefore, optimal control of the process has been very important in practice. In order to stabilize the grinding index and improve grinding capacity in the process,a process model based on population balance model(PBM) is calibrated in this study. The correlation between the mill power and the operating variables in the grinding process is modelled by using the response surface method(RSM), which solves the problem where the traditional power modeling method relies on some unobservable mechanism-related parameters. On this basis, a multi-objective optimization model is established to maximize the useful power of the grinding circuit to improve the throughput of the grinding operation and improve the fraction of –0.074 mm particles in the hydrocyclone overflow to smooth the subsequent flotation operation. The elite non-dominated sorting genetic algorithm-II(NSGA-II) is then employed to solve the multi-objective optimization problem. Finally, subjective and objective weighting methods and integrated multi-attribute decision-making methods are used to select the optimal solution on the Pareto optimal solution set. 11Genetic segregation analysis of unsaturated fatty acids content in the filial generations of high-linolenic-acid rapeseed(Brassica napus)显示文摘Alpha linolenic acid(ALA)is an essential polyunsaturated fatty acids that can improve human health.Rapeseed is the second largest oil crop in the world but the ALA content in its seed fatty acids is only about 10%.Two rapeseed germplasms YH25005 and R8Q10 with high content(up to 21%)ALA were developed by intervarietal crossing.They were used as the maternal parent(P_(1))when crossed with a low ALA parent SW(P2)to produce the seeds of the F1 hybrid,F2,and backcrosses to P1(BCP1)and P2(BCP2).A multigeneration joint segregation analysis was conducted to determine major gene t polygene effects of the content of three major unsaturated fatty acids including oleic(OA),linoleic(LA)and ALA.The results showed that,although some genes favorable to ALA accumulation were not allelic in R8Q10 and YH25005,all the inheritances of OA,LA and ALA in YH25005×SW and R8Q10×SW followed a genetic model of‘two pairs of additive major genes t additive-dominant polygenes’.It is suggested that the contents of OA,LA and ALA are closely related and the major genes in the parent SW containing loss-of-function mutations in FAD2 and FAD3 loci had strong effect to reduce ALA and elevate OA.However,total genetic effect of the polygenes was greater than that of the two major genes,especial on ALA content.It indicated that it is necessary to employ a larger F2 population to find the plant that accumulate enough minor-effect polygenes for high ALA content.The results are useful for high ALA rapeseed breeding and future work of gene mapping.Xiaohan Zhang Jinglong Lian Chunyan Dai Xiaoli Wang Mingzheng Zhang Xing Su Yaohua Cheng Chengyu Yu 2021Oil Crop Science2021,6,4:0

13Involvement of GABAergic pathway on inhibition of subthalamic nucleus high-frequency stimulation of spontaneous firing activity in substantia nigra pars reticulata neurons in a rat model of Parkinson's disease显示文摘BACKGROUND: Subthalamic nucleus-high frequency stimulation (STN-HFS) plays an importantrole in the treatment of Parkinson's disease, but the mechanisms underlying STN-HFS remainunclear. Some studies have demonstrated that STN stimulation inhibits the firing activity ofsubstantia nigra pars reticulata neurons.OBJECTIVE: To investigate the effects of different-frequency STN stimulation andmicroiontophoresis of gamma-aminobutyric acid (GABA) and its antagonist, bicuculline, onspontaneous firing activity in a rat model of Parkinson's disease, and to analyze the action pathwayof high frequency stimulation in firing activity inhibition of substantia nigra pars reticulata neurons.DESIGN, TIME AND SETTING: This neuroelectrophysiological, animal experiment was performedat the Electrophysiology Laboratory of Liaoning Medical University, China from March to August2008.MATERIALS: 6-hydroxydopamine (6-OHDA) (Sigma, USA), A320R isolated stimulus and DAM80preamplifier (World Precision Instruments, USA), 6400A microiontophoresis apparatus (Dagan,USA), and Spike 2 biological signal acquisition system (CED, UK) were used in this study.METHODS: A total of 20 Sprague Dawley rats were used to establish a Parkinson's disease modelvia injection of 6-OHDA into the right striatum. Electrical stimulation (0.06-ms width, 0.4-mA intensity20-200-Hz frequency, 5-second train duration) was delivered to the subthalamic nucleus. Peripheralchannels were separately filled with GABA (pH 3.5, 0.2 mol/L), bicuculline (pH 4.0, 0.01 mol/L), andNaCl (pH 7.0, 3 mol/L). The electrode was positioned with a WK-2 microelectrode propulsion device,and was slowly inserted into the substantia nigra pars reticulata to record spontaneous firing activityof substantia nigra pars reticulata neurons.MAIN OUTCOME MEASURES: The number and firing rate of substantia nigra pars reticulataneurons which were either inhibited or excited were measured.RESULTS: Substantia nigra pars reticulata neurons were inhibited by STN stimulation. Theproportion of inhibited substantia nigra pars reticulata neurons increased with increasing stimulationfrequency. 14Changes in Global DNA Methylation Intensity and DNMT1 Transcription During the Aging Process of Scallop Chlamys farreri显示文摘DNA methylation is an important epigenetic regulatory mechanism that influences genomic stability, gene activation, X-chromosome inactivation and other factors. A change in DNA methylation is usually associated with aging and cellular senescence. DNA methyltransferase 1(DNMT1) is the most abundant DNA methyltransferase, and it plays an important role in maintaining the established methylation pattern during DNA replication in vertebrates. Although the effect of aging on DNA methylation has been well studied in vertebrates, little research has been conducted in invertebrates, especially in marine bivalves. In this study, we examined global DNA methylation levels in four groups of adult Zhikong scallop Chlamys farreri at different ages. The results showed that both the age and tissue type had a strong effect on the DNA methylation. In addition, a significant decrease in DNA methylation with aging(1–4 years) can be detected in mantle, kidney and hepatopancreas. We further measured the change in DNMT1 transcript abundance using quantitative reverse transcription PCR(q RT-PCR), which revealed that DNMT1 transcription significantly decreased with aging in mantle and hepatopancreas and strongly correlated with DNA methylation(R = 0.72).
