| 1 | Microvesicle-delivery miR-150 promotes tumorigenesis by up-regulating VEGF,and the neutralization of miR-150 attenuate tumor development显示文摘Tumor-associated macrophages(TAMs)mostly exhibit M2-like(alternatively activated)properties and play posi-tive roles in angiogenesis and tumorigenesis.Vascular en-dothelial growth factor(VEGF)is a key angiogenic factor.During tumor development,TAMs secrete VEGF and other factors to promote angiogenesis;thus,anti-treatment against TAMs and VEGF can repress cancer development,which has been demonstrated in clinical trials and on an experimental level.In the present work,we show that miR-150 is an oncomir because of its promotional effect on VEGF.MiR-150 targets TAMs to up-regulate their secre-tion of VEGF in vitro.With the utilization of cell-derived vesicles,named microvesicles(MVs),we transferred an-tisense RNA targeted to miR-150 into mice and found that the neutralization of miR-150 down-regulates miR-150 and VEGF levels in vivo and attenuates angiogenesis.There-fore,we proposed the therapeutic potential of neutralizing miR-150 to treat cancer and demonstrated a novel,natu-ral,microvesicle-based method for the transfer of nucleic acids. | Yuchen Liu Luming Zhao Dameng Li Yuan Yin Chen-Yu Zhang Jing Li Yujing Zhang | 2013 | Protein & Cell2013,4,12: | 18 |

| 3 | shRNA-mediated Slc38a1 silencing inhibits migration, but not invasiveness of human pancreatic cancer cells显示文摘Objective:Early metastasis is a major biological feature of pancreatic cancer.The current study examined whether silencing Slc38a1,a gene involved in energy metabolism,using short hairpin RNA(shRNA)could inhibit the growth,migration,and invasiveness of pancreatic cancer cells.Methods:A series of Slc38a1 shRNAs were designed and cloned into the pGPU6/GFP/Neo vectors.An shRNA with the most efficacious inhibitory action on SCL38A1 expression(65%inhibition)upon screening in DH5αbacteria was used to transfect SW1990 human pancreatic cancer cells.Cell growth,migration,and invasiveness were examined using cell counting kit-8,Boyden chamber without and with Matrigel,respectively.Results:Transfection of SW1990 cells with the SLCs38A1 shRNA significantly decreased the proliferation(P<0.0001)and migratory potential(by 46.7%,P=0.0399)of the cancer cells.Invasiveness,however,was not affected.Conclusions:Inhibiting Slc38a1 using shRNA technology could decrease the growth and migration of representative pancreatic cancer cells.However,the fact that invasiveness was not affected suggested that SLC38A1 is unlikely to be responsible for early metastasis. | Jing Xie Zhen Chen Luming Liu Ping Li Xiaoyan Zhu Huifeng Gao Zhiqiang Meng | 2013 | Chinese Journal of Cancer Research2013,25,5: | 5 |
| 4 | Proteasomal deubiquitinase UCH37 inhibits degradation of β-catenin and promotes cell proliferation and motility显示文摘The ubiquitin–proteasome system degrades most cellular proteins in eukaryotes.UCH37,also known as UCH-L5,is a deubiquitinase binding to Rpn13,a receptor for ubiquitinated substrates in the 26 S proteasome.But,it remains unclear how UCH37 influences the proteasomal degradation of the ubiquitinated substrates.Because deletion of UCH37 is embryonically lethal in mice,this study aims to investigate the role of UCH37 in proteasomal degradation by constructing the UCH37-deficient cell lines using CRISPR/Cas9 technology.Our results demonstrated that deletion of UCH37 decreased the levels of proteasomal Rpn13,implying that UCH37 might facilitate incorporation of Rpn13 into the proteasome.Meanwhile,deletion of UCH37 decreased the levels of β-catenin and the early endosomal protein Rab8.β-Catenin interacts with TCF/LEF to control transcription,and is involved in development,tissue homeostasis and tumorigenesis.We further found that deletion of UCH37 increased the levels of the ubiquitinated β-catenin and accelerated the hydrogen peroxide-stimulated degradation of β-catenin.Deletion of UCH37 also down-regulated the transcription of c-Myc,a downstream effector of β-catenin,and inhibited cell proliferation and motility.These results raise the possibility that UCH37 maintains the homeostasis of proteasomal degradation reciprocally by assisting the recruitment of the ubiquitin receptor Rpn13 into the proteasome and by reversing ubiquitination of certain critical substrates of the 26 S proteasome. | Zijian Li Luming Zhou Tianxia Jiang Libin Fan Xiaoying Liu Xiaobo Qiu | 2019 | Acta Biochimica et Biophysica Sinica2019,51,3: | 4 |
| 5 | Anti-tumor effect of the extract from Qingyihuaji formula on pancreatic cancer by down-regulating Notch-4 and Jagged-1显示文摘OBJECTIVE: To investigate, in terms of Notch signaling pathway, the effect on pancreatic cancer of the extract of an anti-tumor prescription — Qingyihuaji formula(QYHJ) — from Traditional Chinese Medicine(TCM).METHODS: Nude mice were implanted subcutaneously with human pancreatic cancer cell line SW1990 and then randomly divided into four groups: Control, QYHJ extract, Gemcitabine, and Combination of QYHJ extract and gemcitabine.Treatments were given for 21 days and tumor growth was evaluated simultaneously. Then, expression of Notch receptors(Notch-1, Notch-2,Notch-3, and Notch-4) and their Jagged ligands(Jagged-1 and Jagged-2) in dissected tumor tissuewere detected by real-time quantitative reverse transcription-polymerase chain reaction and Western blot. Finally, immunohistochemistry was performed to detect CD133, a marker of pancreatic cancer stem cells(CSCs), to evaluate the impact of QYHJ extract on pancreatic CSCs.RESULTS: QYHJ extract treatment effectively inhibited the tumor growth in nude mice. The expression of both Notch-4 and Jagged-1 were decreased significantly in QYHJ treatment groups(P < 0.05), while gemcitabine alone had no significant effect in down-regulating Jagged-1(P > 0.05).No significant difference was observed in the expression of Notch-1, Notch-2, Notch-3, and Jagged-2 between three treatment groups and control group(P > 0.05). Moreover, immunohistochemical analysis showed that the number of CD133 positive cells was significantly reduced by QYHJ treatment(P < 0.05), and the combined treatment was more effective than gemcitabine alone(P < 0.05).CONCLUSION: The role of the extract in pancreatic cancer treatment was associated with down-regulation of Notch-4 and Jagged-1 in Notch signaling pathway. | 7 | Ultrahigh rate capability of 1D/2D polyaniline/titanium carbide(MXene)nanohybrid for advanced asymmetric supercapacitors显示文摘High energy density and enhanced rate capability are highly sought-after for supercapacitors in today's mobile world.In this work,polyaniline/titanium carbide(MXene)(PANI/Ti3C2Tx)nanohybrid is synthesized through a facile and cost-effective self-assembly of.one-dimensional(10)PANI nanofibers and two-dimensional(20)Ti3C2Tx nanosheets.PANl!Ti3C2Tx delivers greatly improved specific capacitance,ultrahigh rate capability(67%capacitance retention from 1 to 100 A·g^(-1))as well as good cycle stability.Electrochemical kinetic analysis reveals that PANI/Ti3C2Tx is featured with surface capacitance-dominated process and has a quasi-reversible kinetics at high scan rates,giving rise to an ultrahigh rate capability.By using PANl!Ti3C2Tx as positive electrode,an 1.8 V aqueous asymmetric supercapacitor(ASC)is successfully assembled,showing a maximum energy density of 50.8 Wh·kg^(-1)·(at 0.9 kW-kg-1)and a power density of 18 kW·kg^(-1)(at 26 Wh·kg^(-1)).Moreover,an 3.0 V organic ASC is also elaborately fabricated,·by using PANI/Ti3C2Tx,achieving an ultrahigh energy density of 67.2 Wh·kg^(-1)(at 1.5 kW·kg^(-1))and a power density of 30 kW·kg^(-1)·(at 26.8 Wh·kg^(-1)).The present work not only improves fundamental understanding of the structure-property relationship towards ultrahigh rate capability electrode materials,but also provides valuable guideline for the rational design of high-performance:energy storage devices with both high energy and power densities. | Jinhua Zhou Qi Kang Shuchi Xu Xiaoge Li Cong Liu Lu Ni Ningna Chen Chunliang Lu Xizhang Wang Luming Peng Xuefeng Guo Weiping Ding Wenhua Hou | 2022 | Nano Research2022,15,1: | 2 |
| 8 | Melon short internode(CmSi)encodes an ERECTA-like receptor kinase regulating stem elongation through auxin signaling显示文摘Plant height is one of the most important agronomic traits that directly determines plant architecture,and compact or dwarf plants can allow for increased planting density and land utilization as well as increased lodging resistance and economic yield.At least four dwarf/semidwarf genes have been identified in different melon varieties,but none of them have been cloned,and little is known about the molecular mechanisms underlying internode elongation in melon.Here,we report map-based cloning and functional characterization of the first semidwarf gene short internode(Cmsi)in melon,which encodes an ERECTA-like receptor kinase regulating internode elongation.Spatial-temporal expression analyses revealed that CmSI exhibited high expression in the vascular bundle of the main stem during internode elongation.The expression level of CmSI was positively correlated with stem length in the different melon varieties examined.Ectopic expression of CmSI in Arabidopsis and cucumber suggested CmSI as a positive regulator of internode elongation in both species.Phytohormone quantitation and transcriptome analysis showed that the auxin content and the expression levels of a number of genes involved in the auxin signaling pathway were altered in the semidwarf mutant,including several well-known auxin transporters,such as members of the ABCB family and PINFORMED genes.A melon polar auxin transport protein CmPIN2 was identified by protein–protein interaction assay as physically interacting with CmSI to modulate auxin signaling.Thus,CmSI functions in an auxin-dependent regulatory pathway to control internode elongation in melon.Our findings revealed that the ERECTA family gene CmSI regulates stem elongation in melon through auxin signaling,which can directly affect polar auxin transport. | Sen Yang Kaige Zhang Huayu Zhu Xiaojing Zhang Wenkai Yan Nana Xu Dongming Liu Jianbin Hu Yufeng Wu Yiqun Weng Luming Yang | 2020 | Horticulture Research2020,7,1: | 2 |

| 15 | Development of branchless watermelon near isogenic lines by marker assisted selection显示文摘Pruning is time-consuming and laborious in watermelon cultivation,which can not meet the needs for simplified cultivation in the future.The development of branchless lines will provide important germplasms for breeding watermelon varieties and is an important method for genetic improvement.In this study,the watermelon accession,Wu Cha Zao(WCZ)is a branchless inbred line that carries the branchless gene Clbl,which was used as the donor parent to develop branchless near isogenic lines(NILs).To construct the NILs of Clbl,WCZ crossed with the normal branching watermelon inbred line WT20 which was used as the recurrent parent.The co-segregating markers dCAPS10 and Indel1 with Clbl were used for foreground selection,and a total of 108 SSR markers was selected with good polymorphism between two parental lines for background selection which had relatively uniform distribution across 11 chromosomes.Using these markers to select individuals from the BC_(1)F_(1),BC_(2)F_(1),and BC_(2)F_(2) generations,three NILs with a proportion of recurrent parent genome(PRPG)>99%were finally obtained.The lateral branch and plant height phenotypes did not significantly differ between the NILs and WCZ,indicating that the NILs of Clbl under the genetic background of WT20 has been successfully developed.These results provide ideal materials for further in-depth analysis of the genetic mechanisms of lateral branch development and ideal plant architecture breeding in watermelon. | Junling Dou Yinping Wang Huihui Yang Huanhuan Niu Dongming Liu Sen Yang Huayu Zhu Shouru Sun Luming Yang | 2022 | Horticultural Plant Journal2022,8,5: | 1 |

| 20 | HD-Zip Transcription Factor is Responsible for No-Lobed Leaf in Watermelon(Citrullus lanatus L.)显示文摘Leaf is a vital organ of plants that plays an essential role in photosynthesis and respiration.As an important agronomic trait in leaf development,leaf shape is classified into lobed,entire(no-lobed),and serrated in most crops.In this study,two-lobed leaf watermelon inbred lines WT2 and WCZ,and a no-lobed leaf watermelon inbred line WT20 were used to create two F_(2)populations.Segregation analysis suggested that lobed leaves were dominant over the no-lobed leaves,and it was controlled by a signal gene.A locus on watermelon chromosome 4 controlling watermelon lobed/no-lobed leaves was identified through BSA-seq strategy combined with linkage analysis.The candidate gene was fine-mapped to a 61.5 kb region between 21,224,481 and 21,285,957 bp on watermelon chromosome 4 using two F_(2)populations.Four functional genes were annotated in the candidate region,while sequences blast showed that there was a single-base deletion(A/-)only in the exon of Cla018360,which resulted in premature termination of translation in the no-lobed leaf lines.Function prediction showed that Cla018360 encodes an HD-Zip protein that has been reported to regulate the development of leaf shape.The single-base deletion also occurred in the HD-Zip domain.We inferred that the Cla018360 gene is the candidate gene for regulating the development of lobed/no-lobed leaves in watermelon.Gene expression analysis showed that Cla018360 was highly expressed in young leaves.Phylogenetic analysis showed that Cla018360 had a close genetic relationship with AtHB51,which had been reported to regulate the formation of leaf shape in Arabidopsis.Furthermore,transcriptome analysis showed that a total of 333 differentially expressed genes were identified between WT2 and WT20,of which 115 and 218 genes were upregulated and downregulated in no-lobed leaved watermelon WT20.This study not only provides a good entry point for studying leaf development but also provides foundational insights into breeding for special plant architecture in watermelon. | Shixiang Duan Yaomiao Guo Yinping Wang Muhammad Jawad Umer Dongming Liu Sen Yang Huanhuan Niu Shouru Sun Luming Yang Junling Dou Huayu Zhu | 2023 | Phyton-International Journal of Experimental Botany2023,92,5: | 1 |
