1Inflammatory response and neuronal necrosis in rats with cerebral ischemia显示文摘In the middle cerebral artery occlusion model of ischemic injury, inflammation primarily occurs in the infarct and peripheral zones. In the ischemic zone, neurons undergo necrosis and apoptosis, and a large number of reactive microglia are present. In the present study, we investigated the pathological changes in a rat model of middle cerebral artery occlusion. Neuronal necrosis appeared 12 hours after middle cerebral artery occlusion, and the peak of neuronal apoptosis appeared 4 to 6 days after middle cerebral artery occlusion. Inflammatory cytokines and microglia play a role in damage and repair after middle cerebral artery occlusion. Serum intercellular cell adhesion molecule-1 levels were positively correlated with the permeability of the blood-brain barrier. These findings indicate that intercellular cell adhesion molecule-1 may be involved in blood-brain barrier injury, microglial activation, and neuronal apoptosis. 3Novel Wx alleles generated by base editing for improvement of rice grain quality显示文摘Amylose content(AC),which is regulated by the Waxy(Wx)gene,is a major indicator of eating and cooking quality(ECQ)in rice(Oryza sativa).Thus far,only a limited number of mutations in the N-terminal domain of Wx were found to have a major impact on the AC of rice grains and no mutations with such effects were reported for other regions of the Wx protein.Here,nucleotide substitutions in the middle region of Wx were generated by adenine and cytosine base editors.The nucleotide substitutions led to changes in 15 amino acid residues of Wx,and a series of novel Wx alleles with ACs of 0.3%-29.43%(wild type with AC of 19.87%)were obtained.Importantly,the waxy~(abe2)allele showed a'soft rice'AC,improved ECQ,favorable appearance,and no undesirable agronomic traits.The transgenes were removed from the waxy~(abe2)progeny,generating a promising breeding material for improving rice grain quality.Xiaorui Huang Fei Su Sheng Huang Fating Mei Xiaomu Niu Changle Ma Hui Zhang Xiaoguo Zhu Jian-Kang Zhu Jinshan Zhang 2021Journal of Integrative Plant Biology2021,63,9:4
4Creation of aromatic maize by CRISPR/Cas显示文摘Aroma is an important quality parameter for breeding in rice(Oryza sativa).For example,the aromatic rice varieties basmati and jasmine rice,with a popcorn-like scent,are popular worldwide and routinely command a price premium.2-acetyl-1-pyrroline(2AP)is a key flavor compound among over 200 volatiles identified in fragrant rice.A naturally fragrant germplasm exists in multiple plant species besides rice,which all exhibit lower activity of BETAINE ALDEHYDE DEHYDROGENASE 2(BADH2).However,no equivalent aromatic germplasm has been described in maize(Zea mays).Here,we characterized the two maize BADH2 homologs,ZmBADH2a and ZmBADH2b.We generated zmbadh2a and zmbadh2b single mutants and the zmbadh2a-zmbadh2b double mutant by CRISPR/Cas in four inbred lines.A popcorn-like scent was only noticeable in seeds from the double mutant,but not from either single mutant or in wild type.In agreement,we only detected 2AP in fresh kernels and dried mature seeds from the double mutant,which accumulated between 0.028 and 0.723 mg/kg 2AP.These results suggest that ZmBADH2a and ZmBADH2b redundantly participate in 2AP biosynthesis in maize,and represent the creation of the world's first aromatic maize by simultaneous genome editing of the two BADH2 genes.Yanxiao Wang Xiaoqin Liu Xiuxiu Zheng Wenxia Wang Xunqing Yin Haifeng Liu Changle Ma Xiaomu Niu Jian-Kang Zhu Fei Wang 2021Journal of Integrative Plant Biology2021,63,9:3
5Kidney injury molecule-1 is a potential receptor for SARS-CoV-2显示文摘COVID-19 patients present high incidence of kidney abnormalities,which are associated with poor prognosis and mortality.The identification of SARS-CoV-2 in the kidney of COVID-19 patients suggests renal tropism of SARS-CoV-2.However,whether there is a specific target of SARS-CoV-2 in the kidney remains unclear.Herein,by using in silico simulation,coimmunoprecipitation,fluorescence resonance energy transfer,fluorescein isothiocyanate labeling,and rational design of antagonist peptides,we demonstrate that kidney injury molecule-1(KIM1),a molecule dramatically upregulated upon kidney injury,binds with the receptor-binding domain(RBD)of SARS-CoV-2 and facilitates its attachment to cell membrane,with the immunoglobulin variable Ig-like(Ig V)domain of KIM1 playing a key role in this recognition.The interaction between SARS-CoV-2 RBD and KIM1 is potently blockaded by a rationally designed KIM1-derived polypeptide AP2.In addition,our results also suggest interactions between KIM1 Ig V domain and the RBDs of SARS-CoV and MERS-CoV,pathogens of two severe infectious respiratory diseases.Together,these findings suggest KIM1 as a novel receptor for SARS-CoV-2 and other coronaviruses.We propose that KIM1 may thus mediate and exacerbate the renal infection of SARS-CoV-2 in a‘vicious cycle’,and KIM1 could be further explored as a therapeutic target.Chen Yang Yu Zhang Xia Zeng Huijing Chen Dong Yang Yuchen Chen Ziwei Shen Xiaomu Wang Xinran Liu Mingrui Xiong Hong Chen Kun Huang 2021Journal of Molecular Cell Biology2021,13,3:2

17RNA-Seq analysis and development of SSR and KASP markers in lentil(Lens culinaris Medikus subsp. culinaris)显示文摘Lentil(Lens culinaris Medikus subsp.culinaris,2 n=14)is a cool-season legume with high production potential for multiple uses.However,limited molecular research has been conducted in this species owing to its large genome,which impedes the generation of genome sequences and the development of molecular markers.In this study,more than 1.37 billion filtered clean reads were collected by RNA-Seq of six diverse lentil accessions and217,836 transcripts and 161,095 unigenes were de novo assembled,yielding respectively 257.1 and 240.6 million nucleotides.The mean transcript length was 1180 bp and the N50 and N90 lengths were respectively 2075 and 479 bp.The mean length of the unigenes was 1494 bp and their N50 and N90 values were respectively 2203 and 714 bp.The unigenes were annotated against seven databases.The FLOWERING LOCUS T(FT)gene homolog in lentil showed high protein sequence similarity to the FT gene homologs of pea and alfalfa.On the basis of the RNA-Seq analysis,26,449 EST-SSR markers were designed in silico,and 276 preliminarily screened markers were selected to evaluate polymorphism in 94 diverse lentil accessions.In total,125(45.29%)of 276 EST-SSR markers were found to be polymorphic.A total of 130,073 SNP loci were detected and 78(61.41%)of 127 SNPs were successfully converted to KASP markers.Population genetic analyses of the lentil accessions with EST-SSR and KASP markers revealed similar genetic structures,suggesting that the RNA-Seq-generated resources and the developed markers are reliable for use in molecular marker-assisted breeding of lentil.Dong Wang Tao Yang Rong Liu Nana Li Xiaomu Wang Ashutosh Sarker Xiaodong Zhang Runfang Li Yanyan Pu Guan Li Yuning Huang Yishan Ji Zhaojun Li Qian Tian Xuxiao Zong Hanfeng Ding 2020The Crop Journal2020,8,6:1
