| 4 | Modeling experimental study on weathering-leaching of Emeishan basalt and its relation with metallogenesis显示文摘The Emeishan basalt has a high Cu background value (170×10-6 on average), and thus provides a ore-forming material source for copper ores. The collected samples were exposed in basalt lavas of the third segment of the Emeishan basalt eruptive cycle. By using a set of automatically cycling glass apparatuses, weathering-leaching experiments by three kinds of rainwater on the collected samples were carried out in the open system (at normal temperature and normal pressure). The analysis results showed that the most intensive export of Cu occurred under acidic rainwater-induced weathering-leaching conditions, almost 2-3 orders of magnitude the export of Cu by mod-ern air rainwater, and 1-2 orders of magnitude higher than the export of Cu by CO2 rainwater. In addition, the total amount of Zn, Cu, U, Ni and Sr exported by acidic rainwater are greater than 1%. | 5 | Unraveling a genetic roadmap for improved taste in the domesticated apple显示文摘Although taste is an important aspect of fruit quality, an understanding of its genetic control remains elusive in apple and other fruit crops. In this study, we conducted genomic sequence analysis of 497 Malus accessions and revealed erosion of genetic diversity caused by apple breeding and possible independent domestication events of dessert and cider apples. Signatures of selection for fruit acidity and size, but not for fruit sugar content, were detected during the processes of both domestication and improvement. Furthermore, we found that single mutations in major genes affecting fruit taste, including Ma1, MdTDT, and MdSOT2, dramatically decrease malate, citrate, and sorbitol accumulation, respectively, and correspond to important domestication events. Interestingly, Ma1 was identified to have pleiotropic effects on both organic acid content and sugar:acid ratio, suggesting that it plays a vital role in determining fruit taste. Fruit taste is unlikely to have been negatively affected by linkage drag associated with selection for larger fruit that resulted from the pyramiding of multiple genes with minor effects on fruit size. | 7 | Distinct contribution of PD-L1 suppression by spatial expression of PD-L1 on tumor and non-tumor cells显示文摘Programmed cell death receptor 1(PD-1)and its ligand,PD-L1,are important immune checkpoint proteins.Although antibodies that block PD-1/PD-L1 have shown promising clinical efficacy in a subset of cancer patients,the detailed cellular and molecular mechanisms behind anti-PD-1 and anti-PD-L1 immunotherapy are not well defined.Specifically,the way in which PD-L1 contributes to immune suppression on tumor and non-tumor cells remains controversial.By selectively blocking PD-L1 on either tumor or non-tumor cells,we demonstrated that PD-L1 from both sources suppressed the anti-tumor T-cell response.Blocking PD-L1 on either tumor cells or non-tumor cells inhibited tumor growth and enhanced immune cell infiltration,as well as the tumor-specific T-cell response.Further,simultaneously blocking tumor-and non-tumor-derived PD-L1 maximized anti-tumor T-cell responses and demonstrated synergy.In addition,the relative contribution of PD-L1 on tumor and non-tumor cells to immune suppression depended on the PD-L1 expression level.Lastly,we found that the F4/80 receptor was involved in the anti-tumor effect of PD-L1 blockade.Taken together,our data indicate that PD-L1 on both tumor and non-tumor cells is critical for T-cell inhibition,which provides new directions for the optimization of PD-L1-blocking antibodies and the development of clinical biomarker strategies. | Xiaoqing Zhang Chen Cheng Jiyan Hou Xinyue Qi Xin Wang Ping Han Xuanming Yang | 2019 | Cellular & Molecular Immunology2019,16,4: | 3 |
| 8 | A post-GWAS confirming the genetic effects and functional polymorphisms of AGPAT3 gene on milk fatty acids in dairy cattle显示文摘Background: People are paying more attention to the healthy and balanced diet with the improvement of their living standards. Milk fatty acids(FAs) have been reported that they were related to some atherosclerosis and coronary heart diseases in human. In our previous genome-wide association study(GWAS) on milk FAs in dairy cattle, 83 genomewide significant single nucleotide polymorphisms(SNPs) were detected. Among them, two SNPs, ARS-BFGL-NGS-109493 and BTA-56389-no-rs associated with C18 index(P = 0.0459), were located in the upstream of 1-acylglycerol-3-phosphate O-acyltransferase 3(AGPAT3) gene. AGPAT3 is involved in glycerol-lipid, glycerol-phospholipid metabolism and phospholipase D signaling pathways. Hence, it was inferred as a candidate gene for milk FAs. The aim of this study was to further confirm the genetic effects of the AGPAT3 gene on milk FA traits in dairy cattle.Results: Through re-sequencing the complete coding region, and 3000 bp of 5′ and 3′ regulatory regions of the AGPA T3 gene, a total of 17 SNPs were identified, including four in 5′ regulatory region, one in 5′ untranslated region(UTR),three in introns, one in 3′ UTR, and eight in 3′ regulatory region. By the linkage disequilibrium(LD) analysis with Haploview4.1 software, two haplotype blocks were observed that were formed by four and 12 identified SNPs,respectively. Using SAS9.2, we performed single locus-based and haplotype-based association analysis on 24 milk FAs in 1065 Chinese Holstein cows, and discovered that all the SNPs and the haplotype blocks were significantly associated with C6:0, C8:0 and C10:0(P < 0.0001–0.0384). Further, with Genomatix, we predicted that four SNPs in 5′ regulatory region(g.146702957 G > A, g.146704373 A > G, g.146704618 A > G and g.146704699 G > A) changed the transcription factor binding sites(TFBSs) for transcription factors SMARCA3, REX1, VMYB, BRACH, NKX26, ZBED4, SP1, USF1, ARNT and FOXA1. | 9 | Sustainable management of Chinese grasslands—issues and knowledge显示文摘China has almost 400 Mhm^2 of grasslands,90% of which is considered degraded to varying degrees,on which 16 million herders depend for their livelihoods and many more indirectly, along the value-adding chain.Since 1950, average stocking rates across China have increased 4-fold. National policies have focused over recent decades on finding ways to rehabilitate the degraded grasslands, to sustain livestock production from them, and to improve the livelihoods of herder households, who are among the poorest people in China. A large collaborative program commenced in the early 2000 s to help find solutions to the sustainable management of grasslands.This paper summarizes key findings of many research projects, identifies where knowledge is weak and argues that the successful rehabilitation of grasslands will also require policies that provide incentives and support for herders as they move from a focus on survival to a focus on production of higher quality products, for which consumers are increasingly willing to pay. A key focus is to emphasize the improvement of animal production per head. When this is done, it naturally leads to lower stocking rates, which in turn provides the opportunities for grasslands to recover. Across a range of experiments, farm demonstrations and analyses using models, in Inner Mongolia and Gansu, a consistent result has been that a 50% reduction in stocking rates, improves net household income and starts the process of grassland rehabilitation.Rather than focusing on stocking rates, better management of grasslands could be achieved by maintaining the grasslands above critical values for herbage mass, values that help optimize botanical composition, reduce soil erosion, optimize animal growth rates and aid ecosystem functions. Managing to critical values for herbage mass is likely to be more effective than efforts to calculate sustainable stocking rates. An early summer rest is valuable for aiding grassland rehabilitation and summer productivity, but a total grazing ban(typically for 5 years)may not achieve its aims as evidence shows it may take 10–15 years to achieve a better grassland state. Lessdesirable plant species often increase in degraded grasslands that are rested and grazing can help manage those species. Surveys of herders indicate they have very mixed views on the benefits of total grazing bans that are unlikely to rehabilitate grasslands to an ideal botanical composition.The current objective is to work with grasslands that herders now have and optimize the existing composition.Grazing grasslands in winter results primarily in weight loss by animals and there is now evidence of how winter grazing reduces grassland growth in the next summer. It is better to keep animals in well-built sheds and feed them better, improved feeding through the cold months is required. | 11 | The LTR of endogenous retrovirus ev21 retains promoter activity and exhibits tissue specific transcription in chicken显示文摘Endogenous viruses integrate into the host genome and influence the expression of neighboring genes through their long terminal repeats (LTRs). In this study, we analyzed the promoter, enhancer and transcriptional activities of the chicken endogenous retrovirus ev21 LTR. The LTR was cloned into pGL3-basic and pGL3-promoter luciferase vectors in forward and reverse orientation separately. The luciferase activities were detected respectively in chicken embryonic fibroblast cell, human embryonic kidney cell line, human lung carcinoma cell line, Chinese hamster ovary cell line, and murine melanoma cell line. Relative luciferase activity analysis indicated that the ev21 LTR retained bi-directional promoter activity but no detectable enhancer activity in these cells. The constructs containing the LTR and F1 region show stronger promoter activity than the constructs containing only LTR. The transcriptional pattern of ev21 LTR varied in tissues of late feathering White Leghorn chicken at post-hatch day one. Skin exhibits the highest expression in the tissues examed. | 12 | Simulating experiment on the enrichment of precious metals in Lower Cambrian black shale series of Hunan and Guizhou provinces显示文摘Adsorption experiments were made at room temperature and neutral pH value on different types of min-erals associated with the Lower Cambrian black shale series polymetallic layers in Hunan and Guizhou provinces on nanometer-sized Pt colloids and PtCl42--bearing ionic solutions with an attempt to constrain the relationship between the different types of minerals in the polymetallic layers and the enrichment of platinum group elements (PGEs). Experimental results showed that the different types of minerals show strong selectivity to the adsorption of nano-meter-sized Pt colloids and PtCl42--bearing ionic solutions. Metallic sulfides, organic matter and clay minerals are the strong adsorbents of PGEs, while quartz, albite, muscovite and other silicate minerals show a week adsorbility to both of them. This phenomenon is well consistent with the geological fact that metallic sulfides, organic matter and clay minerals in the polymetallic layers of the black shale series are the major carrier minerals of PGEs, giving a thorough explanation to the mechanism of enrichment of previous metal elements. Adsorption may be a principal mechanism of enrichment of precious metal elements under lower temperature conditions.
