Reduced function and disassembled microtubules of cultured cardiomyocytes in spaceflight显示文摘Lack of gravity during spaceflight has profound effects on cardiovascular system, but little is known about how the cardiomyocytes respond to microgravity. In the present study, the effects of spaceflight on the structure and function of cultured cardiomyocytes were reported. The primary cultures of neo- natal rat cardiomyocytes were carried on Shenzhou-6 spacecraft and activated at 4 h in orbit. 8 samples were fixed respectively at 4, 48 and 96 h after launching for immunofluorescence of cytoskeleton, and 2 samples remained unfixed to analyze contractile and secretory functions of the cultures. Ground sam- ples were treated in our laboratory in parallel. After 115 h spaceflight, video recordings displayed that the number of spontaneous beating sites in flown samples decreased significantly, and the cells in the beating aggregate contracted in fast frequency without synchrony. Radioimmunoassay of the medium showed that the atrial natriuretic peptide secreted from flown cells reduced by 59.6%. Confocal images demonstrated the time-dependant disassembly of mirotubules versus unchanged distribution and or- ganization of microfilaments. In conclusion, above results indicate reduced function and disorganized cytoskeleton of cardiomyocytes in spaceflight, which might provide some cellular basis for further investigations to probe into the mechanisms underlying space cardiovascular dysfunction. | YANG Fen LI YingHui DING Bai NIE JieLin WANG HongHui ZHANG XiaoYou WANG ChunYan LING ShuKuan NI ChengZhi DAI ZhongQuan TAN YingJun WAN YuMin | 2008 | Chinese Science Bulletin2008,53,8: | 5
Exposing Cu-rich {110} active facets in PtCu nanostars for boosting electrochemical performance toward multiple liquid fuels electrooxidation显示文摘In catalysis, tuning the structural composition of the metal alloy is known as an efficient way to optimize the catalytic activity. This work presents the synthesis of compositional segregated six-armed PtCu nanostars via a facile solvothermal method and their distinct composition-structure-dependent performances in electrooxidation processes. The alloy is shown to have a unique six arms with a Cu-rich dodecahedral core, mainly composed of {110} facets and exhibit superior catalytic activity toward alcohols electrooxidation compared to the hollow counterpart where Cu was selectively etched.Density functional theory (DFT) calculations suggest that the formation of hydroxyl intermediate (OH^*) is crucial to detoxify CO poisoning during the electrooxidation processes. The addition of Cu is found to effectively adjust the d band location of the alloy catalyst and thus enhance the formation of ^*OH intermediate from water splitting, which decreases the coverage of ^*CO intermediate. Our work demonstrates that the unique compositional anisotropy in alloy catalyst may boost their applications in electrocatalysis and provides a methodology for the design of this type catalyst. | Liping Huang Wei Zhang Peng Li Yongbo Song Hongting Sheng Yuanxin Du Yang-Gang Wang Yuen Wu Xun Hong Yanhuai Ding Xiaoyou Yuan Manzhou Zhu | 2019 | Nano Research2019,12,5: | 4
Synthesis and photocatalytic activity of ZnO-Au_(25) nanocomposites显示文摘ZnO-Au_(25) nanocomposites were synthesized by doping Au_(25) nanoclusters into the porous Zn O nanospheres. It was notable that the ultrasmall Au_(25) nanoclusters possessed uniform sizes and fine dispersibility on the porous ZnO supports. A considerable correlation between the loading of Au_(25) nanoclusters and the photocatalytic activity was found. Compared with the pure ZnO nanospheres, the ZnO-Au_(25) nanocomposites exhibited more efficient photocatalytic activity in terms of degradation of Rhodamine B(RhB) in an aqueous solution. In addition, the possible photocatalytic mechanisms are discussed in this work. This strategy may be helpful for preparing other novel hybrid nanocomposites with well-defined structures and superior performances. | Junjie Hu Hai Yuan Peng Li Jian Wang Qiankun Liu Hui Wang Qilin Wang Xiaoyou Yuan | 2016 | Science China Chemistry2016,59,3: | 2
Smart drug delivery systems for precise cancer therapy显示文摘Nano-drug delivery strategies have been highlighted in cancer treatment, and much effort has been made in the optimization of bioavailability, biocompatibility, pharmacokinetics profiles, and in vivo distributions of anticancer nano-drug delivery systems. However, problems still exist in the delicate balance between improved anticancer efficacy and reduced toxicity to normal tissues, and opportunities arise along with the development of smart stimuli-responsive delivery strategies. By on-demand responsiveness towards exogenous or endogenous stimulus, these smart delivery systems hold promise for advanced tumor-specificity as well as controllable release behavior in a spatial-temporal manner. Meanwhile, the blossom of nanotechnology, material sciences, and biomedical sciences has shed light on the diverse modern drug delivery systems with smart characteristics, versatile functions, and modification possibilities. This review summarizes the current progress in various strategies for smart drug delivery systems against malignancies and introduces the representative endogenous and exogenous stimuli-responsive smart delivery systems. It may provide references for researchers in the fields of drug delivery, biomaterials, and nanotechnology. | Xiaoyou Wang Chong Li Yiguang Wang Huabing Chen Xinxin Zhang Cong Luo Wenhu Zhou Lili Li Lesheng Teng Haijun Yu Jiancheng Wang | 2022 | Acta Pharmaceutica Sinica B2022,12,11: | 2

Identification and construction of a novel biomimetic delivery system of paclitaxel and its targeting therapy for cancer显示文摘Dear Editor,Paclitaxel(PTX)is among the frst-line therapeutic agents against diverse malignancies. However, problems still exist concerning its poor water-solubility,multidrug resistance, lack of specific targeting ability and severe toxic effects, and these issues are far from fully resolved despite the various PTX formulations available on.the market,e.g.the gold standard,PTX albumin nanoparticles Abraxane I and PTX liposomes Lipusu. Some studies that try to solve the multiple problems faced by chemotherapy drug delivery, however, fell into the prevailing trap of overcomplicated formulation design which sacrifices the druggability. To better reconcile this paradox,a novel glycosylated liposomal PTX was designed, inspired by the cytomembrane glycosyls with important roles in.maintaining both membrane structure and physiological functions,3 such as enhan-cing membrane stability, evading immunological clearance, and recognizing corresponding receptors.Ginsenosides,as natural amphiphilic molecules, are structurally similar to cholesterol, contain glycosyl moieties, and are easily obtainable. | Xue Wang Wanwei Zheng Qing Shen Yahua Wang Yujen Tseng Zhongguang Luo Xiaoyou Wang Lei Shi Chong Li Jie Liu | 2021 | Signal Transduction and Targeted Therapy2021,6,2: | 1

Vitamin A injection at birth improves muscle growth in lambs显示文摘Vitamin A and its metabolite, retinoic acid(RA)play important roles in regulating skeletal muscle development. This study was conducted to investigate the effects of early intramuscular vitamin A injection on the muscle growth of lambs.A total of 16 newborn lambs were given weekly intramuscular injections of corn oil(control group,n=8)or 7,500 IU vitamin A palmitate(vitamin A group,n=8)from birth to 3 wk of age(4 shots in total). At 3 wk of age and weaning, biceps femoris muscle samples were taken to analyze the effects of vitamin A on the myogenic capacity of skeletal muscle cells. All lambs were slaughtered at 8 months of age.The results suggest that vitamin A treatment accelerated the growth rate of lambs and increased the loin eye area(P<0.05).Consistently,vitamin A increased the diameter of myofibers in longissimus thoracis muscle(P<0.01)and increased the final body weight of lambs(P<0.05).Vitamin A injection did not change the protein kinase B/mammalian target of rapamycin and myostatin signaling(P>0.05).Moreover,vitamin A upregulated the expression of PAX7(P<0.05)and the myogenic marker genes including MYOD and MYOG(P<0.01).The skeletal muscle-derived mononuclear cells from vitamin A-treated lambs showed higher expression of myogenic genes(P<0.05)and formed more myotubes(P<0.01)when myogenic differentiation was induced in vitro. In addition,in vitro analysis showed that RA promoted myogenic differentiation of the skeletal muscle-derived mononuclear cells in the first 3 d(P<0.05)but not at the later stage(P>0.05)as evidenced by myogenic gene expression and fusion index.Taken together, neonatal intramuscular vitamin A injection promotes lamb muscle growth by promoting the myogenic potential of satellite cells. | Pengkang Song Xiaoyou Chen Jiamin Zhao Qiang Li Xinrui Li Yu Wang Bo Wang Junxing Zhao | 2023 | Animal Nutrition2023,,3: | 0
Mediating role of resilience in the relationship between COVID-19 related stigma and mental health among COVID-19 survivors:a cross-sectional study显示文摘Background The global coronavirus disease 2019(COVID-19)has caused many negative effects on physical and mental health of patients who have survived COVID-19.Apart from some long-lasting physical sequelae, those COVID-19 survivors are also suffering stigma and discrimination at different levels around the world. The current study aims to assess the role resilience played in stigma and mental disorders among COVID-19 survivors. Methods The cross-sectional study was carried out among former COVID-19 patients in Jianghan District(Wuhan,China)from June 10 to July 25,2021.The demographic questions, the Impact of Events Scale-Revised,the Generalized Anxiety Disorder Questionnaire, the Patient Health Questionnaire, the Resilience Style Questionnaire and the Short Version of COVID-19 Stigma Scale of 12 items were used to collect relevant information of the participants.Descriptive analyses,Pearson correlation analysis and Structural Equation Modeling were used to make data description and analysis. Results A total of 1541 out of 1601 COVID-19 survivors(887 females and 654 males)were included in the analysis.Perceived stigma of those COVID-19 survivors correlates significantly with anxiety(r=0.335,P<0.001),depression(r=0.325,P<0.001)and post-traumatic stress disorder(PTSD)(r=0.384,P<0.001).It has a direct effect on COVID-19 survivors’anxiety(β=0.326,P<0.001),depression(β=0.314,P<0.001),PTSD(β=0.385,P<0.001)and their resilience(β=−0.114,P<0.01).Resilience partially mediated the association between perceived stigma and anxiety(β=0.020,P<0.01),depression(β=0.020,P<0.01),and PTSD(β=0.014,P<0.01)among COVID-19 survivors. Conclusion Stigma has a significant negative impact on mental health, while resilience plays a mediator role in the relationship between stigma and mental health among COVID-19 survivors.Based on our study,we suggested that when designing psychological interventions for COVID-19 survivors, consideration should be taken into account to reduce stigma and improve resilience. | Weijun Xiao Xiaoyang Liu Hao Wang Yiman Huang Zhenwei Dai Mingyu Si Jiaqi Fu Xu Chen Mengmeng Jia Zhiwei Leng Dan Cui Winnie W.S.Mak Liming Dong Xiaoyou Su | 2023 | Infectious Diseases of Poverty2023,12,2: | 0
Retinal damage after exposure to white light emitting diode lights at different intensities in Sprague-Dawley rats显示文摘Background:The usage of the light emitting diode(LED)has been increasingly applied in the illumination setting and electronic equipment. However, the effect of LED lights on the retina remains unclear. In this study,we observed and analyzed the impact of white LED lights at different intensities on the function and morphology of rat retinas.Methods:Thirty-six Sprague-Dawley rats weighing 150-180 g were randomly divided into six groups(n=6 in each group)including a normal control(NC)group,4 white LED groups at different light intensities(4,000,6,000,7,000,and 10,000 lux),and an ultraviolet B(UVB)lighting group(302 nm,1,000μw/cm2).After 24 hours of continuous illumination,full-field flash electroretinogram(FERG)and pathological examination were performed in each group. Results:As revealed by FERG,the impairment of retinal function gradually worsened with the increase of LED light intensity. In contrast, the UVB group had the most severe retinal function impairment.Particularly, the functional damage of rod cells and inner nuclear layer cells was the main FERG finding in each group. In the NC group, the retina had typical morphologies featured by well-defined structures, clearly visible border between the inner and outer segments, and neatly arranged inner and outer nuclear layer cells. After 24 hours of illumination, the inner and outer parts of the retina in the 4,000 lux group were still neatly arranged, along with a clear border;however, the inner and outer nuclear layers were randomly arranged, and some irregular nuclei and cells were lost. The damage of the internal and external retinal segments and the internal and external nuclear layers became more evident in the 6,000 lux group,7,000 lux group, and 10,000 lux group. The UVB group had a more obviously disordered arrangement of inner and outer nuclear layers and loss of cells. Conclusions:Continuous exposure to white LED light can cause structural and functional damage to rat retinas, and such damage is related to the intensity of illumination. Therefore, the risk of retinal damage should be considered during LED illumination, and proper LED illumination intensity may help to maintain eye health. | Guangsen Wu Xiaoyou Huang He Meng Le Yang Shengjian Lin Yang Gao Yujun Li Yandong Wang | 2019 | Annals of Eye Science2019,,1: | 0
