| 3 | CPT1C-mediated fatty acid oxidation facilitates colorectal cancer cell proliferation and metastasis显示文摘Fatty acid oxidation(FAO)has been proven to be an accomplice in tumor progression.Carnitine palmitoyltransferase 1C(CPT1C),a rate-limiting enzyme in FAO,mainly functions to catalyze fatty acid carnitinylation and guarantee subsequent entry into the mitochondria for FAO in colorectal cancer(CRC).Gene expression data and clinical information extracted from The Cancer Genome Atlas(TCGA)database show significantly higher expression of CPT1C in patients with metastatic CRC(P=0.005).Moreover,overexpression of CPT1C is correlated with worse relapse-free survival in CRC(HR 2.1,P=0.0006),while no statistical significance is indicated for CPT1A and CPT1B.Further experiments demonstrate that downregulation of CPT1C expression leads to a decrease in the FAO rate,suppression of cell proliferation,cell cycle arrest and repression of cell migration in CRC,whereas opposite results are obtained when CPT1C is overexpressed.Furthermore,an FAO inhibitor almost completely reverses the enhanced cell proliferation and migration induced by CPT1C overexpression.In addition,analysis of TCGA data illustrates a positive association between CPT1C expression and HIF1αlevel,suggesting that CPT1C is a transcriptional target of HIF1α.In conclusion,CPT1C overexpression indicates poor relapse-free survival of patients with CRC,and CPT1C is transcriptionally activated by HIF1α,thereby promoting the proliferation and migration of CRC cells. | Jing Li Wanwei Zheng Jie Wu Jun Zhang Bin Lv Wenshuai Li Jie Liu Xin Zhang Tiansheng Huang Zhongguang Luo | 2023 | Acta Biochimica et Biophysica Sinica2023,55,8: | 1 |

| 6 | SIRT6 promotes autophagy through direct interaction with ULK1 and competitive binding to PUMA显示文摘Akt aberrant activation accelerated tumor development and metastasis,and our previous study identified silent information regulator 6(SIRT6)as a novel and critical tumor suppressor in colorectal cancer(CRC)downstream of Akt.1 However,the function of SIRT6 in cancer progression has not been fully elucidated.In the present study,we found that except for inducing apoptosis,SIRT6 could serve as a crucial regulator to initiate autophagy by directly interacting with and activating ULK1.More importantly,we also reported autophagy regulation in a SIRT6-dependent indirect manner,namely,SIRT6 competitively bound to PUMA,which led to the release of ULK1.Of note,autophagy inhibition enhanced the pro-apoptotic effect of Midostaurin both in vitro and in vivo.Summarily,SIRT6 induced apoptosis accompanied by protective autophagy,and SIRT6 promoted autophagy by directly interacting with ULK1 and competitive binding to PUMA.This new insight identified the dual function of SIRT6 on apoptosis and autophagy,which has attractive clinical applications in cancer therapy. | Zhongguang Wu Lanhui Hong Guang Luo Siyao Lu Yanqiu Li Jiangang Wang Yingjie Zhang Lingling Zhang | 2023 | Genes & Diseases2023,10,5: | 0 |
| 7 | Forecasting the development of the COVID-19 epidemic by nowcasting: when did things start to get better?显示文摘Background:Now the coronavirus disease 2019(COVID-19)epidemic becomes a global phenomenon and its development concerns billions of peoples1 lives.The development of the COVID-19 epidemic in China could be used as a reference for the other countries,control strategy.Methods:We used a classical susceptible-infected-recovered(SIR)model to forecast the development of the COVID-19 epidemic in China by nowcasring.The linear regression analyses were employed to predict the COVID-19 epidemic’s inflexion point.Finally,we used a susceptible-exposed-infected-recovered(SEIR)model to simulate the development of the COVID-19 epidemic in China throughout 2020.Results:Our nowcasts show that the COVID-19 transmission rate started to slow down on January 30.The linear regression analyses further show that the inflexion point of this epidemic would arrive between February 17 and 18.The final SEIR model simulation forecasted that the COVID-19 epidemic would probably infect about 82,000 people and last throughout 2020 in China.We also applied our method to USA's and global COVID-19 data and the nowcasts show that the development of COVID-19 pandemic is not optimistic in the rest of 2020.Conclusion:The COVID-19 epidemic's scale in China is much smaller than the previous estimations.After implemented strict control and prevention measures,such as city lockdown,it took a week to slow down the COVID-19 transmission and about four weeks to really mitigate the COVID-19 prevalence in China. | Yuehui Zhang Lin Chen Qili Shi Zhongguang Luo Libing Shen | 2021 | Quantitative Biology2021,9,1: | 0 |
