2Efficacy and safety of low-dose interleukin-2 in combination with methotrexate in patients with active rheumatoid arthritis:a randomized,double-blind,placebo-controlled phase 2 trial显示文摘Rheumatoid arthritis(RA)is an aggressive autoimmune arthritis,and current therapies remain unsatisfactory due to low remission rate and substantially adverse effects.Low-dose interleukin-2(Ld-IL2)is potentially a therapeutic approach to further improve the disease.This randomized,double-blind,placebo-controlled trial was undertaken to evaluate the efficacy and safety of Ld-IL2 in patients with active RA.Patients were randomly assigned(1:1)to receive Ld-IL2,defined as a dose of 1 million IU,or placebo in a 12-week trial with a 12-week follow-up.Three cycles of Ld-IL2 or placebo were administered subcutaneously every other day for 2 weeks(a total of 7 doses),followed by a 2-week break.All patients received a stable dose of methotrexate(MTX).The primary outcomes were the proportion of patients achieving the ACR20,DAS28-ESR<2.6,and the change from baseline in CDAI or SDAI at week 24.Secondary endpoints included other clinical responses and safety.The primary outcomes were achieved in the perprotocol population.The improvements from baseline in CDAI and SDAI were significantly greater across time points for the LdIL2+MTX group(n=17)than for the placebo+MTX group(n=23)(P=0.018 and P=0.015,respectively).More patients achieved ACR20 response in the Ld-IL2+MTX group than those in the placebo+MTX group at week 12(70.6%vs 43.5%)and at week 24(76.5%vs 56.5%)(P=0.014).In addition,low Treg and high IL-21 were associated with good responses to Ld-IL2.Ld-IL-2 treatment was well-tolerated in this study.These results suggested that Ld-IL2 was effective and safe in RA.ClinicalTrials.gov number:NCT 02467504.Xiaoying Zhang Miao Miao Ruijun Zhang Xu Liu Xiaozhen Zhao Miao Shao Tian Liu Yuebo Jin Jiali Chen Huixin Liu Xia Zhang Yun Li Yunshan Zhou Yue Yang Ru Li Haihong Yao Yanying Liu Chun Li Yuhui Li Limin Ren Yin Su Xiaolin Sun Jing He Zhanguo Li 2022Signal Transduction and Targeted Therapy2022,7,4:7
3Combination of ablative fractional carbon dioxide laser and platelet-rich plasma treatment to improve hypertrophic scars:a retrospective clinical observational study显示文摘Background:Hypertrophic scars are one of the main complications that affect the quality of life of patients after burns.Many methods have been shown to be effective in the treatment of hypertrophic scars,such as ablative fractional CO_(2) laser(AFCL)and platelet-rich plasma(PRP).However,there are few studies on the effect of the combined application of these measures.The purpose of this study was to explore the therapeutic effect of AFCL combined with PRP on hypertrophic burn scars.Methods:A retrospective clinical observation study was conducted on 50 patients with hypertrophic burn scars.The AFCL+PRP group included 31 patients who received AFCL combined with PRP treatment;the AFCL group included 19 patients who received AFCL treatment only.The University of North Carolina 4P Scar Scale(UNC4P)and the Vancouver Scar Scale(VSS)scores that were collected before each treatment were used as indicators of the effectiveness of the previous treatment.The scores recorded at the second,fourth and seventh months were analysed.Results:The demographic data of the 2 groups were not significantly different.Before treatment,therewas no difference in the UNC4P and VSS scores between the 2 groups.Therewas a significant decline in the UNC4P and VSS total scores over 6 months in both groups(p<0.05)and scores in the 2 groups were comparable after 3 and 6 months(p<0.05).UNC4P scores in the AFCL+PRP group decreased from a mean of 8.26 to 2.61(p<0.05)with a concomitant drop in VSS scores from a mean of 11.74 to 6.06(p<0.01).In the AFCL group UNC4P and VSS scores decreased from 7.68 to 4.63(p<0.05)and from 10.89 to 8.16(p<0.05),respectively.The sub-items of these 2 assessments were analysed and the results suggest that AFCL combined with PRP can comprehensively improve scarring.Conclusions:This study shows that PRP is an effective adjunct for AFCL in the treatment of hypertrophic burn scars and that the combination of PRP and AFCL proved to be more useful than AFCL alone.This combination may be a new and effective clinical practice for the treatment of scars.However,larger and higher-level clinical studies are still needed to determine its efficacy and possible mechanisms.Zhanzhan Dai Xiaozhen Lou Tuo Shen Yu Sun Yongqiang Xiao Xingfeng Zheng Xuexin Wang Yu Peng Yukun Guo Yibin Guo Jiannan Wen He Fang Bing Ma Zhaofan Xia 2021Burns & Trauma2021,9,1:4
4Enhanced low-temperature performance of slight Mn-substituted LiFePO_4/C cathode for lithium ion batteries显示文摘Low temperature performance of LiFePO4/C cathode was remarkably improved by slight Mn-substitution. Electrochemical measurements showed that about 95% of the discharge capacity of LiFe0.98Mn0.02PO4/C cathode at 20°C was obtained at 0°C, compared to 85% of that of LiFePO4/C cathode. The LiFe0.98Mn0.02PO4/C sample also presented enhanced rate performance at –20°C with the discharge capacities of 124.4 mA h/g (0.1C), 99.8 mA h/g (1C), 80.7mAh/g (2C) and 70 mA h/g (5C), respectively, while pristine LiFePO4/C only delivered capacities of 120.5 mA h/g (0.1C), 90.7 mA h/g (1C), 70.4 mA h/g (2C) and 52.2 mA h/g (5C). Cyclic voltammetry measurements demonstrated an obvious improvement of the lithium insertion-extraction process of the LiFePO4/C cathode by slight Mn-substitution. 5A novel Co(phen)_2/C catalyst for the oxygen electrode in rechargeable lithium air batteries显示文摘A novel Co(phen)2/C catalyst was prepared by coating cobalt(II) phenanthroline (phen) chelate on BP2000 carbon black and then heat treating in an inert atmosphere. The obtained Co(phen)2/C product with 1.0 wt% cobalt loading exhibits similar morphology and porosity characteristics to those of the bare BP2000. X-ray diffraction measurements demonstrate a face-centered cubic (fcc) α-Co phase embedded in the carbon support after pyrolysis. Charge/discharge tests of the lithium-oxygen cells using the prepared Co(phen)2/C catalyst show high discharge capacities of 4870 mAh g-1 (0.05 mA cm-2 ), 3353 mAh g-1 (0.1 mA cm-2 ) and 3220 mAh g 1 (0.15 mA cm-2 ), respectively. The Co(phen) 2 /C cathode exhibits reasonable reversibility with capacity retention of 1401 mAh g-1 ( 0.1 mA cm-2 ) after 10 cycles. 6Optimizing food waste hydrothermal parameters to reduce Maillard reaction and increase volatile fatty acid production显示文摘The occurrence of the Maillard reaction and melanoidins formation during the hydrothermal treatment of food waste can reduce the yield of volatile fatty acids(VFA);however, few studies have investigated the adverse effects of the Maillard reaction. This study identified the impact of hydrothermal treatment parameters on hydrolysis and melanoidins formation and optimized the hydrothermal treatment conditions to enhance VFA production by minimizing the impact of the Maillard reaction. A response surface methodology was employed to optimize the hydrothermal treatment parameters and VFA production was evaluated. Results showed that temperature, reaction time, and pH were significant interacting factors with respect to hydrolysis and melanoidins formation while the C/N ratio and moisture content of food waste had little impact. The optimal conditions for hydrothermal treatment(temperature of 132 °C, reaction time of 27 min, and a pH of 5.6) enhanced VFA production by 22.1%. 9Enhanced endosomal escape of dendrigraft poly-L-lysine polymers for the efficient gene therapy of breast cancer显示文摘Dendrimer,such as dendrigraft poly-L-lysine(DGL)polymers,with high surface charge density,well-defined structure,and narrow poly-dispersity is often employed as a gene vector,but its transfection efficiency is still partially inhibited due to poor endosomal escape ability.Herein,we used a surface modification strategy to enhance the endosomal escape ability of DGL polymers,and thus improved its gene transfection efficiency.A library of phenylboronic acid(PBA)modified DGL polymers(PBA-DGLs)was designed to screen efficient small interfering RNA(siRNA)vectors.The lead candidate screened from the library shown a capability of inducing nearly 90% gene silencing in MDA-MB-231 cells.The study of the transfection mechanism revealed that PBA modification not only improves siRNA cellular uptake,but,more importantly,endows DGL polymers the ability of endosomal escape.One of the top candidates from polyplexes was further shielded with hyaluronic acid to construct targeted nanoparticles,and the yielding nanoparticles significantly suppressed the tumor growth in a breast cancer model by effective siRNA delivery.This research provides a general and effective strategy to enhance the endosomal escape and transfection efficiency of dendrimer.Li Ye Hongmei Liu Xin Fei Ding Ma Xiaozhen He Qianyun Tang Xue Zhao Hanbing Zou Xiaojing Chen Xianming Kong Peifeng Liu 2022Nano Research2022,15,2:1

11Mild Cytokine Elevation, Moderate CD4^+ T Cell Response and Abundant Antibody Production in Children with COVID-19显示文摘Children with Coronavirus Disease 2019(COVID-19) were reported to show milder symptoms and better prognosis than their adult counterparts, but the difference of immune response against SARS-CoV-2 between children and adults hasn’t been reported. Therefore we initiated this study to figure out the features of immune response in children with COVID-19.Sera and whole blood cells from 19 children with COVID-19 during different phases after disease onset were collected.The cytokine concentrations, SARS-CoV-2 S-RBD or N-specific antibodies and T cell immune responses were detected respectively. In children with COVID-19, only 3 of 12 cytokines were increased in acute sera, including interferon(IFN)-cinduced protein 10(IP10), interleukin(IL)-10 and IL-16. We observed an increase in T helper(Th)-2 cells and a suppression in regulatory T cells(Treg) in patients during acute phase, but no significant response was found in the IFN-cproducing or tumor necrosis factor(TNF)-a-producing CD8?T cells in patients. S-RBD and N IgM showed an early induction, while S-RBD and N IgG were prominently induced later in convalescent phase. Potent S-RBD IgA response was observed but N IgA seemed to be inconspicuous. Children with COVID-19 displayed an immunophenotype that is less inflammatory than adults, including unremarkable cytokine elevation, moderate CD4?T cell response and inactive CD8?T cell response, but their humoral immunity against SARS-CoV-2 were as strong as adults. 18Generation and Characterization of a Transgenic Zebrafish Expressing the Reverse Tetracycline Transactivator显示文摘Conditional expression of a target gene during zebrafish development is a powerful approach to elucidate gene functions.The tetracycline-controlled systems have been successfully used in the modulation of gene expression in mammalian cells,but few lines of zebrafish carrying these systems are currently available.In this study,we had generated a stable transgenic zebrafish line that ubiquitously expressed the second-generation of reverse Tet transactivator(rtTA-M2).Southern blotting analysis and high-throughput genome sequencing verified that a single copy of rtTA-M2 gene had stably integrated into the zebrafish genome.After induction with doxycycline(Dox),a strong green fluorescent protein(GFP)was seen in rtTA-transgenic eggs injected with pTREe EGFP plasmids.The fluorescent signal gradually decreased after the withdrawal of Dox and disappeared.However,leaky expression of GFP was undetectable before Doxinduction.Additionally,transgenic embryos expressing rtTA-M2 exhibited no obvious defects in morphological phenotypes,hatching behavior and expression patterns of developmental marker genes,suggesting that rtTA-M2 had little effect on the development of transgenic zebrafish.Moreover,expressed Dickkopf-1(DKK1)in pTRE-DKK1-injected embryos led to alterations in the expression of marker genes associated with Wnt signaling.Thus,this rtTA-transgenic zebrafish can be utilized to dissect functions of genes in a temporal manner.Qilin Gu Xiaojie Yang Xiaozhen He Qing Li Zongbin Cui 2013Journal of Genetics and Genomics2013,40,10:0

20Preparation and performance of LiNi_(0.8)Co_(0.2)O_2 cathode material based on Co-substituted α–Ni(OH)_2 precursor显示文摘Co-substituted α-Ni(OH)2 was synthesized by a novel microwave homogeneous precipitation method in the presence of urea. LiNi0.8Co0.2O2 cathode material was synthesized by calcining Co-substituted α-Ni(OH)2 precursor and LiOH·H2O at 900℃ for 10 h in flowing oxygen. XRD, FTIR, FESEM and electro- chemical tests were used to study the physical and the electrochemical performances of the materials. The results show that the prepared LiNi0.8Co0.2O2 compound has a good layered hexagonal structure. Moreover, the LiNi0.8Co0.2O2 cathode material demonstrates stable cyclability with a high initial specific discharge capacity of 183.9 mAh/g.
