1HY5-HDA9 Module Transcriptionally Regulates Plant Autophagy in Response to Light-to-Dark Conversion and Nitrogen Starvation显示文摘Light is arguably one of the most important environmental factors that determines virtually all aspects of plant growth and development,but the molecular link between light signaling and the autophagy pathway has not been elucidated in plants.In this study,we demonstrate that autophagy is activated during light-to-dark conversion though transcriptional upregulation of autophagy-related genes(ATGs).We showed that depletion of the ELONGATED HYPOCOTYL 5(HY5),a key component of light signaling,leads to enhanced autophagy activity and resistance to extended darkness and nitrogen starvation treatments,contributing to higher expression oiATGs.HY5 interacts with and recruits HISTONE DEACETYLASE 9(HDA9)to ATG5 and ATG8e loci to repress their expression by deacetylation of the Lys9 and Lys27 of histone 3.Furthermore,we found that both darkness and nitrogen depletion induce the degradation of HY5 via 26S proteasome and the concomitant disassociation of HDA9 from ATG5 and ATG8e loci,leading to their depression and thereby activated autophagy.Genetic analysis further confirmed that HY5 and HDA9 act synergistically and function upstream of the autophagy pathway.Collectively,our study unveils a previously unknown transcriptional and epigenetic network that regulates autophagy in response to light-to-dark conversion and nitrogen starvation in plants.Chao Yang Wenjin Shen Lianming Yang Yun Sun Xibao Li Minyi Lai Juan Wei Chaojun Wang Yingchao Xu Faqiang Li Shan Liang Chengwei Yang Shangwei Zhong Ming Luo Caiji Gao 2020Molecular Plant2020,13,3:8
2Pathways related to PMA-differentiated THP1 human monocytic leukemia cells revealed by RNA-Seq显示文摘Previous analyses have reported that the human monocytic cell line THP1 can be differentiated into cells with macrophage-like characteristics by phorbol 12-myristate 13-acetate(PMA). However, little is known about the mechanism responsible for regulating this differentiation process. Here, we performed high-throughput RNA-Seq analysis to investigate the genes differently expressed in THP1 cells treated with and without PMA and examined those that may be responsible for the PMA-induced differentiation of monocytes into macrophages. We found 3,000 genes to be differentially expressed after PMA treatment. Gene ontology analysis revealed that genes related to cellular processes and regulation of biological processes were significantly enriched. KEGG analysis also demonstrated that the differentially expressed genes(DEGs) were significantly enriched in the PI3K/AKT signaling pathway and phagosome pathway. Importantly, we reveal an important role of the PI3K/AKT pathway in PMA-induced THP1 cell differentiation. The identified DEGs and pathways may facilitate further study of the detailed molecular mechanisms of THP1 differentiation. 3MEF2C promotes M1 macrophage polarization and Th1 responses显示文摘The polarization of macrophages to the M1 or M2 phenotype has a pivotal role in inflammation and host defense;however,the underlying molecular mechanism remains unclear.Here,we show that myocyte enhancer factor 2 C(MEF2C)is essential for regulating M1 macrophage polarization in response to infection and inflammation.Global gene expression analysis demonstrated that MEF2C deficiency in macrophages downregulated the expression of M1 phenotypic markers and upregulated the expression of M2 phenotypic markers.MEF2C significantly promoted the expression of interleukin-12 p35 subunit(Il12a)and interleukin-12 p40 subunit(Il12b).Myeloid-specific Mef2c-knockout mice showed reduced IL-12 production and impaired Th1 responses,which led to susceptibility to Listeria monocytogenes infection and protected against DSS-induced IBD in vivo.Mechanistically,we showed that MEF2C directly activated the transcription of Il12a and Il12b.These findings reveal a new function of MEF2C in macrophage polarization and Th1 responses and identify MEF2C as a potential target for therapeutic intervention in inflammatory and autoimmune diseases.Xibao Zhao Qianqian Di Han Liu Jiazheng Quan Jing Ling Zizhao Zhao Yue Xiao Han Wu Zherui Wu Wengang Song Huazhang An Weilin Chen 2022Cellular & Molecular Immunology2022,19,4:4
4Effect of deep cryogenic treatment on mechanical behavior of a Cu-Cr-Zr alloy for electrodes of spot welding显示文摘The effects of deep cryogenic treatment on mechanical behavior of a Cu-Cr-Zr alloy for electrodes of spot weld- ing were investigated employing Brinell-hardness testing unit, abrasion examination machine, electronic almighty testing machine and X-ray stress analyzer. Tensile fracture surfaces of the alloy were characterized by scanning electronic micro- scope (SEM) with energy dispersive X-ray spectroscopy (EDS). 5A Discrete Numerical Study of the Effect of the Thickness and the Porosity of the Sand Cushion on the Impact Response Due to the Rockfall显示文摘The prevention and the reduction of the rockfall are the common measures of the prevention and the reduction of disasters.When the rock-shed resists the impact of the rockfall,the force that acts on the structure consists of the cushion dead load and the impact-induced load,of which the dynamic process of the propagation of the impactinduced load is complex.Therefore,we conducted a numerical study to investigate the impact of the rockfall.Considering the highly discrete characteristic of the sand,we developed a numerical model on the basis of the discrete element method(DEM).The numerical model,which simulation results were validated by the results of real-scale experiments,was used to investigate the dynamic response of the impact force of the rockfall and the transmission of the impact force under the different magnitude of the falling height and the different thickness of the sand cushion.The results of our study indicated that the cushion thickness had little effect on the impact of the rockfall,and the dense sand cushion generated higher impact force than did the loose sand cushion.Although the high thickness enhanced the buffer performance of the sand cushion,the additional force induced by the dead load of sand cushion was significant.Therefore,to determine the appropriate thickness of the sand cushion,we suggested designers consider the buffer performance and the dead load of the sand cushion.The analysis presented in this paper provided a practical estimation of the impact-induced force of the thick sand cushion.Song Yuan Peng Zhao Liangpu Li Xibao Wang Jun Liu Bo Zhang 2022Computer Modeling in Engineering & Sciences2022,,3:2

16Enhanced photocatalytic degradation and H_(2) evolution performance of N-CDs/S-C_(3)N_(4)S-scheme heterojunction constructed byπ-πconjugate self-assembly显示文摘Constructing heterojunction between two semiconductors with matched energy band structure is an effective modification method to obtain excellent photocatalysts.The experimental scheme adopts a simple solvent method to self-assemble nitrogen doped carbon dots(N-CDs)on the surface of sulfur doped carbon nitride(S-C_(3)N_(4))semiconductor throughπ-πconjugate interaction.Based on this,a novel 0D/2D S-scheme heterojunction N-CDs/S-C_(3)N_(4)hybrid was successfully prepared.The degradation kinetic constants of N-CDs/S-C_(3)N_(4)for rhodamine B(RhB)and p-nitrophenol(PNP)reached 0.23522 and 0.01342 min^(−1),repectively,which were 2.72 and 2.65 times that of S-C_(3)N_(4).The highest photocatalytic hydrogen evolution rate was observed under the simulated sunlight irradiation,which was 2.30 times that of S-C_(3)N_(4).The improvement of photocatalytic performance was mainly based on the formation of the S-scheme heterojunction between S-C_(3)N_(4)and N-CDs.The effects of internal electric field,π-πconjugate interaction and band bending promoted the photogenerated h^(+)and e^(−) with low redox ability to recombine and retained the beneficial h+and e−with strong redox ability,which contributed to the production of more active species of h^(+) and•O_(2)−,therefore the photocatalytic degradation and hydrogen evolution performance were significantly enhanced.Xibao Li Qiuning Luo Lu Han Fang Deng Ya Yang Fan Dong 2022Journal of Materials Science & Technology2022,,19:1
