2The transcription factor OpWRKY2 positively regulates the biosynthesis of the anticancer drug camptothecin in Ophiorrhiza pumila显示文摘The limited bioavailability of plant-derived natural products with anticancer activity poses major challenges to the pharmaceutical industry.An example of this is camptothecin,a monoterpene indole alkaloid with potent anticancer activity that is extracted at very low concentrations from woody plants.Recently,camptothecin biosynthesis has been shown to become biotechnologically amenable in hairy-root systems of the natural producer Ophiorrhiza pumila.Here,time-course expression and metabolite analyses were performed to identify novel transcriptional regulators of camptothecin biosynthesis in O.pumila.It is shown here that camptothecin production increased over cultivation time and that the expression pattern of the WRKY transcription factor encoding gene OpWRKY2 is closely correlated with camptothecin accumulation.Overexpression of OpWRKY2 led to a more than three-fold increase in camptothecin levels.Accordingly,silencing of OpWRKY2 correlated with decreased camptothecin levels in the plant.Further detailed molecular characterization by electrophoretic mobility shift,yeast one-hybrid and dual-luciferase assays showed that OpWRKY2 directly binds and activates the central camptothecin pathway gene OpTDC.Taken together,the results of this study demonstrate that OpWRKY2 acts as a direct positive regulator of camptothecin biosynthesis.As such,a feasible strategy for the over-accumulation of camptothecin in a biotechnologically amenable system is presented.Xiaolong Hao Chenhong Xie Qingyan Ruan Xichen Zhang Chao Wu Bing Han Jun Qian Wei Zhou Hans-Wilhelm Nutzmann Guoyin Kai 2021Horticulture Research2021,8,1:7
3Recent advances in wireless epicortical and intracortical neuronal recording systems显示文摘An implantable brain-computer interface(BCI)has proven to be effective in the field of sensory and motor function restoration and in the treatment of neurological disorders.Using a BCI recording system,we can transform current methods of human interaction with machines and the environment,especially to help those with cognitive and mobility disabilities regain mobility and reintegrate into society.However,most reported work has focused on a simple aspect of the whole system,such as electrodes,circuits,or data transmission,and only a very small percentage of systems are wireless.A miniature,lightweight,wireless,implantable microsystem is key to realizing long-term,real-time,and stable monitoring on freely moving animals or humans in their natural conditions.Here,we summarize typical wireless recording systems,from recording electrodes,processing chips and controllers,wireless data transmission,and power supply to the system-level package for either epicortical electrocorticogram(ECoG)or intracortical local field potentials(LFPs)/spike acquisition,developed in recent years.Finally,we conclude with our vision of challenges in next-generation wireless neuronal recording systems for chronic and safe applications.Bowen JI Zekai LIANG Xichen YUAN Honglai XU Minghao WANG Erwei YIN Zhejun GUO Longchun WANG Yuhao ZHOU Huicheng FENG Honglong CHANG Jingquan LIU 2022Science China(Information Sciences)2022,65,4:2

5Nonlinear fusion filters based on prediction and smoothing显示文摘To attenuate white noise, nonstationary noise and impulse noise are important for signal processing. In this letter, we present nonlinear fusion filters (NFF) based on prediction and smoothing. By means of least square fitting of a polynomial, we define and give the operators of left prediction and right prediction, left smoothing and right smoothing, central smoothing and cross-validation smoothing. 7Uncertainties of ENSO-related Regional Hadley Circulation Anomalies within Eight Reanalysis Datasets显示文摘El Nino-Southern Oscillation(ENSO),the leading mode of global interannual variability,usually intensifies the Hadley Circulation(HC),and meanwhile constrains its meridional extension,leading to an equatorward movement of the jet system.Previous studies have investigated the response of HC to ENSO events using different reanalysis datasets and evaluated their capability in capturing the main features of ENSO-associated HC anomalies.However,these studies mainly focused on the global HC,represented by a zonal-mean mass stream function(MSF).Comparatively fewer studies have evaluated HC responses from a regional perspective,partly due to the prerequisite of the Stokes MSF,which prevents us from integrating a regional HC.In this study,we adopt a recently developed technique to construct the three-dimensional structure of HC and evaluate the capability of eight state-of-the-art reanalyses in reproducing the regional HC response to ENSO events.Results show that all eight reanalyses reproduce the spatial structure of HC responses well,with an intensified HC around the central-eastern Pacific but weakened circulations around the Indo-Pacific warm pool and tropical Atlantic.The spatial correlation coefficient of the three-dimensional HC anomalies among the different datasets is always larger than 0.93.However,these datasets may not capture the amplitudes of the HC responses well.This uncertainty is especially large for ENSO-associated equatorially asymmetric HC anomalies,with the maximum amplitude in Climate Forecast System Reanalysis(CFSR)being about 2.7 times the minimum value in the Twentieth Century Reanalysis(20CR).One should be careful when using reanalysis data to evaluate the intensity of ENSO-associated HC anomalies.Yadi LI Xichen LI Juan FENG Yi ZHOU Wenzhu WANG Yurong HOU 2024Advances in Atmospheric Sciences2024,41,1:0
8Rationally designed/constructed MnO_x/WO_3 anode for photoelectrochemical water oxidation显示文摘Photoelectrocatalytic water splitting is an effective way to utilize the solar energy to solve the energy shortage. The valence band edge of WO_3 located at ~3V vs. normal hydrogen electrode(NHE), which can offer enough potential to kinetically oxidize water for oxygen evolution reaction. However, water oxidation reaction kinetics is sluggish when only WO_3 is used as the photoanode. It is highly desirable to use cocatalyst to promote the kinetics. Mn Oxloaded on the WO_3 photoanode through photodeposition methods improves the photoelectrochemical water oxidation performance. A maximum photocurrent density of composite photoanode is achieved with a deposition time of 3 min, which is higher than that of pristine WO_3 photoanode around 40%. Mn O2 is not only a cocatalyst for water splitting but also for improving oxidation selectivity. We tried to use two means to load Mn Oxon WO_3 photoanode material.
