| 5 | The E3 ubiquitin ligase NEDD4-1 protects against acetaminophen-induced liver injury by targeting VDAC1 for degradation显示文摘Acetaminophen(APAP)overdose is a major cause of liver injury.Neural precursor cell expressed developmentally downregulated 4—1(NEDD4-1)is an E3 ubiquitin ligase that has been implicated in the pathogenesis of numerous liver diseases;however,its role in APAP-induced liver injury(AILI)is unclear.Thus,this study aimed to investigate the role of NEDD4-1 in the pathogenesis of AILI.We found that NEDD4-1 was dramatically downregulated in response to APAP treatment in mouse livers and isolated mouse hepatocytes.Hepatocyte-specific NEDD4-1 knockout exacerbated APAP-induced mitochondrial damage and the resultant hepatocyte necrosis and liver injury,while hepatocyte-specific NEDD4-1 overexpression mitigated these pathological events both in vivo and in vitro.Additionally,hepatocyte NEDD4-1 deficiency led to marked accumulation of voltage-dependent anion channel 1(VDAC1)and increased VDAC1 oligomerization.Furthermore,VDAC1 knockdown alleviated AILI and weakened the exacerbation of AILI caused by hepatocyte NEDD4-1 deficiency.Mechanistically,NEDD4-1 was found to interact with the PPTY motif of VDAC1 through its WW domain and regulate K48-linked ubiquitination and degradation of VDAC1.Our present study indicates that NEDD4-1 is a suppressor of AILI and functions by regulating the degradation of VDAC1. | Yiwei Zhu Lin Lei Xinghui Wang Linfang Chen Wei Li Jinxia Li Chenchen Zhao Xiliang Du Yuxiang Song Wenwen Gao Guowen Liu Xinwei Li | 2023 | Acta Pharmaceutica Sinica B2023,13,4: | 1 |

| 7 | Application of sentinel lymph node (SLN) biopsy in breast cancer patients显示文摘Objective: To discuss if the sentinel lymph node (SLN) biopsy is able to reflect the status of the axillary lymph node and the application of this technic in clinic. Methods: Using 99mTc-signed dextran, SLN-biopsy (SLNB) was carried out in 182 cases with breast cancer during May 1999 to September 2006. During the operation, γ-detector was used for orientation. After the SLNB, a modified radical mastectomy or breast conserving surgery were carried out to the patients, then a particular separate pathological examination of the SLN was made. Results: 178 cases of SLNB were carried out successfully, and the success rate was 97.8%, the out-checked SLN of each case ranged from 1 to 4, with an average of 2.5. All SLN was located at the first level of axilla, sensitivity of the SLNB was 93.4%, specificity was 100%, false negative rate was 6.6%, false positive rate was 0, accuracy was 97.8%, positive predictive value was 100.0%, negative predictive value was 96.7%, and Youden’s index was 0.934. Immunohistochemical examination was carried out in 59 cases of SLN, and 14 cases showed the existences of micro-metastasis, however, metastasis had not been found in non-SLN of these cases. Conclusion: SLN is able to reflect the metastasis of the axillary lymph node, and this can suggest the necessity of the axillary dissection in clinic. | 9 | 乳腺癌前哨淋巴结微小转移的检测(英文)显示文摘Objective: To study the best examination method of micrometastases in sentinel lymph node (SLN) of breast cancer and study the related factors with micrometastases. Methods: By step serial sectioning technique and immunohisto- chemistry, 121 SLNs and 44 tumors of 59 cases were examined. Results: Micrometastases was found in 17 SLNs (14%) of 14 (24%) cases. The more sections or examination methods, the more micrometastases were found. Micrometastases was related with the tumor size and the expression of c-erbB2, MMP-2 and VEGF. Conclusion: The best examination method is making sections at two levels with 100 μm interval and combination of HE staining and muc1 immunohistochemistry. We can get the excellent detection/cost by this method. | 11 | Aqueous synthesis of bright near-infrared-emitting Zn-Cu-In-Se quantum dots for multiplexed detection of tumor markers显示文摘Aqueous synthesis is a green but challenging pathway to produce cadmium-free,bright,and near-infrared(NIR)emitting quantum dots(QDs).Herein,we develop a straightforward aqueous phase approach to prepare Zn-Cu-In-Se QDs with tunable emission from 630 to 800 nm through varying the molar ratio of Zn and Cu.The ultrasmall nanoparticle size remains nearly fixed(~2.6 nm)when the QD composition is varied,allowing for the effects of composition on the optical properties of QDs to be decoupled from the impact of particle size.By leveraging the off-stoichiometric effect on photoluminescence(PL),Zn-Cu-In-Se QDs with a Zn:Cu molar ratio of 1.0 show a long PL decay lifetime of 211 ns and an absolute photoluminescence quantum yield(PLQY)of 14.2%.The PLQY is further enhanced by overcoating a ZnS shell,reaching up to 25.8%.Based on the bright,biocompatible,and emission-tunable QDs,nanoprobes with targeting capability for multiple tumor markers are further constructed and employed to simultaneously ascertain targets in the cytoplasm and cell membrane.This study provides a green and effective strategy for achieving bright and biocompatible NIR quantum dots for multiplexed biodetection applications. | Hui Liu Peng Cai Kevin J.McHugh Collin F.Perkinson Laishuo Li Shuna Wang Wei Wang Mingxia Jiao Xiliang Luo Lihong Jing | 2022 | Nano Research2022,15,9: | 0 |
