1Nitrogen deposition alters soil chemical properties and bacterial communities in the Inner Mongolia grassland显示文摘Nitrogen deposition has dramatically altered biodiversity and ecosystem functioning on the earth;however,its effects on soil bacterial community and the underlying mechanisms of these effects have not been thoroughly examined.Changes in ecosystems caused by nitrogen deposition have traditionally been attributed to increased nitrogen content.In fact,nitrogen deposition not only leads to increased soil total N content,but also changes in the NH 4 +-N content,NO 3-N content and pH,as well as changes in the heterogeneity of the four indexes.The soil indexes for these four factors,their heterogeneity and even the plant community might be routes through which nitrogen deposition alters the bacterial community.Here,we describe a 6-year nitrogen addition experiment conducted in a typical steppe ecosystem to investigate the ecological mechanism by which nitrogen deposition alters bacterial abundance,diversity and composition.We found that various characteristics of the bacterial community were explained by different environmental factors.Nitrogen deposition decreased bacterial abundance that is positively related to soil pH value.In addition,nitrogen addition decreased bacterial diversity,which is negatively related to soil total N content and positively related to soil NO 3-N heterogeneity.Finally,nitrogen addition altered bacterial composition that is significantly related to soil NH 4 +-N content.Although nitrogen deposition significantly altered plant biomass,diversity and composition,these characteristics of plant community did not have a significant impact on processes of nitrogen deposition that led to alterations in bacterial abundance,diversity and composition.Therefore,more sensitive molecular technologies should be adopted to detect the subtle shifts of microbial community structure induced by the changes of plant community upon nitrogen deposition.Ximei Zhang Xingguo Han 2012Journal of Environmental Sciences2012,24,8:18
2Relationship between nutrients and plankton biomass in the turbidity maximum zone of the Pearl River Estuary显示文摘Nutrients,dissolved and particulate organic carbon and plankton(bacterio-,phyto-and zoo-)were compared in the turbidity maximum zone(TMZ) and adjacent areas(non-TMZ) in the Pearl River estuary.Our results showed that high levels of suspended substances had marked effect on dynamics of nutrients and plankton in the TMZ.Based on the cluster analysis of total suspended solids(TSS) concentrations,all stations were divided into two groups,TMZ with average TSS of 171 mg/L and non-TMZ of 45 mg/L.Suspended substances adsorbed PO^(3-)_4 and dissolved organic carbon,resulting in higher particulate phosphorus and organic carbon(POC)and lower PO^(3-)_4 and DOC in the TMZ,compared to the non-TMZ.However,suspended substances had limited effect on nitrogenous nutrients.Phytoplankton growth was light-limited due to high concentrations of suspended substances in the TMZ and a peak of phytoplankton abundance appeared in the non-TMZ.In contrast,the highest bacterial abundance occurred in the TMZ,which was likely partly responsible for low DOC levels.Two peaks of zooplankton abundance observed in the TMZ and non-TMZ in the Pearl River estuary were primarily supported by bacteria and phytoplankton,respectively.Our finding implied that high levels of suspended solids in the TMZ affect the trophic balance.Zhen Shi Jie Xu Xiaoping Huang Xia Zhang Zhijian Jiang Feng Ye Ximei Liang 2017Journal of Environmental Sciences2017,29,7:12
3Biological coupling anti-wear properties of three typical molluscan shells—Scapharca subcrenata, Rapana venosa and Acanthochiton rubrolineatus显示文摘Molluscan shells are fascinating examples of highly ordered hierarchical structure and complex organic-inorganic biocomposite material. However, their anti-wear properties were rarely studied especially in the perspective of biological coupling. So in the current study three typical shells, Scapharca subcrenata, Rapana venosa and Acanthochiton rubrolineatus, were selected as coupling models to further study their anti-wear properties. Stereomicroscope and scanning electron microscopic observations showed that all these three shells had specific surface morphologies and complicated section microstructures. Importantly, a special structure, pore canal tubules, was discovered in the shells of Scapharca subcrenata and Acanthochiton rubrolineatus, which probably contributed most to their anti-wear properties. X-ray diffraction and micro-Vikers hardness tester were further adopted to analyze the phase compositions and micro-hardness of the shells. The measured results demonstrated that aragonite was the most extensive phase present in the shell, and possesed a relatively high micro-hardness. In this paper, the shells were described in details in morphology, structure and material with emphasis on the relationship with anti-wear property. 5Targeting BMI-1-mediated epitheliale-mesenchymal transition to inhibit colorectal cancer liver metastasis显示文摘Liver is the most common metastatic site for colorectal cancer(CRC),there is no satisfied approach to treat CRC liver metastasis(CRCLM).Here,we investigated the role of a polycomb protein BMI-1 in CRCLM.Immunohistochemical analysis showed that BMI-1 expression in liver metastases was upregulated and associated with T4 stage,invasion depth and right-sided primary tumor.Knockdown BMI-1 in high metastatic HCT116 and LOVO cells repressed the migratory/invasive phenotype and reversed epithelialemesenchymal transition(EMT),while BMI-1 overexpression in low metastatic Ls174 T and DLD1 cells enhanced invasiveness and EMT.The effects of BMI-1 in CRC cells were related to upregulating snail via AKT/GSK-3βpathway.Furthermore,knockdown BMI-1 in HCT116 and LOVO cells reduced CRCLM using experimental liver metastasis mice model.Meanwhile,BMI-1 overexpression in Ls174 T and DLD1 significantly increased CRCLM.Moreover,sodium butyrate,a histone deacetylase and BMI-1 inhibitor,reduced HCT116 and LOVO liver metastasis in immunodeficient mice.Our results suggest that BMI-1 is a major regulator of CRCLM and provide a potent molecular target for CRCLM treatment.Zhiyao Xu Zhuha Zhou Jing Zhang Feichao Xuan Mengjing Fan Difan Zhou Zhenyu Liuyang Ximei Ma Yiyang Hong Yihong Wang Sherven Sharma Qinghua Dong Guanyu Wang 2021Acta Pharmaceutica Sinica B2021,11,5:8
6High salt diet accelerates the progression of murine lupus through dendritic cells via the p38 MAPK and STAT1 signaling pathways显示文摘The increased incidence of systemic lupus erythematosus(SLE)in recent decades might be related to changes in modern dietary habits.Since sodium chloride(NaCl)promotes pathogenic T cell responses,we hypothesize that excessive salt intake contributes to the increased incidence of autoimmune diseases,including SLE.Given the importance of dendritic cells(DCs)in the pathogenesis of SLE,we explored the influence of an excessive sodium chloride diet on DCs in a murine SLE model.We used an induced lupus model in which bone marrow-derived dendritic cells(BMDCs)were incubated with activated lymphocyte-derived DNA(ALD-DNA)and transferred into C57BL/6 recipient mice.We observed that a high-salt diet(HSD)markedly exacerbated lupus progression,which was accompanied by increased DC activation.NaCl treatment also stimulated the maturation,activation and antigenpresenting ability of DCs in vitro.Pretreatment of BMDCs with NaCl also exacerbated BMDC-ALD-DNA-induced lupus.These mice had increased production of autoantibodies and proinflammatory cytokines,more pronounced splenomegaly and lymphadenopathy,and enhanced pathological renal lesions.The p38 MAPK–STAT1 pathway played an important role in NaClinduced DC immune activities.Taken together,our results demonstrate that HSD intake promotes immune activation of DCs through the p38 MAPK–STAT1 signaling pathway and exacerbates the features of SLE.Thus,changes in diet may provide a novel strategy for the prevention or amelioration of lupus or other autoimmune diseases.Ze Xiu Xiao Xiaojiang Hu Ximei Zhang Zhigang Chen Julie Wang Ke Jin Feng Lin Cao Baoqing Sun Joseph A.Bellanti Nancy Olsen Song Guo Zheng 2020Signal Transduction and Targeted Therapy2020,5,1:6
7Arsenic methylation by an arsenite S-adenosylmethionine methyltransferase from Spirulina platensis显示文摘Arsenic-contaminated water is a serious hazard for human health. Plankton plays a critical role in the fate and toxicity of arsenic in water by accumulation and biotransformation.Spirulina platensis(S. platensis), a typical plankton, is often used as a supplement or feed for pharmacy and aquiculture, and may introduce arsenic into the food chain, resulting in a risk to human health. However, there are few studies about how S. platensis biotransforms arsenic. In this study, we investigated arsenic biotransformation by S. platensis. When exposed to arsenite(As(Ⅲ)), S. platensis accumulated arsenic up to 4.1 mg/kg dry weight.After exposure to As(Ⅲ), arsenate(As(Ⅴ)) was the predominant species making up 64% to86% of the total arsenic. Monomethylarsenate(MMA(Ⅴ)) and dimethylarsenate(DMA(Ⅴ))were also detected. An arsenite S-adenosylmethionine methyltransferase from S. platensis(Sp Ars M) was identified and characterized. Sp Ars M showed low identity with other reported Ars M enzymes. The Escherichia coli AW3110 bearing Spars M gene resulted in As(Ⅲ) methylation and conferring resistance to As(Ⅲ). The in vitro assay showed that Sp Ars M exhibited As(Ⅲ) methylation activity. DMA(Ⅴ) and a small amount of MMA(Ⅴ) were detected in the reaction system within 0.5 hr. A truncated Sp Ars M derivative lacking the last 34 residues still had the ability to methylate As(Ⅲ). The three single mutants of Sp Ars M(C59S, C186 S, and C238S) abolished the capability of As(Ⅲ) methylation, suggesting the three cysteine residues are involved in catalysis. 9Therapeutic effects and safety of olcegepant and telcagepant for migraine A meta-analysis显示文摘OBJECTIVE: To evaluate the therapeutic effects and adverse reactions of olcegepant and telcagepant for the treatment of migraine. DATA RETRIEVAL: We identified studies using Medline (1966-01/2012-06), PubMed (1966-01/2012-06), Scopus (1980-01/2012-06), Cochrane Central Register of Controlled Trials (1980-01/2012-06) and China National Knowledge Infrastructure (1980-01/2012-06). SELECTION CRITERIA: The included studies were double-blind, randomized and placebo-controlled trials of olcegepant or telcagepant for the treatment of single acute migraine in patients with or without aura. Adverse reaction data were also included. Two independent investigators performed quality evaluation and data extraction using Jadad scoring. Meta-analyses were undertaken using RevMan 5.0.25 software. MAIN OUTCOME MEASURES: Pain relief rate, pain-free rate, and incidence of adverse reactions were measured in patients 2 and 24 hours after injection of olcegepant and oral telcagepant. RESULTS: Six randomized, controlled trials were included. Meta-analysis demonstrated that compared with placebo, the pain relief rate (odds ratio, OR = 5.21, 95% confidence interval, CI: 1.91-14.2, P < 0.01) and pain-free rate (OR = 31.11, 95% CI: 3.80-254.98, P < 0.01) significantly increased 2 hours after 2.5 mg/d olcegepant treatment. Pain relief rate and pain-free rate 2 and 24 hours after treatment with telcagepant 150 mg/d and 300 mg/d were superior to placebo (P < 0.01). Moreover, the remission rate of unrelenting headache was higher after 24 hours of 300 mg/d telcagepant treatment compared with 150 mg/d (OR = 0.78, 95% CI: 0.62-0.97, P < 0.05). The incidence of adverse reactions with olcegepant was not significantly greater than placebo (P = 0.28), but within 48 hours of administration of telcagepant 300 mg/d, the incidence of adverse reactions was higher than placebo (OR = 1.21, 95% CI: 1.04-1.42, P < 0.01). Few studies have compared the therapeutic effects of olcegepant and telcagepant. CONCLUSION: The calcitonin-gene-related peptide receptor antagonists olcegepant and telcagepant have shown good therapeutic effects in the treatment of migraine. 10Profiling of circular RNA N^(6)-methyladenosine in moso bamboo(Phyllostachys edulis)using nanopore-based direct RNA sequencing显示文摘N6-methyladenosine(m^(6)A)is a prevalent modification in messenger RNAs and circular RNAs that play important roles in regulating various aspects of RNA metabolism.However,the occurrence of the m^(6)A modification in plant circular RNAs has not been reported.A widely used method to identify m^(6)A modifications relies on m^(6)A-specific antibodies followed by next-generation sequencing of precipitated RNAs(MeRIP-Seq).However,one limitation of MeRIP-Seq is that it does not provide the precise location of m^(6)A at single-nucleotide resolution.Although more recent sequencing techniques such as Nanopore-based direct RNA sequencing(DRS)can overcome such limitations,the technology does not allow sequencing of circular RNAs,as these molecules lack a poly(A)tail.Here,we developed a novel method to detect the precise location of m^(6)A modifications in circular RNAs using Nanopore DRS.We first enriched our samples for circular RNAs,which we then fragmented and sequenced on the Nanopore platform with a customized protocol.Using this method,we identified 470 unique circular RNAs from DRS reads based on the back-spliced junction region.Among exonic circular RNAs,about 10%contained m^(6)A sites,which mainly occurred around acceptor and donor splice sites.This study demonstrates the utility of our antibody-independent method in identifying total and methylated circular RNAs using Nanopore DRS.This method has the additional advantage of providing the exact location of m^(6)A sites at single-base resolution in circular RNAs or linear transcripts from non-coding RNA without poly(A)tails.Yongsheng Wang Huihui Wang Feihu Xi Huiyuan Wang Ximei Han Wentao Wei Hangxiao Zhang Qianyue Zhang Yushan Zheng Qiang Zhu Markus V.Kohnen Anireddy S.N.Reddy Lianfeng Gu 2020Journal of Integrative Plant Biology2020,62,12:3
11Parameter estimates of Heston stochastic volatility model with MLE and consistent EKF algorithm显示文摘Heston model is the most famous stochastic volatility model in finance. This paper considers the parameter estimation problem of Heston model with both known and unknown volatilities. First, parameters in equity process and volatility process of Heston model are estimated separately since there is no explicit solution for the likelihood function with all parameters. Second, the normal maximum likelihood estimation(NMLE) algorithm is proposed based on the It?o transformation of Heston model. The algorithm can reduce the estimate error compared with existing pseudo maximum likelihood estimation. Third, the NMLE algorithm and consistent extended Kalman filter(CEKF) algorithm are combined in the case of unknown volatilities. As an advantage, CEKF algorithm can apply an upper bound of the error covariance matrix to ensure the volatilities estimation errors to be well evaluated. Numerical simulations illustrate that the proposed NMLE algorithm works more efficiently than the existing pseudo MLE algorithm with known and unknown volatilities. Therefore, the upper bound of the error covariance is illustrated. 12Capture efficiency of coal/biomass co-combustion ash in an electrostatic field显示文摘Ash samples from corn stalk and coal co-fired at 0%,5%,10%,15%,20%,and 100%biomass were collected by a 4-stage wire-pipe type electrostatic precipitator (ESP),and the ratio of 14C/12C in ash samples from the 0%,20%,and 100% co-firing scenarios were measured by an accelerator mass spectrometer.The differential capture efficiency in electrostatic fields for coal ash,coal/biomass co-combustion ash (co- ash),and pure biomass ash was studied separately based on the ratio 14C/12C.Other factors that may influence capture efficiency were analyzed,including microscopic morphology,resistivity,dust density, and particle size distribution.The results indicate that co-ash may be efficiently captured by ESP,while pure biomass ash could not.Co-ash capture was mainly concentrated in the first two electrostatic fields in the lab-scale ESP,and the overall capture efficiency exceeded 90%.Biomass addition decreased the resistivity of co-ash,and enhanced its surface adsorption capacity to form agglomerates,facilitating the capture of co-ash.The capture efficiency of coal ash in co-ash was higher than that of pure coal ash in the first electrostatic field of the ESP.Co-firing biomass can aid the removal of both coal ash and biomass ash when using an ESP.Jianyi Lu Lili Fu Ximei Li Eric Eddings 2018Particuology2018,16,5:2
13Stochastic processes play more important roles in driving the dynamics of rarer species显示文摘Aims One major goal of modern community ecology is to understand how deterministic and stochastic processes combine to drive community assembly.However,little empirical knowledge is known about how their relative importance varies between common and rare species.Methods We exploited two 30-year data sets of plant communities in a temperate steppe using two different methods.One is a null model method,and the other is a recently developed direct-calculation method.Important Findings We found that stochastic processes tended to be more important in influencing rare than common species.This finding suggests that stochastic forces may play a more important role in structuring communities with more rare species,providing a possible solution to the debate on the varied importance of deterministic and stochastic processes among different communities.Ximei Zhang Zhichao Pu Yuanheng Li Xingguo Han 2016Journal of Plant Ecology2016,9,3:2
14Credit Scoring Based on the Set-Valued Identification Method显示文摘Credit scoring is one of the key problems in financial risk managements.This paper studies the credit scoring problem based on the set-valued identification method,which is used to explain the relation between the individual attribute vectors and classification for the credit worthy and credit worthless lenders.In particular,system parameters are estimated by the set-valued identification algorithm based on a given recognition criteria.In order to illustrate the efficiency of the proposed method,practical experiments are conducted for credit card applicants of Australia and credit card holders from Taiwan,respectively.The empirical results show that the set-valued model has a higher prediction accuracy on both small and large numbers of data set compared with logistic regression model.Furthermore,parameters estimated by the set-valued identification method are more stable,which provide a meaningful and logical explanation for extracting factors that influence the borrowers’credit scorings.WANG Ximei HU Min ZHAO Yanlong DJEHICHE Boualem 2020Journal of Systems Science & Complexity2020,33,5:2

16Transcriptome and QTL analyses reveal candidate genes for fiber quality in Upland cotton显示文摘With increasing demand for high-quality cotton,it is desirable to identify genes involved in fiber development for molecular improvement of cotton.In this study,780 differentially expressed genes(DEGs)were identified in developing fibers at 10 days post-anthesis(DPA)in Gossypium hirsutum acc.DH962 and G.hirsutum cv.Jimian 5 using RNA-seq.Of 15 stable QTL for fiber quality identified in the same two parents in previous studies,4,3,6,1,and 1 QTL were associated with fiber length(FL),fiber strength(FS),micronaire(MIC),fiber elongation(FE)and fiber length uniformity ratio(FU),respectively.Integration of DEGs and QTL allowed the identification of 31 genes in 9 QTL regions,of which 25 were highly expressed in fibers based on the transcriptome datasets and 9 were preferentially expressed in different stages of fiber development.Gh_A01G0453(GhDTX19),Gh_D07G1581 and Gh_D04G0942 were expressed specifically in 5 and 10 DPA fibers,with Gh_D04G0942 showing low expression in other tissues except pistil.Gh_D07G1799(GhGAUT9),Gh_D11G0326(GhVPS29),Gh_D11G0333(GhTCP14),and Gh_D11G0334(GhNRP2)were preferentially expressed in 5 or 10 DPA fibers;Gh_A01G0397(GhABCG10)and Gh_D07G1744 were expressed specifically in 20 and 25 DPA fibers.These results suggest candidate genes for molecular improvement of cotton fiber quality.Hantao Wang Ruiting Zhang Chao Shen Ximei Li De Zhu Zhongxu Lin 2020The Crop Journal2020,8,1:2
