1Near-infrared responsive 5-fluorouracil and indocyanine green loaded MPEG-PCL nanoparticle integrated with dissolvable microneedle for skin cancer therapy显示文摘The prevalence of skin cancer is rising along with the rapid population aging in recent years.Traditional therapies,such as surgical treatment,radiotherapy,chemotherapy,photodynamic therapy,and immunotherapy,may accompany serious side effects,limiting their clinical benefits.According to the biological characteristics of skin cancer,we have already established two kinds of synergetic systems of photothermal therapy(microneedle)and chemotherapy,containing gold nanorods(GNR).Although the microneedle system exhibited great potential for skin cancer treatment,the system could be still improved further.So,we designed a near-infrared lightresponsive 5-fluorouracil(5-Fu)and indocyanine green(ICG)loaded monomethoxy-poly(ethylene glycol)-polycaprolactone(MPEG-PCL)nanoparticle(5-Fu-ICG-MPEG-PCL),and then 5-Fu-ICG-MPEG-PCL was integrated with a hyaluronic acid dissolvable microneedle system(HA MN)to get 5-Fu-ICG-MPEG-PCL loaded HA MN for treating skin cancers,including human epidermoid cancer and melanoma.In this system,hyaluronic acid,the microneedle carrier,possesses good skin penetration ability and is approved by FDA as a pharmaceutical adjuvant;5-Fu is recommended by FDA for skin cancer treatment;ICG,a photothermal agent,possesses a strong photothermal ability and is approved by FDA for its use in the human body.We hypothesized that 5-Fu-ICG-MPEG-PCL could be delivered by the dissolvable microneedle through the skin,and the release behavior of the drug in the nanoparticle could be controlled by near-infrared light for achieving a single-dose cure of skin cancer,improving the cure rate of skin cancer and providing a new idea and possibility for the clinical treatment of skin cancer.Ying Hao YuWen Chen XinLong He Fan Yang RuXia Han ChengLi Yang Wei Li ZhiYong Qian 2020Bioactive Materials2020,5,3:17

4Third-order nonlinear optical properties of WTe2 films synthesized by pulsed laser deposition显示文摘The prominent third-order nonlinear optical properties of WTe2 films are studied through the Z-scan technique using a femtosecond pulsed laser at 1030 nm.Open-aperture(OA)and closed-aperture(CA)Z-scan measurements are performed at different intensities to investigate the nonlinear absorption and refraction properties of WTe2 films.OA Z-scan results show that WTe2 films always hold a saturable absorption characteristic without transition to reverse saturable absorption.Further,a large nonlinear absorption coefficientβis determined to be-3.37×10^3 cm∕GW by fitting the OA Z-scan curve at the peak intensity of 15.603 GW∕cm^2.In addition,through the slow saturation absorption model,the ground state absorption cross section,excited state absorption cross section,and absorber's density were found to be 1.4938×10^-16 cm^2,1.2536×10^-16 cm^2,and 6.2396×10^20 cm^-3,respectively.CA Z-scan results exhibit a classic peak–valley shape of the CA Z-scan signal,which reveals a self-defocusing optical effect of WTe2 films under the measured environment.Furthermore,a considerable nonlinear refractive index value n2 can be obtained at-1.629×10^-2 cm^2∕GW.Ultimately,the values of the real and imaginary parts of the third-order nonlinear susceptibility of WTe2 are roughly estimated to be-5.93×10^-9 and-1.01×10^-8 esu with the help of first-principles calculations.WTe2 possesses sizeable thirdorder nonlinear coefficients compared with other materials,which suggests that it has great potential to improve performance in nonlinear optical devices.MI HE YEQUAN CHEN LIPENG ZHU HUAN WANG XUEFENG WANG XINLONG XU ZHANYU REN 2019Photonics Research2019,7,12:1
5The construction of a lymphoma cell-based, DC-targeted vaccine, and its application in lymphoma prevention and cure显示文摘In recent years,Non-Hodgkin lymphoma(NHL)has been one of the most fast-growing malignant tumor diseases.NHL poses severe damages to physical health and a heavy burden to patients.Traditional therapies(chemotherapy or radiotherapy)bring some benefit to patients,but have severe adverse effects and do not prevent relapse.The relevance of emerging immunotherapy options(immune-checkpoint blockers or adoptive cellular methods)for NHL remains uncertain,and more intensive evaluations are needed.In this work,inspired by the idea of vaccination to promote an immune response to destroy tumors,we used a biomaterial-based strategy to improve a tumor cell-based vaccine and constructed a novel vaccine named Man-EG7/CH@CpG with antitumor properties.In this vaccine,natural tumor cells are used as a vector to load CpG-ODN,and following lethal irradiation,the formulations were decorated with mannose.The study of the characterization of the doubleimproved vaccine evidenced the enhanced ability of DCs targeting and improved immunocompetence,which displayed an antitumor function.In the lymphoma prevention model,the Man-EG7/CH@CpG vaccine restrained tumor formation with high efficiency.Furthermore,unlike the non-improved vaccine,the double-improved vaccine elicited an enhanced antitumor effect in the lymphoma treatment model.Next,to improve the moderate therapeutic effect of the mono-treatment method,we incorporated a chemotherapeutic drug(doxorubicin,DOX)into the process of vaccination and devised a combination regimen.Fortunately,a tumor inhibition rate of~85%was achieved via the combination therapy,which could not be achieved by mono-chemotherapy or mono-immunotherapy.In summary,the strategy presented here may provide a novel direction in the establishment of a tumor vaccine and is the basis for a prioritization scheme of immuno-chemotherapy in enhancing the therapeutic effect on NHL.Tianlin Zhou Jinrong Penga Ying Hao Kun Shi Kai Zhou Yun Yang Chengli Yang Xinlong He Xinmian Chen Zhiyong Qian 2021Bioactive Materials2021,6,3:1

10Tumor-targeted/reduction-triggered composite multifunctional nanoparticles for breast cancer chemo-photothermal combinational therapy显示文摘Breast cancer has become the most commonly diagnosed cancer type in the world.A combination of chemotherapy and photothermal therapy(PTT) has emerged as a promising strategy for breast cancer therapy.However,the intricacy of precise delivery and the ability to initiate drug release in specific tumor sites remains a challenging puzzle.Therefore,to ensure that the therapeutic agents are synchronously delivered to the tumor site for their synergistic effect,a multifunctional nanoparticle system(PCRHNs) is developed,which is grafted onto the prussian blue nanoparticles(PB NPs) by reductionresponsive camptothecin(CPT) prodrug copolymer,and then modified with tumor-targeting peptide cyclo(Asp-D-Phe-Lys-Arg-Gly)(cRGD) and hyaluronic acid(HA).PCRHNs exhibited nano-sized structure with good monodispersity,high load efficiency of CPT,triggered CPT release in response to reduction environment,and excellent photothermal conversion under laser irradiation.Furthermore,PCRHNs can act as a photoacoustic imaging contrast agent-guided PTT.In vivo studies indicate that PCRHNs exhibited excellent bio compatibility,prolonged blood circulation,enhanced tumor accumulation,allow tumor-specific che mo-photo thermal therapy to achieve synergistic antitumor effects with reduced systemic toxicity.Moreover,hyperthermia-induced upregulation of heat shock protein 70 in the tumor cells could be inhibited by CPT.Collectively,PCRHNs may be a promising therapeutic way for breast cancer therapy.Yun Yang Danrong Hu Yi Lu Bingyang Chu Xinlong He Yu Chen Yao Xiao Chengli Yang Kai Zhou Liping Yuan Zhiyong Qian 2022Acta Pharmaceutica Sinica B2022,12,6:1
11Single-Cell Microwell Platform Reveals Circulating Neural Cells as a Clinical Indicator for Patients with Blood-Brain Barrier Breakdown显示文摘Central nervous system diseases commonly occur with the destruction of the blood-brain barrier.As a primary cause of morbidity and mortality,stroke remains unpredictable and lacks cellular biomarkers that accurately quantify its occurrence and development.Here,we identify NeuN^(+)/CD45^(−)/DAPI^(+)phenotype nonblood cells in the peripheral blood of mice subjected to middle cerebral artery occlusion(MCAO)and stroke patients.Since NeuN is a specific marker of neural cells,we term these newly identified cells as circulating neural cells(CNCs).We find that the enumeration of CNCs in the blood is significantly associated with the severity of brain damage in MCAO mice(p<0:05).Meanwhile,the number of CNCs is significantly higher in stroke patients than in negative subjects(p<0:0001).These findings suggest that the amount of CNCs in circulation may serve as a clinical indicator for the real-time prognosis and progression monitor of the occurrence and development of ischemic stroke and other nervous system disease.Yu Zhang Antony R.Warden Khan Zara Ahmad Yanlei Liu Xijun He Minqiao Zheng Xinlong Huo Xiao Zhi Yuqing Ke Hongxia Li Sijia Yan Wenqiong Su Deng Cai Xianting Ding 2021Research2021,,1:0
12Macrophage Membrane-Camouflaged shRNA and Doxorubicin:A pH-Dependent Release System for Melanoma Chemo-Immunotherapy显示文摘mproving the eficacy of melanoma treatment remains an important global challenge.Here,we combined chemotherapy with protein tyrosine phosphatase nonreceptor type 2(Ptpn2)based immunotherapy in an effort to address this challenge.Shorthairpin RNA(shRNA)targeting Ptpn2 was coencapsulated with doxorubicin(DOX)in the cell membrane of MI macrophages(MIHD@RPR).The prepared nanoparticles(NPs)were effectively phagocytosed by B16F10 cells and MI macrophages,but not by MOmacrophages.Hence,NPevasion from the reticuloendothelial system(RES)was improved and NPenrichment in tumor sites increased.MIHD@RPR can directly kill tumor cells and stimulate immunogenic cell death(ICD)by DOX and downregulate Ptpn2.It can promote Ml macrophage polarization and dendritic cell maturation and increase the proportion of CD8+T cells.MIHD@RPR killed and inhibited the growth of primary melanoma and lung metastatic tumor celis without harming the surrounding tssue.These findings establish MIHD@RPR as a safe multifunctional nanoparticle capable of effectively combining chemotherapy and gene immunotherapies against melanoma.Chengli Yang Yang Ming Kai Zhou Ying Hao Danrong Hu Bingyang Chu Xinlong He Yun Yang Zhiyong Qian 2022Research2022,,2:0

15S-doped porous carbon fibers with superior electrode behaviors in lithium ion batteries and fuel cells显示文摘The orientation construction of S-doped porous carbon fibers(SPCFs)is realized by the facile template-directed methodology using asphalt powder as carbon source.The unique fiber-like morphology without destruction can be well duplicated from the template by the developed methodology.MgSO4 fibers serve as both templates and S dopant,realizing the in-situ S doping into carbon frameworks.The effects of different reaction temperatures on the yield and S doping level of SPCFs are investigated.The S doping can not only significantly enhance the electrical conductivity,but also introduce more defects or disorders.As anode material for lithium ion batteries(LIBs),SPCFs electrode delivers better rate capability than undoped PCFs.And the capacity of SPCFs electrode retains around 90%after 300 cycles at 2 A g1,exhibiting good cycling stability.As the electrocatalysts for fuel cells,the onset potentials of SPCFs obtained at 800 and 900C are concentrated at 0.863 V,and the higher kinetic current densities at 0.4 V of them are larger than that of PCFs,demonstrating the superior electrocatalytic performance.Due to the synergistic effect of abundant pore channels and S doping,SPCFs electrode exhibits superior electrochemical performances as anode for LIBs and elecctrocatalyst for fuel cells,respectively.Additionally,the oriented conversion of asphalt powder into high-performance electrode material in this work provides a new way for the high value application of asphalt.Peng Jin Long Li Xiaohu Gu Yanshao Hu Xiaojing Zhang Xiongchao Lin Xinlong Ma Xing He 2022Materials Reports(Energy)2022,2,4:0
