2Bioengineered miR-27b-3p and miR-328-3p modulate drug metabolism and disposition via the regulation of target ADME gene expression显示文摘Drug-metabolizing enzymes, transporters, and nuclear receptors are essential for the absorption, distribution, metabolism, and excretion(ADME) of drugs and xenobiotics. MicroRNAs participate in the regulation of ADME gene expression via imperfect complementary Watson–Crick base pairings with target transcripts. We have previously reported that Cytochrome P450 3A4(CYP3A4) and ATP-binding cassette sub-family G member 2(ABCG2) are regulated by miR-27b-3p and miR-328-3p,respectively. Here we employed our newly established RNA bioengineering technology to produce bioengineered RNA agents(BERA), namely BERA/miR-27b-3p and BERA/miR-328-3p, via fermentation. When introduced into human cells, BERA/miR-27b-3p and BERA/miR-328-3p were selectively processed to target miRNAs and thus knock down CYP3A4 and ABCG2 mRNA and their protein levels,respectively, as compared to cells treated with vehicle or control RNA. Consequently, BERA/miR-27b-3p led to a lower midazolam 10-hydroxylase activity, indicating the reduction of CYP3A4 activity. Likewise,BERA/miR-328-3p treatment elevated the intracellular accumulation of anticancer drug mitoxantrone, a classic substrate of ABCG2, hence sensitized the cells to chemotherapy. 4Differential Inhibition of Nav1.7 and Neuropathic Pain by Hybridoma-Produced and Recombinant Monoclonal Antibodies that Target Nav1.7显示文摘The voltage-gated Na^+ channel subtype Nav1.7 is important for pain and itch in rodents and humans. We previously showed that a Nav1.7-targeting monoclonal antibody(SVmab) reduces Na+ currents and pain and itch responses in mice. Here, we investigated whether recombinant SVmab(rSVmab) binds to and blocks Nav1.7 similar to SVmab. ELISA tests revealed that SVmab was capable of binding to Nav1.7-expressing HEK293 cells,mouse DRG neurons, human nerve tissue, and the voltagesensor domain Ⅱ of Nav1.7. In contrast, rSVmab showed no or weak binding to Nav1.7 in these tests. Patch-clamp recordings showed that SVmab, but not rSVmab, markedly inhibited Na+ currents in Nav1.7-expressing HEK293 cells. Notably, electrical field stimulation increased the blocking activity of SVmab and rSVmab in Nav1.7-expressing HEK293 cells. SVmab was more effective than rSVmab in inhibiting paclitaxel-induced mechanical allodynia. SVmab also bound to human DRG neurons and inhibited their Na^+ currents. 13Portable quantitative phase microscope for material metrology and biological imaging显示文摘Quantitative phase microscopy(QPM)has emerged as an important tool for material metrology and biological imaging.For broader adoption in those applications,we have proposed and demonstrated a new portable off-axis QPM method,which works in both transmission and reflection modes to meet different sample measurement requirements.The temporal and spatial sensitivities of our system,as quantified by optical path-length difference values,are 0.65 nm and 1.04 nm,respectively.To demonstrate its applicability for a wide range of applications,we deployed our system for profiling transistor gold electrode samples,observing red blood cell membrane fluctuations,imaging living cells flowing in a microfluidic chip,etc.Our portable QPM system has a low-cost design and involves a simple and robust phase-retrieval algorithm that we envision will allow for broader deployment at different environmental settings,including in resource-limited sites and integration with other metrology or imaging modalities.Mengxuan Niu Gang Luo Xin Shu Fuyang Qu Shuang Zhou Yi-Ping Ho Ni Zhao Renjie Zhou 2020Photonics Research2020,8,7:1
14Theoretical and Experimental Studies of Schottky Diodes that Use Aligned Arrays of Single-Walled Carbon Nanotubes显示文摘We present theoretical and experimental studies of Schottky diodes that use aligned arrays of single-walled carbon nanotubes. A simple physical model, taking into account the basic physics of current rectification, can adequately describe the single-tube and array devices. We show that for as-grown array diodes, the rectification ratio, defined by the maximum-to-minimum-current-ratio, is low due to the presence of metallic-single-walled nanotube (SWNT) shunts. These tubes can be eliminated in a single voltage sweep resulting in a high rectification array device. 15A dual-functional BODIPY-based molecular rotor probe reveals different viscosity of protein aggregates in live cells显示文摘Aberrant protein aggregation leads to various human diseases,but little is known about the physical chemical properties of these aggregated proteins in cells.Herein,we developed a boron-dipyrromethene(BODIPY)-based HaloTag probe,whose conjugation to HaloTag-fused proteins allows us to study protein aggregates using both fluorescence intensity and lifetime.Modulation of BODIPY fluorophore reveals key structural features to attain the dual function.The optimized probe exhibits increased fluorescence intensity and elongated fluorescence lifetime in protein aggregates.Fluorescence lifetime imaging using this probe indicates that protein aggregates afford different viscosity in the forms of soluble oligomers and insoluble aggregates in live cells.The strategy presented in this work can be extended to enable a wide class of HaloTag probes that can be used to study a variety of physical properties of protein aggregates,thus helping unravel the pathogenic mechanism and develop therapeutic strategy.Baoxing Shen Kwan Ho Jung Songtao Ye Conner A.Hoelzel Charles H.Wolstenholme He Huang Yu Liu Xin Zhang 2023Aggregate2023,4,3:0
16Facile designing a superhydrophobic anti-icing surface applied for reliable long-term deicing显示文摘While superhydrophobic coatings have shown promise as potential anti-icing coatings, the surface roughness of these coatings is prone to damage during repeated icing-deicing cycles. Herein, two kinds of superhydrophobic anti-icing coatings are prepared from organic resin and micro-nano particles using two strategies, and their excellent anti-icing properties are also investigated. However, superhydrophobic surface Ⅰ(SF1), prepared by first strategy, cannot be used for extended periods of time due to irreversible damage to the surface roughness during the icing–deicing process. Finite element simulations and experimental studies are preformed to investigate the fatal issue of such roughness damage. In contrast,the anti-icing properties of superhydrophobic surface Ⅱ(SF2), prepared by second strategy, can easily regain through a simple sandpaper abrasion treatment even the surface roughness was damaged during the icing–deicing process. 17Factors early in life associated with hepatic steatosis显示文摘BACKGROUND The rise in prevalence of non-alcoholic fatty liver disease(NAFLD)mirrors the obesity epidemic.NAFLD is insidious but may gradually progress from simple steatosis to steatohepatitis,fibrosis and cirrhosis and/or hepatocellular carcinoma.Intervention strategies to ameliorate developmental programming of NAFLD may be more efficacious during critical windows of developmental plasticity.AIM To review the early developmental factors associated with NAFLD.METHODS Databases MEDLINE via PubMed,and EMBASE and Reference Citation Analysis were searched and relevant publications up to April 30,2021 were assessed.Original research studies that included risk factors associated with early development of NAFLD in human subjects were included.These factors include:Maternal factors,intrauterine and prenatal factors,post-natal factors,genetic and ethnic predisposition,childhood and adolescence environmental factors.Studies were excluded if they were review articles or animal studies,case reports or conference abstracts,or if NAFLD was not clearly defined and assessed radiologically.RESULTS Of 1530 citations identified by electronic search,420 duplicates were removed.Of the 1110 citations screened from title and abstract,80 articles were included in the final analysis.Genetic polymorphisms such as patatin-like phospholipase domain-containing protein 3(PNPLA3)and membrane-bound O-acyltransferase domain-containing protein 7(MBOAT7)were associated with increased risk of NAFLD.Familial factors such as maternal obesogenic environment and parental history of hepatic steatosis was associated with offspring NAFLD.Longer duration of exclusive breastfeeding in infancy was associated with a lower risk of developing NAFLD later in life while metabolic dysfunction and/or obesity in adolescence was associated with increased risk of NAFLD.Studies relating to socioeconomic factors and its association with NAFLD reported confounding results.CONCLUSION Maternal metabolic dysfunction during pregnancy,being exclusively breastfed for a longer time postnatally,diet and physical activity in childhood and adolescence are potential areas of intervention to decrease risk of NAFLD.Sabrina Xin Zi Quek Eunice Xiang-Xuan Tan Yi Ping Ren Mark Muthiah Evelyn Xiu Ling Loo Elizabeth Huiwen Tham Kewin Tien Ho Siah 2022World Journal of Hepatology2022,14,6:0
18The modulatory effect of oolong tea polyphenols on intestinal flora and hypothalamus gene expression in a circadian rhythm disturbance mouse model显示文摘The interaction between host circadian rhythm and gut microbes through the gut-brain axis provides new clues for tea polyphenols to improve host health.Our present research showed that oolong tea polyphenols(OTP)improved the structural disorder of the intestinal flora caused by continuous darkness,thereby modulating the production of metabolites related to pyruvate metabolism,glycolysis/gluconeogenesis,and tryptophan metabolism to alleviate the steady-state imbalance.After fecal microbiota transplantation from the OTP group,the single-cell transcriptomic analysis revealed that OTP significantly increased the number of hypothalamus cell clusters,up-regulated the number of astrocytes and fibroblasts,and enhanced the expression of circadian rhythm genes Cry2,Per3,Bhlhe41,Nr1d1,Nr1d2,Dbp and Rorb in hypothalamic cells.Our results confirmed that OTP can actively improve the intestinal environmental state as well as internal/peripheral circadian rhythm disorders and cognitive impairment,with potential prebiotic functional characteristics to notably contribute to host health.Ruonan Yan Chi-Tang Ho Yanan Liu Shengnan Zhan Zufang Wu Xin Zhang 2024Food Science and Human Wellness2024,13,2:0
19Modulatory effects in circadian-related diseases via the reciprocity of tea polyphenols and intestinal microbiota显示文摘Tea is a widespread functional plant resource.Phytochemicals such as tea polyphenols(TP)can interact with the intestinal flora and participate in regulating the expression and rhythm of biological clock genes.Circadian rhythm controls a variety of behaviors and physiological processes,and circadian misalignment has been found to be closely related to multiple metabolic diseases.Interestingly,the gut microbiota also has diurnal fluctuations,which can be affected by diet composition and feeding rhythm,and play a role in maintaining the host’s circadian rhythm.The two-way relationship between the host’s circadian rhythm and intestinal microbiota confirms the possibility that prebiotics or probiotic can be used to adjust the intestinal environment and microbiome composition to improve the host health.This article reviews the relationship between the host’s circadian rhythm and microbiota and its influence on metabolic diseases.The beneficial effects of the interaction between TP and gut microbiota on diseases related to rhythm disorders are emphasized to improve the theories of disease prevention and treatment.Ruonan Yan Chi-Tang Ho Xin Zhang 2022Food Science and Human Wellness2022,11,3:0
20Regenerated polymer optical fiber Bragg gratings with thermal treatment for high temperature measurements显示文摘We report for the first time, to the best of our knowledge, regenerated polymer optical fiber Bragg gratings(RPOFBGs) in ZEONEX-based polymer optical fibers(POFs). The regeneration temperature can be adjusted using a heat treatment process on the POF before FBG inscription, enabling a scalable improvement of the thermal stability of the RPOFBGs. Thermal sustainability of the RPOFBGs at high temperature conditions was investigated for their prolonged use in diverse environments. Furthermore, these RPOFBGs can withstand strain levels up to 2.8% while maintaining a good linearity, even at temperature of 110℃. The RPOFBGs are capable of short-term operation at elevated temperatures of up to 132℃, which is the standard temperature for steam sterilization with at least a 4 min exposure period. The distinction in the morphologies of the two grades of ZEONEX(E48R and 480R, ZEON Corp.) used to fabricate the optical fiber together with the characteristics of UV irradiated and regenerated gratings is explained using micro-Raman spectroscopy.
