1The chromosome-level Hemerocallis citrina Borani genome provides new insights into the rutin biosynthesis and the lack of colchicine显示文摘Hemerocallis citrina Borani(huang hua cai in Chinese)is an important horticultural crop whose flower buds are widely consumed as a delicious vegetable in Asia.Here we assembled a high-quality reference genome of H.citrina using single-molecule sequencing and Hi-C technologies.The genome assembly was 3.77 Gb and consisted of 3183 contigs with a contig N50 of 2.09 Mb,which were further clustered into 11 pseudochromosomes.A larger portion(3.25 Gb or 86.20%)was annotated as a repetitive content and 54,295 protein-coding genes were annotated in the genome.Genome evolution analysis showed that H.citrina experienced a recent whole-genome duplication(WGD)event at~15.73 million years ago(Mya),which was the main factor leading to many multiple copies of orthologous genes.We used this reference genome to predict 20 genes involved in the rutin biosynthesis pathway.Moreover,our metabolomics data revealed neither colchicine nor its precursors in H.citrina,challenging the long-standing belief that this alkaloid causes poisoning by the plant.The results of our disruptive research are further substantiated by our genomic finding that H.citrina does not contain any genes involved in colchicine biosynthesis.The high-quality genome lays a solid foundation for genetic research and molecular breeding of H.citrina.Zhixing Qing Jinghong Liu Xinxin Yi Xiubin Liu Guoan Hu Jia Lao Wei He Zihui Yang Xiaoyan Zou Mengshan Sun Peng Huang Jianguo Zeng 2021Horticulture Research2021,8,1:6
2Covalent organic frameworks derived hollow structured N-doped noble carbon for asymmetric-electrolyte Zn-air battery显示文摘We report a relatively low-temperature molten salt strategy to prepare hollow structured N-doped noble carbon(h-NNC) with highly desirable features of ultra-large surface area(1957 m^2 g^(-1)) and high graphitization, endowing the h-NNC with high activity toward catalysis of oxygen reduction reaction in acidic medium. 4Implementing a “free” tuberculosis (TB) care policy under the integrated model in Jiangsu, China: practices and costs in the real world显示文摘Background:In the 1990s,China introduced a“free”tuberculosis(TB)care policy under the national TB control program.Recently,as a part of a new TB diagnosis and treatment model,it has been recommended that the integrated model scale up.This paper examines whether or not TB designated hospitals in the selected project sites have provided TB care according to the national and local guidelines,and analyzes the actual practices and expenditures involved in completing TB treatment.It also explores the reasons why“free”TB care in China cannot be effectively implemented under the integrated model.Methods:This study was conducted in three counties of Zhenjiang city,Jiangsu province.Mixed methods were used,which comprised reviewing the national and local TB control guidelines,conducting TB patient surveys,collecting TB inpatient and outpatient hospital records,and conducting qualitative interviews with stakeholders.Descriptive statistics were used for quantitative data analysis across counties and in order to compare patients who received only outpatient care and those who received both outpatient and inpatient care.The chi-square test and analysis of variance were performed where necessary.Qualitative data were analyzed using the framework approach.Results:Although the national TB care guidelines recommend outpatient care as a basis for TB treatment in China,we found high hospital admission rates for TB patients ranging from 39%in Yangzhong county to 83%in Dantu county.Almost all outpatient TB patients paid for lab tests and over 80%paid for liver protection drugs and around 70%paid for image examinations.These three components accounted for three-quarters of the total outpatient expenditure.For patients who received only outpatient care,the total expenditure upon completion of TB treatment was on average 1,135 Chinese yuan.For patients who received outpatient and inpatient care,the total expenditure upon completion of TB treatment was 11,117 Chinese yuan.Conclusion:The“free”TB care policy under the integrated model has not been effectively implemented in China.There has been substantial spending on non-recommended services,examinations,and drugs for TB treatment.Xinxin Jia Jiaying Chen Siyuan Zhang Bing Dai Qian Long Shenglan Tang 2016Infectious Diseases of Poverty2016,5,1:4

6Divergence, evolution and adaptation in ray-finned fish genomes显示文摘With the rapid development of next-generation sequencing technologies and bioinformatics, over 50 ray-finned fish genomes by far have been sequenced with high quality. The genomic work provides abundant genetic resources for deep understanding of divergence, evolution and adaptation in the fish genomes. They are also instructive for identification of candidate genes for functional verification, molecular breeding, and development of novel marine drugs. As an example of other omics data, the Fish-T1 K project generated a big database of fish transcriptomes to integrate with these published fish genomes for potential applications. 7Coarsening kinetics of γ' precipitates in a Re-containing Ni-based single crystal superalloy during long-term aging显示文摘The morphological evolution and coarsening kinetics ofγ'precipitates in a Re-containing Ni-based single crystal superalloy were investigated during isothermal aging at 900,950 and 1000℃.After heat treatment,well-defined cuboidalγ'precipitates with low misfit was obtained within the experimental alloy.Then coarsening rate constants and particle size distribution(PSD)ofγ'phases were calculated and specified based on the measured precipitate sizes for va rying periods of aging times from 100 to 2000 h.After aging for 2000 h,γ'precipitates maintained cubical shape at 900℃,while exhibited sphere at 950 and 1000℃.Coarsening models based on diffusion-controlled process with a functional relationship of r^(3) vs.t(classic Lifshitz-Slyozov-Wagner coarsening model)and interface-controlled model with a function of r^(2) vs.t(trans-interface diffusion-controlled coarsening model)were investigated to fit between the experimental results and theoretical analysis.It was found that Re as the slowest diffusing solute in the alloy constituted the rate-limited step for coarsening based on LSW model,while the process limiting coarsening as governed by an interface diffusion process could possibly be related to the Al diffusion through theγ/γ'interface.The PSDs and coarsening exponent were discussed by comparing the experimental data with predictions of LSW and TIDC models.Finally,coarsening mechanism could be divided into four regimes:(i)coarsening by diffusion-controlled;(ii)coarsening by diffusion and interface cocontrolled;(iii)coarsening by interface-controlled;(iv)coarsening by interface-controlled accompanied withγ'coalescence.Jiachen Zhang Lin Liu Taiwen Huang Jia Chen Kaili Cao Xinxin Liu Jun Zhang Hengzhi Fu 2021Journal of Materials Science & Technology2021,,3:2

12Quantification of groundwater recharge and evapotranspiration along a semi-arid wetland transect using diurnal water table fluctuations显示文摘Groundwater is a vital water resource in arid and semi-arid areas.Diurnal groundwater table fluctuations are widely used to quantify rainfall recharge and groundwater evapotranspiration(ET_(g)).To assess groundwater resources for sustainable use,we estimated groundwater recharge and ET_(g) using the diurnal water table fluctuations at three sites along a section with different depths to water table(DWT)within a wetland of the Mukai Lake in the Ordos Plateau,Northwest China.The water table level was monitored at an hourly resolution using a Keller DCX-22 A data logger that measured both the total pressure and barometric pressure,so that the effect of barometric pressure could be removed.At this study site,a rapid water table response to rainfall was observed in two shallow wells(i.e.,Obs1 and Obs2),at which diurnal water table fluctuations were also observed over the study period during rainless days,indicating that the main factors influencing water table variation are rainfall and ET_(g).However,at the deep-water table site(Obs3),the groundwater level only reacted to the heaviest rainfalls and showed no diurnal variations.Groundwater recharge and ET_(g) were quantified for the entire hydrological year(June 2017–June 2018)using the water table fluctuation method and the Loheide method,respectively,with depth-dependent specific yields.The results show that the total annual groundwater recharge was approximately 207 mm,accounting for 52%of rainfall at Obs1,while groundwater recharge was approximately 250 and 21 mm at Obs2 and Obs3,accounting for 63%and 5%of rainfall,respectively.In addition,the rates of groundwater recharge were mainly determined by rainfall intensity and DWT.The daily mean ET_(g) at Obs1 and Obs2 over the study period was 4.3 and 2.5 mm,respectively,and the main determining factors were DWT and net radiation.JIA Wuhui YIN Lihe ZHANG Maosheng ZHANG Xinxin ZHANG Jun TANG Xiaoping DONG Jiaqiu 2021Journal of Arid Land2021,13,5:1

16A genome-wide association study on growth traits in orangespotted grouper(Epinephelus coioides) with RAD-seq genotyping显示文摘The orange-spotted grouper, Epinephelus coioides, is one of the most popular fish in China and Southeast Asian countries because of its important economic value. However, molecular mechanism underlying the growth of orange-spotted grouper has never been fully understood. Herein, we performed a genome-wide association study(GWAS) on a natural population of 198 individuals aiming to screen the whole genome of orange-spotted grouper for identification of growth-related loci by restrictionsite associated DNA sequencing. In this research, 261,366 single nucleotide polymorphisms(SNPs) were developed, in which110 SNPs were identified to be correlated with growth and 20 SNPs were further confirmed to be associated with both body weight and total length. From these identified SNPs, we annotated a total of 34 genes, including adgrb2, csnkza1, cers5, col22 a1,creb5, dnd1, dzank1, dnai1, npy2 r, fat3, lrrk2, lrp5, map3 k9, and so on. Among these candidate genes, npy2 r(neuropeptide Y receptor Y2) was reported to play a critical role in growth of the orange-spotted grouper. In addition, population structure,principal component analysis, kinship matrix and linkage disequilibrium were examined to verify the accuracy and reliability of our GWAS results. 19Extraction of straight field roads between farmlands based on agricultural vehicle-mounted LiDAR显示文摘The application of autonomous agricultural vehicles is gaining popularity as a way to increase production efficiency and lower operational costs.To achieve high performance,perception tasks(such as obstacle detection,road extraction,and drivable area extraction)are of great importance.Compared with structured roads,field roads between farmlands,including unstructured roads and semi-structured roads,are unfavorable for autonomous agricultural vehicle driving due to their bumpiness and unstructured nature.This study proposed an extraction method for the straight field roads between farmlands.The proposed method was based on the point cloud data acquired by LiDAR(Velodyne VLP-16)mounted on a John Deere 12046B-1204 tractor.The proposed method has three aspects:Euclidean Clustering-based extraction,boundary-based extraction,and road point cloud curve segment modification.Firstly,Euclidean Clustering with K-Dimensional(KD)-Tree data structure was adopted to extract the road curve segments close to the LiDAR composed of road points.Secondly,the boundary lines constraint was constructed to extract the distant road curve segments.Thirdly,the local distance ratio was used to modify the extracted road curve segments.The average extraction accuracy for both semi-structured and unstructured roads exceeded 98%,and the false positive rate(FPR)was less than 0.5%.These experimental findings demonstrated that the proposed road extraction method was precise and effective.The proposed method of this study can be applied to enhance the perception ability of autonomous agricultural vehicles thereby increasing the efficiency and safety of field road driving.Lili Yang Yuanyuan Xu Yajie Liang Jia Qin Yuanbo Li Xinxin Wang Weixin Zhai Long Wen Zhibo Chen Caicong Wu 2022International Journal of Agricultural and Biological Engineering2022,15,5:1
20Cloning and Functional Characterization of Chalcone Isomerase Genes Involved in Anthocyanin Biosynthesis in Clivia miniata显示文摘Chalcone isomerase(CHI),catalyzing isomerization of chalcones,is a crucial enzyme in flavonoid biosynthesis.Three CHI genes were isolated from Clivia miniata and designated as CmCHI1,CmCHI2 and CmCHI3,respectively.Multiple sequence alignments and phylogenetic analysis showed that CmCHI1 and CmCHI2 were members of type I CHI proteins,whereas CmCHI3 belonged to type IV CHI proteins.Subcellular localization analysis found that all three CmCHIs had diffused distribution in the cytoplasm similar to green fluorescent protein(GFP).Anthocyanin biosynthesis and gene expression analysis demonstrated that CmCHIs were highly expressed in anthocyanin accumulated tissues.To further functionally characterize the role of CmCHIs,an in vitro enzymatic activity assay was carried out using the purified recombinant proteins.Results showed that CmCHI1 and CmCHI2 could completely convert the substrate naringenin chalcone(NC)into the product naringenin(NA),whereas CmCHI3 seemed nonfunctional as no increment of NA was detected.Further genetic transformation of Arabidopsis tt5-1 mutant validated that CmCHI1 and CmCHI2 rather than CmCHI3 could complement the chi deficient phenotypes.In summary,CmCHI1 and CmCHI2 are the real active CHI genes in Clivia miniata.The results not only broaden our knowledge on flavonoid biosynthesis in C.miniata but also lay a new foundation for further flavonoid modification in C.miniata.Yue Liu Xinxin Xue Chunli Zhao Jia Zhang Meng Liu Xiangyu Li Yueqing Li Xiang Gao 2021Ornamental Plant Research2021,1,1:1
