| 1 | Kinsenoside attenuates osteoarthritis by repolarizing macrophages through inactivating NF-κB/MAPK signaling and protecting chondrocytes显示文摘The objective was to investigate the effect of kinsenoside(Kin) treatments on macrophage polarity and evaluate the resulting protection of chondrocytes to attenuate osteoarthritis(OA) progression.RAW264.7 macrophages were polarized to M1/M2 subtypes then administered with different concentrations of Kin. The polarization transitions were evaluated with quantitative real-time polymerase chain reaction(q RT-PCR), confocal observation and flow cytometry analysis. The mechanism of Kin repolarizing M1 macrophages was evaluated by Western blot. Further, macrophage conditioned medium(CM) and IL-1β were administered to chondrocytes. Micro-CT scanning and histological observations were conducted in vivo on anterior cruciate ligament transection(ACLT) mice with or without Kin treatment. We found that Kin repolarized M1 macrophages to the M2 phenotype. Mechanistically, Kin inhibited the phosphorylation of IκBα, which further reduced the downstream phosphorylation of P65 in nuclear factor-κB(NF-κB) signaling. Moreover, Kin inhibited mitogen-activated protein kinases(MAPK) signaling molecules p-JNK, p-ERK and p-P38. Additionally, Kin attenuated macrophage CM and IL-1β-induced chondrocyte damage. In vivo, Kin reduced the infiltration of M1 macrophages,promoted M2 macrophages in the synovium, inhibited subchondral bone destruction and reduced articular cartilage damage induced by ACLT. | 2 | Degradation of chlorpyrifos in laboratory soil and its impact on soil microbial functional diversity显示文摘Degradation of chlorpyrifos at different concentrations in soil and its impact on soil microbial functional diversity were investigated under laboratory condition.The degradation half-live of chlorpyrifos at levels of 4,8,and 12 mg/kg in soil were calculated to be 14.3,16.7,and 18.0 d,respectively.The Biolog study showed that the average well color development(AWCD) in soils was significantly(P < 0.05) inhibited by chlorpyrifos within the first two weeks and thereafter recovered to a similar level as the control.A similar variation in the diversity indices(Simpson index 1/D and McIntosh index U) was observed,but no significant difference among the values of the Shannon-Wiener index H was found in chlorpyrifos-treated soils.With an increasing chlorpyrifos concentration,the half-life of chlorpyrifos was significantly(P 0.05) extended and its inhibitory effect on soil microorganisms was aggravated.It is concluded that chlorpyrifos residues in soil had a temporary or short-term inhibitory effect on soil microbial functional diversity. | FANG Hua YU Yunlong CHU Xiaoqiang WANG Xiuguo YANG Xiaoe YU Jingquan | 2009 | Journal of Environmental Sciences2009,21,3: | 25 |
| 3 | Carbendazim induces a temporary change in soil bacterial community structure显示文摘The effect of carbendazim applications on the diversity and structure of a soil bacterial community was studied under field conditions using temperature gradient gel electrophoresis (TGGE) and partial sequence analysis of PCR-amplified 16S rRNA gene. After four successive introductions of carbendazim at a level of 0.94 kg active ingredient (a.i.)/ha, the genetic diversity (expressed as Shannon index, H ) decreased from 1.43 in the control to 1.29 in treated soil. This harmful effect seems to increase with the concentration of carbendazim. The value of H in the soil treated with carbendazim at 4.70 kg a.i./ha was reduced to 1.05 (P 0.05). The structure of soil bacterial community was also affected after four repeated applications of carbendazim at levels of 0.94, 1.88 and 4.70 kg a.i./ha, as seen in the relative intensities of the individual band. However, the bacterial community in carbendazim-treated soil recovered to that in the control 360 d after the first treatment. | 4 | Highly Efficient Photoelectrocatalytic Reduction of CO2 to Methanol by a p–n Heterojunction CeO2/CuO/Cu Catalyst显示文摘Photoelectrocatalytic reduction of CO2 to fuels has great potential for reducing anthropogenic CO2 emissions and also lessening our dependence on fossil fuel energy.Herein,we report the successful development of a novel photoelectrocatalytic catalyst for the selective reduction of CO2 to methanol,comprising a copper catalyst modified with flower-like cerium oxide nanoparticles(CeO2 NPs)(a n-type semiconductor)and copper oxide nanoparticles(CuO NPs)(a p-type semiconductor).At an applied potential of−1.0 V(vs SCE)under visible light irradiation,the CeO2 NPs/CuO NPs/Cu catalyst yielded methanol at a rate of 3.44μmol cm^−2 h^−1,which was approximately five times higher than that of a CuO NPs/Cu catalyst(0.67μmol cm^−2 h^−1).The carrier concentration increased by^108 times when the flower-like CeO2 NPs were deposited on the CuO NPs/Cu catalyst,due to synergistic transfer of photoexcited electrons from the conduction band of CuO to that of CeO2,which enhanced both photocatalytic and photoelectrocatalytic CO2 reduction on the CeO2 NPs.The facile migration of photoexcited electrons and holes across the p–n heterojunction that formed between the CeO2 and CuO components was thus critical to excellent light-induced CO2 reduction properties of the CeO2 NPs/CuO NPs/Cu catalyst.Results encourage the wider application of composite semiconductor electrodes in carbon dioxide reduction. | Zhengbin Pan Ershuan Han Jingui Zheng Jing Lu Xiaolin Wang Yanbin Yin Geoffrey INWaterhouse Xiuguo Wang Peiqiang Li | 2020 | Nano-Micro Letters2020,12,2: | 3 |

| 13 | ASF1 regulates asexual and sexual reproduction in Stemphylium eturmiunum by DJ-1 stimulation of the PI3K/AKT signaling pathway显示文摘Most fungi display a mixed mating system with both asexual and sexual reproduction.The timing of the two modes of reproduction must be carefully coordinated through signal perception and coordination in the cell along with chromatin modification.Here,we investigated coordination of reproductive output by investigating the function of the histone chaper-one anti-silencing factor 1(ASF1)in a fungal species amenable to characterization of both asexual and sexual reproduction.We used knockout approach to show that SeASF1 influenced asexual and sexual reproduction in Stemphylium eturmiunum.SeASF1-deleted strains failed to produce pseudothecia,but produce abnormal conidia and showed an irregular distribution of nuclei in mycelium.Transcriptome sequencing was then used to identify genes with altered expression in the SeASF1-deleted strains.The transcriptional expression of the identified SeDJ-1 was strongly regulated by SeASF1.The interaction of SeDJ-1 and SeASF1 was confirmed using Y2H,Co-IP,and pull-down.Due to some components of phosphatidylinositol 3-kinase/protein kinase B(PI3K/AKT)signaling pathway were known to interact with DJ-1 in mammals,we verified SePI3K,an element of PI3K/AKT signaling pathway in S.eturmiunum,was directly linked to SeDJ-1 and then these two proteins were defined as a coordinator of reproduction.However,knockout of SeDJ-1 or SePI3K altered the asexual and sexual repro-duction,but SePI3K recovered the asexual and sexual development of∆Sedj-1.The SeDJ-1-M6 segment of SeDJ-1 was essential for its interaction with SePI3K and played a critical role in restoring sexual reproduction in the∆Sepi3k,providing a deep understanding of the regulatory mechanism of SeDJ-1 in S.eturmiunum development.Summarily,SeASF1 is able to trigger SeDJ-1 and SeDJ-1can also activate SePI3K,which is orchestrally involved in asexual and sexual reproduction in S.eturmiunum.All these results reveal that SeASF1 manipulates asexual and sexual reproduction in S.eturmiunum by SeDJ-1 perception of PI3K/AKT signaling pathway.These data highlight the deep similarities in coordinating asexual and sexual processes in both fungi and eukaryotes in general. | Shi Wang Xiaoman Liu Chenlin Xiong Susu Gao Wenmeng Xu Lili Zhao Chunyan Song Xiaoyong Liu Timothy Y.James Zhuang Li Xiuguo Zhang | 2023 | Fungal Diversity2023,,6: | 0 |
| 14 | Effects of Post-expanding Lower Temperature Granulating Process on Growth Performance of Piglets显示文摘[Objective] The paper was to compare the effects of traditional granulating process and post-expanding lower temperature granulating process on starch gelatinization degree and in vitro protein digestibility of conversation feed, and growth performance of piglets. [Method] Sixty weaned piglets, 25 days old, were randomly divided into two groups, six replicates each group, and each replicate contained five piglets. Piglets in control group were fed with the conversation feed developed by traditional granulating process, and those in experimental group were fed with the conservation feed developed by post-expanding lower temperature granulating process. The trial lasted 28 d. [Result] Compared to traditional granulating process, post-expanding lower temperature granulating process significantly enhanced starch gelatinization degree of conservation feed by56.9%(P<0.05), and significantly improved in vitro protein digestibility of conservation feed by 11.48%(P<0.05). Compared to the control group, the feed intake of piglets in experimental group was increased by 21.63%(P<0.05); the average daily gain was increased by 27.61%(P<0.05); the diarrhea rate was lowered; and the feed gain ratio was improved by 4.51%(P>0.05). The results suggested that post-expanding lower temperature granulating process effectively enhanced feed digestion and absorption of piglets. The aim of this study was to analyze the overall composition of the gut virome in coronary heart disease(CHD) patients, and try to discover the potential link between the human gut virome and CHD. Viral metagenomics methods were performed to detect the viral sequences in fecal samples collected from CHD inpatients and healthy persons as controls. We present the analysis of the virome composition in these CHD patients and controls. Our data shows that the virome composition may be linked to daily living habits and the medical therapy of CHD.Virgaviridae and Microviridae were the two dominant types of viruses found in the enteric virome of CHD patients. Fourteen divergent viruses belonging to the family Microviridae were found, twelve of which were grouped into the subfamily Gokushovirinae, while the remaining two strains might represent two new subfamilies within Microviridae, according to the phylogenetic analysis. | 18 | Characterization of tick viromes collected from dogs in China显示文摘In this study,we explored the viral diversity and community structures of the ticks collected from dogs and detected possible tick-borne viruses(TBVs)using metagenomic analysis.Additionally,full-length sequences and the structural,phylogenetic andmolecular features of the possible TBVs were characterized using bioinformatics tools.The sequence run produced 12,254,268 reads and 6,667,259 unique reads from the tick pools.Further analysis of the viral reads revealed that 92.73%were similar to ssRNA viruses,and 90.5%of the total viral sequences belonged to Non-classified viral families.Of the 29 classified viral families,most virome sequenceswere homologouswith vertebrate viruses from Circoviridae,plant viruses from Virgaviridae,phages fromMicroviridae,insect viruses from Baculoviridae,and giant viruses from Mimiviridae.In the process of assembly of reads,large contigs representing four virus families were identified,including Phenuiviridae,Chuviridae,Parvoviridae and Non-classified families.Some sequences of Non-classified family shared a highly divergent amino acid sequence identities from existing virus sequences.The importance of these newly identified virus contigs to public and veterinary health needs additional researches.Because of the lack of the acquired viral reads from these families,we only demonstrated the complete sequences of Circoviridae in detail.The complete canine circovirus(CaCV)-NC21 genome detected from police dogs was 2,063 bp long with a GC content of 53.6%.Our results suggested that tick viromes collected fromdogs contained diverse sequences with a broad range of animal,insect,plant,and phage viruses,which may reflect the ecological characteristics of the dogs and their ticks.Furthermore,our study revealed the existence of the partial contigs belonging to the four virus families in ticks.More researches are needed to verify their importance to public and veterinary health.As well,our detection of CaCv-NC21 demonstrated that ticks can harbor and potentially transmit canine circoviruses. | Seongjin Wang Tingting Zhao Xiangqian Yu Zhibing Lin Xiuguo Hua Li Cui | 2020 | Biosafety and Health2020,2,2: | 0 |
