1Human ESC-derived expandable hepatic organoids enable therapeutic liver repopulation and pathophysiological modeling of alcoholic liver injury显示文摘We report the generation of human ESC-derived,expandable hepatic organoids(hEHOs)using our newly established method with wholly defined(serum-free,feeder free)media.The hEHOs stably maintain phenotypic features of bipotential liver stem/progenitor cells that can differentiate into functional hepatocytes or cholangiocytes.The hEHOs can expand for 20 passages enabling large scale expansion to cell numbers requisite for industry or clinical programs.The cells from hEHOs display remarkable repopulation capacity in injured livers of FRG mice following transplantation,and they differentiate in vivo into mature hepatocytes.If implanted into the epididymal fat pads of immune-deficient mice,they do not generate non-hepatic lineages and have no tendency to form teratomas.We further develop a derivative model by incorporating human fetal liver mesenchymal cells(hFLMCs)into the hEHOs,referred to as hFLMC/hEHO,which can model alcoholic liver disease-associated pathophysiologic changes,including oxidative stress generation,steatosis,inflammatory mediators release and fibrosis,under ethanol treatment.Our work demonstrates that the hEHOs have considerable potential to be a novel,ex vivo pathophysiological model for studying alcoholic liver disease as well as a promising cellular source for treating human liver diseases.Shuyong Wang Xuan Wang Zuolong Tan Yuxin Su Juan Liu Mingyang Chang Fang Yan Jie Chen Tao Chen Chuanjiang Li Jie Hu Yunfang Wang 2019Cell Research2019,29,12:20
2Analysis and Numerical Simulation of Rolling Contact between Sphere and Cone显示文摘In non-conforming rolling contact, the contact stress is highly concentrated in the contact area. However, there are some limitations of the special contact model and stress model used for the theoretical study of the phenomenon, and this has prevented in-depth analysis of the associated friction, wear, and failure. This paper is particularly aimed at investigating the area of rolling contact between a sphere and a cone, for which purpose the boundary is determined by the Hertz theory and the geometries of the non-conforming surfaces. The phenomenon of stick-slip contact is observed to occur in the contact area under the condition of no-full-slip(Q < μ·P). Using the two-dimensional rolling contact theory developed by CARTER, the relative positions of the stick and slip regions and the distribution of the tangential force over the contact area are analyzed. Furthermore, each stress component is calculated based on the Mc Ewen theory and the idea of narrow band. The stress equations for the three-dimensional rolling contact between the sphere and the cone are obtained by the principle of superposition, and are used to perform some numerical simulations. The results show that the stress components have a large gradient along the boundary between the stick and slip regions, and that the maximum stress is inversely proportional to the contact coefficient and proportional to the friction coefficient. 4Inheritance of Rice Seed Germination Ability under Salt Stress显示文摘A population of recombinant inbred lines(RILs,F2:9),derived from a cross between IR26(Oryza sativa subsp.indica)and Jiucaiqing(Oryza sativa subsp.japonica),was used to identify seed germination ability of rice under 100 mmol/L NaCl for 10 days.Six germination traits including imbibition rate,germination rate,germination index,root length,shoot length and vigor index were investigated.A mixed major gene and polygene inheritance model was applied to conduct genetic analysis for germination ability.Significant differences were detected in all the germination traits under salt stress among RILs in rice,and the early germination stage(0-5 days)might be the salt sensitive stage.The frequency distributions of the germination traits under salt stress in the RIL population showed continuous segregation,suggesting that they were quantitative traits controlled by several genes.The germination traits under salt stress were regulated by two or three major genes plus polygene,and mainly dominated by major genes with high heritability values,accounting for 12.5%-99.0%of the total phenotypic variation.Each trait was controlled by the specific genetic model:imbibition rate was controlled by two major genes,germination index and vigor index by two major genes plus polygene,germination rate and shoot length by three major genes plus polygene,and root length by two major genes or two major genes plus polygene.WANG Zhou-fei WANG Jian-fei BAO Yong-mei Wu Yun-yu Su Xuan ZHANG Hong-sheng 2010Rice science2010,17,2:14
5Molecular phylogenetic relationships of China Seas groupers based on cytochrome b gene fragment sequences显示文摘The classification and evolutionary relationships are important issues in the study of the groupers. Cytochrome b gene fragment of twenty-eight grouper species within six genera of subfamily Epinephelinae was amplified using PCR techniques and the sequences were analyzed to derive the phylogenetic relationships of the groupers from the China Seas. Genetic information indexes, including Kimura-2 parameter genetic distance and Ts/Tv ratios, were generated by using a variety of biology softwares. With Niphon spinosus, Pagrus major and Pagrus auriga as the designated outgroups, phylogenetic trees, which invoke additional homologous sequences of other Epinephelus fishes from GenBank, were constructed based on the neighbor-joining (NJ), maximum-parsimony (MP), maximum-likelihood (ML) and minimum-evolution (ME) methods. 8Effects of Tanreqing Capsule on the negative conversion time of nucleic acid in patients with COVID-19:A retrospective cohort study显示文摘Objective:Traditional Chinese medicine plays a significant role in the treatment of the pandemic of coronavirus disease 2019(COVID-19).Tanreqing Capsule(TRQC)was used in the treatment of COVID-19 patients in the Shanghai Public Health Clinical Center.This study aimed to investigate the clinical efficacy of TRQC in the treatment of COVID-19.Methods:A retrospective cohort study was conducted on 82 patients who had laboratory-confirmed mild and moderate COVID-19;patients were treated with TRQC in one designated hospital.The treatment and control groups consisted of 25 and 57 cases,respectively.The treatment group was given TRQC orally three times a day,three pills each time,in addition to conventional Western medicine treatments which were also administered to the control group.The clinical efficacy indicators,such as the negative conversion time of pharyngeal swab nucleic acid,the negative conversion time of fecal nucleic acid,the duration of negative conversion of pharyngeal-fecal nucleic acid,and the improvement in the level of immune indicators such as T-cell subsets(CD3,CD4 and CD45)were monitored.Results:COVID-19 patients in the treatment group,compared to the control group,had a shorter negative conversion time of fecal nucleic acid(4 vs.9 days,P=0.047)and a shorter interval of negative conversion of pharyngeal-fecal nucleic acid(0 vs.2 days,P=0.042).The level of CD3+T cells increased in the treatment group compared to the control group([317.09±274.39]vs.[175.02±239.95]counts/l L,P=0.030).No statistically significant differences were detected in the median improvement in levels of CD4+T cells(173 vs.107 counts/l L,P=0.208)and CD45+T cells(366 vs.141 counts/l L,P=0.117)between the treatment and control groups.Conclusion:Significant reductions in the negative conversion time of fecal nucleic acid and the duration of negative conversion of pharyngeal-fecal nucleic acid were identified in the treatment group as compared to the control group,illustrating the potential therapeutic benefits of using TRQC as a complement to conventional medicine in patients with mild and moderate COVID-19.The underlying mechanism may be related to the improved levels of the immune indicator CD3+T cells.Xing Zhang Yan Xue Xuan Chen Jia-min Wu Zi-jian Su Meng Sun Lu-jiong Liu Yi-bao Zhang Yi-le Zhang Gui-hua Xu Miao-yan Shi Xiu-ming Song Yun-fei Lu Xiao-rong Chen Wei Zhang Qi Chen 2021Journal of Integrative Medicine2021,19,1:8
9Application of deep learning in ecological resource research:Theories, methods, and challenges显示文摘Ecological resources are an important material foundation for the survival,development,and self-realization of human beings.In-depth and comprehensive research and understanding of ecological resources are beneficial for the sustainable development of human society.Advances in observation technology have improved the ability to acquire long-term,cross-scale,massive,heterogeneous,and multi-source data.Ecological resource research is entering a new era driven by big data.Traditional statistical learning and machine learning algorithms have problems with saturation in dealing with big data.Deep learning is a method for automatically extracting complex high-dimensional nonlinear features,which is increasingly used for scientific and industrial data processing because of its ability to avoid saturation with big data.To promote the application of deep learning in the field of ecological resource research,here,we first introduce the relationship between deep learning theory and research on ecological resources,common tools,and datasets.Second,applications of deep learning in classification and recognition,detection and localization,semantic segmentation,instance segmentation,and graph neural network in typical spatial discrete data are presented through three cases:species classification,crop breeding,and vegetation mapping.Finally,challenges and opportunities for the application of deep learning in ecological resource research in the era of big data are summarized by considering the characteristics of ecological resource data and the development status of deep learning.It is anticipated that the cooperation and training of cross-disciplinary talents may promote the standardization and sharing of ecological resource data,improve the universality and interpretability of algorithms,and enrich applications with the development of hardware.Qinghua GUO Shichao JIN Min LI Qiuli YANG Kexin XU Yuanzhen JU Jing ZHANG Jing XUAN Jin LIU Yanjun SU Qiang XU Yu LIU 2020Science China Earth Sciences2020,63,10:7

133D-Printed Superhydrophobic and Magnetic Device That Can Self-Powered Sense A Tiny Droplet Impact显示文摘Three-dimensional(3D)-printed magnetic soft architectures have attracted extensive attention and research from the engineering and material fields.The force-driven shape deformation of such architectures causes a change in the magnetic field distribution,indicating the capability to convert mechanical energy to electricity.Herein,we fabricate a flexible superhydrophobic and magnetic device by integrating two kinds of 3D printing approaches.The 3D-printed magnetic device(3DMD)exhibits a long-term stable mechanoelectrical conversion capacity under consecutive water droplet dripping.The output current of the 3DMD is higher than that of records in the existing literature.Combined with Maxwell numerical simulation,the mechanoelectrical conversion mechanism of the 3DMD is investigated,further guiding regulation of the diverse parameters.Moreover,three 3DMDs are integrated to light up a commercial light-emitting diode(LED)by a stream of collected rainwater.Such a combined design incorporating energy conversion is believed to promisingly motive advances in the 3D printing field.Xuan Zhang Qi Wang Ruiping Zou Bo Song Chunze Yan Yusheng Shi Bin Su 2022Engineering2022,8,8:4

16Early therapeutic interventions of traditional Chinese medicine in COVID-19 patients:A retrospective cohort study显示文摘Objective:To observe the early interventions of traditional Chinese Medicine(TCM)on the conversion time of nucleic acid in patients with coronavirus disease 2019(COVID-19),and find possible underlying mechanisms of action.Methods:A retrospective cohort study was conducted on 300 confirmed COVID-19 patients who were treated with TCM,at a designated hospital in China.The patients were categorized into three groups:TCM1,TCM2 and TCM3,who respectively received TCM interventions within 7,8–14,and greater than15 days of hospitalization.Different indicators such as the conversion time of pharyngeal swab nucleic acid,the conversion time of fecal nucleic acid,length of hospital stay,and inflammatory markers(leukocyte count,and lymphocyte count and percentage)were analyzed to observe the impact of early TCM interventions on these groups.Results:The median conversion times of pharyngeal swab nucleic acid in the three groups were 5.5,7 and16 d(P<0.001),with TCM1 and TCM2 being statistically different from TCM3(P<0.01).TCM1(P<0.05)and TCM3(P<0.01)were statistically different from TCM2.The median conversion times of fecal nucleic acid in the three groups were 7,9 and 17 d(P<0.001).Conversion times of fecal nucleic acid in TCM1 were statistically different from TCM3 and TCM2(P<0.01).The median lengths of hospital stay in the three groups were 13,16 and 21 d(P<0.001).TCM1 and TCM2 were statistically different from TCM3(P<0.01);TCM1 and TCM3 were statistically different from TCM2(P<0.01).Both leucocyte and lymphocyte counts increased gradually with an increase in the length of hospital stay in TCM1 group patients,with a statistically significant difference observed at each time point in the group(P<0.001).Statistically significant differences in lymphocyte count and percentage in TCM2(P<0.001),and in leucocyte count(P=0.043)and lymphocyte count(P=0.038)in TCM3 were observed.The comparison among the three groups showed a statistically significant difference in lymphocyte percentage on the third day of admission(P=0.044).Conclusion:In this study,it was observed that in COVID-19 patients treated with a combination of Chinese and Western medicines,TCM intervention earlier in the hospital stay correlated with faster conversion time of pharyngeal swab and fecal nucleic acid,as well as shorter length of hospital stay,thus helping promote faster recovery of the patient.The underlying mechanism of action may be related to improving inflammation in patients with COVID-19.Miao-yan Shi Shi-qi Sun Wei Zhang Xing Zhang Gui-hua Xu Xuan Chen Zi-jian Su Xiu-ming Song Lu-jiong Liu Yi-bao Zhang Yi-le Zhang Meng Sun Qi Chen Yan Xue Hua Lü Wei-an Yuan Xiao-rong Chen Yun-fei Lu 2021Journal of Integrative Medicine2021,19,3:4
17Dual activated NIR-Ⅱ fluorescence and photoacoustic imaging-guided cancer chemo-radiotherapy using hybrid plasmonic-fluorescent assemblies显示文摘Multimodal imaging in the second near-infrared window(NIR-II)guided cancer therapy is a highly precise and efficient cancer theranostic strategy.However,it is still a challenge to develop activated NIR-II optical imaging and therapy agents.In this study,we develop a pH-responsive hybrid plasmonic-fluorescent vesicle by self-assembly of amphiphilic plasmonic nanogapped gold nanorod(AuNNR)and fluorescent down-conversion nanoparticles(DCNP)(AuNNR-DCNP Ve),showing remarkable and activated NIR-II fluorescence(FL)/NIR-II photoacoustic(PA)imaging performances.The hybrid vesicle also exhibited superior loading capacity of doxorubicin as a superior drug carrier and efficient radiosensitizer for X-ray-induced radiotherapy.Interestingly,the accumulated hybrid AuNNR-DCNP Ve in the tumor resulted in a recovery of NIR-II FL imaging signal and a variation in NIR-II PA imaging signal.Dual activated NIR-II PA and FL imaging of the hybrid vesicle could trace drug release and precisely guided cancer radiotherapy to ultimately reduce the side effects to healthy tissue.Tao Chen Lichao Su Xiaoguang Ge Wenmin Zhang Qingqing Li Xuan Zhang Jiamin Ye Lisen Lin Jibin Song Huanghao Yang 2020Nano Research2020,13,12:4

20Fuzzy-Immune PID Control for AMB Systems显示文摘In order to improve the dynamic performance of active magnetic bearing systems with highly nonlinear and naturally unstable dynamics, a new nonlinear fuzzy-immune proportional-integral-derivative (PID) controller is proposed by combining the immune feedback law with linear PID control. This controller consists of a PID controller and a basic immune proportional controller in cascaded connection, the nonlinear function of the immune proportional controller is realized by using fuzzy reasoning.
