1Human ESC-derived expandable hepatic organoids enable therapeutic liver repopulation and pathophysiological modeling of alcoholic liver injury显示文摘We report the generation of human ESC-derived,expandable hepatic organoids(hEHOs)using our newly established method with wholly defined(serum-free,feeder free)media.The hEHOs stably maintain phenotypic features of bipotential liver stem/progenitor cells that can differentiate into functional hepatocytes or cholangiocytes.The hEHOs can expand for 20 passages enabling large scale expansion to cell numbers requisite for industry or clinical programs.The cells from hEHOs display remarkable repopulation capacity in injured livers of FRG mice following transplantation,and they differentiate in vivo into mature hepatocytes.If implanted into the epididymal fat pads of immune-deficient mice,they do not generate non-hepatic lineages and have no tendency to form teratomas.We further develop a derivative model by incorporating human fetal liver mesenchymal cells(hFLMCs)into the hEHOs,referred to as hFLMC/hEHO,which can model alcoholic liver disease-associated pathophysiologic changes,including oxidative stress generation,steatosis,inflammatory mediators release and fibrosis,under ethanol treatment.Our work demonstrates that the hEHOs have considerable potential to be a novel,ex vivo pathophysiological model for studying alcoholic liver disease as well as a promising cellular source for treating human liver diseases.Shuyong Wang Xuan Wang Zuolong Tan Yuxin Su Juan Liu Mingyang Chang Fang Yan Jie Chen Tao Chen Chuanjiang Li Jie Hu Yunfang Wang 2019Cell Research2019,29,12:20
2Thermal evolution effects on the properties of converting Cs-polluted soil into pollucite-base glass-ceramics for radioactive cesium immobilization显示文摘The Fukushima nuclear accident in Japan on March 11,2011,produced large amounts of Cs-polluted soil which must be controlled to prevent the spread of hazardous Cs into the environment.In this paper,the effects of heat treatment on the structure and properties of Cs-containing glass-ceramics,as a simulated nuclear waste form,were systematically investigated.Cesium atoms are chemically bonded in the pollucite structure,and the amorphous phase further encapsulates the pollucite crystals in the glassceramics,thus providing an extra protective layer for the immobilized Cs.XRD analysis,Raman and FT-IR studies on the glass-ceramics synergistically indicated that the optimum crystallization temperature for pollucite is around 1000℃.The pollucite is predominantly the main crystalline phase with a narrow crystal size distribution between 0.5 and 2 mm.Standard leaching test results show that the leaching rate of Cs was very low(3.0×10^(-3) g/(m^(2)·d)).The study offers a practical method for immobilizing Cs in pollucite-base glass-ceramics.Moreover,the experimentally obtained data may provide some important references for converting Cs-polluted soil into pollucite-base glass-ceramics waste form.Yong Yang Xin Cao Lifen Shi Zhengyi Zhang Pingping Wang Jinwei Li Yangshan Sun Shuyong Chen Tianhe Wang Liyun Ma Shou Peng 2021Journal of Materiomics2021,7,6:2
3Stannous oxalate: An efficient catalyst for poly(trimethylene terephthalate) synthesis显示文摘A complete study on the catalytic activity of stannous oxalate for poly(trimethylene terephthalate) (PTT) synthesis via esterification method is carried out by comparison to the well known catalysts (tetrabutyl titanate (TBT), dibutyltin oxide (Bu2SnO), and stannous octoate (SOC)). Their catalytic activity in the esterification process is monitored by measuring the amount of water generated, while intrinsic viscosity (IV) and content of terminal carboxyl groups (CTCG) are used as the index in the polycondensation process. Stannous oxalate shows higher activity than the other catalysts. Decrease in reaction time and improvements in PTT property are observed. 4A new approach of proration-injection allocation for water-flooding mature oilfields显示文摘This paper presents a new method of injection-production allocation estimation for water-flooding mature oilfields.The suggested approach is based on logistic growth rate functions and several type-curve matching methods.Using the relationship between these equations,oil production and water injection rate as well as injection-production ratio can be easily forecasted.The calculation procedure developed and outlined in this paper requires very few production data and is easily implemented.Furthermore,an oilfield case has been analyzed.The synthetic and field cases validate the calculation procedure,so it can be accurately used in forecasting production data,and it is important to optimize the whole injection-production system.Shuyong Hu Yongkai Li Ziwei Wang Guoqiang Hu 2015Petroleum2015,1,1:2

8Preemptive analgesic effects of low-dose ketamine on growth-associated protein expression in dorsal root ganglion of chronic constriction injury model rats显示文摘BACKGROUND: Ketamine is a noncompetitive N-methyl-D-aspartate (NMDA) receptor antagonists and plays an important role in the treatment of pain. OBJECTIVE: To analyze the preemptive analgesic effects of different doses of ketamine on growth-associated protein-43 (GAP-43) expression in dorsal root ganglion in a rat model of chronic sciatic nerve constricted injury, and to study the differences between high-dose and low-dose ketamine DESIGN: Randomized controlled animal study. SETTING: Medical College of Shantou University. MATERIALS: Thirty-five adult male Sprague Dawley rats were provided by the Experimental Animal Center of Guangzhou University of Traditional Chinese Medicine. Ketamine hydrochloride injection was provided by Hengrui Pharmaceutical Co., Ltd., Jiangsu. METHODS: This study was performed at the Immunological Laboratory, Medical College of Shantou University from September to December 2006. Model of chronic sciatic nerve constricted injury: after anesthesia, the right sciatic nerve was exposed and ligated 1-cm distal to the ischiadic tuberosity with a No. 3-0 cat gut suture. Grouping and intervention: 35 rats were randomly divided into 4 groups: normal control group (n = 5), chronic constriction injury (CCI) group (n = 10), low-dose ketamine group (n = 10), and high-dose ketamine group (n = 10). Rats in the normal control group did not undergo any surgery or drug intervention. Rats in the CCI group received intraperitoneal injection of saline (1 mL), and their sciatic nerves were ligated after 10 minutes. Rats in the low-dose ketamine group underwent intraperitoneal injection of ketamine (25 mg/kg) 10 minutes prior to ligation of sciatic nerve; while, rats in the high-dose ketamine group were given intraperitoneal injection of ketamine (50 mg/kg) 10 minutes prior to ligation of sciatic nerve. On the third and the seventh days after surgery, dorsal root ganglion were resected from the sciatic nerve and cut into sections. MAIN OUTCOME MEASURES: GAP-43 expression in dorsal root ganglion was detected by immunohistochemistry and image analysis system, as well as semi-quantitative analysis. RESULTS: Thirty-five Sprague Dawley rats were included in the final analysis. Qualitative analysis: GAP-43 expression in the CCI group was higher than in the normal control group. Quantitative analysis: after three post-operative days, GAP-43 expression in the CCI group was significantly higher than in the normal control group (t = 22.919, 7.319, P < 0.05). GAP-43 expression in the low-dose and high-dose ketamine group was significantly lower than in the CCI group (t = 11.166, 26.474, P < 0.05). After seven postoperative days, GAP-43 expression in the low-dose and high-dose ketamine groups was significantly lower than in the CCI group (t = 2.382, 5.016, P < 0.05). 10Study on Soil Moisture Distributing Characteristics of Artificial Forest in Mountainous Region显示文摘Physical characteristics of soil structure and soil moisture of five kinds of artificial forests were researched in Shengquan Temple Forestry Farm, in the southern mountainous area of Jinan City, in order to offer scientific management and nurture of forest vegetation in mountainous area of North China. Results indicated that: (1) In these artificial forests researched, some indexes expressing soil pedonydrological physical character, such as soil density, porosity, infiltration rate, were much better than those in bare land, and these indexes showed this order: mixed forest > pure forest, Kalopanax pictus > Koelreuteria paniculata > Pistacia chinensis. (2) In dry and rainy seasons, with the increasing of soil depth, soil water content decreased in most parts of forest, and their extent of change was less than those in bare land. (3) With the increasing of soil depth, soil saturated water content decreased in all kinds of forests. Water storing capacity of surface soil layer was the best in the mixed forest, while differences were little in other forest. Water storing and adjusting capacity was good in the mixed forest, while water offering capacity had little discrepancy in all forests. Conclusions show that judging from water conservation and water use efficiency, mixed forest is better than pure forest, and in pure forest K. pictus is better than K. paniculata. Therefore, it is proposed that in the future, mixed forest should be fostered and managed firstly, the species being made up of K. pictus and P. chinensis. In the pure forest, K. pictus is much better than others. Nowadays K. paniculata, and P. chinensis, especially their soil surface vegetation and litter, should be protected. As to poor forest, closing hill for afforestation should be adopted. 12Fractal features and infiltration characteristics of the soil from different land uses in a small watershed in a rocky,mountainous area in Shandong Province显示文摘We studied infiltration and fractal mechanisms on sloping farmlands in a small watershed in Shandong Province in the rocky mountain areas of northern China.We studied soil fraction and soil water retention curves,and developed a soil infiltration model to analyze its quantitative relationship with soil particle size and pore dimensions under four types of land use,including sloping farmland,abandoned land,natural forest,and forest plantation(Malus pumila,Crataegus pinnatifida).Soil stability infiltration rate ranked as sloping farmland>abandoned land>natural forest>forest plantation.The sequence of soil particle size and pore dimension ranked as natural forest>forest plantation>abandoned land>sloping farmland.There were significant positive correlations between soil particle size and pore dimension,and both were positively correlated with the percent volume of silt and clay.They were negatively correlated with soil infiltration rate.The Horton model and the power function model were more suitable for simulating soil infiltration and the infiltration rate.We concluded that the soil in this area displayed typical fragments of rocky mountainous regions due to the loss of silt and clay caused by cultivation on sloping farmland.The uniformity and fractal dimensions of soil particle sizes and pore distribution decreased,thereby enhancing soil infiltration capacity and decreasing soil water retention capacity.Lei Wang Rui Yang Shuyong Zhang Guangcan Zhang Xia Liu 2020Journal of Forestry Research2020,31,3:1

17Critical effects on the photosynthetic efficiency and stem sap flow of poplar in the Yellow River Delta in response to soil water显示文摘To explore the critical relationships of photosynthetic efficiency and stem sap flow to soil moisture,two-year-old poplar saplings were selected and a packaged stem sap flow gauge,based on the stem-heat balance method,and a CIRAS-2 portable photosynthesis system were used.The results show that photosynthetic rates(P_(n)),transpiration rates(T_(r)),instantaneous water use efficiency(WUE)and the stem sap flow increased initially and then decreased with decreasing soil water,but their critical values were different.The turning point of relative soil water content(W_(r))from stomatal limitation to nonstomatal limitation of P_(n)was 42%,and the water compensation point of P_(n)was 13%.Water saturation points of P_(n)and T_(r)were 64%and 56%,respectively,and the WUE was 71%.With increasing soil water,the apparent quantum yield(AQY),light saturation point(LSP)and maximum net photosynthetic rate(P_(n)max)increased first and then decreased,while the light compensation point(LCP)decreased first and then increased.When W_(r)was 64%,LCP reached a lower value of 30.7µmol m^(-2)s^(-1),and AQY a higher value of 0.044,indicating that poplar had a strong ability to utilize weak light.When W_(r)was 74%,LSP reached its highest point at 1138.3µmol·m^(-2)s^(-1),indicating that poplar had the widest light ecological amplitude and the highest light utilization efficiency.Stem sap flow and daily sap flow reached the highest value(1679.7 g d^(-1))at W_(r)values of 56%and 64%,respectively,and then declined with increasing or decreasing W_(r),indicating that soil moisture significantly affected the transpiration water-consumption of poplar.Soil water was divided into six threshold grades by critical values to maintain photosynthetic efficiency at different levels,and a W_(r)of 64-71%was classified to be at the level of high productivity and high efficiency.In this range,poplar had high photosynthetic capacity and efficient physiological characteristics for water consumption.The saplings had characteristics of water tolerance and were not drought resistant.Full attention should be given to the soil water environment in the Yellow River Delta when planting Populus.Changxi Wang Huanyong Liu Jiangbao Xia Xianshuang Xing Shuyong Zhang 2021Journal of Forestry Research2021,32,6:0
18Targeting LncRNA LLNLR-299G3.1 with antisense oligonucleotide inhibits malignancy of esophageal squamous cell carcinoma cells in vitro and in vivo显示文摘Accumulating evidence has indicated that long non-coding RNAs(lncRNAs)play critical roles in the development and progression of cancers,including esophageal squamous cell carcinoma(ESCC).However,the mechanisms of lncRNAs in ESCC are still incompletely understood and therapeutic attempts for in vivo targeting cancer-associated lncRNA remain a challenge.By RNA-sequencing analysis,we identified that LLNLR-299G3.1 was a novel ESCC-associated lncRNA.LLNLR-299G3.1 was up-regulated in ESCC tissues and cells and promoted ESCC cell proliferation and invasion.Silencing of LLNLR-299G3.1 with ASO(antisense oligonucleotide)resulted in opposite effects.Mechanistically,LLNLR-299G3.1 bound to cancerassociated RNA binding proteins and regulated the expression of cancer-related genes,including OSM,TNFRSF4,HRH3,and SSTR3.ChIRP-seq(chromatin isolation by RNA purification and sequencing)revealed that these genes contained enriched chromatin binding sites for LLNLR-299G3.1.Rescue experiments confirmed that the effects of LLNLR-299G3.1 on ESCC cell proliferation were dependent on interaction with HRH3 and TNFRSF4.Therapeutically,intravenous delivery of placental chondroitin sulfate A binding peptide-coated nanoparticles containing antisense oligonucleotide(pICSA-BP-ANPs)strongly inhibited ESCC tumor growth and significantly improved animal survival in vivo.Overall,our results suggest that LLNLR-299G3.1 promotes ESCC malignancy through regulating gene-chromatin interactions and targeting ESCC by pICSA-BP-ANPs may be an effective strategy for the treatment of lncRNA-associated ESCC.LI TIAN YONGYI HUANG BAOZHEN ZHANG YI SONG LIN YANG QIANQIAN CHEN ZHENG WANG YILING WANG QIHAN HE WENHAN YANG SHUYONG YU TIANYU LU ZICHEN LIU KAIPING GAO XIUJUN FAN JIAN SONG RIHONG ZHAI 2023Oncology Research2023,31,4:0
