| 2 | Full-thickness tissue engineered skin constructed with autogenic bone marrow mesenchymal stem cells显示文摘To explore the feasibility of repairing clinical cutaneous deficiency, autogenic bone marrow mesen-chymal stem cells (BMSCs) were isolated and differentiated into epidermal cells and fibroblasts in vitro supplemented with different inducing factors and biomaterials to construct functional tissue- engineered skin. The results showed that after 72 h induction, BMSCs displayed morphologic changes such as typical epidermal cell arrangement, from spindle shape to round or oval; tonofibrils, melano-somes and keratohyaline granules were observed under a transmission electronic microscope. The differentiated cells expressed epidermal stem cell surface marker CK19 (59.66% ± 4.2%) and epidermal cells differentiation marker CK10. In addition, the induced epidermal cells acquired the anti-radiation capacity featured by lowered apoptosis following exposure to UVB. On the other hand, the collagen microfibrils deposition was noticed under a transmission electronic microscope after differentiating into dermis fibroblasts; RT-PCR identified collagen type I mRNA expression in differentiated cells; radioimmunoassay detected the secretion of interleukin-6 (IL-6) and interleukin-8 (IL-8) (up to 115.06 pg/mL and 0.84 ng/mL, respectively). Further in vivo implanting BMSCs with scaffold material short-ened skin wound repair significantly. | 3 | Authentic compound-free strategy for simultaneous determination of primary coumarins in Peucedani Radix using offline high performance liquid chromatography–nuclear magnetic resonance spectroscopy–tandem mass spectrometry显示文摘Herein, a strategy is proposed for the simultaneous determination of primary coumarins in Peucedani Radix(Chinese name: Qianhu). The methodology consists of three consecutive steps: 1) Semi-preparative LC in combination with a home-made automated fraction collection module to fragment the universal metabolome standard into ten fractions(Frs. I–X); 2) LC–accurate MS/MS and quantitative1 H NMR spectroscopy conducted in parallel to acquire the qualitative and quantitative data of each fraction; 3) Robust identification and quantification of components by use of LC coupled to multiple reaction monitoring. In this final step, the most significant fractions(Frs. III–X) were pooled to serve as the pseudo-mixed standard solution. Meticulous online parameter optimization was performed to obtain the optimal parameters, including ion transitions and collision energies. Concerns were particularly paid onto pursuing the parameters being capable of monitoring regiospecific isomers, notably praeruptorin E vs. 3′-isovaleryl-4′-angeloylkhellactone. The quantitative performance of the method was validated according to diverse assays. Eleven primary coumarins(1–11) were unambiguously identified and absolutely quantified, even though no external reference compound was used. | 4 | In vitro differentiation of human adipose-derived mesenchymal stem cells into endothelial-like cells显示文摘The neovascularization of ischemic tis- sue is a crucial initial step for the functional rehabili- tation and wound healing. However, the short of seed cell candidate for the foundation of vascular network is still a big issue. Human adipose tissue derived mesenchymal stem cells (hADSCs), which possess multilineage potential, are capable of adipogenic, osteogenic, and chondrogenic differentiation. We examined whether this kind of stem cells could dif- ferentiate into endothelial-like cells and participate in blood vessel formation, and whether they could be used as an ideal cell source for therapeutic angio- genesis in ischemic diseases or vascularization of tissue constructs. The results showed that hADSCs, grown under appropriately induced conditions, dis- played characteristics similar to those of vessel en- dothelium. The differentiated cells expressed endo- thelial cell markers CD34 and vWF, and had high metabolism of acetylated low-density lipoprotein and prostacyclin. In addition, the induced cells were able to form tube-like structures when cultured on matrigel. Our data indicated that induced hADSCs could ex- hibit characteristics of endothelial cells. | 7 | Proliferation and differentiation into endothelial cells of human bone marrow mesenchymal stem cells (MSCs) on poly DL-lactic-co-glycolic acid (PLGA) films显示文摘The functional realization is the most important problem in vascular tissue engineering. The small-caliber blood vessel substitutes are prone to thrombi, which results in functional loss of blood vessels. However, this is probably due to the imper- fection of endothelial layer in the substitutes. In this study, MSCs were seeded on a series of porous PLGA films with various porosity and pore size made by sodium chloride (NaCl) particulate leaching, and cell proliferation on each film was inspected. The film made of the 75% (w/w) particulate proportion and 30―50 μm pore size maximized the proliferation rate and was chosen as the scaffolds for the differentia- tion of MSCs into endothelial cells. The induced cells expressed endothelial cells specific Flk-1, Ⅷ factor and CD34, possessed endothelial cells specific Weible-palade (W-P) body, and had the abilities of ingesting low density lipoprotein and secreting prostacyclin (PGI2). | 8 | Low temperature preparation of flower-like BiOCl film and its photocatalytic activity显示文摘The BiOCl thin film with flower-like sphere structure was prepared at a low temperature by the alcoholysis-coating method using BiCl 3 as precursor. The obtained thin film was characterized by X-ray diffraction (XRD), scanning electron microscopy (SEM), electronic energy spectrum (EDS) and ultraviolet-visible diffuse reflectance spectroscopy (UV-vis DRS). And the results showed that the obtained BiOCl film without calcination was composed of flower-like sphere structure with tetragonal phase and had a good absorption for ultraviolet. The photocatalytic activity of BiOCl thin film was also evaluated by the degradation of methyl orange in water under UV light irradiation. The degradation experimental results confirmed that the film prepared at low temperature possessed a high photocatalytic activity and could achieve 97% degradation to 10 mg/L methyl orange solution after 150 min UV light irradiation. The stability of the obtained BiOCl thin film was also good and its photocatalytic activity still remained an above 94% removal of methyl orange after being used four times. The citric acid was served as a chelating and reductive agent for the preparation of Ag-decorated Bi OBr photocatalysts(named as Ag/Bi OBr-2). The as-synthesized samples were characterized by X-ray diffraction(XRD), X-ray photoelectron spectroscopy(XPS), scanning electron microscopy(SEM), transmission electron microscopy(TEM), and UV-Vis diffuse reflection spectroscopy(DRS). The Ag/Bi OBr-2 photocatalyst exhibited excellent and stable photocatalytic activities on MO and phenol degradation under simulated sunlight irradiation. The enhanced photocatalytic activity could be ascribed to the smaller size, rough surface, and the surface plasma resonance(SPR) effect of Ag. Also, the Schottky junction, between the surface of the Bi OBr and silver nanoparticles, accelerated the efficient transfer and separation of photoinduced electron-hole pairs and promoted the photocatalytic performance. The active species tests indicated that the superoxide radical(·O-2) was responsible for the enhanced photocatalytic performance of Ag/Bi OBr-2. | 20 | The 9L^(LUC)/Wistar rat glioma model is not suitable for immunotherapy显示文摘The availability of a well-characterized animal brain tumor model will play an important role in identifying treatments for human brain tumors. Wistar rats bearing 9L glioma cells can develop solid, well-circumcised tumors, and may be a useful animal model for the evaluation of various therapeutic approaches for gliosarcomas. In this study, the 9L/Wistar rat glioma model was produced by intracerebral implantation of 9LLUC glioma cells syngenic to Fischer 344 (F344) rats. Bioluminescence imaging showed that tumors progressively grew from day 7 to day 21 in 9LLUC/F344 rats, and tumor regression was found in some 9LLUC/Wistar rats. Hematoxylin-eosin staining verified that intracranial tumors were gliomas. Immunohistochemistry results demonstrated that no CD4- and CD8-positive cells were found in the syngeneic 9LLUC/F344 model. However, many infiltrating CD4- and CD8-positive cells were observed within the tumors of the 9LLUC/Wistar model.
