1Artificial intelligence in drug design显示文摘Thanks to the fast improvement of the computing power and the rapid development of the computational chemistry and biology,the computer-aided drug design techniques have been successfully applied in almost every stage of the drug discovery and development pipeline to speed up the process of research and reduce the cost and risk related to preclinical and clinical trials.Owing to the development of machine learning theory and the accumulation of pharmacological data, the artificial intelligence(AI) technology, as a powerful data mining tool, has cut a figure in various fields of the drug design, such as virtual screening,activity scoring, quantitative structure-activity relationship(QSAR) analysis, de novo drug design, and in silico evaluation of absorption, distribution, metabolism, excretion and toxicity(ADME/T) properties. Although it is still challenging to provide a physical explanation of the AI-based models, it indeed has been acting as a great power to help manipulating the drug discovery through the versatile frameworks. 2Changing the strategy and culture of stroke awareness education in China: implementing Stroke 1-2-0显示文摘This project implemented the Stroke1-2-0 stroke awareness programme across China and investigated its impact over a 2-year period.We initiated the Stroke1-2-0 educational campaign and Stroke1-2-0 special task forces(STF)across the nation.Massive media coverage,community-based educational sessions with videos and other related materials and induction of Stroke1-2-0 STF were the major means of promotion.We delivered a survey at the end of 2016 and 2018 to evaluate the impact of our effort.A total of 3066 participants responded to the first survey in 2016,and 15207 participants responded in 2018 across China.The acceptance rate for Stroke1-2-0 versus FAST(an English-language stroke awareness tool)was 50.2%versus 19.1%in 2016,and changed significantly to 82.2%versus 8.0%in 2018(p<0.001).Stroke1-2-0 was well accepted by all ages and by people with different academic qualifications.Only 6.5%of survey respondents were aware that there was a therapeutic window for thrombolytic therapy in 2016,but this awareness increased significantly to 32.8%in 2018.Only 12.6%of people in 2016 indicated that they would send patients with stroke to the nearest hospital capable of performing thrombolytic therapy,but there was a nearly threefold increase(52.5%)in this number by 2018.More than 1000 major hospitals joined the Stroke1-2-0 STF,and more than 20000‘stroke warriors’have joined our stroke awareness improvement effort so far.Stroke1-2-0 stroke awareness programme is well-implemented and accepted,and is generating profound improvement in stroke awareness in China.Jing Zhao Xutong Li Xiaochuan Liu Yuming Xu Jihong Xu Anding Xu Yongjun Wang Renyu Liu 2020Stroke & Vascular Neurology2020,5,4:6
3A Post-domestication Mutation, Dt2, Triggers Systemic Modification of Divergent and Convergent Pathways Modulating Multiple Agronomic Traits in Soybean显示文摘The semi-determinate stem growth habit in leguminous crops,similar to the“green revolution”semi-dwarf trait in cereals,is a key plant architecture trait that affects several other traits determining grain yield.In soybean semi-determinacy is modulated by a post-domestication gain-of-function mutation in the gene,Dt2,which encodes an MADS-box transcription factor.However,its role in systemic modification of stem growth and other traits is unknown.In this study,we show that Dt2 functions not only as a direct repressor of Dt1,which prevents terminal flowering,but also as a direct activator of putative floral integrator/identity genes including GmSOC1,GmAP1,and GmFUL,which likely promote flowering.We also demonstrate that Dt2 functions as a direct repressor of the putative drought-responsive transcription factor gene GmDREB1D,and as a direct activator of GmSPCH and GmGRP7,which are potentially associated with asymmetric division of young epidermal cells and stomatal opening,respectively,and may affect the plant's water-use efficiency(WUE).Intriguingly,Dt2 was found to be a direct activator or repressor of the precursors of eight microRNAs targeting genes potentially associated with meristem maintenance,flowering time,stomatal density,WUE,and/or stress responses.This study thus reveals the molecular basis of pleiotropy associated with plant productivity,adaptability,and environmental resilience.Dajian Zhang Xutong Wang Shuo Li Chaofan Wang Michael JGosney Michael VMickelbart Jianxin Ma 2019Molecular Plant2019,12,10:4
4Effects of Rat Cytomegalovirus on the Nervous System of the Early Rat Embryo显示文摘The purpose of the study was to investigate the impact of rat cytomegalovirus(RCMV) infection on the development of the nervous system in rat embryos,and to evaluate the involvement of Wnt signaling pathway key molecules and the downstream gene neurogenin 1(Ngn1) in RCMV infected neural stem cells(NSCs).Infection and control groups were established,each containing 20 pregnant Wistar rats.Rats in the infection group were inoculated with RCMV by intraperitoneal injection on the first day of pregnancy.Rat E20 embryos were taken to evaluate the teratogenic rate.NSCs were isolated from E13 embryos,and maintained in vitro.We found:1) Poor fetal development was found in the infection group with low survival and high malformation rates.2) The proliferation and differentiation of NSCs were affected.In the infection group,NSCs proliferated more slowly and had a lower neurosphere formation rate than the control.The differentiation ratio from NSCs to neurons and glial cells was significantly different from that of the control,showed by immunofluorescence staining.3) Ngn1 mRNA expression and the nuclear β-catenin protein level were significantly lower than the control on day 2 when NSCs differentiated.4) The Morris water maze test was performed on 4-week pups,and the infected rats were found worse in learning and memory ability.In a summary,RCMV infection caused abnormalities in the rat embryonic nervous system,significantly inhibited NSC proliferation and differentiation,and inhibited the expression of key molecules in the Wnt/β-catenin signaling pathway so as to affect NSCs differentiation.This may be an important mechanism by which RCMV causes embryonic nervous system abnormalities.Xiuning Sun YingJun Guan Fengjie Li Xutong Li Xiaowen Wang Zhiyu Guan Kai Sheng Li Yu Zhijun Liu 2012Virologica Sinica2012,27,4:4

9Drug target inference by mining transcriptional data using a novel graph convolutional network framework显示文摘A fundamental challenge that arises in biomedicine is the need to characterize compounds in a relevant cellular context in order to reveal potential on-target or offtarget effects.Recently,the fast accumulation of gene transcriptional profiling data provides us an unprecedented opportunity to explore the protein targets of chemical compounds from the perspective of cell transcriptomics and RNA biology.Here,we propose a novel Siamese spectral-based graph convolutional network(SSGCN)model for inferring the protein targets of chemical compounds from gene transcriptional profiles.Although the gene signature of a compound perturbation only provides indirect clues of the interacting targets,and the biological networks under different experiment conditions further complicate the situation,the SSGCN model was successfully trained to learn from known compound-target pairs by uncovering the hidden correlations between compound perturbation profiles and gene knockdown profiles.On a benchmark set and a large time-split validation dataset,the model achieved higher target inference accuracy as compared to previous methods such as Connectivity Map.Further experimental validations of prediction results highlight the practical usefulness of SSGCN in either inferring the interacting targets of compound,or reversely,in finding novel inhibitors of a given target of interest.Feisheng Zhong Xiaolong Wu Ruirui Yang Xutong Li Dingyan Wang Zunyun Fu Xiaohong Liu XiaoZhe Wan Tianbiao Yang Zisheng Fan Yinghui Zhang Xiaomin Luo Kaixian Chen Sulin Zhang Hualiang Jiang Mingyue Zheng 2022Protein & Cell2022,13,4:1
10Genomic mosaicism due to homoeologous exchange generates extensive phenotypic diversity in nascent allopolyploids显示文摘Allopolyploidy is an important process in plant speciation,yet newly formed allopolyploid species typically suffer from extreme genetic bottlenecks.One escape from this impasse might be homoeologous meiotic pairing,during which homoeologous exchanges(HEs)generate phenotypically variable progeny.However,the immediate genome-wide patterns and resulting phenotypic diversity generated by HEs remain largely unknown.Here,we analyzed the genome composition of 202 phenotyped euploid segmental allopolyploid individuals from the fourth selfed generation following chromosomal doubling of reciprocal F1 hybrids of crosses between rice subspecies,using whole-genome sequencing.We describe rampant occurrence of HEs that,by overcoming incompatibility or conferring superiority of hetero-cytonuclear interactions,generate extensive and individualized genomic mosaicism across the analyzed tetraploids.We show that the resulting homoeolog copy number alteration in tetraploids affects known-function genes and their complex genetic interactions,in the process creating extraordinary phenotypic diversity at the population level following a single initial hybridization.Our results illuminate the immediate genomic landscapes possible in a tetraploid genomic environment,and underscore HE as an important mechanism that fuels rapid phenotypic diversification accompanying the initial stages of allopolyploid evolution.Ying Wu Fan Lin Yao Zhou Jie Wang Shuai Sun Bin Wang Zhibin Zhang Guo Li Xiuyun Lin Xutong Wang Yue Sun Qianli Dong Chunming Xu Lei Gong Jonathan F.Wendel Zhiwu Zhang Bao Liu 2021National Science Review2021,8,5:1
11Transcriptomic analysis of interstockinduced dwarfism in Sweet Persimmon(Diospyros kaki Thunb.)显示文摘Growth monitoring indicated that the height of‘Kanshu’plants with‘Nantong-xiaofangshi’as an interstock was significantly shorter than that of‘Kanshu’plants with no interstock.A transcriptome analysis of the two graft combinations(‘Kanshu’/Diospyros lotus and‘Kanshu’/‘Nantong-xiaofangshi’/Diospyros lotus)was conducted to explore the dwarfing genes related to the use of the‘Nantong-xiaofangshi’interstock.Hormone levels and water conductance were also measured in these two graft combinations.The results indicated that the levels of both IAA and GA were lower in‘Kanshu’that had been grafted onto the‘Nantong-xiaofangshi’interstock than in‘Kanshu’with no interstock;additionally,the water conductance was lower in grafts with interstocks than in grafts without interstocks.The expression of AUX/IAA and auxin-responsive GH3 genes was enhanced in scions grafted on the interstock and was negatively correlated with the IAA content and growth of scions.The expression of GA2ox,DELLA,and SPINDLY genes were also upregulated and associated with a decrease in the level of GA in scions grafted on the interstock.Since one of the GA2ox unigenes was annotated as DkGA2ox1 in Diospyros kaki,but was not functionally validated,a functional analysis was conducted in transgenic tobacco.Overexpression of DkGA2ox1 in transgenic plants resulted in a dwarf phenotype that could be recovered by the exogenous application of GA3.We conclude that the‘Nantong-xiaofangshi’interstock affects the water conductance and expression of genes related to the metabolism and transduction of IAA and GA in the grafted scion and thus regulates phytohormone levels,producing dwarfing.Yanying Shen Weibing Zhuang Xutong Tu Zhihong Gao Aisheng Xiong Xinyi Yu Xuehan Li Feihong Li Shenchun Qu 2019Horticulture Research2019,6,1:1
12Experimental Investigations on NO_(x) Emission and Combustion Dynamics in an Axial Fuel Staging Combustor显示文摘Axial Fuel Staging(AFS)technology is an advanced low-emission combustion method in modem gas turbine,which divides the combustor into two axially arranged combustion zones.For revealing the characteristics of axial staged combustion,an industrial-grade combustor was designed and built.The distribution of temperature and velocity field in the combustor was presented with numerical simulation.And an Atmospheric Combustor Test Rig for axial staged combustion was built.The flow resistance characteristics of the combustor were measured at first.Then the effects of the equivalent ratio and the preheating temperature on the pollutant emission and combustion instability were investigated.The results show that the total pressure recovery coefficient in cold state is always above 98%;starting the secondary combustion at low load can reduce NO emissions by 50%,and can suppress the combustion oscillation amplitude of the combustor.At the design point with φ=0.62 and preheating temperature=400°C,NO emission and CO emission are 15.68 and 4.22 mg/m^(3)(@15%O_(2)).LI Yuze JIA Yuliang JIN Ming ZHU Xutong GE Bing MAO Ronghai REN Lilei CHEN Mingmin JIAO Guangyun 2022Journal of Thermal Science2022,31,1:0

15Cognitive Translation of Paintings of Lijiang Painting School Under the Guidance of Rewriting Theory显示文摘It is an important task to communicate Chinese culture with other countries in the more conflicting global world.Lijiang Painting School plays a very important part in Chinese art,and it greatly influences the painters of various periods in China.In this context,some representative paintings of Lijiang Painting School in different periods were collected in this paper as the research objects.The rewriting theory was applied in the study because Chinese readers and English readers have different cultural backgrounds and diverse thinking.Based on the rewriting theory,these representative paintings were translated and the explained in English.In the study,it is found that as an intersemiotic translation,the translation of the representative paintings of Lijiang Painting School should be focused on those target readers who can’t comprehend directly from the paintings.In addition,the artistic integrity should be considered first before details in paintings.Therefore,the overall perception of target readers gaining from the translations should be paid more attention to rather than metaphrase.The translation under the guidance of rewriting theory will be helpful for the target readers to understand the connotation of source culture with some cultural images of target readers.QIN Kai LI Xutong 2020Journal of Literature and Art Studies2020,10,7:0
