1Collagen-binding basic fibroblast growth factor improves functional remodeling of scarred endometrium in uterine infertile women: a pilot study显示文摘Intrauterine adhesion(IUA) is a common cause of uterine infertility and one of the most severe clinical features is endometrial fibrosis namely endometrial scarring for which there are few cures currently. Blocked angiogenesis is the main pathological change in the scarred endometrium. The fibroblast growth factor 2(b FGF), a member of FGF family, is usually applied to promote healing of refractory ulcer and contributes to angiogenesis of tissues. In this study, the sustained-release system of b FGF100 μg was administrated around scarred endometrium guiding by ultrasound every 4 weeks in 18 patients(2–4 times). Results showed that after treatment, the menstrual blood volume, endometrial thickness and the scarred endometrial area were improved.Histological study showed blood vessel density increased obviously. Three patients(3/18) achieved pregnancy over 20 gestational weeks. 3Neurogranin as an important regulator in swimming training to improve the spatial memory dysfunction of mice with chronic cerebral hypoperfusion显示文摘Background:Vascular cognitive impairment caused by chronic cerebral hypoperfusion(CCH)has become a hot issue worldwide.Aerobic exercise positively contributes to the preservation or restoration of cognitive abilities;however,the specific mechanism has remained inconclusive.And recent studies found that neurogranin(Ng)is a potential biomarker for cognitive impairment.This study aims to investigate the underlying role of Ng in swimming training to improve cognitive impairment.Methods:To test this hypothesis,the clustered regularly interspaced short palindromic repeats(CRISPR)-associated protein 9(Cas9)system was utilized to construct a strain of Ng conditional knockout(Ng cKO)mice,and bilateral common carotid artery stenosis(BCAS)surgery was performed to prepare the model.In Experiment 1,2-month-old male and female transgenic mice were divided into a control group(wild-type littermate,n=9)and a Ng cKO group(n=9).Then,2-month-old male and female C57BL/6 mice were divided into a sham group(C57BL/6,n=12)and a BCAS group(n=12).In Experiment 2,2-month-old male and female mice were divided into a sham group(wild-type littermate,n=12),BCAS group(n=12),swim group(n=12),BCAS+Ng cKO group(n=12),and swim+Ng cKO group(n=12).Then,7 days after BCAS,mice were given swimming training for 5 weeks(1 week for adaptation and 4 weeks for training,5 days a week,60 min a day).After intervention,laser speckle was used to detect cerebral blood perfusion in the mice,and the T maze and Morris water maze were adopted to test their spatial memory.Furthermore,electrophysiology and Western blotting were conducted to record long-term potential and observe the expressions of Ca^(2+)pathway-related proteins,respectively.Immunohistochemistry was applied to analyze the expression of relevant markers in neuronal damage,inflammation,and white matter injury.Results:The figures showed that spatial memory impairment was detected in Ng cKO mice,and a sharp decline of cerebral blood flow and an impairment of progressive spatial memory were observed in BCAS mice.Regular swimming training improved the spatial memory impairment of BCAS mice.This was achieved by preventing long-term potential damage and reversing the decline of Ca^(2+)signal transduction pathway-related proteins.At the same time,the results suggested that swimming also led to improvements in neuronal death,inflammation,and white matter injury induced by CCH.Further study adopted the use of Ng cKO transgenic mice,and the results indicated that the positive effects of swimming training on cognitive impairments,synaptic plasticity,and related pathological changes caused by CCH could be abolished by the knockout of Ng.Conclusion:Swimming training can mediate the expression of Ng to enhance hippocampal synaptic plasticity and improve related pathological changes induced by CCH,thereby ameliorating the spatial memory impairment of vascular cognitive impairment.Huawei Lin Jiayong Zhang Yaling Dai Huanhuan Liu Xiaojun He Lewen Chen Jing Tao Chaohui Li Weilin Liu 2023Journal of Sport and Health Science2023,12,1:1

5Research on the Activity Characteristics of Small Earthquake Swarms in the Muli Area of Sichuan Province显示文摘In this article,we relocated the seismic source location of the earthquakes in the Muli area of Sichuan,inverted the focal mechanism of the larger earthquakes and analyzed the relationship between the water level of the Jinping reservoir and the frequency of the earthquake swarm. The results show that:( 1) The epicenters of the relocated small earthquake swarms are distributed in a seismic zone,and the earthquake focal depths were in the range of 0- 12 km.( 2) By analyzing the earthquake swarm spatial distribution,we found that the swarms were generated by one branch fault on the west of Xiaojinhe fault.( 3) The focal mechanism of the three earthquakes with magnitude greater than 4. 0 is significantly different,with the shallow source thrust events affected by vertical stress,and the strike-slip events are related to regional stress tectonic activity.Cai Yichuan Dai Shigui Wei Yaling 2016Earthquake Research in China2016,30,2:0
6Long-term Outcomes after Second-Generation Cryoballoon Ablation of Atrial Fibrillation and Analysis of Risk Factors Related to Recurrence显示文摘Objective:This study is aimed to analyze the long-term safety and effectiveness of second-generation cryoballoon(CB2)ablation in the treatment of atrial fibrillation(AF).Methods:Data from 760 consecutive patients in the Department of Cardiology,General Hospital of Northern Theater Command from August 2016 to December 2018 with drug-refractory symptomatic AF undergoing pulmonary vein isolation(PVI)using CB2 were assessed.Procedure-related safety and freedom from AF and atrial flutter/atria tachycardia through 3 years were determined.The risk factors related to atrial tachyarrhythmia recurrence were analyzed.Results:Acute PVI was achieved in 100%of the 760 patients.Radiofrequency application for additional focal ablation was needed in 11(1.4%)patients and for 14 pulmonary veins(0.5%,14/(760×4))to achieve PVI.A total of 748 patients,including 539 with paroxysmal AF(PAF)and 209 with persistent AF(SAF)completed the follow-up,and only 12(1.6%)patients were lost.The mean follow-up duration was(19±8)months.The rate of major complications was 0.9%,including 0.8%of right phrenic nerve injury,which resolved before discharge.Freedom from all tachyarrhythmias was achieved in 75.0%,69.4%,and 63.2%of patients with PAF,respectively,at 12-,24-,and 36-month follow-up,and in 75.1%,67.4%,and 60.9%for SAF,with no significant differences between the PAF and SAF groups.AF course and the rate of body weight gain were independent risk factors for recurrence at 12 months after ablation(P=0.001 and P=0.009,respectively).Conclusion:PVI using CB2 has a high acute success rate and good safety in the treatment of PAF and SAF.Long course of AF and weight gain after ablation were independent risk factors for recurrence.Yufan Dai Chenyuan Wang Zulu Wang Ming Liang Guitang Yang Zhiqing Jin Jian Ding Ping Zhang Yaling Han 2022Cardiology Discovery2022,2,3:0
7Inside-out assembly of viral antigens for the enhanced vaccination显示文摘Current attempts in vaccine delivery systems concentrate on replicating the natural dissemination of live pathogens,but neglect that pathogens evolve to evade the immune system rather than to provoke it.In the case of enveloped RNA viruses,it is the natural dissemination of nucleocapsid protein(NP,core antigen)and surface antigen that delays NP exposure to immune surveillance.Here,we report a multi-layered aluminum hydroxide-stabilized emulsion(MASE)to dictate the delivery sequence of the antigens.In this manner,the receptor-binding domain(RBD,surface antigen)of the spike protein was trapped inside the nanocavity,while NP was absorbed on the outside of the droplets,enabling the burst release of NP before RBD.Compared with the natural packaging strategy,the inside-out strategy induced potent type I interferon-mediated innate immune responses and triggered an immune-potentiated environment in advance,which subsequently boosted CD40+DC activations and the engagement of the lymph nodes.In both H1N1 influenza and SARS-CoV-2 vaccines,rMASE significantly increased antigen-specific antibody secretion,memory T cell engagement,and Th1-biased immune response,which diminished viral loads after lethal challenge.By simply reversing the delivery sequence of the surface antigen and core antigen,the inside-out strategy may offer major implications for enhanced vaccinations against the enveloped RNA virus.Fengqiang Cao Sha Peng Yaling An Kun Xu Tianyi Zheng Lianpan Dai Kenji Ogino To Ngai Yufei Xia Guanghui Ma 2023Signal Transduction and Targeted Therapy2023,8,6:0
8Durable and enhanced immunity against SARS-CoV-2 elicited by manganese nanoadjuvant formulated subunit vaccine显示文摘Dear editor,Vaccines are the most efficient and effective means to prevent infectious diseases,but improving the long-term protective efficacy is still a major challenge in contemporary vaccine development.1 The waning immunity varies depending on the diversification of the pathogen and the number of booster doses.1 Strategies to overcome this warrant is using adjuvants that amplify the immune response,and drive the production of memory B and T cells or long-lived plasma cells that recognize the pathogen for durable protection.2–4 Although existing adjuvants have achieved promising results,research on generating durable protective immunity is lacking in promoting vaccine development and staying ahead of global pandemics such as coronavirus disease 2019(COVID-19).The precisely designed nanoadjuvants can enhance lymph node targeting and increase antigenpresenting cell(APCs)uptake,achieving the co-delivery of adjuvants and antigens and activating innate and adaptive immune responses.5 Previously,we reported a manganese nanoadjuvant(MnARK)and receptor-binding domain(RBD)monomer antigen formulated nanovaccine.6 MnARK transported antigens to lymph nodes,activated the STING pathway,elicited strong neutralizing abilities and increased immune memory T cell percentage against the infection of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2).6 Regarding the long-term protection potential of MnARK for subunit vaccine development,we further explored the durable immune regulation abilities of MnARK to a SARS-CoV-2 RBD dimer antigen,which has been used in an approved COVID-19 subunit vaccine ZF2001 with aluminum adjuvant(alum).7,8 TEM result revealed that RBD dimer could interact with BSA on MnARK surface and epitope can be well preserved(Supplementary Fig.1a).The size and zeta potential of MnARK-RBD dimer nanovaccine was~58 nm and-14 mV,respectively(Supplementary Fig.1b,c).Mengyu Guo Mingjing Cao Jiufeng Sun Ziwei Chen Xin Wang Lianpan Dai George F.Gao Yuliang Zhao Yaling Wang Chunying Chen 2024Signal Transduction and Targeted Therapy2024,9,1:0
