| 1 | Characterization and allergic role of IL-33-induced neutrophil polarization显示文摘Neutrophils are involved in the pathogenesis of allergy.However,the contribution of the different functionally polarized neutrophils in allergy needs to be clarified.We sought to define the characteristics of interleukin(IL)-33-induced neutrophils and the involvement of this subset of polarized neutrophils in allergic pathogenesis.Freshly isolated neutrophils were treated with different cytokines and the cytokine expression levels were detected by realtime PCR.The gene expression profile of IL-33-induced neutrophils was determined by microarray assay.Adoptive transfer assay was used to investigate the function of IL-33-induced neutrophils in an ovalbumin(OVA)-induced allergic asthma model.IL-33-treated neutrophils selectively produced IL-4,IL-5,IL-9 and IL-13(referred as to N(IL-33)cells)and displayed a distinctive gene expression profile in sharp contrast to resting and lipopolysaccharide(LPS)-treated neutrophils.IL-33-induced neutrophils expressed high Levels of IL-1R2 on cell surface,whereas resting and LPS-treated neutrophils did not,indicating IL-1R2 might be used as a biomarker for N(IL-33)cells.Importantly,N(IL-33)neutrophils exist in the lungs of OVA-induced allergic asthma mice.Adoptive transfer of N(IL-33)neutrophils significantly promotes the severity of the lung pathogenesis in this model.IL-33 induces neutrophil polarization through c-Jun N-terminal kinase-and nuclear factor-κB-dependent pathways.A previously unappreciated neutrophil polarization driven by IL-33 with unique cell surface markers and cytokine/chemokineproducing gene profile was defined.The newly identified N(IL-33)subpopulation may have significant contribution to IL-33-related pathogenesis. | Bo Sun Linnan Zhu Yaling Tao Hai-Xi Sun Yang Li Peng Wang Yuzhu Hou Yang Zhao Xiaodong Zhang Lianfeng Zhang Ning Na Yong Zhao | 2018 | Cellular & Molecular Immunology2018,15,8: | 9 |
| 2 | Implementation of an efficient segregated algorithm-IDEAL on 3D collocated grid system显示文摘The segregated algorithm-IDEAL (inner doubly-iterative efficient algorithm for linked-equations) is an efficient and stable algorithm. In this algorithm, there exist inner doubly-iterative processes for pres-sure equation, which almost completely overcome two approximations in SIMPLE algorithm. Thus the coupling between velocity and pressure is fully guaranteed, greatly enhancing the convergence rate and stability of iteration process. In this paper, implementation of the IDEAL algorithm on a 3D collo-cated grid system is conducted. The interface velocity is calculated by the modified momentum inter-polation method (MMIM), by which the converged result is independent of the under-relaxation factor. Finally, five three-dimensional incompressible fluid flow and heat transfer problems are provided to compare the convergence rate and robustness between the IDEAL and three other most widely-used algorithms (SIMPLER, SIMPLEC and PISO). | 3 | Mapping and analysis of a spatiotemporal H3K27ac and gene expression spectrum in pigs显示文摘The limited knowledge of genomic noncoding and regulatory regions has restricted our ability to decipher the genetic mechanisms underlying complex traits in pigs. In this study, we characterized the spatiotemporal landscape of putative enhancers and promoters and their target genes by combining H3K27ac-targeted Ch IP-Seq and RNA-Seq in fetal(prenatal days 74–75) and adult(postnatal days 132–150) tissues(brain, liver, heart, muscle and small intestine) sampled from Asian aboriginal Bama Xiang and European highly selected Large White pigs of both sexes. We identified 101,290 H3K27ac peaks, marking 18,521promoters and 82,769 enhancers, including peaks that were active across all tissues and developmental stages(which could indicate safe harbor locus for exogenous gene insertion) and tissue-and developmental stage-specific peaks(which regulate gene pathways matching tissue-and developmental stage-specific physiological functions). We found that H3K27ac and DNA methylation in the promoter region of the XIST gene may be involved in X chromosome inactivation and demonstrated the utility of the present resource for revealing the regulatory patterns of known causal genes and prioritizing candidate causal variants for complex traits in pigs. In addition, we identified an average of 1,124 super-enhancers per sample and found that they were more likely to show tissue-specific activity than ordinary peaks. | 9 | Neurogranin as an important regulator in swimming training to improve the spatial memory dysfunction of mice with chronic cerebral hypoperfusion显示文摘Background:Vascular cognitive impairment caused by chronic cerebral hypoperfusion(CCH)has become a hot issue worldwide.Aerobic exercise positively contributes to the preservation or restoration of cognitive abilities;however,the specific mechanism has remained inconclusive.And recent studies found that neurogranin(Ng)is a potential biomarker for cognitive impairment.This study aims to investigate the underlying role of Ng in swimming training to improve cognitive impairment.Methods:To test this hypothesis,the clustered regularly interspaced short palindromic repeats(CRISPR)-associated protein 9(Cas9)system was utilized to construct a strain of Ng conditional knockout(Ng cKO)mice,and bilateral common carotid artery stenosis(BCAS)surgery was performed to prepare the model.In Experiment 1,2-month-old male and female transgenic mice were divided into a control group(wild-type littermate,n=9)and a Ng cKO group(n=9).Then,2-month-old male and female C57BL/6 mice were divided into a sham group(C57BL/6,n=12)and a BCAS group(n=12).In Experiment 2,2-month-old male and female mice were divided into a sham group(wild-type littermate,n=12),BCAS group(n=12),swim group(n=12),BCAS+Ng cKO group(n=12),and swim+Ng cKO group(n=12).Then,7 days after BCAS,mice were given swimming training for 5 weeks(1 week for adaptation and 4 weeks for training,5 days a week,60 min a day).After intervention,laser speckle was used to detect cerebral blood perfusion in the mice,and the T maze and Morris water maze were adopted to test their spatial memory.Furthermore,electrophysiology and Western blotting were conducted to record long-term potential and observe the expressions of Ca^(2+)pathway-related proteins,respectively.Immunohistochemistry was applied to analyze the expression of relevant markers in neuronal damage,inflammation,and white matter injury.Results:The figures showed that spatial memory impairment was detected in Ng cKO mice,and a sharp decline of cerebral blood flow and an impairment of progressive spatial memory were observed in BCAS mice.Regular swimming training improved the spatial memory impairment of BCAS mice.This was achieved by preventing long-term potential damage and reversing the decline of Ca^(2+)signal transduction pathway-related proteins.At the same time,the results suggested that swimming also led to improvements in neuronal death,inflammation,and white matter injury induced by CCH.Further study adopted the use of Ng cKO transgenic mice,and the results indicated that the positive effects of swimming training on cognitive impairments,synaptic plasticity,and related pathological changes caused by CCH could be abolished by the knockout of Ng.Conclusion:Swimming training can mediate the expression of Ng to enhance hippocampal synaptic plasticity and improve related pathological changes induced by CCH,thereby ameliorating the spatial memory impairment of vascular cognitive impairment. | Huawei Lin Jiayong Zhang Yaling Dai Huanhuan Liu Xiaojun He Lewen Chen Jing Tao Chaohui Li Weilin Liu | 2023 | Journal of Sport and Health Science2023,12,1: | 1 |

| 16 | Predicting Lung Cancers Using Epidemiological Data:A Generative-Discriminative Framework显示文摘Predictive models for assessing the risk of developing lung cancers can help identify high-risk individuals with the aim of recommending further screening and early intervention.To facilitate pre-hospital self-assessments,some studies have exploited predictive models trained on non-clinical data(e.g.,smoking status and family history).The performance of these models is limited due to not considering clinical data(e.g.,blood test and medical imaging results).Deep learning has shown the potential in processing complex data that combine both clinical and non-clinical information.However,predicting lung cancers remains difficult due to the severe lack of positive samples among follow-ups.To tackle this problem,this paper presents a generative-discriminative framework for improving the ability of deep learning models to generalize.According to the proposed framework,two nonlinear generative models,one based on the generative adversarial network and another on the variational autoencoder,are used to synthesize auxiliary positive samples for the training set.Then,several discriminative models,including a deep neural network(DNN),are used to assess the lung cancer risk based on a comprehensive list of risk factors.The framework was evaluated on over 55000 subjects questioned between January 2014 and December 2017,with 699 subjects being clinically diagnosed with lung cancer between January 2014 and August 2019.According to the results,the best performing predictive model built using the proposed framework was based on DNN.It achieved an average sensitivity of 76.54%and an area under the curve of 69.24%in distinguishing between the cases of lung cancer and normal cases on test sets. | Jinpeng Li Yaling Tao Ting Cai | 2021 | IEEE/CAA Journal of Automatica Sinica2021,8,5: | 1 |
