1Specific Regulation of m^(6)A by SRSF7 Promotes the Progression of Glioblastoma显示文摘Serine/arginine-rich splicing factor 7(SRSF7),a known splicing factor,has been revealed to play oncogenic roles in multiple cancers.However,the mechanisms underlying its oncogenic roles have not been well addressed.Here,based on N6-methyladenosine(m^(6)A)co-methylation network analysis across diverse cell lines,we find that the gene expression of SRSF7 is positively correlated with glioblastoma(GBM)cell-specific m^(6)A methylation.We then indicate that SRSF7 is a novel m^(6)A regulator,which specifically facilitates the m^(6)A methylation near its binding sites on the mRNAs involved in cell proliferation and migration,through recruiting the methyltransferase complex.Moreover,SRSF7 promotes the proliferation and migration of GBM cells largely dependent on the presence of the m^(6)A methyltransferase.The two m^(6)A sites on the mRNA for PDZ-binding kinase(PBK)are regulated by SRSF7 and partially mediate the effects of SRSF7 in GBM cells through recognition by insulin-like growth factor 2 mRNA-binding protein 2(IGF2BP2).Together,our discovery reveals a novel role of SRSF7 in regulating m^(6)A and validates the presence and functional importance of temporal-and spatial-specific regulation of m^(6)A mediated by RNA-binding proteins(RBPs).Yixian Cun Sanqi An Haiqing Zheng Jing Lan Wenfang Chen Wanjun Luo Chengguo Yao Xincheng Li Xiang Huang Xiang Sun Zehong Wu Yameng Hu Ziwen Li Shuxia Zhang Geyan Wu Meisongzhu Yang Miaoling Tang Ruyuan Yu Xinyi Liao Guicheng Gao Wei Zhao Jinkai Wang Jun Li 2023Genomics, Proteomics & Bioinformatics2023,21,4:1

3How do lateral septum projections to the ventral CA1 influence sociability?显示文摘Social dysfunction is a risk factor for several neuropsychiatric illnesses.Previous studies have shown that the lateral septum(LS)-related pathway plays a critical role in mediating social behaviors.Howeve r,the role of the connections between the LS and its downstream brain regions in social behavio rs remains unclea r.In this study,we conducted a three-chamber test using electrophysiological and chemogenetic approaches in mice to determine how LS projections to ventral CA1(vCA1)influence sociability.Our res ults showed that gamma-aminobutyric acid(GABA)-e rgic neuro ns were activated following social experience,and that social behavio rs were enhanced by chemogenetic modulation of these neurons.Moreover,LS GABAergic neurons extended their functional neural connections via vCA1 glutamatergic pyramidal neurons,and regulating LSGABA→vCA1Gluneural projections affected social behaviors,which were impeded by suppressing LSprojecting vCA1 neuronal activity or inhibiting GABAAreceptors in vCA1.These findings support the hypothesis that LS inputs to the vCA1 can control social prefe rences and social novelty behaviors.These findings provide new insights rega rding the neural circuits that regulate sociability.Dan Wang Di Zhao Wentao Wang Fengai Hu Minghu Cui Jing Liu Fantao Meng Cuilan Liu Changyun Qiu Dunjiang Liu Zhicheng Xu Yameng Wang Yu Zhang Wei Li Chen Li 2024Neural Regeneration Research2024,19,8:0
4Filamentation initiated by Cas2 and its association with the acquisition process in cells显示文摘Cas1-and-Cas2-mediated new spacer acquisition is an essential process for bacterial adaptive immunity.The process is critical for the ecology of the oral microflora and oral health.Although molecular mechanisms for spacer acquisition are known,it has never been established if this process is associated with the morphological changes of bacteria.In this study,we demonstrated a novel Cas2-induced filamentation phenotype in E.coli that was regulated by co-expression of the Cas1 protein.A 30 amino acid motif at the carboxyl terminus of Cas2 is necessary for this function.By imaging analysis,we provided evidence to argue that Cas-induced filamentation is a step coupled with new spacer acquisition during which filaments are characterised by polyploidy with asymmetric cell division.This work may open new opportunities to investigate the adaptive immune response and microbial balance for oral health.Lei Wang Xin Yu Mengjie Li Guiqin Sun Lin Zou Tiansheng Li Linlin Hou Yameng Guo Danfeng Shen Di Qu Xunjia Cheng Li Chen 2019International Journal of Oral Science2019,11,4:0
5Hydroxyapatite reinforced inorganic-organic hybrid nanocomposite as high-performance adsorbents for bilirubin removal in vitro and in pig models显示文摘Highly efficient removal of bilirubin from whole blood directly by hemoperfusion for liver failure therapy remains a challenge in the clinical field due to the low adsorption capacity,poor mechanical strength and low biocompatibility of adsorbents.In this work,a new class of nanocomposite adsorbents was constructed through an inorganic-organic co-crosslinked nanocomposite network between vinyltriethoxysilane(VTES)-functionalized hydroxyapatite nanoparticles(V-Hap)and non-ionic styrene-divinylbenzene(PS-DVB)resins(PS-DVB/V-Hap)using suspension polymerization.Notably,our adsorbent demonstrated substantially improved mechanical performance compared to the pure polymer,with the hardness and modulus increasing by nearly 3 and 2.5 times,respectively.Moreover,due to the development of a mesoporous structure,the prepared PS-DVB/V-Hap3 exhibited an ideal adsorption capacity of 40.27 mg·g^(-1).More importantly,the obtained adsorbent beads showed outstanding blood compatibility and biocompatibility.Furthermore,in vivo extracorporeal hemoperfusion verified the efficacy and biosafety of the adsorbent for directly removing bilirubin from whole blood in pig models,and this material could potentially prevent liver damage and improve clinical outcomes.Taken together,the results suggest that PS-DVB/V-Hap3 beads can be used in commercial adsorption columns to threat hyperbilirubinemia patients through hemoperfusion,thus replacing the existing techniques where plasma separation is initially required.Yamin Chai Zhuang Liu Yunzheng Du Lichun Wang Jinyan Lu Qian Zhang a Wenyan Han Tingting Wang Yameng Yu Lisha Sun Lailiang Ou 2021Bioactive Materials2021,6,12:0
6Direct observation of the plasmon-enhanced palladium catalysis with single-molecule fluorescence microscopy显示文摘Plasmonic nanostructures have been proved effective not only in catalyzing chemical reactions,but also in improving the activity of non-plasmonic photocatalysts.It is essential to reveal the synergy between the plasmonic structure and the non-plasmonic metal photocatalyst for expounding the underlying mechanism of plasmon-enhanced catalysis.Herein,the enhancement of resazurin reduction at the heterostructure of silver nanowire(AgNW)and palladium nanoparticles(PdNPs)is observed in situ by single-molecule fluorescence microscopy.The catalysis mapping results around single AgNW suggest that the catalytic activity of PdNPs is enhanced for~20 times due to the excitation of localized surface plasmon resonance(LSPR)in the vicinity of the AgNW.This catalysis enhancement is also highly related to the wavelength and polarization of the excitation light.In addition,the palladium catalysis is further enhanced by~10 times in the vicinity of a roughened AgNW or a AgNW-AgNW nanogap because of the improvement of catalytic hotspots.These findings clarify the contribution of plasmon excitation in palladium catalysis at microscopic scale,which will help to deepen the understanding of the plasmon-enhanced photocatalysis and provide a guideline for developing highly efficient plasmon-based photocatalysts.Zhuoyao Li Rajkumar Devasenathipathy Junjie Wang Liuyingzi Yu Yan Liang Huixiang Sheng Yameng Zhu Hai Li Hiroshi Uji-i Xiao Huang Gang Lu 2023Nano Research2023,16,7:0
