| 1 | Clinical features of seizures after cerebral venous sinus thrombosis and its effect on outcome among Chinese Han population显示文摘background and purpose Clinical features of epileptic seizures after cerebral venous sinus thrombosis(CVST)among Chinese patients are not known,and it is still controversial whether seizures would affect the outcome of CVST.Methods In a Chinese hospital-based study of consecutive patients with CVST between 2003 and 2015,we described the clinical features of seizures and determined the predictors of seizure onset using multivariable logistic regression analysis.We also compared the in-hospital case-fatality and short-term functional outcome(modified Rankin Scale(mRS)at discharge)in patients with versus without seizures using ordinal regression analysis.results Among 151 patients with CVST,52(34.4%)presented seizures,of which 42(80.8%)were generalised seizures.Male gender(OR 6.32,95% CI 2.06 to 19.35,p=0.001),motor deficits(OR 4.89,95% CI 1.52 to 15.68,p=0.008),intracerebral haemorrhage(OR 3.93,95% CI 1.16 to 13.26,p=0.027),cerebral infarction(OR 3.78,95% CI 1.15 to 12.36,p=0.029)and superior sagittal sinus thrombosis(OR 3.38,95% CI 91.16 to 9.86,p=0.026)were independent predictors for seizures.The overall in-hospital case-fatality rate was 2.0%(3/151),and 21(13.9%)had mRS>2 at discharge.Compared with patients without seizures,patients with seizures were more likely to have a worse outcome(p=0.02)at discharge,independent of age,gender,clinical presentation,clot burden and presence of parenchymal lesions.conclusions In Chinese Han patients,compared with patients without seizures,patients with seizures after CVST had a worse outcome.Risk factors such as male gender,paresis,parenchymal lesion and superior sagittal sinus thrombosis were independently associated with seizure onset after CVST.Generalised seizure was the main form of seizures after CVST,which was obviously different to seizures after strokes of arterial origin. | Hongyan Ding Yanan Xie Linxin Li Heling Chu Yuping Tang Qiang Dong Mei Cui | 2017 | Stroke & Vascular Neurology2017,2,4: | 8 |
| 2 | Hypoxia-stressed cardiomyocytes promote early cardiac differentiation of cardiac stem cells through HIF-1α/Jagged1/Notch1 signaling显示文摘Hypoxia is beneficial for the differentiation of stem cells transplanted for myocardial injury,but mechanisms underlying this benefit remain unsolved. Here, we report the impact of hypoxia-induced Jagged1 expression in cardiomyocytes(CMs) for driving the differentiation of cardiac stem cells(CSCs).Forced hypoxia-inducible factor 1α(HIF-1α) expression and physical hypoxia(5% O_2) treatment could induce Jagged1 expression in neonatal rat CMs. Pharmacological inhibition of HIF-1α by YC-1 attenuated hypoxia-promoted Jagged1 expression in CMs. An ERK inhibitor(PD98059), but not inhibitors of JNK(SP600125), Notch(DAPT), NF-κB(PTDC), JAK(AG490), or STAT3(Stattic) suppressed hypoxiainduced Jagged1 protein expression in CMs. c-Kit^+ CSCs isolated from neonatal rat hearts using a magnetic-activated cell sorting method expressed GATA4, SM22α or vWF, but not Nkx2.5 and cTnI.Moreover, 87.3% of freshly isolated CSCs displayed Notch1 receptor expression. Direct co-culture of CMs with BrdU-labeled CSCs enhanced CSCs differentiation, as evidenced by an increased number of BrdU^+/Nkx2.5^+ cells, while intermittent hypoxia for 21 days promoted co-culture-triggered differentiation of CSCs into CM-like cells. | 4 | by a cascade optical field modulation strategy facilitating selective multispectral narrow-band near-infrared photodetection显示文摘Since selective detection of multiple narrow spectral bands in the near-infrared(NIR)region still poses a fundamental challenge,we have,in this work,developed NIR photodetectors(PDs)using photon upconversion nanocrystals(UCNCs)combined with perovskite films.To conquer the relatively high pumping threshold of UCNCs,we designed a novel cascade optical field modulation strategy to boost upconversion luminescence(UCL)by cascading the superlensing effect of dielectric microlens arrays and the plasmonic effect of gold nanorods,which readily leads to a UCL enhancement by more than four orders of magnitude under weak light irradiation.By accommodating multiple optically active lanthanide ions in a core-shell-shell hierarchical architecture,developed PDs on top of this structure can detect three well-separated narrow bands in the NIR region,i.e.,those centered at 808,980,and 1540 nm.Due to the large UCL enhancement,the obtained PDs demonstrate extremely high responsivities of 30.73,23.15,and 12.20 AW^(−1) and detectivities of 5.36,3.45,and 1.91×10^(11) Jones for 808,980,and 1540 nm light detection,respectively,together with short response times in the range of 80-120 ms.Moreover,we demonstrate for the first time that the response to the excitation modulation frequency of a PD can be employed to discriminate the incident light wavelength.We believe that our work provides novel insight for developing NIR PDs and that it can spur the development of other applications using upconversion nanotechnology. | Yanan Ji Wen Xu Nan Ding Haitao Yang Hongwei Song Qingyun Liu Hans Agren Jerker Widengren Haichun Liu | 2020 | Light(Science & Applications)2020,9,1: | 6 |
| 5 | Microstructure and performance evaluations on Q&P hot stamping parts of several UHSS sheet metals显示文摘Hot stamping(press hardening) is widely used to fabricate safety components such as door beams and B pillars with increased strength via quenching. However, parts that are hot-stamped from ultra-high-strength steel(UHSS) have very limited elongation,i.e., low ductility. In the present study, a novel variant of hot stamping technology called quenching-and-partitioning(Q&P) hot stamping was developed. This approach was tested on several UHSS sheet metals, and it was confirmed that this method can be used to overcome the drawbacks associated with conventional hot stamping. The applicability of Q&P hot stamping to each of these steels was also assessed. The part properties and performances of three widely used ultra-high-strength sheet metals, B1500 HS,27 SiMn, and TRIP780, were evaluated through tensile testing and microstructural observations. The results demonstrated that the ductility of Q&P hot-stamped sheet metals was notably higher than that of the conventionally hot-stamped parts because Q&P hot stamping gives rise to a dual-phase structure of both martensite and austenite. Further, material tests demonstrated that the Q&P treatment had positive effects on all three selected materials, of which TRIP780 had the best ductility and the highest value of the product of strength and plasticity. | 7 | Single-cell RNA sequencing of meiocytes and microspores reveals the involvement of the Rf4 gene in redox homeostasis of CMS-C maize显示文摘Normal microsporogenesis is determined by both nuclear and mitochondrial genes. In maize C-type cytoplasmic male sterility, it is unclear how the development of meiocytes and microspores is affected by the mitochondrial sterility gene and the nuclear restorer gene. In this study, we sequenced the transcriptomes of single meiocytes(tetrad stage) and early mononucleate microspores from sterile and restorer lines. The numbers of expressed genes varied in individual cells and fewer than half of the expressed genes were common to the same cell types. Four comparisons revealed 3379 differentially expressed genes(DEGs), with 277 putatively associated with mitochondria, 226 encoding transcription factors,and 467 possibly targeted by RF4. KEGG analysis indicated that the DEGs in the two lines at the tetrad stage were involved predominantly in carbon metabolism and in amino acid biosynthesis and metabolism, whereas the DEGs during the transition from the tetrad stage to the early mononucleate stage were associated mostly with regulation of protein metabolism, fatty acid metabolism, and anatomical structure morphogenesis. | 8 | Geniposide, the component of the Chinese herbal formula Tongluojiunao, protects amyloid-β peptide(1–42)-mediated death of hippocampal neurons via the non-classical estrogen signaling pathway显示文摘Tongluojiunao(TLJN) is an herbal medicine consisting of two main components, geniposide and ginsenoside Rg1. TLJN has been shown to protect primary cultured hippocampal neurons. However, its mechanism of action remains unclear. In the present study, primary cultured hippocampal neurons treated with Aβ1–42(10 μmol/L) significantly increased the release of lactate dehydrogenase, which was markedly reduced by TLJN(2 μL/mL), specifically by the component geniposide(26 μmol/L), but not ginsenoside Rg1(2.5 μmol/L). The estrogen receptor inhibitor, ICI182780(1 μmol/L), did not block TLJN- or geniposide-mediated decrease of lactate dehydrogenase under Aβ1–42-exposed conditions. However, the phosphatidyl inositol 3-kinase or mitogen-activated protein kinase pathway inhibitor, LY294002(50 μmol/L) or U0126(10 μmol/L), respectively blo cked the decrease of lactate dehydrogenase mediated by TLJN or geniposide. LiCoO2 nanomaterials are synthesized via hydrothermal reaction with consequent calcination process. And the particle size increases as the calcination temperature rises.LCO-650 sample with the largest particle size displays the maximum capacitances of 817.5 Fg-1with the most outstanding capacity retention rate of 96.8% after 2000 cycles. It is shown that large particle size is beneficial to the electrochemical and structural stability of Li CoO2 materials. We speculate that the micron-sized waste LiCoO2 materials have great potential for supercapacitor application. | 19 | A simple method to improve the dissolution of repaglinide and exploration of its mechanism显示文摘In the present study,a simple and rapid method was developed to improve the in vitro dissolution of repaglinide,an oral antidiabetic drug,which was based on addition of meglumine in 50%(v/v)ethanol to dissolve repaglinide,and the drug dissolved in meglumine/50%ethanol was used directly with a binder to prepare tablets.The mechanism of solubilization of repaglinide by meglumine was studied by using infrared spectrum(IR),ultraviolet(UV)measurement through dual wavelength,differential scanning calorimetry(DSC)and X-ray powder diffraction methods.Dissolution tests of repaglinide tablets were performed in the media with different pH values and the repaglinide concentrations were analyzed by High Performance Liquid Chromatography(HPLC)method.The solubility data showed that with the meglumine concentration increasing,the solubility of repaglinide was increased.Meanwhile,tablets with the molar ratio of repaglinide and meglumine 1:2(n/n)resulted in a significant increase in dissolution compared to the repaglinide tablets without using meglumine,and nearly equal to the commercial preparations of Novo-Norm^(®),which concluded that meglumine had a great role in promoting the dissolution of repaglinide.The results of IR and UV dual wavelength methods suggested the formation of repaglinideemeglumine(REPeMEG)molecular complex.DSC results showed that the melting peak of repaglinide disappeared in the REPeMEG coprecipitate,which indicated that repaglinide was stable when existing at amorphous or molecular state.The experiment of X-ray powder diffraction showed that with the solubilization of meglumine,the crystal diffraction peak of repaglinide disappeared,which further inferred that repaglinide was formed complexes with meglumine.It was demonstrated that the method of improving repaglinide with meglumine was reliable and could be suitable for repaglinide tablets production in industry.This study also provides a feasible way to enhance the dissolution of drugs with low solubility,which will be leading to improved bioavailability of these drugs. | Zhaolu Zhu Tianzhi Yang Yanan Zhao Nannan Gao Donglei Leng Pingtian Ding | 2014 | Asian Journal of Pharmaceutical Sciences2014,9,4: | 1 |
