1Large-scale analysis of small RNAs derived from traditional Chinese herbs in human tissues显示文摘Plant-derived microRNAs have recently been reported to function in human blood and tissues. Controversy was immediately raised due to possible contamination and the lack of large sample sizes. Here, we report thousands of unique small RNAs derived from traditional Chinese medicine(TCM) herbs found in human blood cells and mouse lung tissues using a large-scale analysis.We extracted small RNAs from decoctions of 10 TCM plants(Ban Zhi Lian, Chai Hu, Chuan Xin Lian, Di Ding Zi Jin, Huang Qin, Jin Yin Hua, Lian Qiao, Pu Gong Ying, Xia Ku Cao, and Yu Xing Cao) and obtained millions of RNA sequences from each herb. We also obtained RNA-Seq data from the blood cells of humans who consumed herbal decoctions and from the lung tissues of mice administered RNAs from herbal decoctions via oral gavage. We identified thousands of unique small RNA sequences in human blood cells and mouse lung tissues. Some of these identified small RNAs from Chuan Xin Lian and Hong Jing Tian could be mapped to the genomes of the herbs, confirming their TCM plant origin. 2A comprehensive analysis of the Bencao(herbal)small RNA Atlas reveals novel RNA therapeutics for treating human diseases显示文摘Cross-kingdom herbal mi RNA was first reported in 2012.Using a modified herbal extraction protocol,we obtained 73,677,287sequences by RNA-seq from 245 traditional Chinese Medicine(TCM),of which 20,758,257 were unique sequences.We constructed a Bencao(herbal)small RNA(s RNA)Atlas(http://bencao.bmicc.cn),annotated the sequences by sequence-based clustering,and created a nomenclature system for Bencao s RNAs.The profiles of 21,757 mi RNAs in the Atlas were highly consistent with those of plant mi RNAs in mi RBase.Using software tools,our results demonstrated that all human genes might be regulated by s RNAs from the Bencao s RNA Atlas,part of the predicted human target genes were experimentally validated,suggesting that Bencao s RNAs might be one of the main bioactive components of herbal medicines.We established roadmaps for oligonucleotide drugs development and optimization of TCM prescriptions.Moreover,the decoctosome,a lipo-nano particle consisting of 0.5%–2.5%of the decoction,demonstrated potent medical effects.We propose a Bencao(herbal)Index,including small-molecule compounds(SM),protein peptides(P),nucleic acid(N),non-nucleic and non-proteinogenic large-molecule compounds(LM)and elements from Mendeleev's periodic table(E),to quantitatively measure the medical effects of botanic medicine.The Bencao s RNA Atlas is a resource for developing gene-targeting oligonucleotide drugs and optimizing botanical medicine,and may provide potential remedies for the theory and practice of one medicine.Yinghao Cao Yexuan Lin Na Sun Xinyi Du Yixin Dong Song Mei Xingyu Deng Xiaobei Li Shaoting Guo Kegong Tang Jiaqi Liu Xiangyu Qiao Dandan Zhao Yuhao Qin Cong Zhang Tianyi Xin Xiaohu Shi Congzhao Zhou Tao Dong De-an Guo Benedikt MKessler Dong Xu Jingyuan Song Fengming Huang Xiaoyue Wang Chengyu Jiang 2023Science China(Life Sciences)2023,66,10:2
3Orally administered BZL-s RNA-20 oligonucleotide targeting TLR4 effectively ameliorates acute lung injury in mice显示文摘The global COVID-19 pandemic emerged at the end of December 2019.Acute respiratory distress syndrome(ARDS)and acute lung injury(ALI)are common lethal outcomes of bacterial lipopolysaccharide(LPS),avian influenza virus,and SARS-Co V-2.Toll-like receptor 4(TLR4)is a key target in the pathological pathway of ARDS and ALI.Previous studies have reported that herbal small RNAs(s RNAs)are a functional medical component.BZL-s RNA-20(Accession number:B59471456;Family ID:F2201.Q001979.B11)is a potent inhibitor of Toll-like receptor 4(TLR4)and pro-inflammatory cytokines.Furthermore,BZLs RNA-20 reduces intracellular levels of cytokines induced by lipoteichoic acid(LTA)and polyinosinic-polycytidylic acid(poly(I:C)).We found that BZL-s RNA-20 rescued the viability of cells infected with avian influenza H5N1,SARS-Co V-2,and several of its variants of concern(VOCs).Acute lung injury induced by LPS and SARS-Co V-2 in mice was significantly ameliorated by the oral medical decoctosome mimic(bencaosome;sphinganine(d22:0)+BZL-s RNA-20).Our findings suggest that BZLs RNA-20 could be a pan-anti-ARDS/ALI drug.Dandan Zhao Yuhao Qin Jiaqi Liu Kegong Tang Shuaiyao Lu Zirui Liu Yexuan Lin Cong Zhang Fengming Huang Jiahui Chang Chang Li Mingyao Tian Yiming Ma Xiaoyun Li Congzhao Zhou Xiao Li Xiaozhong Peng Ningyi Jin Chengyu Jiang 2023Science China(Life Sciences)2023,66,7:1

5IMPACTS OF DIFFERENT TYPES OF LAND USE ON PROCESSES OF SOIL AND WATER LOSS OVER PURPLE SOIL SLOPELAND显示文摘Based on natural precipitation observations, impacts of different types of land use on processes of soil and water loss over purple soil related slopeland were studied by simulated rainfall experiments. Measurement data revealed that rainstorms and slope length are the essential factors accountable for soil and water loss on purple soil slopeland for intense rill erosion can be caused on 10 meter long purple soil slopes by high intensity rainfall. Under circumanstances of rainstorms, annual hedge plants grown on slopeland of 25 degrees can cause a reduction of runoff by 22 43 percent and that of erosion induced sand content by 94 98 percent. Stone bund horizontal terraces can lead to a runoff reduction by 62 67 percent in comparison with steep slopelands and that of erosion induced sediment by 97.8 99 percent. Soil and water loss can be substantially decreased on steep slopes by hedge plants with a cost of only 10 20 percent that of the stone bund horizontal terraces. 6Liver Fibrosis Scoring Systems as Novel Tools for Predicting Recurrent Cardiovascular Events in Patients with a Prior Cardiovascular Event显示文摘Objective:Regarding the secondary prevention of cardiovascular disease(CVD),there is great interest in preventing recurrent cardiovascular events(RCVEs).The prognostic importance of liver fibrosis scores(LFSs)has previously been reported in variousCVDs.We hypothesized that LFSs might also be useful predictors for RCVEs in patients with prior cardiovascular events(CVEs).Herein,we aimed to evaluate the associations of LFSs with RCVEs in a large,real-world cohort of coronary artery disease(CAD)patients with a prior CVE.Methods:In this multicenter prospective study,6527 consecutive patients with angiography-diagnosed CAD who had experienced a prior CVE(acute coronary syndrome,stroke,percutaneous coronary intervention,or coronary artery bypass grafting)were enrolled.LFSs were computed according to the published formulas:non-alcoholic fatty liver disease fibrosis score(NFS)includes age,body mass index(BMI),impaired fasting glycemia or diabetes mellitus(DM),aspartate aminotransferase(AST)/alanine aminotransferase(ALT)ratio,platelets,and albumin;fibrosis-4(FIB-4)includes age,AST,ALT,and platelets;Forns score includes age,gamma-glutamyltransferase(GGT),and platelets;BARD includes BMI,AST/ALT ratio,and DM;GGT/platelet ratio includes GGT and platelets;AST/ALT ratio includes AST and ALT;and AST/platelet ratio index includes AST and platelets.The originally reported cutoffs were used for the categorization of low-,intermediate-,and high-score subgroups.All patients were followed up for the occurrence of RCVEs(comprising cardiovascular death,non-fatal myocardial infarction,and stroke).Cox and Poisson regression analyses were used to assess the relationship of baseline LFSs with the risk of RCVE.Results:During a mean follow-up of(54.67±18.80)months,532(8.2%)RCVEs were recorded.Intermediate and high NFS,FIB-4,Forns,and BARD scores were independently associated with an increased risk of RCVE(hazard ratios ranging from 1.42 to 1.75 for intermediate scores and 1.35 to 2.52 for high scores).In the subgroup analyses of sex,age,BMI,DM,and hypertension status,the increased risk of RCVEs with high LFSs(NFS,FIB-4,Forns,and BARD)was maintained across the different subgroups(all P<0.05).Conclusion:This study showed that LFSs are indeed independently associated with RCVEs,suggesting that LFSs may be used as novel tools for risk stratification in CAD patients with a prior CVE.Huihui Liu Yexuan Cao Jinglu Jin Yuanlin Guo Chenggang Zhu Naqiong Wu Qi Hua Yanfang Li Lifeng Hong Qian Dong Jianjun Li 2021Cardiology Discovery2021,1,4:1

9Coherent Raman scattering imaging of lipid metabolism in cancer显示文摘Cancer cells dysregulate lipid metabolism to accelerate energy production and biomolecule synthesis for rapid growth.Lipid metabolism is highly dynamic and intrinsically heterogeneous at the single cell level.Although°uorescence microscopy has been commonly used for cancer research,bulky°uorescent probes can hardly label small lipid molecules without perturbing their biological activities.Such a challenge can be overcome by coherent Raman scattering(CRS)microscopy,which is capable of chemically selective,highly sensitive,submicron resolution and high-speed imaging of lipid molecules in single live cells without any labeling.Recently developed hyperspectral and multiplex CRS microscopy enables quantitative mapping of various lipid metabolites in situ.Further incorporation of CRS microscopy with Raman tags greatly increases molecular selectivity based on the distinct Raman peaks well separated from the endogenous cellular background.Owing to these unique advantages,CRS microscopy sheds new insights into the role of lipid metabolism in cancer development and progression.This review focuses on the latest applications of CRS microscopy in the study of lipid metabolism in cancer.Shuo Zhang Yexuan He Shuhua Yue 2023Journal of Innovative Optical Health Sciences2023,16,3:0
