| 4 | Surrogate production of genome-edited sperm from a different subfamily by spermatogonial stem cell transplantation显示文摘The surrogate reproduction technique,such as inter-specific spermatogonial stem cells(SSCs)transplantation(SSCT),provides a powerful tool for production of gametes derived from endangered species or those with desirable traits.However,generation of genome-edited gametes from a different species or production of gametes from a phylogenetically distant species such as from a different subfamily,by SSCT,has not succeeded.Here,using two small cyprinid fishes from different subfamilies,Chinese rare minnow(gobiocypris rarus,for brief:Gr)and zebrafish(danio rerio),we successfully obtained Gr-derived genome-edited sperm in zebrafish by an optimized SSCT procedure.The transplanted Gr SSCs supported the host gonadal development and underwent normal spermatogenesis,resulting in a reconstructed fertile testis containing Gr spermatids and zebrafish testicular somatic cells.Interestingly,the surrogate spermatozoa resembled those of host zebrafish but not donor Gr in morphology and swimming behavior.When pou5f3 and chd knockout Gr SSCs were transplanted,Gr-derived genome-edited sperm was successfully produced in zebrafish.This is the first report demonstrating surrogate production of gametes from a different subfamily by SSCT,and surrogate production of genome-edited gametes from another species as well.This method is feasible to be applied to future breeding of commercial fish and livestock. | Fenghua Zhang Yongkang Hao Xianmei Li Yi Li Ding Ye Ru Zhang Xiaosi Wang Mudan He Houpeng Wang Zuoyan Zhu Yonghua Sun | 2022 | Science China(Life Sciences)2022，65，5： | 7 |

| 6 | DEK43 is a P-type pentatricopeptide repeat(PPR)protein responsible for the Cis-splicing of nad4 in maize mitochondria显示文摘Mitochondria, the main energy transducers in plant cells, require the proper assembly of respiratory chain complexes Ⅰ–Ⅴ for their function. The NADH dehydrogenase4(nad4) gene encodes mitochondrial respiratory chain complex Ⅰ subunit Ⅳ, but the mechanism underlying nad4 transcript splicing is unclear. Here, we report that the P-type pentatricopeptide repeat(PPR) protein DEFECTIVE KERNEL 43(DEK43) is responsible for cis-splicing of the nad4 transcript in maize. We demonstrate that DEK43 localizes to both the nucleus and mitochondria. The mutation of Dek43 resulted in embryo-lethal and light-colored defective kernels. Among the 22 mitochondrial group Ⅱ introns, the splicing efficiency of nad4 introns 1 and 3 was reduced by up to 50% compared to the wild type. The levels of complex Ⅰ and supercomplex Ⅰ+Ⅲ2 were also reduced in dek43. Furthermore, in-gel NADH dehydrogenase assays indicated that the activities of these complexes were significantly reduced in dek43. Further, the mitochondrial ultrastructure was altered in the mutant. | 8 | Effect of thermal exposure on the stress-rupture life and microstructure of a low Re-containing single crystal alloy显示文摘In this paper, the stress-rupture tests of a low Re-containing single crystal alloy IC21 before and after thermal exposure at 1100 ℃for various periods of time were conducted under the test condition of 1100 ℃/137 MPa, and the microstructure of the tested specimens was characterized by SEM and TEM. The experimental results showed that the stress rupture life of this alloy was over 150 h after the standard heat treatment of1320 ℃, 10 h/AC t 870, 32 h/AC, however the stress rupture life decreased with the increase of exposure time due to the microstructure degradation. The TEM analysis revealed that the interface mismatch dislocation networks were well established. | 14 | Effects of mild moxibustion on intestinal microbiome and NLRP3 inflammasome in rats with 5-fluorouracil-induced intestinal mucositis显示文摘Objective:The present study investigated how mild moxibustion treatment affects the intestinal Microbiome and expression of NLRP3-related immune factors in a rat model of intestinal mucositis(IM)induced with 5-fluorouracil(5-Fu).Methods:Forty male Sprague-Dawley rats were randomly divided into control,chemotherapy,moxibustion and probiotics groups.The IM rat model was established by intraperitoneal injection of 5-Fu.Mild moxibustion treatment and intragastric probiotic administration were provided once daily for 15 days.Tissue morphology,serum levels of inflammatory factors and the expression levels of tight junction proteins,caspase-1,gasdermin D and NLRP3 were evaluated in colon tissue,through hematoxylin and eosin staining,electron microscopy,enzyme-linked immunosorbent assay,Western blotting,quantitative realtime reverse transcription polymerase chain reaction and immunofluorescence.Gut microbiome profiling was conducted through 16 S rRNA amplicon sequencing.Results:Moxibustion and probiotic treatments significantly increased the expression levels of tight junction proteins,reduced cell apoptosis and the expression levels of caspase-1,gasdermin D and NLRP3;they also decreased the serum levels of tumor necrosis factor-α,interleukin(IL)-6,IL-1βand IL-18,while increasing serum levels of IL-10.Moxibustion and probiotic treatments also corrected the reduction inα-diversity andβ-diversity in IM rats,greatly increased the proportion of the dominant bacterial genus Lactobacillus and reduced the abundance of the genera Roseburia and Escherichia in chemotherapytreated rats to levels observed in healthy animals.We also found that these dominant genera were firmly correlated with the regulation of pyroptosis-associated proteins and inflammatory factors.Finally,moxibustion and probiotic treatments elicited similar effects in regulating intestinal host-microbial homeostasis and the expression of NLRP3 inflammasome-related factors.Conclusion:Moxibustion exerts its therapeutic effect on IM by ameliorating mucosal damage and reducing inflammation.Moreover,moxibustion modulates the gut microbiota,likely via decreasing the expression levels of the NLRP3 inflammasome. | Bing-rong Li Shi-yun Shao Long Yuan Ru Jia Jian Sun Qing Ji Hua Sui Li-hong Zhou Yi Zhang Hui Liu Qi Li Yan Wang Bi-meng Zhang | 2021 | Journal of Integrative Medicine2021，19，2： | 2 |

| 16 | Subdivision and age of the Yanchang Formation and the Middle/Upper Triassic boundary in Ordos Basin, North China显示文摘The Yanchang Formation is extensively developed in the Ordos Basin and its surrounding regions. As one of the best terrestrial Triassic sequences in China and the major oil-gas bearing formations in the Ordos Basin, its age determination and stratigraphic assignment are important in geological survey and oil-gas exploration. It had been attributed to the Late Triassic and regarded as the typical representative of the Upper Triassic in northern China for a long time, although some scholars had already proposed that the lower part of this formation should be of the Middle Triassic age in the mid-late 20 th century. In this paper, we suggest that the lower and middle parts of the Yanchang Formation should be of the Ladinian and the bottom possibly belongs to the late Anisian of the Middle Triassic, mainly based on new fossils found in it and high resolution radiometric dating results. The main source rocks, namely the oil shales and mudstones of the Chang-7, are of the Ladinian Age. The upper part of the Yanchang Formation, namely the Chang-6 and the above parts, belongs to the Late Triassic. The uppermost of the Triassic is missed in most parts of the Ordos Basin. The Middle-Upper Triassic Series boundary lies in the Yanchang Formation, equivalent to the boundary between Chang-7 and Chang-6. The Ladinian is an important palaeoenvironmental turning point in the Ordos Basin. Palaeoenvironmental changes in the basin are coincidence with that of the Sichuan Basin and the main tectonic movement of the Qinling Mountains.
