| 1 | Exploring the history of cultural exchange in prehistoric Eurasia from the perspectives of crop diffusion and consumption显示文摘The history of cultural exchange in prehistoric Eurasia(CEPE) has been widely investigated. Based on archaeological evidence, this process is thought to date back to at least the early Bronze Age, although details about timings and routes remain unclear. It is likely that CEPE promoted the spread and exchange of crops that originated in different parts of Eurasia; since these remains can be definitely identified and directly dated, they provide ideal research materials to explore the history of CEPE. In this paper, we review the available archaeobotanical evidence and direct radiocarbon dates for crop remains, alongside carbon isotopic data from human bones unearthed from prehistoric sites in Eurasia, in order to investigate the history of the spread of millet crops, and wheat and barley, that were first domesticated in the eastern and western parts of Eurasia during prehistoric times.In combination with other archaeological evidences, we discuss the history of CEPE. Our results suggest that wheat and barley were domesticated in western Asia around 10500 a BP, spread into Europe and western Central Asia before 8000 a BP, and reaching eastern Central Asia and northwestern China between 4500 and 4000 a BP. Data show that both broomcorn and foxtail millet were domesticated in eastern Asia before 7700 a BP, spread into eastern Central Asia between 4500 and 4000 a BP, and into western Asia and Europe prior to 3500 a BP. Wheat, barley, and millet crops were first utilized together in eastern Kazakhstan within Central Asia around 4400 a BP, the region where earliest CEPE is likely to have taken place. These crops were mixedly used mainly in eastern central Asia and northwest China between 4500 and 3500 a BP, and then across the Eurasia before 2200 a BP. | 2 | Dose-dependent Inhibition of Gynecophoral Canal Protein Gene Expression in Vitro in the Schistosome(Schistosomajaponicum)by RNA Interference显示文摘The gynecophoral canal protein gene SjGCP of Schistosoma japonicum that is necessary for the pairing between the male and female worms is specifically expressed in the adult male worm. This protein is widely distributed in the adult female worm after pairing. Reverse transcription-polymerase chain reaction (RT-PCR) and immunofluorescence were employed to analyze the relationship between the RNAi effect and dsRNA dosage in the parasites. The results revealed that the inhibition of SjGCP expression by siRNA is dose-dependent. | 3 | Characteristics of Long Air Gap Discharge Current Subjected to Switching Impulse显示文摘Measuring the pre-breakdown current of long sparks in air is important for investigating the discharge mechanism.Since the breakdown of long air gaps is conducted by a series of streamer-leader processes,the corresponding current signals cover a bandwidth of 0 to more than 20 MHz.Measurement accuracy of the current from the high voltage side is affected by the displacement current and impulse electromagnetic interference.In this paper,a coaxial current sensor with a DC bandwidth of 74.45 MHz is developed.A displacement current-restrained electrode structure is proposed to reduce the equivalent capacitance between the current sensor and the ground over 30 times.Combined with the digital optical fiber synchronous acquisition unit,a current measurement system for long air gap discharge is established.For the purpose of the UHV system’s external insulation optimization design,the discharge current waveform of a 6 m rod-plane air gap under positive switching impulse voltage with 250µs and 1000µs time to crest is obtained.Discharge images and stressed voltage are combined to analyze the continuous feature of a current waveform under critical time to crest impulse and discontinuous feature under long front duration impulse.For the purposes of a lightning protection study,the current waveform of a 10 m rod-plane air gap is subjected to negative switching impulse.Finally,the pulse characteristics of the current corresponding to the single channel and branching stepped negative leader are discussed. | Yishi Yue Hengxin He Weijiang Chen Junjia He Chuanqi Wu Xiangen Zhao Feng Huo | 2015 | CSEE Journal of Power and Energy Systems2015,1,3: | 13 |
| 4 | Inhibition of genes expression of SARS coronavirus by synthetic small interfering RNAs显示文摘RNA interference (RNAi) is triggered by the presence of a double-stranded RNA (dsRNA), and results in the silencing of homologous gene expression through the specific degradation of an mRNA containing the same sequence. dsRNAmediated RNAi can be used in a wide variety of eucaryotes to induce the sequence-specific inhibition of gene expression.Synthetic 21-23 nucleotide (nt) small interfering RNA (siRNA) with 2 nt 3' overhangs was recently found to mediate efficient sequence-specific mRNA degradation in mammalian cells. Here, we studied the effects of synthetic siRNA duplexes targeted to SARS coronavirus structural proteins E, M, and N in a cell culture system. Among total 26 siRNA duplexes, we obtained 3 siRNA duplexes which could sequence-specifically reduce target genes expression over 80% at the concentration of 60 nM in Vero E6 cells. The downregulation effect was in correlation with the concentrations of the siRNA duplexes in a range of 0~60 nM. Our results also showed that many inactive siRNA duplexes may be brought to life simply by unpairing the 5' end of the antisense strands. | 5 | Establishment of a Screening System for Selection of siRNA Target Sites Directed against Hepatitis B Virus Surface Gene显示文摘To study the inhibitory effects of plasmid-derived small interfering RNA (siRNA) and synthetic siRNA on the expression of the hepatitis B virus surface (HBs) gene, three plasmid-derived siRNAs and one synthetic siRNA that complement the coding region of the HBs gene were prepared. The HBs expression plasmid pHBs-EGFP was also constructed. HeLa cells were co-transfected with pHBs-EGFP and the above siRNAs. The HBs mRNA quantities were measured by reverse-transcription PCR, and the level of HBs-EGFP fusion protein was quantified by fluorescent microscope. The concentrations of the hepatitis B virus surface antigen (HBsAg) derived from the culture supernatant of transfected HepG2.2.15 cells were measured by an enzyme-linked immunosorbent assay (ELISA) kit. The results showed that the three plasmid-derived siRNAs and the synthetic siRNA can effectively reduce the quantities of HBs mRNA and protein. The plasmid-derived siRNA psiRNAl was found to be the most effective inhibitor of HBs expression. It can inhibit HBs-EGFP expression by 63.3% and suppress HBs mRNA by 75.6%. To further substantiate the above observations, psiRNAl was transfected into HepG2.2.15 cells (an HBV secreting cell line). The transfections resulted in almost complete blockage of HBsAg production, whereas control vectortransfected cells secreted high levels of HBsAg 7 days post-transfection. | 6 | Multiple evidences indicate no relationship between prehistoric disasters in Lajia site and outburst flood in upper Yellow River valley, China显示文摘All around the world, ancient legends exist about the Great Flood, and the influence of the Great Flood in human evolutionary history is a hotly debated topic. In China, the legend of the prehistoric Great Flood and Yu the Great's flood control has a long history and is considered to be closely linked to the establishment of the Xia Dynasty. However, there is a lack of solid scientific evidence. Against this issue, some scholars have proposed that an earthquake around 1920 BCE in the Guanting Basin along the upper reaches of the Yellow River led to the creation of a lake by damming in the Jishi Gorge and that the outburst of water from that lake led to the massive flood in the Yellow River region. These studies provided new scientific evidence for the Chinese legend. In this paper, we date skeletal samples embedded in earthquake sand blasting from the Lajia site, analyze remains from natural disasters(such as earthquakes and floods) and also archaeological remains. In addition, we compared the skeletal samples dating with that of sediments from the dammed-up lake. Our results are inconsistent with those of previous scholars. The earthquake at the Lajia site occurred no earlier than 1800 BCE, and the dammed-up lake in Jishi Gorge had disappeared by 3600 BCE. Thus, the formation and outburst of the dammed lake, the sudden death of ancient humans at the Lajia site and the ancient earthquake were independent events occurring at different times. In addition, the massive flood in the upper reaches of the Yellow River did not actually happen. | 8 | Refined chronology of prehistoric cultures and its implication for re-evaluating human-environment relations in the Hexi Corridor,northwest China显示文摘The reconstruction of high-resolution chronologies for prehistoric cultures is a prerequisite for understanding the history of human evolution and its relationship with environmental change, and is valuable for exploring the trajectory of transcontinental cultural exchanges in prehistoric time. The Hexi Corridor of northwest China was one of the earliest centers for long-distance culture exchange in the prehistoric world. The timing and sequence of cultural changes in this area remains poorly understood resulting from the lack of radiometric age control. This paper presents a refined radiocarbon(14 C) chronology to resolve the timing of human occupation and cultural evolution in the Hexi Corridor. Radiocarbon dating of crop remains, which have an annual life cycle, has the advantage of eliminating problems such as the carbon stored in ‘old wood’. As a result,14C dates from crop remains are used to test the validity of the14C dates derived from charcoal, bone and plant remains. The resultant database of ages is modelled using the Bayesian approach. The updated chronology presented in this paper shows generally good agreement with the original dates used to constrain cultural evolution in the Hexi Corridor. However, the occupation of the corridor by the Shajing and Shanma Late Bronze Age-Early Iron Age cultures appears to have taken place ~300 years later than previously thought. | 11 | Unraveling the mechanisms of synapse formation and axon regeneration: the awesome power of C. elegans genetics显示文摘Since Caenorhabditis elegans was chosen as a model organism by Sydney Brenner in 1960's, genetic studies in this organism have been instrumental in discovering the function of genes and in deciphering molecular signaling network. The small size of the organism and the simple nervous system enable the complete reconstruction of the first connectome. The stereotypic developmental program and the anatomical reproducibility of synaptic connections provide a blueprint to dissect the mechanisms underlying synapse formation. Recent technological innovation using laser surgery of single axons and in vivo imaging has also made C. elegans a new model for axon regeneration. Importantly, genes regulating synaptogenesis and axon regeneration are highly conserved in function across animal phyla. This mini-review will summarize the main approaches and the key findings in understanding the mechanisms underlying the development and maintenance of the nervous system. | 13 | Large System Analysis of Downlink C-RAN with Phase Noise and Fronthaul Compression显示文摘This paper studies the effect of phase noise and fronthaul compression on a downlink cloud radio access network(C-RAN), where several remote radio heads(RRHs) are coordinated to communicate with users by a baseband unit(BBU) on the cloud server. In the system, the baseband signals are precoded at BBU, and then compressed before being transmitted to RRHs through capacity-limited fronthaul links which results in the compressive quantization noise. We assume the regularized zero-forcing precoding is performed with an imperfect channel state information and a compression strategy is applied at BBU. The effect of phase noise arising from nonideal local oscillators both at RRHs and users is considered. We propose an approximate expression for the downlink ergodic sum-rate of considered C-RAN utilizing large dimensional random matrix theory in the large-system regime. | 14 | TLR4 mediates LPS-induced HO-1 expression in mouse liver:Role of TNF-α and IL-1β显示文摘AIM: Heme oxygenase (HO)-I catalyzes the conversion of heme to biliverdin, iron and carbon monoxide. HO-1 is induced by many stimuli including heme, Hb, heat stress,lipopolysaccharide (LP5) and cytokines. Previous studies demonstrated that LP5 induced HO-1 gene activation and HO-1 expression in liver. However, the mechanisms of LPS-induced HO-1 expression in liver remain unknown. The effect of toll-like receptor-4 (TLR4) on LPS-induced liver HO-1 expression and the role of TNF-α and IL-1β in this condition were determined.METHODS: HO-1 expression was determined by immunofluorescent staining and immunoblotting. Double immunofluorescent staining was performed to determine the cell type of HO-1 expression in liver.RESULTS: A low dose of LPS significantly increased HO-1 expression in the liver which was localized in Kupffer cells only. Furthermore, HO-1 expression was enhanced by three doses of LPS. HO-1 expression was significantly inhibited in the liver of TLR4 mutant mice. While the liver HO-1 expression in TNF KO mice was much lower than that in C57 mice following the same LPS treatment, IL-1β KO had a slight influence on liver HO-1 expression following LPS treatment.CONCLUSION: The preserfl: results confirm that macrophages are the major source of HO-1 in the liver induced by LPS.This study demonstrates that TLR4 plays a dominant role in mediating HO-1 expression following LPS. | 15 | Particulate matter 2.5 triggers airway inflammation and bronchial hyperresponsiveness in mice by activating the SIRT2-p65 pathway显示文摘Exposure to particulate matter 2.5(PM2.5)potentially triggers airway inflammation by activating nuclear factor-κB(NF-κB).Sirtuin 2(SIRT2)is a key modulator in inflammation.However,the function and specific mechanisms of SIRT2 in PM2.5-induced airway inflammation are largely understudied.Therefore,this work investigated the mechanisms of SIRT2 in regulating the phosphorylation and acetylation of p65 influenced by PM2.5-induced airway inflammation and bronchial hyperresponsiveness.Results revealed that PM2.5 exposure lowered the expression and activity of SIRT2 in bronchial tissues.Subsequently,SIRT2 impairment promoted the phosphorylation and acetylation of p65 and activated the NF-κB signaling pathway.The activation of p65 triggered airway inflammation,increment of mucus secretion by goblet cells,and acceleration of tracheal stenosis.Meanwhile,p65 phosphorylation and acetylation,airway inflammation,and bronchial hyperresponsiveness were deteriorated in SIRT2 knockout mice exposed to PM2.5.Triptolide(a specific p65 inhibitor)reversed p65 activation and ameliorated PM2.5-induced airway inflammation and bronchial hyperresponsiveness.Our findings provide novel insights into the molecular mechanisms underlying the toxicity of PM2.5 exposure.Triptolide inhibition of p65 phosphorylation and acetylation could be an effective therapeutic approach in averting PM2.5-induced airway inflammation and bronchial hyperresponsiveness. | Manling Liu Zhaoling Shi Yue Yin Yishi Wang Nan Mu Chen Li Heng Ma Qiong Wang | 2021 | Frontiers of Medicine2021,15,5: | 2 |
| 16 | Efficient Virtual Network Embedding Algorithm Based on Restrictive Selection and Optimization Theory Approach显示文摘Network virtualization(NV) is widely considered as a key component of the future network and promises to allow multiple virtual networks(VNs) with different protocols to coexist on a shared substrate network(SN). One main challenge in NV is virtual network embedding(VNE). VNE is a NPhard problem. Previous VNE algorithms in the literature are mostly heuristic, while the remaining algorithms are exact. Heuristic algorithms aim to find a feasible embedding of each VN, not optimal or sub-optimal, in polynomial time. Though presenting the optimal or sub-optimal embedding per VN, exact algorithms are too time-consuming in smallscaled networks, not to mention moderately sized networks. To make a trade-off between the heuristic and the exact, this paper presents an effective algorithm, labeled as VNE-RSOT(Restrictive Selection and Optimization Theory), to solve the VNE problem. The VNERSOT can embed virtual nodes and links per VN simultaneously. The restrictive selection contributes to selecting candidate substrate nodes and paths and largely cuts down on the number of integer variables, used in the following optimization theory approach. The VNE-RSOT fights to minimize substrate resource consumption and accommodates more VNs. To highlight the efficiency of VNERSOT, a simulation against typical and stateof-art heuristic algorithms and a pure exact algorithm is made. Numerical results reveal that virtual network request(VNR) acceptance ratio of VNE-RSOT is, at least, 10% higher than the best-behaved heuristic. At the 2 nd International Symposium on Tribbles and Diseases held on May 7–9, 2018 in Beijing, China, a group of leading Tribbles scientists reported their findings and ongoing studies about the effects of the different TRIB proteins in the areas of immunity, metabolism, fundamental cell biology and cancer. Here, we summarize important and insightful overviews from 4 keynote lectures, 13 plenary lectures and 8 short talks that took place during this meeting.
