1Disruption of splicing-regulatory elements using CRISPR/Cas9 to rescue spinal muscular atrophy in human iPSCs and mice显示文摘We here report a genome-editing strategy to correct spinal muscular atrophy(SMA).Rather than directly targeting the pathogenic exonic mutations,our strategy employed Cas9 and guide-sg RNA for the targeted disruption of intronic splicing-regulatory elements.We disrupted intronic splicing silencers(ISSs,including ISS-N1 and ISS+100)of survival motor neuron(SMN)2,a key modifier gene of SMA,to enhance exon 7 inclusion and full-length SMN expression in SMA iPSCs.Survival of splicing-corrected iPSC-derived motor neurons was rescued with SMN restoration.Furthermore,co-injection of Cas9 mRNA from Streptococcus pyogenes(SpCas9)or Cas9 from Staphylococcus aureus(SaCas9)alongside their corresponding sgRNAs targeting ISS-N1 into zygotes rescued 56%and 100%of severe SMA transgenic mice(Smn-/-,SMN2tg/-).The median survival of the resulting mice was extended to>400 days.Collectively,our study provides proof-of-principle for a new strategy to therapeutically intervene in SMA and other RNA-splicing-related diseases.Jin-Jing Li Xiang Lin Cheng Tang Ying-Qian Lu Xinde Hu Erwei Zuo He Li Wenqin Ying Yidi Sun Lu-Lu Lai Hai-Zhu Chen Xin-Xin Guo Qi-Jie Zhang Shuang Wu Changyang Zhou Xiaowen Shen Qifang Wang Min-Ting Lin Li-Xiang Ma Ning Wang Adrian R.Krainer Linyu Shi Hui Yang Wan-Jin Chen 2020National Science Review2020,7,1:1
2Syndecan-4 functionalization of tissue regeneration scaffolds improves interaction with endothelial progenitor cells显示文摘Key to most implanted cell free scaffolds for tissue regeneration is the ability to sequester and retain undifferentiated mesenchymal stem cells at the repair site.In this report,syndecan-4,a heparan sulfate containing proteoglycan,was investigated as a unique molecule for use in scaffold functionalization.An electrospun hybrid scaffold comprised of poly(glycerol)sebacate(PGS),silk fibroin and type I collagen(PFC)was used as a model scaffold to develop a procedure and test the hypothesis that functionalization would result in increased scaffold binding of endothelial progenitor cells(EPCs).For these studies both Syndecan-4 and stromal derived factor-1a(SDF-1a)were used in functionalization PFC.Syndecan-4 functionalized PFC bound 4.8 fold more SDF-1a compared to nonfunctionalized PFC.Binding was specific as determined by heparin displacement studies.After culture for 7 days,significantly,more EPCs were detected on PFC scaffolds having both syndecan-4 and SDF-1a compared to scaffolds of PFC with only syndecan-4,or PFC adsorbed with SDF-1a,or PFC alone.Taken together,this study demonstrates that EPCs can be bound to and significantly expanded on PFC material through syndecan-4 mediated growth factor binding.Syndecan-4 with a multiplicity of binding sites has the potential to functionalize and expand stem cells on a variety of scaffold materials for use in tissue regeneration.Harleigh Warner Yidi Wu William D.Wagner 2021Regenerative Biomaterials2021,8,6:1

4In situ formation of lithiophilic Li_(22)Sn_(5) alloy and high Li-ion conductive Li_(2)S/Li_(2)Se via metal chalcogenide SnSSe for dendrite-free Li metal anodes显示文摘Lithium metal has gained extensive attention as the most ideal candidate for next-generation battery anode owing to the ultrahigh specific capacity and the lowest electrochemical potential.However,uncontrollable dendrite growth and huge volume variation extremely restrict the future deployment of lithium metal batteries.Herein,we report metal chalcogenide SnSSe with unique nanoplate stacking structure as a robust substrate for stable Li metal anode.During the initial Li plating process,lithiophilic Li_(22)Sn_(5) alloy and Li_(2)S/Li_(2)Se sites are obtained via in-situ electrochemical reaction of Li metal and SnSSe.Density functional theory(DFT)calculation demonstrates that the formed Li_(2)S/Li_(2)Se achieves low Li diffusion energy barrier,ensuring rapid Li~+migration.Li_(22)Sn_(5) alloy provides strong nucleation sites,promoting uniform Li nucleation.Furthermore,in-situ optical microscopy analysis suggests that the synthesized effect fundamentally inhibits lithium dendrite growth.Consequently,SnSSe modified Cu foil delivered an ultralow nucleation overpotential,superior cycling stability with 450 cycles(Coulombic efficiency,>98%),and excellent plating/stripping behavior over 2200 h at 0.5 mA cm^(-2).Moreover,the brilliant reversible cycles and rate capability were also realized in Li@SnSSe//LiFePO_(4)(LFP)full cell,shedding light on the feasibility of SnSSe for stable and dendrite-free lithium metal anode.Yaya Wang Yang Guo Jiang Zhong Meng Wang Lei Wang Shengyang Li Song Chen Hongli Deng Yong Liu Yidi Wu Jian Zhu Bingan Lu 2022Journal of Energy Chemistry2022,31,10:0
5Application of nitrogen loaded biochar in purifying agricultural wastewater and as a nitrogen releaser for rice production显示文摘Herein a new approach to the application of agricultural eutrophic wastewater for rice plant cultivation is described.Biochar was used as a medium for the sorption of ammonia from simulated wastewater and subsequently as a nitrogen(N)releaser in the cultivation of rice plants.The main goals of this approach were to isolate ammonia from simulated wastewater and transfer it into rice cultivation,and or explore how exogenous N promoted the growth of rice.The results demonstrate that according to X-ray diffraction phase analysis,most of the properties of biochar were retained before and after loading NH_(4)^(+)-N.Compared with biochar,the crystal peak of AlOOH in N-loaded biochar(NLB)disappeared and the intensity of the crystal peak of CuCaSe_(2)decreased,which was the important mechanism allowing it to adsorb 30.8%of the N present in simulated low N-concentration agricultural wastewater.The soil N content in NLB treatments was higher than in Non-NLB treatments during the critical tillering and reproductive growth stages.Moreover,the N adsorption-desorption process of NLB matched the N requirements of the rice plant,and thus greatly increased the tiller number by 11.9%and rice yield by 7.5%.These results indicated that the indirect use of ammonia derived from wastewater using biochar as a sorption and releasing medium for rice plant cultivation was promising.This is the first time that biochar was used for possibly indirect reuse of agricultural eutrophic wastewater and enhancement of rice plant growth.Hongyang Chen Yidi Sun Yang Sun Yanzhi Wang Yanqi Li Qi Wu Daocai Chi 2023International Journal of Agricultural and Biological Engineering2023,16,4:0
6Recent progress in co-detection of single-cell transcripts and proteins显示文摘Cellular heterogeneity is a universal property of living systems,and the interrogation of single cells facilitates in-depth understanding of distinct cellular states and functions in various biological processes.Co-analysis of transcripts and proteins from the same single cells opens the way to decipher complex RNA regulatory frameworks and phenotypes,facilitating the understanding of cellular fate and function regulations,discovery of novel cell types,and construction of a high-resolution cell atlas.Herein,we review the state-of-art advances in the development of methodologies for co-analysis of single-cell transcripts and proteins.First,imaging-based methods are summarized with particular emphasis on optical and mass spectrometry imaging.Next,sequencing-based approaches for high-throughput and sensitive co-analysis of single-cell transcripts and proteins are described,including droplet-,microwell-,and split-pool-based platforms.Subsequently,combined methods with more flexibility and universality are discussed.These methods commonly employ different strategies or reactions to convert transcripts and proteins of single cells into distinct signals simultaneously,which can be detected by different instruments or platforms.Lastly,some perspectives on the future challenges and development trends in this field are presented.Shanqing Huang Qian Fan Yidi Wang Zhi Huang Weixiong Shi Yanli Gong Ting Yang Jie Wang Lingling Wu Chaoyong Yang 2024Nano Research2024,17,1:0
7Fast period estimation of X-ray pulsar signals using an improved fast folding algorithm显示文摘An accurate period is important to recover the pulse profile from a recorded photon event series of an X-ray pulsar and to estimate the pulse time of arrival,which is the measurement of X-ray pulsar navigation.Epoch folding is a classical period estimation method in the time domain;however,its computational complexity grows as the number of trail periods increases.In order to reduce the computational complexity,this paper improves the fast folding algorithm through segment correlation and amplitude accumulation,which is based on the post-order traversal of a binary tree.Compared with epoch folding,the improved fast folding algorithm can achieve a similar accuracy at the cost of a lower computational burden.Compared with the original fast folding algorithm,the improved algorithm can be applied to detectors with a much smaller effective area.The performance of the method is investigated by simulation data and observation data from the Neutron star Interior Composition Explorer(NICER).Minzhang SONG Yidi WANG Wei ZHENG Liansheng LI Yusong WANG Xiaowei HU Yulong WU 2023Chinese Journal of Aeronautics2023,36,10:0
