Preoperative neutrophil-to-lymphocyte ratio plus platelet-to-lymphocyte ratioin predicting survival for patients with stage Ⅰ-Ⅱgastric cancer显示文摘Background:The preoperative neutrophil-to-lymphocyte ratio(NLR) and the platelet-to-lymphocyte ratio(PLR) are associated with poor prognosis of gastric cancer. We aimed to determine whether the combination of NLR and PLR(NLR-PLR) could better predict survival of patients after curative resection for stage Ⅰ-Ⅱ gastric cancer. Methods:We collected data from the medical records of patients with stage Ⅰ-Ⅱ gastric cancer undergoing curative resection between December 2000 and November 2012 at the Sun Yat-sen Cancer Center. The preoperative NLRPLR was calculated as follows:patients with both elevated NLR(≥2.1) and PLR(≥ 120) were given a score of 2,and patients with only one or neither were given a score of 1 or 0,respectively.Results:Kaplan-Meier analysis and log-rank tests revealed significant differences in overall survival(OS) among patients with NLR-PLR scores of 0,1 and 2(P < 0.001).Multivariate analysis showed that OS was independently associated with the NLR-PLR score[hazard ratio(HR) = 1.51,95%confidence interval(CI) 1.02-2.24,P = 0.039]and TNM stage(HR = 1.36,95%CI 1.01-1.83,P= 0.041).However,other systemic inflammation-based prognostic scores, including the modified Glasgow prognostic score, the prognostic nutritional index, and the combination of platelet count and NLR,were not.In TNM stage-stratified analysis, the prognostic significance of NLR-PLR was maintained in patients with stage Ⅰ(P < 0.001) and stage Ⅱ cancers(P= 0.022).In addition, the area under the receiver operating characteristic curve for the NLR-PLR score was higher than those of other systemic inflammation-based prognostic scores(P = 0.001).Conclusion:The preoperative NLR-PLR score is a useful predictor of postoperative survival in the patients with stage l-ll gastric cancer and may help identify high-risk patients for rational therapy and timely follow-up.Xiaowei Sun Xuechao Liu Jianjun Liu Shangxiang Chen Dazhi Xu Wei Li Youqing Zhan Yuanfang Li Yingbo Chen Zhiwei Zhou 2016Chinese Journal of Cancer2016,35,7:25
Prognostic nutritional index is an independent prognostic factor for gastric cancer patients with peritoneal dissemination显示文摘Objective: The predictive and prognostic role of prognostic nutritional index(PNI) in gastric cancer patients with peritoneal dissemination remains unclear. This study aims to explore the role of the PNI in predicting outcomes of gastric cancer patients with peritoneal dissemination. Methods: A total of 660 patients diagnosed with gastric adenocarcinoma with peritoneal metastasis between January 2000 and April 2014 at Sun Yat-sen University Cancer Center and the Sixth Affiliated Hospital of Sun Yatsen University were retrospectively analyzed. The clinicopathologic characteristics and clinical outcomes of patients with peritoneal dissemination were analyzed. Results: Compared with PNI-high group, PNI-low group was correlated with advanced age(P=0.036), worse performance status(P<0.001), higher frequency of ascites(P<0.001) and higher frequency of multisite distant metastasis(P<0.001). Kaplan-Meier survival curves showed that PNI-high group had a significantly longer median overall survival than PNI-low group(13.13 vs. 9.03 months, P<0.001). Multivariate survival analysis revealed that Borrmann type IV(P=0.014), presence of ascites(P=0.017) and lower PNI(P=0.041) were independent poor prognostic factors, and palliative surgery(P<0.001) and first-line chemotherapy(P<0.001) were good prognostic factors. For patients receiving palliative surgery, the postoperative morbidity rates in the PNI-low group and PNIhigh group were 9.1% and 9.9%, respectively(P=0.797). The postoperative mortality rate was not significantly different between PNI-low and PNI-high groups(2.3% vs. 0.9%, P=0.362).Conclusions: PNI is a useful and practical tool for evaluating the nutritional status of gastric cancer patients with peritoneal dissemination, and is an independent prognostic factor for these patients.Runcong Nie Shuqiang Yuan Shi Chen Xiaojiang Chen Yongming Chen Baoyan Zhu Haibo Qiu Zhiwei Zhou Junsheng Peng Yingbo Chen 2016Chinese Journal of Cancer Research2016,28,6:19
A discussion on gas sources of the Feixianguan Formation H_2S-rich giant gas fields in the northeastern Sichuan Basin显示文摘In recent years several H2S-rich oolite giant gas pools have been discovered in the Lower Triassic Feixianguan Formation of the northeastern Sichuan basin, and their explored gas reserves have been over 5000×108 m3. However, gas sources remain unsolved due to multiple source horizons with high maturity in this area and TSR alterations. By integrating analytical data of natural gas samples with conprehensive investigations on many factors, such as oil-gas geology, distribution and evolution of source rocks, charging and adjustment of gas pools, mixture of natural gases and secondery alterations, the present study concluded that the dominant source for the Feixianguan Fr. gas pools is the Permian Longtan Fr. source rock and secondly the Silurian Longmaxi Fr. source rock. Natural gases from the various gas pools differ genetically due to the matching diversity of seal configurations with phases of hydrocarbon generation and expulsion by different source rocks, among which natural gases in Puguang Gasfield are dominated by the trapped gas generated from the Longtan Fr. source rock and commingled with the gas cracked from the Silurian crude oil, while those in Dukouhe, Tieshangpo and Luojiazhai Gasfields are composed mainly of the Silurian oil-cracking gas and commingled with the natural gas derived from the Longtan Fr. source rock.ZHANG ShuiChang ZHU GuangYou CHEN JianPing LIANG YingBo 2007Chinese Science Bulletin2007,52,A01:12
Gossypol: phytoalexin of cotton显示文摘Sesquiterpenoids are a class of 15-carbon secondary metabolites that play diverse roles in plant adaptation to environment.Cotton plants accumulate a large amount of sesquiterpene aldehydes(including gossypol)as phytoalexins against pathogens and herbivores. They are stored in pigment glands of aerial organs and in epidermal layers of roots.Several enzymes of gossypol biosynthesis pathway have been characterized, including 3-hydroxy-3-methylglutaryl coenzyme A reductase(HMGR)and farnesyl diphosphate synthase(FPS)that catalyze the formation of the precursor farnesyl diphosphate(FPP),(+)-δ-cadinene synthase(CDN)which is the first enzyme committed to gossypol biosynthesis, and the downstream enzymes of CYP706B1 and methyltransferase.Expressions of these genes are tightly regulated during cotton plants development and induced by jasmonate and fungi elicitors. The transcription factor Ga WRKY1 has been shown to be involved in gossypol pathway regulation.Recent development of new genomic platforms and methods and releases of diploid and tetraploid cotton genome sequences will greatly facilitate the elucidation of gossypol biosynthetic pathway and its regulation.Xiu Tian Juxin Ruan Jinquan Huang Xin Fang Yingbo Mao Lingjian Wang Xiaoya Chen Changqing Yang 2016Science China(Life Sciences)2016,59,2:10

Analysis and external validation of a nomogram to predict peritoneal dissemination in gastric cancer显示文摘Objective:Peritoneal dissemination is difficult to diagnose by conventional imaging technologies. We aimed to construct a nomogram to predict peritoneal dissemination in gastric cancer(GC)patients. Methods:We retrospectively analyzed 1,112 GC patients in Sun Yat-sen University Cancer Center between2001 and 2010 as the development set and 474 patients from The Sixth Affiliated Hospital,Sun Yat-sen University between 2010 and 2016 as the validation set.The clinicopathological variables associated with gastric cancer with peritoneal dissemination(GCPD)were analyzed. We used logistic regression analysis to identify independent risk factors for peritoneal dissemination. Then,we constructed a nomogram for the prediction of GCPD and defined its predictive value with a receiver operating characteristic(ROC)curve.External validation was performed to validate the applicability of the nomogram. Results:In total,250 patients were histologically identified as having peritoneal dissemination.Logistic regression analysis demonstrated that age, sex, tumor location, tumor size,signet-ring cell carcinoma(SRCC),T stage,N stage and Borrmann classification IV(Borrmann IV)were independent risk factors for peritoneal dissemination. We constructed a nomogram consisting of these eight factors to predict GCPD and found an optimistic predictive capability, with a C-index of 0.791,an area under the curve(AUC)of 0.791,and a 95%confidence interval(95%CI)of 0.762-0.820.The results found in the external validation set were also promising. Conclusions:We constructed a highly sensitive nomogram that can assist clinicians in the early diagnosis of GCPD and serve as a reference for optimizing clinical management strategies.Xijie Chen Shi Chen Xinyou Wang Runcong Nie Dongwen Chen Jun Xiang Yijia Lin Yingbo Chen Junsheng Peng 2020Chinese Journal of Cancer Research2020,32,2:6
A nomogram to predict prognosis for gastric cancer with peritoneal dissemination显示文摘Objective: To identify independent prognostic factors to be included in a nomogram to predict the prognosis of gastric cancer patients with peritoneal dissemination. Methods: This is a retrospective study on 684 patients with a histological diagnosis of gastric cancer with peritoneal dissemination from the Sun Yat-sen University Cancer Center as the development set, and 62 gastric cancer patients from the Sixth Affiliated Hospital of Sun Yat-sen University as the validation group. Chi-square test and Cox regression analysis were used to compare the clinicopathological variables and the prognosis of gastric cancer patients with peritoneal dissemination. The Harrell's concordance index(C-index) and calibration curve were determined for comparisons of predictive ability of the nomogram. Results: Univariate and multivariate analyses showed that serum carcinoembryonic antigen(CEA) level(P=0.032), ascites grading(P=0.008), presence of extraperitoneal metastasis(P<0.001), seeding status(P=0.016) and performance status(P=0.009) were independent prognostic factors for gastric cancer patients with peritoneal dissemination in the development set. The nomogram model was constructed using these five factors. Internal validation showed that the C-index of the model was 0.641. For the external validation, the C-index of this model was 0.709.Conclusions: We developed and validated a nomogram to predict the prognosis for gastric cancer patients with peritoneal dissemination. This nomogram may play an important clinical role in guiding palliative therapy for these types of patients, although it may need more data for optimization.Shi Chen Xijie Chen Runcong Nie Liying Ou Yang Aihong Liu Yuanfang Li Zhiwei Zhou Yingbo Chen Junsheng Peng 2018Chinese Journal of Cancer Research2018,30,4:5
Transcriptome analysis of three cotton pests reveals features of gene expressions in the mesophyll feeder Apolygus lucorum显示文摘The green mirid bug Apolygus lucorum is an agricultural pest that is known to cause damage to more than 150 plant species. Here,we report the transcriptomes of A.lucorum at three different developmental stages(the second and fifth instar nymphs and adults).A total of 98,236 unigenes with an average length of l,335 nt was obtained,of which 50,640 were annotated, including those encoding digestive enzymes and cytochrome P450 s.Comparisons with cotton bollworm and cotton aphid transcriptomes revealed distinct features of A.lucorum as a mesophyll feeder. The gene expression dynamics varied during development from young nymphs to adults. The high-quality transcriptome data and the gene expression dynamics reported here provide valuable data for a more comprehensive understanding of the physiology and development of mirid bugs, and for mining targets for their control.Dianyang Chen Fangyan Chen Chunyu Chen Xiaoya Chen Yingbo Mao 2017Science China(Life Sciences)2017,60,8:4
Prognostic potential of an immune score based on the density of CD8+ T cells, CD20+ B cells, and CD33+/p-STAT1+ double-positive cells and HMGB1 expression within cancer nests in stage ⅢA gastric cancer patients显示文摘Objective: There is heterogeneity in the prognosis of gastric cancers staged according to the tumornodes-metastasis(TNM) system. This study evaluated the prognostic potential of an immune score system to supplement the TNM staging system. Methods: An immunohistochemical analysis was conducted to assess the density of T cells, B cells, and myeloid-derived suppressor cells(MDSCs) in cancer tissues from 100 stage ⅢA gastric cancer patients; the expression of the high-mobility group protein B1(HMGB1) was also evaluated in cancer cells. The relationship between the overall survival(OS), disease-free survival(DFS), and immunological parameters was analyzed. Results: An immune score system was compiled based on the prognostic role of the density of T cells, B cells, MDSCs, and the expression of HMGB1 in cancer tissues. The median 5-year survival of this group of patient was 32%.However, the 5-year survival rates of 80.0%, 51.7%, 0%, 5.8%, and 0% varied among the patients with an immune score of 4 to those with an immune score of 0 based on the immune score system, respectively.Similarly, differences in DFS rates were observed among the immune score subgroups. Conclusions: An immune score system could effectively identify the prognostic heterogeneity within stage ⅢA gastric cancer patients, implying that this immune score system may potentially supplement the TNM staging system, and help in identifying a more homogeneous group of patients who on the basis of prognosis can undergo adjuvant therapy.Jun Dong Jiao Li Shiming Liu Xingyu Feng Shi Chen Zhiwei Zhou Yingbo Chen Xiaoshi Zhang 2016Chinese Journal of Cancer Research2016,28,5:3
Synthesis of novel hierarchical porous polymers with a nanowire-interconnected network structure from core-shell polymer nanoobjects显示文摘Design and fabrication of the micro/nanostructures of the network units is a critical issue for porous nanonetwork structured materials. Significant progress has been attained in construction of the network units with zero-dimensional spherical shapes. However, owing to the limitations of synthetic methods, construction of porous building blocks in one dimension featuring high aspect ratios for porous nanonetwork structured polymer(PNSP) remains largely unexplored. Here we present the successful design and preparation of PNSP with a novel type of one-dimensional network unit, i.e., microporous heterogeneous nanowire. Well-defined core-shell polymer nanoobjects prepared from a gelable block copolymer, poly(3-(triethoxysilyl)propyl methacrylate)-block-polystyrene are employed as building blocks, and facilely transformed into PNSP via hypercrosslinking of polystyrene shell. The as-prepared PNSP exhibits unique three-dimensional hierarchical nanonetwork morphologies with large surface area. These findings could provide a new avenue for fabrication of unique well-defined PNSP, and thus generate valuable breakthroughs in many applications.yeru liang yingbo ruan junlong huang bo peng chen-yang liu ruowen fu mingqiu zhang yongming chen dingcai wu 2017Science China Chemistry2017,60,8:2
Experimental investigation and prediction model for UCS loss of unsaturated sandstones under freeze-thaw action显示文摘Sandstone is widely distributed in cold regions and the freeze-thaw deterioration of them has caused many geological engineering disasters. As an important and direct index of frost resistance, the strength loss of sandstones under freeze-thaw actions should be investigated to provide a guidance for the stability assessment of geological engineering. In this research, the UCS(Uniaxial compressive strength)loss of six typical sandstones with different water contents after 0,20,40 and 60 freeze-thaw cycles was measured in the laboratory. The experimental results indicated that the freeze-thaw damage was more serious in sandstones containing high water contents, and the critical saturations for causing a significant loss of UCS under freeze-thaw were 60%-80%for these sandstones.Below this critical saturation, the UCS loss of the sandstones was mainly caused by water weakening rather than freeze-thaw damage.Besides,a developed strength prediction model was proposed by combining the exponential decay function and multiple linear regression method. The initial porosity, elastic modulus and tensile strength of fresh sandstones were a good parameter combination to accurately determine the decay constant in this developed model. The main novelty of this model is that it can accurately and easily estimate the UCS loss of sandstones after any freeze-thaw cycle only using the initial parameters of fresh sandstones, but it does not need to perform freeze-thaw and mechanical strength experiments. This study not only provides an accurate prediction model of UCS under freeze-thaw,but also makes a contribution to better understanding the frost resistance mechanism of sandstones.Shibing Huang Yingbo He Shilin Yu Chen Cai 2022International Journal of Mining Science and Technology2022,32,1:2

Aryl hydrocarbon receptor activation drives polymorphonuclear myeloid-derived suppressor cell response and efficiently attenuates experimental Sjögren’s syndrome显示文摘Myeloid-derived suppressor cells(MDSCs)comprise heterogeneous myeloid cell populations with immunosuppressive capacity that contribute to immune regulation and tolerance induction. We previously reported impaired MDSC function in patients with primary Sjögren’s syndrome(pSS)and mice with experimental SS(ESS).However, the molecular mechanisms underlying MDSC dysfunction remain largely unclear. In this study,we first found that aryl hydrocarbon receptor(AhR)was highly expressed by human and murine polymorphonuclear MDSCs(PMN-MDSCs).Indole-3-propionic acid(IPA),a natural AhR ligand produced from dietary tryptophan, significantly promoted PMN-MDSC differentiation and suppressive function on CD4^(+)T cells. In contrast, feeding a tryptophan-free diet resulted in a decreased PMN-MDSC response,a phenotype that could be reversed by IPA supplementation. The functional importance of PMN-MDSCs was demonstrated in ESS mice by using a cell-depletion approach.Notably,AhR expression was reduced in PMN-MDSCs during ESS development, while AhR antagonism resulted in exacerbated ESS pathology and dysregulated T effector cells, which could be phenocopied by a tryptophan-free diet.Interferon regulatory factor 4(IRF4),a repressive transcription factor, was upregulated in PMN-MDSCs during ESS progression.Chromatin immunoprecipitation analysis revealed that IRF4 could bind to the promoter region of AhR,while IRF4 deficiency markedly enhanced AhR-mediated PMN-MDSC responses.Furthermore, dietary supplementation with IPA markedly ameliorated salivary glandular pathology in ESS mice with restored MDSC immunosuppressive function.Together, our results identify a novel function of AhR in modulating the PMN-MDSC response and demonstrate the therapeutic potential of targeting AhR for the treatment of pSS.Yanxia Wei Na Peng Chong Deng Futao Zhao Jie Tian Yuan Tang Sulan Yu Yacun Chen Yu Xue Fan Xiao Yingbo Zhou Xiaomei Li Hejian Zou Ke Rui Xiang Lin Liwei Lu 2022Cellular & Molecular Immunology2022,19,12:1
