| 1 | Differential gene expression profile of Buyanghuanwu decoction in rats with ventricular remodeling post-myocardial infarction显示文摘OBJECTIVE: To investigate the effect of Buyanghuanwu decoction(BYHWD) on gene expression in ventricular remodeling post-myocardial infarction in rats.METHODS: Animal models of myocardial infarction were established by permanent ligation of the left anterior descending coronary artery. Echocardiography measurements were performed after the treatment of BYHWD(18 g·kg-1 collagen was observ·d-1) for 90 days.Myocardialed by mallory trichrome staining. Capillary density was quantified by using Factor rentially expⅧre immunohistochemical staining.Diffessed genes were explored by a short-read sequencing technology combined with a tag-based digital gene expression profiling(DGE)system. Real-time quantitative polymerase chain reaction detecting system(q PCR) was used to validate the sequencing results. After assembling the gene information from Sham, model and BYHWD groups, we constructed three DGE libraries based on each group. The sequencing of three libraries generated 66 000-73 000 unique tags, which were mapped to reference sequences for annotation of expressed genes.RESULTS: Among them, 511 and 352 differentially expressed genes were found in comparison with sham/model and model/BYHWD, respectively. Fifty-five genes exhibited reversed direction of gene expression differences between Sham/Model and Model/BYHWD groups. We found that transforming growth factor beta receptor-1, junctophilin-2,monocyte chemotactic protein 1, neuropeptide Y,arachidonate 5-Lipoxygenase, arachidonate 15-Lipoxygenase were significantly modulated, which suggested the involvement of these genes in BYHWD treatment.CONCLUSION: The DGE profiling data provide comprehensive gene expression information at the transcriptional level that could facilitate our understanding of the pharmacological mechanisms of BYHWD in ventricular remodeling post-myocardial infarction. | Zhang Tao Hua Yue Luo Hao Chen Hongmei Shao Meng Fu Xiuqiong John Man-Tak Chu Huang Guiqiong Liu Bin Zhou Yingchun | 2017 | Journal of Traditional Chinese Medicine2017,37,3: | 1 |
| 2 | Research Progress on the Problem of Fluid,Heat and Energy Distribution near the Earthquake Source Area显示文摘As the basic problems in seismology, fluid, heat and energy distribution near earthquake sources during earthquake generation have been the leading subjects of concern to seismologists. Currently, more and more research shows fluid around earthquake source areas, which plays an important role in the process of earthquake preparation and generation. However, there is considerable controversy over the source of fluid in the deep crust. As for the problem of heat around earthquake source areas, different models have been proposed to explain the stress heat flow paradox. Among them, the dynamic weakening model has been thought to be the key to solving the heat flow paradox issue. After large earthquakes, energy distribution is directly related to friction heat. It is of timely and important practical significance to immediately implement deep drilling in-site surveying to gain understanding of fluid, friction heat and energy distribution during earthquake generation. | 3 | Effect of vortex dynamics and instability characteristics on the induced drag of trailing vortices显示文摘In this paper, trailing vortices generated by three wingtip configurations, namely the M6wing and the M6 wing with a blended or split winglet, are experimentally investigated using the Stereo Particle Image Velocimetry(SPIV) technology. Then, linear stability analysis is performed to investigate instability characteristics. Three corresponding trailing vortex patterns, including the isolated trailing vortex without wake(pattern v) and with wake(pattern v-w), co-rotating vortex pair(pattern v-v), are observed in experiments. The strength of trailing vortices, characterized by circulation, is reduced after installing winglets as expected, and the strength of pattern v-v can be further suppressed compared with pattern v-w. Moreover, instability characteristics, such as the eigenvalue spectrum and perturbation mode, are distinctive among these three vortex patterns.The distribution of eigenvalue spectrums indicates that pattern v and pattern v-w are temporally“marginally stable”, while pattern v-v is temporally “unstable.” The primary perturbation mode of pattern v and pattern v-w is the m =-1 helical mode, while |m|>1 for the case of pattern v-v.The effect of vortex dynamics and instability characteristics can be concluded in two aspects.Firstly, the value of induced drag is polluted by about 3% from vortex wandering since vortex wandering affects the tangential velocity and streamwise vorticity of trailing vortices. Secondly, the growth rate and penetration depth perturbation mode affect trailing vortex evolution and further affect induced drag. Specifically, the larger the growth rate and penetration depth are, the more turbulence injects inside the vortex core, thus leading to a quicker and more intense attenuation of trailing vortex, as well as a smaller induced drag. | 4 | Long non-coding RNA-AK138945 regulates myocardial ischemia-reperfusion injury via the miR-1-GRP94 signaling pathway显示文摘Objective:Myocardial ischemia-reperfusion injury(MIRI)is one of the leading causes of death from cardiovascular disease in humans,especially in individuals exposed to cold environments.Long non-coding RNAs(lncRNAs)regulate MIRI through multiple mechanisms.This study explored the regulatory effect of lncRNA-AK138945 on myocardial ischemia-reperfusion injury and its mechanism.Methods:In vivo,8-to 12-weeks-old C57BL/6 male mice underwent ligation of the left anterior descending coronary artery for 50 minutes followed by reperfusion for 48 hours.In vitro,the primary cultured neonatal mouse ventricular cardiomyocytes(NMVCs)were treated with 100μmol/L hydrogen peroxide(H_(2)O_(2)).The knockdown of lncRNA-AK138945 was evaluated to detect cardiomyocyte apoptosis,and a glucose-regulated,endoplasmic reticulum stress-related protein 94(GRP94)inhibitor was used to detect myocardial injury.Results:We found that the expression level of lncRNA-AK138945 was reduced in MIRI mouse heart tissue and H2O2-treated cardiomyocytes.Moreover,the proportion of apoptosis in cardiomyocytes increased after lncRNA-AK138945 was silenced.The expression level of Bcl2 protein was decreased,and the expression level of Bad,Caspase 9 and Caspase 3 protein was increased.Our further study found that miR-1a-3p is a direct target of lncRNA-AK138945,after lncRNA-AK138945 was silenced in cardiomyocytes,the expression level of miR-1a-3p was increased while the expression level of its downstream protein GRP94 was decreased.Interestingly,treatment with a GRP94 inhibitor(PU-WS13)intensified H2O2-induced cardiomyocyte apoptosis.After overexpression of FOXO3,the expression levels of lncRNA-AK138945 and GRP94 were increased,while the expression levels of miR-1a-3p were decreased.Conclusion:LncRNA-AK138945 inhibits GRP94 expression by regulating miR-1a-3p,leading to cardiomyocyte apoptosis.The transcription factor Forkhead Box Protein O3(FOXO3)participates in cardiomyocyte apoptosis induced by endoplasmic reticulum stress through up-regulation of lncRNA-AK138945. | Yanying Wang Jian Huang Han Sun Jie Liu Yingchun Shao Manyu Gong Xuewen Yang Dongping Liu Zhuo Wang Haodong Li Yanwei Zhang Xiyang Zhang Zhiyuan Du Xiaoping Leng Lei Jiao Ying Zhang | 2024 | Frigid Zone Medicine2024,4,1: | 0 |
