| 1 | Complementation of Wild-Type and Drug-Resistant Hepatitis B Virus Genomes to Maintain Viral Replication and Rescue Virion Production under Nucleos(t)ide Analogs显示文摘As the open reading frames of hepatitis B virus(HBV)genomes are overlapping,resistance mutations(MTs)in HBV polymerase may result in stop codon MTs in hepatitis B surface proteins,which are usually detected as a mixed population with wild-type(WT)HBV.The question was raised how the coexistence of nucleos(t)ide analogs(NAs)resistance MTs and WT sequences affects HBV replication.In the present study,HBV genomes with frequently detected reverse transcriptase(RT)/surface truncation MTs,rtA181T/sW172^*,rtV191I/sW182^*and rtM204I/sW196^*,were phenotypically characterized alone or together with their WT counterparts in different ratios by transient transfection in the absence or presence of Nas.In the absence of Nas,RT/surface truncation MTs impaired the expression and secretion of HBV surface proteins,and had a dose-dependent negative effect on WT HBV virion secretion.However,in the presence of Nas,coexistence of MTs with WT maintained viral replication,and the presence of WT was able to rescue the production of MT HBV virions.Our findings reveal that complementation of WT and MT HBV genomes is highly effective under drug treatment. | Chunchen Wu Baolin Li Xiaoyong Zhang Kaitao Zhao Yingshan Chen Yifei Yuan Yan Liu Rongjuan Chen Dongping Xu Xinwen Chen Mengji Lu | 2019 | Virologica Sinica2019,34,4: | 4 |

| 3 | How do GPM and TRMM precipitation products perform in alpine regions?A case study in northwestern China's Qilian Mountains显示文摘Satellite technologies provide valuable areal precipitation datasets in alpine mountains.However,coarse resolution still limits the use of satellite precipitation datasets in hydrological and meteorological research.We evaluated different time scales and precipitation magnitudes of Tropical Rainfall Measurement Mission 3B43 V7(TRMM)and Global Precipitation Measurement(GPM)products for alpine regions using ground precipitation datasets from January 2015 to June 2019 obtained from 25 national meteorological stations and 11 sets of T-200B weighing precipitation gauges in the Qilian Mountains.The results indicated that GPM outperformed TRMM at all temporal scales at an elevation<3500 m with a higher probability of detection(POD),false alarm ratio(FAR),and frequency bias index(FBI)and performed best at 3000 m;TRMM performed better than GPM at an elevation>3500 m,with the best performance at 4000 m.GPM and TRMM had the best estimation accuracy in areas with monthly precipitation of 30 mm and 40 mm,respectively.Both TRMM and GPM products underestimated mid to large daily precipitation and overestimated light daily precipitation averaging<2 mm/d.This research not only emphasizes the superiority of GPM/TRMM in different regions but also indicates the limitations of precipitation algorithms. | SUN Weijun CHEN Rensheng WANG Lei WANG Yingshan HAN Chuntan HUAI Baojuan | 2022 | Journal of Geographical Sciences2022,32,5: | 1 |
| 4 | Grafting Propagation Techniques of Red Sandalwood(Pterocarpus santalinus),a Precious Hongmu Tree Species显示文摘[Objectives] This article aimed to study the grafting propagation techniques of red sandalwood ( Pterocarpus santalinus ), so as to provide technical support for the effective propagation of seedlings of this precious Hongmu species.[Methods] Based on the analysis and comparison of the growth performance of the Hongmu tree species introduced to China, taking 1 to 2-year-old Pterocarpus indicus seedlings as rootstocks and single-bud stems of P. santalinus as scions, grafting was carried out from January to April using three grafting propagation methods, cut grafting propagation, skin grafting propagation and oblique grafting propagation.[Results] The results of several years of experiments show that in early spring, P. santalinus could be propagated well using the grafting propagation methods of skin grafting propagation and oblique grafting propagation, with survival rate over 80%. For rootstocks with young age or thin stems, the joint unions of the oblique grafting propagation healed well, without apparent protrusions, unlike those of skin grafting propagation. The survival rate of oblique grafting propagation that is fast and reliable was higher than that of cut grafting propagation.[Conclusions] It is feasible to use the grafting propagation methods to propagate P. santalinus seedlings. | 9 | Obesity-induced inflammatory miR-133a mediates apoptosis of granulosa cells and causes abnormal folliculogenesis显示文摘Obesity has been reported to promote disordered folliculogenesis,but the exact molecular mechanisms are still not fully understood.In this study,we find that miR-133a is involved in obesity-induced follicular development disorder.After feeding with a high-fat diet(HFD)and fructose water for nine weeks,the mouse body weight is significantly increased,accompanied by an inflammatory state and increased expression of miR-133a in the adipose tissues and ovaries as well as accelerated follicle depletion.Although miR-133a is increased in the fat and ovaries of HFD mice,the increased miR-133a in the HFD ovaries is not derived from exosome transferred from obese adipose tissues but is synthesized by ovarian follicular cells in response to HFD-induced inflammation.In vivo experiments show that intrabursal injection of miR-133a agomir induces a decrease in primordial follicles and an increase in antral follicles and atretic follicles,which is similar to HFD-induced abnormal folliculogenesis.Overexpression of miR-133a modestly promotes granulosa cell apoptosis by balancing the expression of anti-apoptotic proteins such as C1QL1 and XIAP and pro-apoptotic proteins such as PTEN.Overall,this study reveals the function of miR-133a in obesityinduced ovarian folliculogenesis dysfunction and sheds light on the etiology of female reproductive disorders. | Ruizhi Chen Xueqing Wu Han Qiu Baiming Yang Yao Chen Xiang Chen Yingshan Li Shaochun Yuan Dan Liu Luanjuan Xiao Yanhong Yu | 2023 | Acta Biochimica et Biophysica Sinica2023,55,8: | 0 |
| 10 | Comparison of viral propagation and drug response among SARS-CoV-2 VOCs using replicons capable of recapitulating virion assembly and release显示文摘Several variants of concern(VOCs)have emerged since the WIV04 strain of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)was first isolated in January 2020.Due to mutations in the spike(S)protein,these VOCs have evolved to enhance viral infectivity and immune evasion.However,whether mutations of the other viral proteins lead to altered viral propagation and drug resistance remains obscure.The replicon is a noninfectious viral surrogate capable of recapitulating certain steps of the viral life cycle.Although several SARS-CoV-2 replicons have been developed,none of them were derived from emerging VOCs and could only recapitulate viral genome replication and subgenomic RNA(sgRNA)transcription.In this study,SARS-CoV-2 replicons derived from the WIV04 strain and two VOCs(the Beta and Delta variants)were prepared by removing the S gene from their genomes,while other structural genes remained untouched.These replicons not only recapitulate viral genome replication and sgRNA transcription but also support the assembly and release of viral-like particles,as manifested by electron microscopic assays.Thus,the S-deletion replicon could recapitulate virtually all the post-entry steps of the viral life cycle and provides a versatile tool for measuring viral intracellular propagation and screening novel antiviral drugs,including inhibitors of virion assembly and release.Through the quantification of replicon RNA released into the supernatant,we demonstrate that viral intracellular propagation and drug response to remdesivir have not yet substantially changed during the evolution of SARS-CoV-2 from the WIV04 strain to the Beta and Delta VOCs. | Lingqian Tian Qiuhong Liu Rongjuan Pei Yingshan Chen Chonghui Xu Jielin Tang Hao Sun Kunpeng Liu Qi Yang Lei Yang Leshan Li Yongli Zhang Yuan Zhou Chao Shan Xue Hu Xinwen Chen Yun Wang | 2022 | Virologica Sinica2022,37,5: | 0 |
