1Loss of C-terminal α-helix decreased SDF-1α-mediated signaling and chemotaxis without influencing CXCR4 internalization显示文摘AIM: To investigate the possibility that a novel α-helix-defective mutant of stromal cell-derived factor-1α (SDF- 1α) (SDF-1/54R) acts as an antagonist of CXC chemokine receptor 4 (CXCR4). METHODS: According to the genetic sequence of natural SDF-1α, a recombinant α-helix-defective mutant of SDF-1α was designed and some biologic characteristics of this mutant were demonstrated. The migration of Jurkat cells was assessed with chemo- tactic assay. ERK phosphorylation was analyzed by Western blot with a specific anti-phospho-ERK1/2 antibody. Intracellular calcium influx was examined by flow cytometer with a calcium indicator dye Fluo-3AM. The CXCR4 on the cell surface was detected by flow cytometer with a PE conjoined anti-human CXCR4 antibody. RESULTS: Compared with native SDF-1α, SDF-1/54R displayed apparent decrease in chemotactic ability, ERK1/2 activation, and intracellular calcium influx in Jurkat cells. However, the binding to CXCR4 and inducing CXCR4 internalization of SDF-1/54R did not change outstandingly. Moreover, a competitive inhibitory effect of SDF-1/54R on the migration of Jurkat cells induced by native SDF-1α was confirmed. CONCLUSION: α-helix-defective mutant of SDF-1α, SDF-1/54R that remained both the N-terminus and the central β-sheet region, decreased SDF-1α-medi- ated signaling and chemotaxis but did not influence CXCR4 internalization, which suggested that SDF-1/54R might be developed as an anti-CHIV inhibitor with high biological potency and low side-effect.Shao-huiCAI YiTAN Xian-daREN Xiao-hongLI Shao-xiCAI JunDU 2004Acta Pharmacologica Sinica2004,25,2:12
2Patterning catalyst via inkjet printing to grow single-walled carbon nanotubes显示文摘Controlled growth of patterned single-walled carbon nanotubes (SWNTs) is an important issue in many applications. Herein, we demonstrated a method to pattern catalyst via inkjet printing for the growth of SWNTs, using metal salt solutions as the inks and an ordinary office-use printer. We printed water solutions of cobalt acetate on hydrophilic Si substrates and grew high quality SWNT films. 10Influence of Early Pregnancy Termination by Focused Ultrasound Beams on Menstrual Recovery of Macaques显示文摘Objective To explore the effects on macaques’ menstrual recovery after terminating early pregnancy by focused ultrasound beams (FUB) Methods FUB was used to terminate early pregnancy in 5 macaques with gestation duration ranging from 37-66 d. Two circles after the recovery of menstruation, color Doppler flow imaging (CDFI) and magnetic resonance imaging (MRI) were used to estimate the shape, size and blood flow of uterus, and pathological examinations were performed to check against any lesions to uterine endometrium and ovary. Results Forty days after FUB abortion, menstruation recovered and the volume and duration of each macaque’s menstruation were not changed compared with those be- fore gestation. CDFI and MRI suggested that the size and shape of uterus were normal. The endometrial line was clear and no lesions were found in adjacent organs. 11Material patterning on substrates by manipulation of fluidic behavior显示文摘Patterned materials on substrates are of great importance for a wide variety of applications.In solution-based approaches to material patterning;fluidic flow is inevitable.Here we demonstrate not only the importance of fluidic behavior but also the methodology of engineering the flow pattern to guide the material crystallization and assembly.We show by both experiment and simulation that substrate heating;which is generally used to accelerate evaporation,produces irregular complex vortexes.Instead;a top-heating-bottom-cooling(THBC)set-up offers an inverse temperature gradient and results in a single Marangoni vortex,which is desired for ordered nanomaterial patterning near the contact line.We then realize the fabrication of large-scale patterns of iodide perovskite crystals on different substrates under THBC conditions.We further demonstrate that harnessing the flow behavior is a general strategy with great feasibility to pattern various functional materials ranging from inorganic;organic,hybrid to biological categories on different substrates,presenting great potential for practical applications.Yitan Li Hao Wang Henglu Xu Shiting Wu Xuemei Li Jiapeng Yu Chaoyu Huang Zeyao Zhang Hao Sun Lu Han Meihui Li Anyuan Cao Zhenhai Pan Yan Li 2019National Science Review2019,6,4:0

14Effects of maturity of citrus fruits on their stalks cutting force显示文摘When the citrus harvesting robot harvests citruses,the mechanical properties of citrus stalks have an important influence on the success rate of the citrus harvesting robot.During the harvesting,the maturity of citrus fruits not only determined the harvesting time of citrus fruits but also affected the mechanical properties of citrus fruit stalks.In this study,the changes in the cutting force of citrus fruit stalks were described during the maturity of citrus fruits,and the effect of the maturity on the cutting force of stalks was clarified,so as to determine the harvesting time with the minimum cutting force required for harvesting citrus fruits by the harvesting robot.During the maturity,the relevant parameters of fruit maturity,such as the hardness,pH,and solid solution content of citrus fruits,were monitored.The results showed that there is a significant correlation between the hardness,pH,the solid solution content of citrus fruits,and the cutting force of citrus fruit stalks during maturity.The single-factor mechanical model of hardness,pH,solid solution content of citrus fruits,and the cutting force of citrus fruit stalks were established based on the data of 2019,which were verified through tests in 2020.The test results are as follows:during the ripening period of citrus fruits,the fruit hardness varies in the range of 0.13-0.31 MPa,the hardness changes by 0.02 MPa,and the cutting force changes by about 2.0-6.0 N;the pH of the citrus fruits changes in the range of 2.8-4.0,and the cutting force changes by about 1.5-2.2 N for every 0.1 change in the pH;the variation range of fruit solid solution content is 6.5%-9.0%,and for every 0.2%change in solid solution,the cutting force of citrus fruit stalks changes by about 1.25-2.0 N.The mechanical models can predict the cutting force required to cut off citrus fruit stalks according to the relevant parameters of citrus fruit maturity and can provide a reference for effectively evaluating the required cutting force.Yi Wang Dun Liu Yali Li Hongmei Zhao Changhui Yang Yitan Zhang 2022International Journal of Agricultural and Biological Engineering2022,15,6:0
