| 4 | A two-step lineage reprogramming strategy to generate functionally competent human hepatocytes from fibroblasts显示文摘Terminally differentiated cells can be generated by lineage reprogramming,which is,however,hindered by incomplete conversion with residual initial cell identity and partial functionality. Here, we demonstrate a new reprogramming strategy by mimicking the natural regeneration route, which permits generating expandable hepatic progenitor cells and functionally competent human hepatocytes. Fibroblasts were first induced into human hepatic progenitor-like cells (hHPLCs), which could robustly expand in vitro and efficiently engraft in vivo. Moreover, hHPLCs could be efficiently induced into mature human hepatocytes (hiHeps) in vitro, whose molecular identity highly resembles primary human hepatocytes (PHHs). Most importantly, hiHeps could be generated in large quantity and were functionally competent to replace PHHs for drug-metabolism estimation, toxicity prediction and hepatitis B virus infection modeling. Our results highlight the advantages of the progenitor stage for successful lineage reprogramming. | 6 | Long-term outcomes of staged recanalization for concurrent chronic total occlusion in patients with ST-segment elevation myocardial infarction after primary percutaneous coronary intervention显示文摘Background In patients with acute ST-segment elevation myocardial infarction(STEMI)who undergo primary percutaneous coronary intervention(PCI),approximately 10%are concomitant with a chronic total occlusion(CTO)in a non-culprit vessel.However,the impact of staged CTO recanalization on prognosis in this cohort remains disputable.This study aimed to compare the long-term outcomes of staged CTO recanalization versus medical therapy in patients with STEMI after primary PCI.Methods Between January 2005 and December 2016,a total of 287 patients were treated with staged CTO-PCI(n=91)or medical therapy(n=196)after primary PCI in our center.The primary endpoint was major adverse cardiovascular and cerebrovascular event(MACCE),defined as a composite of all-cause death,nonfatal myocardial infarction(MI),stroke or unplanned revascularization.After propensity-score matching,77 pairs of well-balanced patients were identified.Results The mean follow-up period was 6.06 years.Overall,the incidence of the primary endpoint of MACCE was significantly lower in staged CTO-PCI group than that in medical therapy group in both overall population(22.0%vs.46.9%;hazard ratio(HR)=0.48,95%CI:0.29-0.77)and propensity-matched cohorts(22.1%vs.42.9%;HR:0.48,95%CI:0.27-0.86).In addition,staged CTO-PCI was also associated with reduced risk of the composite of cardiac death,nonfatal MI or stroke compared with medical therapy in both overall population(9.9%vs.26.5%;hazard ratio(HR)=0.39,95%CI:0.19-0.79)and propensity-matched cohorts(9.1%vs.22.1%;HR:0.40,95%CI:0.16-0.96).After correction of the possible confounders,staged CTO-PCI was independently associated with reduced risks of MACCE(adjusted HR:0.46,95%CI:0.28-0.75),the composite of cardiac death,nonfatal MI or stroke(adjusted HR:0.45,95%CI:0.22-0.94)and all-cause mortality(adjusted HR:0.32,95%CI:0.13-0.83).Moreover,the results of sensitivity analysis were almost concordant with the overall analysis.Conclusions In patients with STEMI and a concurrent CTO who undergo primary PCI,successful staged recanalization of CTO in the non-culprit vessels is associated with better clinical outcomes during long-term follow-up. | Kong-Yong CUI Fei YUAN Hong LIU Feng XU Min ZHANG Wei WANG Ming-Duo ZHANG Yun-Lu WANG Dong-Feng ZHANG Xiao ZHANG Jin-Fan TIAN Shu-Zheng LYU | 2020 | Journal of Geriatric Cardiology2020,17,1: | 8 |
| 7 | Spatial Variation of Dissolved Organic Carbon in Soils of Riparian Wetlands and Responses to Hydro-geomorphologic Changes in Sanjiang Plain, China显示文摘Spatial variation of dissolved organic carbon(DOC) in soils of riparian wetlands and responses to hydro-geomorphologic changes in the Sanjiang Plain were analyzed through in situ collecting soil samples in the Naoli River and the Bielahong River. The results showed that the average contents of DOC for soil layer of 0–100 cm were 730.6 mg/kg, 250.9 mg/kg, 423.0 mg/kg and 333.1 mg/kg respectively from riverbed to river terrace along the transverse directions of the Naoli watershed. The content of the soil DOC was the highest in the riverbed, lower in the high floodplain and much lower in the river terrace, and it was the lowest in the low floodplain. The difference in the content and vertical distribution of DOC between the riverbed and the three riparian wetlands was significant, while it was not significant among the low floodplain, the high floodplain and the river terrace. The variability of soil DOC was related to the hydrological connectivity between different landscape position of the riparian wetlands and the adjacent stream. Extremely significant correlations were observed between DOC and total organic carbon(TOC), total iron(TFe), ferrous iron(Fe(II)) whose correlation coefficients were 0.819, –0.544 and –0.709 in riparian wetlands of the Naoli River. With the increase of wetland destruction, soil p H increased and soil DOC content changed. The correlation coefficients between soil DOC and TOC, TFe, Fe(II) also changed into 0.759, –0.686 and –0.575 respectively in the Bielahong River. Under the impact of drainage ditches, the correlations between soil DOC and TFe, Fe(II) were not obvious, while the soil p H was weakly alkaline and was negatively correlated with soil DOC in the previous high floodplain. | 9 | Intratympanic dexamethasone injection for sudden sensorineural hearing loss in pregnancy显示文摘BACKGROUND As sudden sensorineural hearing loss(SSNHL)rarely occurs in pregnant women,there is a lack of knowledge and relevant research on its management.AIM To investigate the effect of intratympanic dexamethasone injection in the treatment of pregnant patients with SSNHL.METHODS A retrospective chart review was made for the period between June 2017 and August 2019 at our Department of Otorhinolaryngology-Head and Neck Surgery.Pregnant women who met the criteria for SSNHL were included and grouped based on the therapeutic modalities.The treatment group received intratympanic dexamethasone(2.5 mg)q.o.d.for a total of four times,while the control group received no medication other than bed rest and medical observations.All the patients were under close care of obstetricians.Pure-tone audiograms were performed before and after treatment.RESULTS Eleven patients who met the inclusion criteria were assigned to the treatment group(n=7)and the control group(n=4).The mean age of patients was 31.2±3.8 years;the right ear was affected in seven(63.64%)cases.Two patients(18.2%)suffered from vertigo,10(90.9%)suffered from tinnitus and 6(54.5%)suffered from aural fullness.The time from onset to clinic visit was relatively short,with a mean time of 1.3±0.9 d.All the women were within the second or third trimester;the average gestation period was 26.0±6.2 wk.The pure-tone averages at onset between the two groups were similar.After one wk of therapy,the treatment group had a curative rate of 57.1%and a significantly better hearing threshold and greater improvement compared to the control group(all P<0.05).Some patients experienced transient discomfort from intratympanic injections that disappeared after getting rest,while none had permanent complications.All patients delivered healthy full-term neonates with an average Apgar score of 9.7±0.5.CONCLUSION Intratympanic dexamethasone injections can be used as a first-line therapy in pregnant women with SSNHL. | Yan-Lu Lyu Fan-Qian Zeng Zhou Zhou Min Yan Wei Zhang Ming Liu Zhao-Yang Ke | 2020 | World Journal of Clinical Cases2020,8,18: | 4 |

| 13 | Profiling and targeting of cellular mitochondrial bioenergetics: inhibition of human gastric cancer cell growth by carnosine显示文摘L-Carnosine (β-alanyl-L-histidine) is a naturally occurring dipeptide distributed in various organs of mammalians. We previously showed that carnosine inhibited proliferation of human gastric cancer cells through targeting both mitochondrial bioenergetics and glycolysis pathway. But the mechanism underlying carnosine action on mitochondrial bioenergetics of tumor cells remains unclear. In the current study we investigated the effect of carnosine on the growth of human gastric cancer SGC-7901 cells in vitro and in vivo. We firstly showed that hydrolysis of carnosine was not a prerequisite for its anti-gastric cancer effect. Treatment of SGC-7901 cells with carnosine (20?mmol/L) significantly decreased the activities of mitochondrial respiratory chain complexes I–IV and mitochondrial ATP production, and downregulated 13 proteins involved in mitochondrial bioenergetics. Furthermore, carnosine treatment significantly suppressed the phosphorylation of Akt, while inhibition of Akt activation with GSK690693 significantly reduced the localization of prohibitin-1 (PHB-1) in the mitochondria of SGC-7901 and BGC-823 cells. In addition, we showed that silencing of PHB-1 gene with shRNA markedly reduced the mitochondrial PHB-1 in SGC-7901 cells, and significantly decreased the colony formation capacity and growth rate of the cells. In SGC-7901 cell xenograft nude mice, administration of carnosine (250?mg?kg/d, ip, for 3 weeks) significantly inhibited the tumor growth and decreased the expression of mitochondrial PHB-1 in tumor tissue. | 14 | Spatial variations in CO2 fluxes in a subtropical coastal reservoir of Southeast China were related to urbanization and land-use types显示文摘Carbon dioxide(CO_(2))emissions from aquatic ecosystems are important components of the global carbon cycle,yet the CO_(2)emissions from coastal reservoirs,especially in developing countries where urbanization and rapid land use change occur,are still poorly understood.In this study,the spatiotemporal variations in CO_(2)concentrations and fluxes were investigated in Wenwusha Reservoir located in the southeast coast of China.Overall,the mean CO_(2)concentration and flux across the whole reservoir were 41.85±2.03μmol/L and 2.87±0.29 mmol/m2/h,respectively,and the reservoir was a consistent net CO_(2)source over the entire year.The land use types and urbanization levels in the reservoir catchment significantly affected the input of exogenous carbon towater.The mean CO_(2)fluxwasmuch higher from waters adjacent to the urban land(5.05±0.87 mmol/m2/hr)than other land use types.Sites with larger input of exogenous substance via sewage discharge and upstream runoff were often the hotspots of CO_(2)emission in the reservoir.Our results suggested that urbanization process,agricultural activities,and large input of exogenous carbon could result in large spatial heterogeneity of CO_(2)emissions and alter the CO_(2)biogeochemical cycling in coastal reservoirs.Further studies should characterize the diurnal variations,microbial mechanisms,and impact of meteorological conditions on reservoir CO_(2)emissions to expand our understanding of the carbon cycle in aquatic ecosystems. | Yifei Zhang Min Lyu Ping Yang Derrick Y.F.Lai Chuan Tong Guanghui Zhao Ling Li Yuhan Zhang Hong Yang | 2021 | Journal of Environmental Sciences2021,33,11: | 4 |

| 16 | Hybrid rice yield response to potted-seedling machine transplanting and slow-release nitrogen fertilizer application combined with urea topdressing显示文摘Machine transplanting and the application of slow-release nitrogen(N) fertilizer(SRNF) have played vital roles in the modernization of rice production. We aimed to determine the effects of potted-seedling transplanting—a new machine-transplanting method—and SRNF on hybrid rice yields. A 2-year splitplot experiment(2016–2017) was conducted in Meishan, Sichuan province, China, using two machinetransplanting methods(potted-seedling and blanket-seedling) and three N treatments. Total green leaf area, high-effective leaf area and its rate at heading, net photosynthetic rate of flag leaves 7 days after heading, glutamate synthase(GOGAT) and glutamine synthase(GS) activity after heading, dry matter production, and N accumulation at heading and maturity increased under the potted-seedling method or 70% SRNF as a base + 30% urea application at the panicle initiation stage(SBUP). Stem diameter and number of small and of all vascular bundles at the neck–panicle node in potted-seedling plants increased as a result of increasing numbers of effective panicles, secondary branches, and spikelets. | 17 | Influence of Acupuncture on Microcirculation Perfusion of Pericardium Meridian and Heart in Acute Myocardial Ischemia Model Rats显示文摘Objective: To observe the influence of acupuncture on microcirculation perfusion of the pericardium meridian and heart in acute myocardial ischemia(AMI) rats and evaluate whether acupuncture can simultaneously affect the meridians and corresponding viscera. Additionally, acupoints at different meridians were compared and whether they exert the same effects was discussed. Methods: Totally 32 Sprague-Dawley rats were subjected to left anterior descending(LAD) ligation to develop an AMI model.Rats were divided into 4 groups, including AMI, acupuncture Neiguan(PC 6), Lieque(LU 7) and Qiansanli(LI 10) groups(n=8). Eight rats received only thoracotomy(sham-operated group). The rats in the acupuncture groups received manual acupuncture at PC 6, LU 7 and LI 10 acupoints for 15 min, respectively. The microcirculation perfusion of pericardium meridian and heart was monitored by laser speckle perfusion imager(LSPI) before, during and after acupuncture manipulation for 15 min. Subsequently, the perfusion unit(PU)was calculated and analyzed by PSI System. Results: After LAD, compared to pre-acupuncture stage, the heart microcirculation perfusion(HMP) in the AMI group decreased continuously at during-acupuncture(P>0.05) and post-acupuncture stages(P<0.05), and the pericardium meridian microcirculation perfusion(PMP) showed no significant differences at 3 stages(P>0.05). Compared to pre-acupuncture stage, the PMP and HMP in PC 6 group significantly increased during acupuncture manipulation(both P<0.05), and PMP decreased obviously after acupuncture(P<0.05). The PMP in the LU 7 and LI 10 groups were slightly elevated(both P>0.05);however, they were significantly reduced after acupuncture manipulation(both P<0.05). Additionally, HMP of LI 10 group was decreased significantly during acupuncture, especially compared to pre-acupuncture stage(P<0.05). Conclusions: Acupuncture at PC 6 obviously increased the PMP and HMP in AMI rats, and the effects were superior to at LU 7 and LI 10 acupoints.
