Formation mechanism of the Lidang circular structure in the Guangxi Province显示文摘The Lidang circular structure in the center of the Guangxi Province is about 8 km in diameter. This structure appears as an abnormal shallow depression that has disturbed the rather harmonic regional joint systems. Its unique occurrence in the whole region, the circular morphology, negative topography, and the spatial distribution of interior and exterior strata are all consistent with those of impact craters that are formed by asteroidal or cometary collision. To test the impact hypothesis, we carried out both field investigation and remote sensing study of this structure. Regional geological history suggests that if the impact hypothesis were correct, the impact event should have occurred at or after the Early Permian. Field investigation found that the strata inside and outside the crater are dominated by parallel stacks of Lower and Upper Permian limestone that have various thicknesses and different mud contents. The layers of limestone within and outside the circular structure have identical attitudes;no structural disturbances were visible in the outcrops. Field investigations provide conclusive evidence against the impact cratering hypothesis. A high-resolution digital elevation model shows that the spatial distribution of rounded mountains within the structure is controlled by faint but continual extension of joints, suggesting that the crater interior has gone through a much higher degree of erosion. Therefore, regional joints that had once existed within the crater are preserved less well than exterior terrains, forming the abruptly disrupted circular depression. Differential erosion, as the possible formation mechanism of the Lidang structure, is consistent with the different mud contents found between the interior and exterior limestone. The circular outline of this structure may correspond to the shape of the original deposition basin. In conclusion, the Lidang circular structure is a polje formed by karstification, not an astrobleme. | Pan Yan ZhiYong Xiao YiZhen Ma YiChen Wang Jiang Pu | 2019 | Earth and Planetary Physics2019,3,4: | 2
An invasive zone in human liver cancer identified by Stereo-seq promotes hepatocyte–tumor cell crosstalk, local immunosuppression and tumor progression显示文摘Dissecting and understanding the cancer ecosystem, especially that around the tumor margins, which have strong implications for tumor cell infiltration and invasion, are essential for exploring the mechanisms of tumor metastasis and developing effective new treatments.Using a novel tumor border scanning and digitization model enabled by nanoscale resolution-SpaTial Enhanced REsolution Omics-sequencing(Stereo-seq),we identified a 500µm-wide zone centered around the tumor border in patients with liver cancer, referred to as“the invasive zone”.We detected strong immunosuppression, metabolic reprogramming, and severely damaged hepatocytes in this zone.We also identified a subpopulation of damaged hepatocytes with increased expression of serum amyloid A1 and A2(referred to collectively as SAAs)located close to the border on the paratumor side.Overexpression of CXCL6 in adjacent malignant cells could induce activation of the JAK-STAT3 pathway in nearby hepatocytes, which subsequently caused SAAs’overexpression in these hepatocytes.Furthermore, overexpression and secretion of SAAs by hepatocytes in the invasive zone could lead to the recruitment of macrophages and M2 polarization, further promoting local immunosuppression, potentially resulting in tumor progression.Clinical association analysis in additional five independent cohorts of patients with primary and secondary liver cancer(n=423)showed that patients with overexpression of SAAs in the invasive zone had a worse prognosis.Further in vivo experiments using mouse liver tumor models in situ confirmed that the knockdown of genes encoding SAAs in hepatocytes decreased macrophage accumulation around the tumor border and delayed tumor growth. The identification and characterization of a novel invasive zone in human cancer patients not only add an important layer of understanding regarding the mechanisms of tumor invasion and metastasis, but may also pave the way for developing novel therapeutic strategies for advanced liver cancer and other solid tumors. | Liang Wu Jiayan Yan Yinqi Bai Feiyu Chen Xuanxuan Zou Jiangshan Xu Ao Huang Liangzhen Hou Yu Zhong Zehua Jing Qichao Yu Xiaorui Zhou Zhifeng Jiang Chunqing Wang Mengnan Cheng Yuan Ji Yingyong Hou Rongkui Luo Qinqin Li Liang Wu Jianwen Cheng Pengxiang Wang Dezhen Guo Waidong Huang Junjie Lei Shang Liu Yizhen Yan Yiling Chen Sha Liao Yuxiang Li Haixiang Sun Na Yao Xiangyu Zhang Shiyu Zhang Xi Chen Yang Yu Yao Li Fengming Liu Zheng Wang Shaolai Zhou Huanming Yang Shuang Yang Xun Xu Longqi Liu Qiang Gao Zhaoyou Tang Xiangdong Wang Jian Wang Jia Fan Shiping Liu Xinrong Yang Ao Chen Jian Zhou | 2023 | Cell Research2023,33,8: | 1
The protective effect of oleic acid on Lactiplantibacillus plantarum during freeze-drying显示文摘Membrane characteristics are determined by the fatty acids composition, which affects survival rates after freeze-drying.However,it is unknown which composition provides the greatest effect. In this study,we found that the addition of Tween 80 and Tween 20 significantly increased survival rates of Lactiplantibacillus plantarum, which reached a maximum of 93.1%.Conversely,Tween 60 caused a significant decrease. We further found that the difference between the effects of adding Tween 80 and Tween 60 was the change in oleic acid contents. To verify the role of oleic acid,we used CRISPR-Cas9 to knock-out the key synthesis gene cla-er.The survival rates of L.plantarum AR113Δcla-er declined to 5.48%.The addition of oleic acid restored the rates to those of wild type strains. Moreover, the membrane integrity and fl uidity of knockout strains markedly decreased. This is the fi rst confi rmation that Tween 80 or Tween 20 increases the survival rate by increasing the content of oleic acid in the cell membrane. These fi ndings also indicated that oleic acid in cell membranes has a substantial protective effect on L.plantarum during freeze-drying. | Yongjun Xia Yizhen Wang Jing Pu Yan Wu Zhiqiang Xiong Xin Song Hui Zhang Lianzhong Ai Guangqiang Wang | 2023 | Food Science and Human Wellness2023,12,6: | 0
Mechanisms of imperatorin on regulating Pglycoprotein in blood-brain barrier based on network pharmacology and in vitro experiment显示文摘OBJECTIVE To explore mecha⁃nisms of imperatorin on regulating P-glycoprotein(P-gp)in blood-brain barrier(BBB)based on net⁃work pharmacology combined with in vitro experi⁃ment.METHODS Drug targets were predicted using the Pharmapper and Swiss targets data⁃bases;disease targets were obtained through the Genecards database;intersections between drugs and disease targets were screened by Cytoscape software;the obtained core targets were used to construct protein-protein interaction(PPI)network, gene ontology(GO)functions, and Kyoto encyclopedia of genes and genomes(KEGG)pathway enrichment analysis. The effects of imperatorin(20,50,100μmol·L^(-1))on P-gp activity were monitored in hCMEC/D3 in vitro BBB model, and the effects of imperatorin on the expression of target proteins were verified using Western blot method.RESULTS 55 drug targets and 3102 disease targets were obtained from the network pharmacology screening, and 37 core targets were obtained after the combination.Enrichment analysis showed that core targets were closely related to chemical synaptic trans⁃mission regulation, neurotransmitter receptor activity, protein kinase regulation activity,G proteincoupled receptor signaling pathway, neural active ligand receptor interaction pathway,PI3K-Akt sig⁃naling pathway,VEGF signaling pathway,etc..In vitro experimental validation suggested that all tested concentration groups of imperatorin signifi⁃cantly reduced the activity and expression of P-gp,which were achieved by significantly downregu⁃lating the phosphorylation levels of PI3K and Akt,and repressing the expression of VEGFR2 pro⁃tein.CONCLUSION Network pharmacology was used to predict the core targets and signaling pathways of imperatorin on regulating P-gp in BBB and relevant validation was conducted through in vitro experiments, providing a refer⁃ence basis for further exploration of the mecha⁃nisms of imperatorin on regulating P-gp in BBB. | SONG Yizhen YIN Wanxin MA Yicong AN Yufan HUANG Jiaqi YAN Zhongjie WU Xiuwen WANG Yuanyu | 2023 | 中国药理学与毒理学杂志2023,37,S01: | 0
Virtualization of Real Estate Industry and Housing Bubbles --Empirical Evidence from China显示文摘In the last twenty years, China has experienced a rapid development in the real estate industry. The continuous rising house price causes many social and economic problems and draws a lot of attentions from academia, business world and government. Many researchers shed light on this problem from different angles, but till today, it hasn’t been fully resolved. In this paper, we try to examine the real estate bubble from a virtual economic perspective. We think that the virtual feature of real estate alters its economic role from a purely living and working place to a financing instrument. We use the newest monthly data of China real estate market from 2006 to 2018 and estimate the relationship between the housing bubble and real estate industry virtualization. We identify a significant positive association between house price index and the purchase price of land in real estate investment. This result suggests that virtual feature of real estate has deepen the bubble problem in the industry and its virtualization degree appears to be closely related to the virtual feature of land and its price proportion to real estate investment. | Beibei Yan Yizhen Wang | 2019 | 经济管理学刊(中英文版)2019,8,1: | 0
A Cobalt(Ⅲ)Corrole with a Tethered Imidazole for Boosted Electrocatalytic Oxygen Reduction Reaction显示文摘Developing electrocatalysts with high activity and selectivity for the oxygen reduction reaction(ORR)is vital to promote the performance of the next-generation energy technologies, which depend on the efficiency of the catalytic reduction of dioxygen. In the structure of cytochrome c oxidases(CcOs),a histidine imidazole residue binding to the axial position of Fe plays a crucial role in facilitating the selective reduction of O_(2)to water.Inspired by nature,we herein report on the synthesis of CoIII corrole 1 tethered with an imidazole ligand as well as its electrocatalytic ORR and O_(2)binding features. As compared to the imidazolium-free analogue, complex 1 displayed remarkably boosted activity for the selective four-electron/four-proton(4e-/4H+)ORR with a half-wave potential of E1/2=0.82 V versus reversible hydrogen electrode(RHE)in 0.1 mol/L KOH solutions.Importantly,we demonstrate that the tethered axial imidazole ligand improves the O_(2)binding ability of 1 thermodynamically and dynamically, which is crucial to boost electrocatalytic ORR performance. This work presents an example to improve electrocatalytic ORR activity and selectivity of Co corroles by introducing an axial imidazole ligand to enhance the O_(2)binding and activation. | Yimei Gao Benxing Mei Yizhen Wu Qian Zhao Zijia Bao Haonan Qin Yuhan Xu Haoyuan Lv Xinyang Peng Yan He Ting Luo Ran Yao Wei Zhang Haitao Lei Rui Cao | 2023 | Chinese Journal of Chemistry2023,41,21: | 0
