1Effects of endostatin-vascular endothelial growth inhibitor chimeric recombinant adenoviruses on antiangiogenesis显示文摘AIM: To investigate the inhibitory effects of endostatinvascular endothelial growth inhibitor (VEGI151) recombinant adenoviruses on neovascularization.METHODS: We used recombinant adenoviruses to treat human vascular endothelial cell line ECV304, human hepatocellular carcinoma cell line HepG2, and murine fibroblast cell line L929, in order to study the chimeric gene expression in these cell lines. Chick choriallantic membrane(CAM) model, rabbit inflammatory corneal neovascularization(CNV) model, and liver cancer-bearing nude mice model were employed to investigate the negative biological effect of fusion molecules on neovascularization in vivo.RESULTS: Western blot showed that the molecular weight of fusion protein was about 41kD after infection of ECV304,HepG2 and L929 cells with supernatant of AdhENDO-VEGI151.The fusion protein showed a specific inhibitory effect on the proliferation of ECV304 cells, but no inhibitory effect on the growth of HepG2 and L929 cells (F=13112.13, P=0.0001). In the chick choriallantic membrane (CAM) assay, the expressed fusion protein significantly inhibited neovascularization. Rabbit inflammatory corneal neovascularization (CNV) induced by intrastromal sutures resulted in a uniform neovascular response. In this model, direct subconjunctival injection of AdhENDO-VEGI151 expressed the fusion protein in vivo and suppressed the development of CNV. Topical application of AdhENDO-VEGI151 led to a significant suppression of CNV (F=1413.11, P=0.0001),as compared with the control group of AdLacZ. Immunohistochemical staining showed the fusion protein dominantly expressed in corneal epithelium. Compared with the control group of AdLacZ (4075.9±1849.9 mm^3), the average tumor size of group AdhENDO-VEGI151 reduced in size (487.7±241.2 mm^3) (F=14.80, P=0.0085), with an inhibition rate of 88.03%. Immunohistochemical staining showed the adenoviruses carried the fusion gene expressed on liver cancer cell membrane. MVD decreased more significantly in treated mice (30.75±3.31%) than in AdLacZ control(50.25±8.65%) (F=17.72, P=0.0056) with an inhibition rate of 39%.CONCLUSION: Fusion protein expressed by recombinant adenoviruses has a significant inhibitory effect on neovascularization.XinPan YongWang MinZhang WeiPan Zhong-TianQi Guang-WenCao 2004World Journal of Gastroenterology2004,10,10:19

3Effect of manganese on the catalytic performance of an iron-manganese bimetallic catalyst for light olefin synthesis显示文摘A systematic study was carried out to investigate the promotion effect of manganese on the performance of a coprecipitated iron-manganese bimetallic catalyst for the light olefins synthesis from syngas. The catalyst samples were characterized by N2 physisorption, transmission electron microscopy (TEM), X-ray photoelectron spectroscopy (XPS), powder X-ray diffraction (XRD), Mo¨ssbauer spectroscopy, H2-differential thermogravimetric analysis (H2-DTG), CO temperature-programmed reduction (CO-TPR) and CO2 temperature-programmed desorption (CO2-TPD). The Fischer-Tropsch synthesis (FTS) performance of the catalyst was measured at 1.5 MPa, 250 C and syngas with H2 /CO ratio of 2.0. The characterization results indicated that the addition of manganese decreases the catalyst crystallite size, and improves the catalyst BET surface area and pore volume. The presence of manganese suppresses the catalyst reduction and carburization in H2 , CO and syngas, respectively. The addition of manganese improves the catalytic activity of water-gas shift reaction and suppresses the oxidation of iron carbides in the FTS reaction. The incorporation of manganese improves the catalyst surface basicity and results in a significant improvement in the selectivities to light olefins and heavy hydrocarbons (C5+ ), and furthermore an inhibition of methane formation in FTS. The pure iron catalyst (Mn-00) has the highest initial FTS catalytic activity (65%) and the lowest selectivity (17.35 wt%) to light olefins (C=2-C=4 ). 4Effect of aerial spray adjuvant applying on the efficiency of small unmanned aerial vehicle for wheat aphids control显示文摘Small unmanned aerial vehicles(UAVs)have been widely used in different aspects of modern farming management,including pest and disease control in China in recent years.In this study,the spray performance of a small plant protection UAV at low volume spray was evaluated by adjusting the pesticide dosage and adding aerial spraying adjuvants.Droplet deposition,droplet density,coverage,control effect and pesticide residue from field trials were assessed.In addition,the residue and control effect of UAV spray were compared to manual knapsack at high volume spray.The results showed that,the adjuvant applying improved the efficiency of UAV spray.Also,the adjuvant applying reduced the dosage of imidacloprid by 20%.However,there was no significant difference on initial residue between UAV spray and knapsack spray.Thus,plant protection UAV spraying pesticide by adding appropriate adjuvant showed the ability of improving the pesticide effectiveness by improving the control efficiency,reducing the pesticide dosage and residue.Yanhua Meng Yubin Lan Guiying Mei Yongwang Guo Jianli Song Zhiguo Wang 2018International Journal of Agricultural and Biological Engineering2018,11,5:13
5Inhibitory effect of adeno-associated virus-mediated gene transfer of human endostatin on hepatocellular carcinoma显示文摘AIM: To investigate the effect of adeno-associated virusmediated gene transfer of human endostatin on the growth of hepatocellular carcinoma (HCC).METHODS: HCC cell line Hep3B was infected with recombinantadeno-associated virus containing human endostatin gene (rAAV2-hEndo). The results of transfection were detected by RT-PCR and SDS-PAGE assay. MTT assay was used to observe the effects of supernatant of transfected cells on ECV304 cell proliferation. An animal model of HCC was established by injecting Hep3B cells subcutaneously into the back of nude mice. Intratumoral injection of rAAV2hEndo, empty virus and phosphate-buffered saline were given sequentially. Serum endostatin was determined byELISA, the inhibitory effect of endostatin on the growth of xenograft was assessed in 3 wk.RESULTS: The results of RT-PCR and SDS-PAGE assay confirmed that rAAV2-hEndo successfully transfected Hep3B cells, and endostatin was secreted from Hep3B cells to medium. The supernatant of transfected cells markedly inhibited the proliferation of ECV304 cells (P<0.01). Intratumoral injection of rAAV2-hEndo (2×1010v.g.) led to a sustained serum endostatin level ofapproximately (86.71±5.19) ng/mL. 8The Discovery of Diamonds in Chromitites of the Hegenshan Ophiolite,Inner Mongolia,China显示文摘Diamond,moissanite and a variety of other minerals,similar to those reported from ophiolites in Tibet and northern Russia,have recently been discovered in chromitites of the Hegenshan ophiolite of the Central Asian Orogenic Belt,north China. The chromitites are small,podiform and vein-like bodies hosted in dunite,clinopyroxene-bearing peridotite,troctolite and gabbro. All of the analysed chromite grains are relatively Al-rich,with Cr# [100Cr/(Cr+Al)] of about 47–53. Preliminary studies of mainly disseminated chromitite from ore body No. 3756 have identified more than 30 mineral species in addition to diamond and moissanite. These include oxides(mostly hematite,magnetite,rutile,anatase,cassiterite,and quartz),sulfides(pyrite,marcasite and others),silicates(magnesian olivine,enstatite,augite,diopside,uvarovite,pyrope,orthoclase,zircon,sphene,vesuvianite,chlorite and serpentine) and others(e.g.,calcite,monazite,glauberite,iowaite and a range of metallic alloys). 10Gene Transcription Profile in Mice Vaccinated with Ultraviolet-attenuated Cercariae of Schistosoma japonicum Reveals Molecules Contributing to Elevated IFN-γ Levels显示文摘AVaccination with ultraviolet-attenuated cercariae of Schistosoma japonicum induced protective immunity against challenge infection in experimental animal models. Our preliminary study on .he transcription levels of IFN-γ and IL-4 in splenic CD4~ T cells revealed that attenuated cercariae elicited predominantly a Thl response in mice at the early stage, whereas normal cercariae stimulated primarily Th2-dependent responses. Further analysis on the gene profile of the skin-draining lymph nodes demonstrated that the levels of IFN-γ were significantly higher in vaccinated mice than those in infected mice at day 4, 7 and 14 post-vaccination or post-infection. However, for IL-12 and IL-4, the potent inducers of Th1 and Th2 responses, respectively, as well as IL-10, there were no differences over the course of the experiment between the infected and vaccinated mice. To explore the underlying factors that may potentially contribute to elevated IFN-y in vaccinated mice, the mRNA profiles of the skin-draining lymph nodes at day 4 postexposure were compared using oligonucleotide microarrays. Within the 847 probe sets with increased signal values, we focused on chemokines, cytokines and relevant receptors, which were validated by semi-quantitative RT-PCR. 11Substrate matters:The influences of substrate layers on the performances of thin-film composite reverse osmosis membranes显示文摘Thin-film composite(TFC) reverse osmosis(RO) membranes are playing the dominating role in desalination.Tremendous efforts have been put in the studies on the polyamide selective layers. However, the effect of the substrate layers is far less concerned. In this review, we summarize the works that consider the impacts of the substrates, including pore sizes, surface hydrophilicity, on the processes of interfacial polymerization and consequently on the morphologies of the active layers and on final RO performances of the composite membranes. All the works indicate that the pore sizes and surface hydrophilicity of the substrate evidently influence the RO performances of the composite membranes. Unfortunately, we find that the observations and understandings on the substrate effect are frequently varied from case to case because of the lack of substrates with uniform pores and surface chemistries. We suggest using track-etched membranes or anodized alumina membranes having relatively uniform pores and functionalizable pore walls as model substrates to elucidate the substrate effect.Moreover, we argue that homoporous membranes derived from block copolymers have the potential to be used as substrates for the large-scale production of high-performances TFC RO membranes.Jie Li,Mingjie Wei YongWang 2017Chinese Journal of Chemical Engineering2017,25,11:5
12Distinct Overexpression of Fas Ligand on T Lymphocytes in Aplastic Anemia显示文摘Increased expression of Fas by hematopoietic progenitors in aplastic anemia (AA) suggests that Fas/Fas ligand (FasL) system plays a key role in the formation of severe pancytopenia. To further confirm the above hypothesis, T cells from 8 patients with AA were systematically studied for their FasL's distribution pattern,releasing manner and proapoptotic activity, compared with normal resting T cells and artificially activated Tcell blasts. The results demonstrated that AA T cells abnormally expressed low levels of membrane-bound FasL and contained high levels of intracellular FasL which could be triggered to release by high-dose phytohemagglutinin (PHA) pulse-stimulation. The supernatants from the PHA-stimulated AA T cells had apparent cytotoxicity against FasL-sensitive Jurkat cells, which could be significantly inhibited by monoclonal antibody against FasL in a dose-dependent manner, or nearly completely abrogated by ultracentrifugation. The above phenomena also appeared on artificially activated T cell blasts, but this was not the case on normal resting Tcells. These results indicate that AA T cell is a type of 'preactivated' T lymphocyte, characterized by overexpression of FasL, especially intracellular FasL which can be stimulated to release in bioavtive exosomesbound form. 14Effects of Ti, Al and Hf on niobium silcides formation in the Nb-Si in situ composites显示文摘The effects of alloying elements Ti, Al and Hf on niobium silicides formation in the Nb-Si in situ composites have been investigated by scanning electron microscopy (SEM), X-ray diffraction (XRD) and transmission electron microscopy (TEM). The binary, ternary and multicomponent alloys have been fabricated by vacuum non-consumable arc melting method. The results show that Ti tends to stabilize Nb3Si phase, while Al promotes the direct formation of β-Nb5Si3 phase with a tetrahedral D8m structure. Exceptionally, it seems that Hf is beneficial to the formation of γ-Nb5Si3 phase with a hexangular D88 structure. 17ILF2 cooperates with E2F1 to maintain mitochondrial homeostasis and promote small cell lung cancer progression显示文摘Objective:Mitochondria play multifunctional roles in carcinogenesis.Deciphering uncertainties of molecular interactions within mitochondria will promote further understanding of cancer.Interleukin enhancer binding factor 2(ILF2)is upregulated in several malignancies,however,much remains unknown regarding ILF2 in small cell lung cancer(SCLC).In the current study,we explored ILF2's role in SCLC and demonstrated its importance in mitochondria quality control.Methods:Colony formation,cell proliferation,cell viability and xenograft studies were performed to examine ILF2's role on SCLC progression.Glucose uptake,lactate production,cellular oxygen consumption rate and extracellular acidification rate were measured to examine the effect of ILF2 on glucose metabolism.RNA-sequencing was utilized to explore genes regulated by ILF2.E2 F1 transcriptional activity was determined by dual luciferase reporter assay.Mitochondria quantification and mitochondrial membrane potential assays were performed to examine mitochondrial quality.Gene expression was determined by RT-qPCR,Western blot and IHC assay.Results:ILF2 promotes SCLC tumor growth in vitro and in vivo.ILF2 elevates oxidative phosphorylation expression and declines glucose intake and lactate production.Genome-wide analysis of ILF2 targets identified a cohort of genes regulated by E2 F1.In consistent with this,we found ILF2 interacts with E2 F1 in SCLC cells.Further studies demonstrated that suppression of E2 F1 expression could reverse ILF2-induced tumor growth and enhanced mitochondria function.Significantly,expression of ILF2 is progressively increased during SCLC progression and high ILF2 expression is correlated with higher histologic grades,which indicates ILF2's oncogenic role in SCLC.Conclusions:Our results demonstrate that ILF2 interacts with E2 F1 to maintain mitochondria quality and confers SCLC cells growth advantage in tumorigenesis.Meng Zhao Yahui Liu Jiao Chang Jin Qi Ran Liu Yongwang Hou Yanhui Wang Xinwei Zhang Lu Qiao Li Ren 2019Cancer Biology & Medicine2019,16,4:3

20Activation pressure studies with an iron-based catalyst for slurry Fischer-Tropsch synthesis显示文摘Fischer-Tropsch synthesis(FTS) was carried out with an industrial iron-based catalyst(100Fe/5Cu/6K/16SiO 2,by weight) under the baseline conditions in a stirred tank slurry reactor(STSR) . The effects of activation pressure on the catalyst activity and selectivity were investigated. It was found that iron phase compositions,textural properties,and FTS performances of the catalysts were strongly dependent on activation pressure. The high activation pressure retards the carburization. M ssbauer effect spectroscopy(MES) results indicated that the contents of the iron carbides clearly decrease with the increase of activation pressure,especially for the activation pressure increasing from 1.0 MPa to 1.5 MPa,and the reverse trend is observed for superparamagnetic Fe 3+(spm) . The higher content of Fe 3+(spm) results in the higher amount of CO 2 in tail gas when the catalyst is reduced at higher pressure. The catalyst activity decreases with the increase of activation pressure. The high quantity of iron carbides is necessary to obtain high FTS activity. However,the activity of the catalyst activated in syngas can not be predicted solely from the fraction of the carbides.
