3Unsteady Turbulent Simulation and Pressure Fluctuation Analysis for Centrifugal Pumps显示文摘The pressure fluctuation in the flow passage of both impeller and casing is addressed on design condition.The initial conditions for the unsteady turbulent simulation are resulted from the steady calculations, and the three dimensional unsteady turbulent simulation concerning the rotor-stator interaction is executed by a Navier-Stoke solver embedded with k-ε turbulence model and with appropriate moving interface boundary conditions.Detecting points are distributed in the flow passage in different radial and circumferential positions to capture the static pressure fluctuation character for one cycle of the impeller.The time-domain spectrums show that the static pressure curves are periodic and have five peaks and five valleys.With the radius increasing, the pressure fluctuation peak-to-peak values in the impeller are increasing, and reach the maximum value on the interface.In the casing flow passage, those values are about 7% of local static pressure except some ones near the tongue.The values become decreasingly in the diffuser pipe.The frequency spectrums transformed by fast Fourier transform(FFT) show that the dominant frequency is approximate with the blade passing frequency, and the pressure fluctuations in impeller passage have high frequency content while those in casing ones have no such information.YUAN Shouqi NI Yongyan PAN Zhongyong YUAN Jianping 2009Chinese Journal of Mechanical Engineering2009,22,1:48
4DETECTION OF CAVITATION IN CENTRIFUGAL PUMP BY VIBRATION METHODS显示文摘For the purpose of detecting the cavitation of centrifugal pump onsite and real time,the vibration signals on varied operation conditions of both cavitation and non-cavitation obtained through acceleration sensors were analyzed.When cavitation occurs,the cavities near the leading edge of the blade will appear periodic oscillating,which will induce quasi-synchronous vibration.The frequency of the quasi-synchronous vibration symmetrically appears on the two sides of the blade passing frequency,by which the cavitation incipiency can be detected.During the developing process of the cavitation,as the severe complexity of the unsteady flow,it is very difficult to detect the development of cavitation by classical analysis methods.Fractal method of Higuchi is successfully used for detecting the incipiency,fully development of cavitation and the development between them.NI Yongyan YUAN Shouqi PAN Zhongyong YUAN Jianping 2008Chinese Journal of Mechanical Engineering2008,21,5:15
5Hepatitis B Virus Down-Regulates Expressions of MHC Class I Molecules on Hepatoplastoma Cell Line显示文摘Chronic HBV infection is associated with a 100-fold high risk of developing hepatocellular carcinoma. Tumor recognition is of the most importance during the immune surveillance process that prevents cancer development in humans. In the present study, the expressions of MHC class I molecules on hepatoplastoma cell line HepG2.2.15 were investigated to indicate the possible effects of HBV on the immune recognition during HBV-associated hepatocellular carcinoma. It was found that the expressions of MHC class I molecules HLA-ABC, HLA-E and MICA were much lower in HepG2.2.15 cells compared with HepG2 cells. The expressing HBV in human hepatoplastoma cell line significantly down-regulated the expressions of MHC class I molecules. Additionally, it was observed that in murine chronic HBsAg carriers the expression of classical MHC-I molecule on hepatocytes was down-regulated. These results demonstrated that HBV might affect the immune recognition during HBV- associated hepatocellular carcinoma such as the recognition of CD8+ T, NK-CTL and NK cells and prevent the immune surveillance against tumors. 6Innate lymphocytes:pathogenesis and therapeutic targets of liver diseases and cancer显示文摘The liver is a lymphoid organ with unique immunological properties,particularly,its predominant innate immune system.The balance between immune tolerance and immune activity is critical to liver physiological functions and is responsible for the sensitivity of this organ to numerous diseases,including hepatotropic virus infection,alcoholic liver disease,nonalcoholic fatty liver disease,autoimmune liver disease,and liver cancer,which are major health problems globally.In the past decade,with the discovery of liver-resident natural killer cells,the importance of innate lymphocytes with tissue residency has gradually become the focus of research.In this review,we address the current knowledge regarding hepatic innate lymphocytes with unique characteristics,including NK cells,ILC1/2/3s,NKT cells,γδ T cells,and MAIT cells,and their potential roles in liver homeostasis maintenance and the progression of liver diseases and cancer.A better understanding of the immunopathogenesis of hepatic innate lymphocytes will be helpful for proposing effective treatments for liver diseases and cancer.Yongyan Chen Zhigang Tian 2021Cellular & Molecular Immunology2021,18,1:9
7Tumor microenvironments self-activated nanoscale metal-organic frameworks for ferroptosis based cancer chemodynamic/photothermal/chemo therapy显示文摘Ferroptosis,as a newly discovered cell death form,has become an attractive target for precision cancer therapy.Several ferroptosis therapy strategies based on nanotechnology have been reported by either increasing intracellular iron levels or by inhibition of glutathione(GSH)-dependent lipid hydroperoxidase glutathione peroxidase 4(GPX4).However,the strategy by simultaneous iron delivery and GPX4 inhibition has rarely been reported.Herein,novel tumor microenvironments(TME)-activated metal-organic frameworks involving Fe&Cu ions bridged by disulfide bonds with PEGylation(FCSP MOFs)were developed,which would be degraded specifically under the redox TME,simultaneously achieving GSH-depletion induced GPX4 inactivation and releasing Fe ions to produce ROS via Fenton reaction,therefore causing ferroptosis.More ROS could be generated by the acceleration of Fenton reaction due to the released Cu ions and the intrinsic photothermal capability of FCSP MOFs.The overexpressed GSH and H2O2 in TME could ensure the specific TME self-activated therapy.Better tumor therapeutic efficiency could be achieved by doxorubicin(DOX)loading since it can not only cause apoptosis,but also indirectly produce H2O2 to amplify Fenton reaction.Remarkable anti-tumor effect of obtained FCSP@DOX MOFs was verified via both in vitro and in vivo assays.Yu Liang Li Zhang Chao Peng Shiyu Zhang Siwen Chen Xin Qian Wanxian Luo Qing Dan Yongyan Ren Yingjia Li Bingxia Zhao 2021Acta Pharmaceutica Sinica B2021,11,10:7
8Mitochondrial tRNA^(Glu) A14693G variant may modulate the phenotypic manifestation of deafness-associated 12S rRNA A1555G mutation in a Han Chinese family显示文摘Mutations in mitochondrial 12S rRNA gene are one of the most important causes of aminoglycoside-induced and nonsyndromic hear- ing loss. Here we report the characterization of one Han Chinese pedigree with aminoglycoside-induced and nonsyndromic hearing loss. This Chinese family carrying the 12S rRNA A1555G mutation exhibited high penetrance and expressivity of hearing impairment. In par- ticular, penetrances of hearing loss in this family pedigree were 43.8% and 25%, respectively, when aminoglycoside-induced hearing loss was included or excluded. Mutational analysis of entire mitochondrial genomes in this family showed the homoplasmic A1555G mutation and a set of variants belonging to haplogroup Y2. Of these, the A14693G variant occurred at the extremely conserved nucleotide (conven- tional position 54) of the TΨC-loop of tRNAGlu and was absent in 156 Chinese controls. Nucleotides at position 54 of tRNAs are often modified, thereby contributing to the structural formation and stabilization of functional tRNAs. Thus, the structural alteration of tRNA by the A14693G variant may lead to a failure in tRNA metabolism and impair mitochondrial protein synthesis, thereby worsening mito- chondrial dysfunctions altered by the A1555G mutation. 9Differential regulation of the anthocyanin profile in purple kiwifruit(Actinidia species)显示文摘Anthocyanins are a group of secondary metabolites that colour fruit and flowers orange,red,purple or blue depending on a number of factors,such as the basic structure,co-pigmentation,metal ion complexation and vacuolar pH.The biosynthesis of anthocyanin is regulated at the transcriptional level by a group of transcription factors,the MYB–bHLH–WD40(MBW)complex.In this study,the purple colouration in several kiwifruit(Actinidia)species was identified and characterised as red cyanidin-based and blue delphinidin-based anthocyanins.The differential pigmentation in the skin and flesh can be attributed to the differential ratio of cyanidin and delphinidin derivatives accumulated in the total anthocyanin profile.The expression of anthocyanin biosynthetic genes chalcone synthase(CHS),flavonoid 3-O-glucosyltransferase(F3GT),flavonoid 3′-hydroxylase(F3′H)and flavonoid 3′5′-hydroxylase(F3′5′H)is crucial for anthocyanin accumulation.However,the balance of expression of the F3′H and F3′5′H genes appears responsible for the ratio of cyanidin and delphinidin derivatives,while a lack of CHS,F3GT and MYB110 expression is responsible for a lack of total anthocyanins.The transcriptional regulation of the F3′H and F3′5′H promoters by the R2R3 MYB transcription factor MYB110 is markedly different in tobacco transient assays.When kiwifruit MYB10 or MYB110 are over-expressed in Actinidia chinensis both cyanidin-based and delphinidin-based anthocyanins are elevated,but F3′H and F3′5′H genes are not strongly correlated with MYB expression.These results suggest that the core kiwifruit anthocyanin pathway genes are dependent on characterised MYB transcription factors,while other regulatory proteins are more directly responsible for the expression of the F3′H and F3′5′H genes.Yongyan Peng Kui Lin-Wang Janine M.Cooney Tianchi Wang Richard V.Espley Andrew C.Allan 2019Horticulture Research2019,6,1:6
10A combination extract of Renshen(Panax Ginseng),Yinyanghuo(Herba Epimedii Brevicornus),Yuanzhi(Radix Palygalae) and Jianghuang(Rhizoma Curcumae Longae) decreases glycogen synthase kinase 3β expression in brain cortex of APPV717I transgenic mice显示文摘OBJECTIVE:To investigate the neuroprotective mechanism of combination extract of Renshen(Panax Ginseng),Yinyanghuo(Herba Epimedii Brevicornus),Yuanzhi(Radix Palygalae) and Jianghuang(Rhizoma Curcumae Longae)(GEPT) in treating Alzheimer's disease on the target of glycogen synthase kinase 3β(GSK-3β).METHODS:Three-month-old APPV717I transgenic mice were randomly divided into ten groups(n=12 per group) and intragastrically administrated vehicle or medicines:APP group was given 0.5% CMC,donepezil group was given donepezil(APP + D group)(0.92 mg/kg-1·day-1),and GEPT groups were given small dose of GEPT(APP+Gs group)(0.075 g/kg-1·day-1),medium dose(APP+Gm group)(0.15 g/kg-1·day-1),and large dose(APP+Gl group)(0.30 g/kg-1·day-1) for 4 or 8 months,respectively.Three-month-old C57BL/6J mice as vehicle controls(n=12) were given 0.5% CMC for 4 or 8 months as well.The GSK-3β expression in the cortex of 7-and 11-month-old APPV717I transgenic mice with and without GEPT or donepezil treatment and normal C57BL/6J mice were measured via Western blotting and Immunohistochemistry.RESULTS:Immunohistochemistry analysis showed significant increase of GSK-3β in the cerebral cortex of 7-month-old APP group(compare to control group P=0.003),while the GSK-3β expression of donepezil or GEPT group were all significantly decreased(Donepezil vs APP:P=0.041;Gl vs APP:P= 0.049;Gm vs APP:P=0.029;Gh vs APP:P=0.036).Western blot analysis showed similar results.The densitometric measures of GSK-3β in APP mice increased significantly as compared with the control group(P=0.008).And the GSK-3β expression in donepezil and GEPT groups were all decreased.There was significant difference between Gh group or donepezil group and the control group(P=0.05).Similar findings were shown in the 11-month-old mice in each group,except for greater decrease of GSK-3β in the GEPT group.CONCLUSION:GEPT can effectively decrease the level of GSK-3β expression in the brain cortex of APPV717I transgenic mice,and such effect is more significant in 11-month-old mice.This partially explains the neuroprotecting mechanism of GEPT in preventing and treating of AD.Jing Shi Jinzhou Tian Xuekai Zhang Chuiyou Zeng Mingqing Wei Pengwen Wang Yongyan Wang 2013Journal of Traditional Chinese Medicine2013,33,2:6
11Corrosion behavior of Mg-Zn-Ca amorphous alloys with Nd addition in simulated body fluids显示文摘The effects of Nd addition on corrosion behavior of Mg66Zn30Ca4 amorphous alloys in simulated body f luids(SBF) were studied in this paper. Electrochemical properties of the samples before and after corrosion were determined. Surface morphologies of samples after immersion in SBF at 37℃ for different times were observed under scanning electron microscope(SEM). Results show that the corrosion resistance of Mg-based alloys in SBF is improved with the addition of Nd element. The electrochemical properties indicate that microalloying Nd element to the alloys leads to an ennoblement in the open circuit potentials of the alloys and a decrease in the anodic current density in SBF, especially for the Mg66-xZn30Ca4 Ndx alloys with Nd content of 1.0at.%-1.5at.%. It was observed that the surface morphologies of the alloys immersed in SBF change with the Nd addition. 13Retroperitoneal Fibrosis: A Retrospective Clinical Data Analysis of 30 Patients in a 10-year Period显示文摘Background: Retroperitoneal fibrosis (RPF) is an uncommon disease that is characterized by development of fibrosclerotic tissues involving retroperitoneal structures. This study aimed to investigate the clinical features of 30 patients with RPF in a single center in Beijing in a 10-year period. Methods: We retrospectively analyzed clinical data on demographic characteristics, clinical manifestations, laboratory findings, radiological findings, modalities of treatments, outcomes and prognosis of 30 patients with RPF. Patients were treated in Beijing Chao-Yang Hospital between January 2003 and December 2013. Results: The mean age of patients with RPF was 56.7 ± 14.4 years. Twenty-three patients were men and seven patients were women. Acute phase reactants were elevated in most patients. Rheumatic factor was positive in 4/25 (16.0%) patients, and antinuclear antibody was positive in 6/22 (27.3%) patients. Elevation oflgG4 was observed in 9/22 (40.9%) patients. The most common type was Ⅰ+ Ⅲ (17 = 13), followed by Ⅰ + Ⅱ+ Ⅲ(n = 12). Five patients undertook an 18F-fluoro-deoxy-D-glucose positron emission tomography examination and increased uptake was detected in four patients. Eight patients received combination therapy with glucocorticoids and tamoxifen. Surgical intervention treatments included intraureteral double-J stent implantation (n = 26), percutaneous nephrostomy (n = 2), open ureterolysis and intraperitonealization of the ureters (n = 5) and laparoscopic ureterolysis and intraperitonealization of the ureters (n = 5). Three patients underwent hemodialysis because of renal failure. Conclusions: Clinical characteristics of RPF patients in our study are similar to those previously reported. Steroids and immunosuppressive therapy combined with ureterolysis could be a viable choice of treatment for RPF. 14Deployment of Sensors in WSN: An Efficient Approach Based on Dynamic Programming显示文摘Efficient sensor node deployment is extremely important in wireless sensor networks. It earns great practical meanings through using fewer sensor nodes as far as possible to satisfy different requirements such as the requirement on coverage and overcoming the potential sensor node failures and the adverse influence from the environment. We propose an efficient approach for the deployment of sensor nodes in wireless networks, termed as EDSNDA, which is excellent in taking both the requirements of sensor coverage and network connectivity into consideration when minimizing the number of necessary sensor nodes to the best of its ability. We proposed a new coverage model of sensor node. Based on the sensor coverage model, we establish four dynamic programming models in four different practical situations, respectively. The algorithms are then proposed which are used for solving the corresponding dynamic programming models. The validity of the method is justified by simulation studies in which the method is compared with the current representative methods. 15Synthesis of HTPB based PU-PS IPN Membrane for Pervaporation Recovery of Butanol显示文摘Hydroxyl terminated polybutadiene(HTPB) based polyurethane(PU)-polystyrene(PS) interpenetrating polymer network(IPN) was prepared and characterized by FTIR, TGA, WCA, swelling experiments, and SEM. The IPN was used for pervaporation(PV) recovery of butanol. Both the permeation flux and separation factor increased with feed temperature, and both water and butanol fluxes increased with feed concentration while no obvious effect of concentration on separation factor was found. Through the formation of IPN structure, the total flux of HTPB based PU increased greatly with the decrease of separation factor. 16Natural variations of ZmSRO1d modulate the trade-off between drought resistance and yield by affecting ZmRBOHC-mediated stomatal ROS production in maize显示文摘While crop yields have historically increased,drought resistance has become a major concern in the context of global climate change.The trade-off between crop yield and drought resistance is a common phenomenon;however,the underlying molecular modulators remain undetermined.Through genome-wide association study,we revealed that three non-synonymous variants in a drought-resistant allele of ZmSRO1d-R resulted in plasma membrane localization and enhanced mono-ADP-ribosyltransferase activity of ZmSRO1d toward ZmRBOHC,which increased reactive oxygen species(ROS)levels in guard cells and promoted stomatal closure.ZmSRO1d-R enhanced plant drought resilience and protected grain yields under drought conditions,but it led to yield drag under favorable conditions.In contrast,loss-of-function mutants of ZmRBOHC showed remarkably increased yields under well-watered conditions,whereas they showed compromised drought resistance.Interestingly,by analyzing 189 teosinte accessions,we found that the ZmSRO1d-R allele was present in teosinte but was selected against during maize domestication and modern breeding.Collectively,our work suggests that the allele frequency reduction of ZmSRO1d-R in breeding programs may have compromised maize drought resistance while increased yields.Therefore,introduction of the ZmSRO1d-R allele into modern maize cultivars would contribute to food security under drought stress caused by global climate change.Huajian Gao Junjun Cui Shengxue Liu Shuhui Wang Yongyan Lian Yunting Bai Tengfei Zhu Haohao Wu Yijie Wang Shiping Yang Xuefeng Li Junhong Zhuang Limei Chen Zhizhong Gong Feng Qin 2022Molecular Plant2022,15,10:2

18Single-cell characterization of monolayer cultured human dental pulp stem cells with enhanced differentiation capacity显示文摘Human dental pulp stem cells(hDPSCs)are easily obtained multipotent cells,however,their potential value in regenerative medicine is hindered by the phenotypic and functional changes after conventional monolayer expansion.Here,we employed single-cell RNA sequencing(scRNA-seq)to comprehensively study the transcriptional difference between the freshly isolated and monolayer cultured DPSCs.The cell cluster analysis based on our scRNA-seq data showed that monolayer culture resulted in a significant cellular composition switch compared to the freshly isolated DPSCs.However,one subpopulation,characterized as MCAM(+)JAG(+)PDGFRA(−),maintained the most transcriptional characteristics compared to their freshly isolated counterparts.Notably,immunofluorescent staining revealed that the MCAM(+)JAG(+)PDGFRA(−)hDPSCs uniquely located in the perivascular region of human dental pulp tissue.Flow-cytometry analysis confirmed that their proportion remained relatively stable(~2%)regardless of physiological senescence or dental caries.Consistent with the annotation of scRNA-seq data,MCAM(+)JAG(+)PDGFRA(−)hDPSCs showed higher proliferation capacity and enhanced in vitro multilineage differentiation potentials(osteogenic,chondrogenic and adipogenic)compared with their counterparts PDGFRA(+)subpopulation.Furthermore,the MCAM(+)JAG(+)PDGFRA(−)hDPSCs showed enhanced bone tissue formation and adipose tissue formation after 4-week subcutaneous implantation in nude mice.Taken together,our study for the first time revealed the cellular composition switch of monolayer cultured hDPSCs compared to the freshly isolated hDPSCs.After in vitro expansion,the MCAM(+)JAG(+)PDGFRA(−)subpopulation resembled the most transcriptional characteristics of fresh hDPSCs which may be beneficial for further tissue regeneration applications.Yu Cui Wei Ji Yongyan Gao Yao Xiao Huan Liu Zhi Chen 2021International Journal of Oral Science2021,13,4:2

20Soluble CD163 and CD163 Expression on Monocytes Associated with Chronic Hepatitis B Inflammation and HBsAg Loss显示文摘Background and Aims:Monocyte/macrophage-associat-ed CD163 is an indicator of the severity of liver inflam-mation and cirrhosis,but the difference of soluble CD163(sCD163)levels in chronic hepatitis B(CHB)patients and hepatitis B surface antigen(HBsAg)-loss patients is un-clear.Herein,we aimed to compare the sCD163 levels in CHB patients and HBsAg-loss patients with or without an-tiviral treatment.Methods:sCD163 and CD163 expres-sion on monocytes were compared among four groups,healthy subjects,treatment-naïve CHB patients,sponta-neous HBsAg-loss patients,and treatment-related HBsAg-loss patients.The correlation between sCD163 levels and clinical parameters in CHB patients was analyzed.A group of 80 patients with hepatitis B virus(HBV)infection and liver biopsy were recruited.Results:sCD163 levels were higher in the CHB group than in the other three groups.sCD163 levels were higher in treatment-related HBsAg-loss patients than in spontaneous HBsAg-loss patients.sCD163 levels were negatively correlated with hepatitis B e-antigen(HBeAg)and HBsAg levels in HBeAg-positive patients.Liv-er biopsy results further demonstrated that sCD163 levels were elevated in CHB patients with substantial inflamma-tion(A≥2)or fibrosis(F≥2).The sCD163 model was more sensitive in predicting inflammation than other noninvasive models.Its levels were higher in patients with normal ala-nine aminotransferase levels and significant inflammation(A≥2)than in patients with no or mild inflammation.Con-clusions:sCD163 and CD163 expression on monocytes were associated with CHB inflammation and HBsAg loss,and may be used as markers to predict HBV-specific im-mune activation.Peilin Xie Bilian Yao Dao Huang Yongyan Chen Qiming Gong Xinxin Zhang 2022Journal of Clinical and Translational Hepatology2022,10,6:2
