1Gene admixture in ethnic populations in upper part of Silk Road revealed by mtDNA polymorphism显示文摘To evaluate the gene admixture on the current genetic landscape in Gansu Corridor (GC) in China, the upper part of the ancient Silk Road which connects the Eastern and Central Asia, we examined mitochondrial DNA (mtDNA) polymorphisms of five ethnic populations in this study. Using PCR-RFLP and sequencing, we analyzed mtDNA haplotypes in 242 unrelated samples in three ethnic populations from the GC region and two ethnic populations from the adjacent Xinjiang Uygur Autonomous Region of China. We analyzed the data in comparison with the previously reported data from Eastern, Central and Western Asia and Europe. We found that both European-specific haplogroups and Eastern Asian-specific haplogroups exist in the Gansu Corridor populations, while a modest matrilineal gene flow from Europeans to this region was revealed. The Gansu Corridor populations are genetically located between Eastern Asians and Central Asians, both of who contributed significantly to the maternal lineages of the GC populations. 2Application of Avaatech X-ray fluorescence core-scanning in Sr/Ca analysis of speleothems显示文摘Application of X-ray fluorescence core-scanning(XRF-CS) on both marine and lake sediments has achieved remarkable results. However, its application has not been widely extended to the research on speleothems. In this study, we measure the Sr abundance and the Sr/Ca ratios of three stalagmites(two aragonite stalagmites, one calcite stalagmite) using the state-of-the-art fourth-generation Avaatech high-resolution XRF core scanner. Through comparisons among different scan paths and among different scan resolutions, as well as comparisons with inductively coupled plasma optical emission spectrometer(ICP-OES), Itrax XRF, and Artax XRF results, we confirm that the Avaatech XRF core scanner could precisely, quickly, and nondestructively analyze the high-resolution Sr abundance of speleothems. 3Parallel selection of distinct Tof5 alleles drove the adaptation of cultivated and wild soybean to high latitudes显示文摘Photoperiod responsiveness is a key factor limiting the geographic distribution of cultivated soybean and its wild ancestor.In particular,the genetic basis of the adaptation in wild soybean remains poorly understood.In this study,by combining whole-genome resequencing and genome-wide association studies we identified a novel locus,Time of Flowering 5(Tof5),which promotes flowering and enhances adaptation to high latitudes in both wild and cultivated soybean.By genomic,genetic and transgenic analyses we showed that Tof5 en-codes a homolog of Arabidopsis thaliana FRUITFULL(FUL).Importantly,further analyses suggested that different alleles of Tof5 have undergone parallel selection.The Tof5H1 allele was strongly selected by humans after the early domestication of cultivated soybean,while Tof5H2 allele was naturally selected in wild soybean,and in each case facilitating adaptation to high latitudes.Moreover,we found that the key flowering repressor E1 suppresses the transcription of Tof5 by binding to its promoter.In turn,Tof5 physically associates with the promoters of two important FLOWERING LOCUS T(FT),FT2a and FT5a,to upregulate their transcription and promote flowering under long photoperiods.Collectively,ourfindings provide insights into how wild soybean adapted to high latitudes through natural selection and indicate that cultivated soybean underwent changes in the same gene but evolved a distinct allele that was artificially selected after domestication.Lidong Dong Qun Cheng Chao Fang Lingping Kong Hui Yang Zhihong Hou Yongli Li Haiyang Nan Yuhang Zhang Qingshan Chen Chunbao Zhang Kun Kou Tong Su Lingshuang Wang Shichen Li Haiyang Li Xiaoya Lin Yang Tang Xiaohui Zhao Sijia Lu Baohui Liu Fanjiang Kong 2022Molecular Plant2022,15,2:5
4CMIP5/AMIP GCM Simulations of East Asian Summer Monsoon显示文摘The East Asian summer monsoon(EASM) is a distinctive component of the Asian climate system and critically influences the economy and society of the region. To understand the ability of AGCMs in capturing the major features of EASM, 10 models that participated in Coupled Model Intercomparison Project/Atmospheric Model Intercomparison Project(CMIP5/AMIP), which used observational SST and sea ice to drive AGCMs during the period 1979–2008, were evaluated by comparing with observations and AMIP II simulations. The results indicated that the multi-model ensemble(MME) of CMIP5/AMIP captures the main characteristics of precipitation and monsoon circulation, and shows the best skill in EASM simulation, better than the AMIP II MME. As for the Meiyu/Changma/Baiyu rainbelt, the intensity of rainfall is underestimated in all the models. The biases are caused by a weak western Pacific subtropical high(WPSH) and accompanying eastward southwesterly winds in group I models, and by a too strong and west-extended WPSH as well as westerly winds in group II models. Considerable systematic errors exist in the simulated seasonal migration of rainfall, and the notable northward jumps and rainfall persistence remain a challenge for all the models. 5Agronomical selection on loss-of-function of GIGANTEA simultaneously facilitates soybean salt tolerance and early maturity显示文摘Salt stress and flowering time are major factors limiting geographic adaptation and yield productivity in soybean(Glycine max).Although improving crop salt tolerance and latitude adaptation are essential for efficient agricultural production,whether and how these two traits are integrated remains largely unknown.Here,we used a genome-wide association study to identify a major salt-tolerance locus controlled by E2,an ortholog of Arabidopsis thaliana GIGANTEA(GI).Loss of E2 function not only shortened flowering time and maturity,but also enhanced salt-tolerance in soybean.E2 delayed soybean flowering by enhancing the transcription of the core flowering suppressor gene E1,thereby repressing Flowering Locus T(FT)expression.An E2 knockout mutant e2^(CR) displayed reduced accumulation of reactive oxygen species(ROS)during the response to salt stress by releasing peroxidase,which functions in ROS scavenging to avoid cytotoxicity.Evolutionary and population genetic analyses also suggested that loss-of-function e2 alleles have been artificially selected during breeding for soybean adaptation to high-latitude regions with greater salt stress.Our findings provide insights into the coupled selection for adaptation to both latitude and salt stress in soybean;and offer an ideal target for molecular breeding of early-maturing and salt-tolerant cultivars.Lidong Dong Zhihong Hou Haiyang Li Zhaobo Li Chao Fang Lingping Kong Yongli Li Hao Du Tai Li Lingshuang Wang Milan He Xiaohui Zhao Qun Cheng Fanjiang Kong Baohui Liu 2022Journal of Integrative Plant Biology2022,64,10:3
6Safety of endovascular therapy for symptomatic intracranial artery stenosis:a national prospective registry显示文摘Introduction The safety outcomes of endovascular therapy for intracranial artery stenosis in a real-world stetting are largely unknown.The Clinical Registration Trial of Intracranial Stenting for Patients with Symptomatic Intracranial Artery Stenosis(CRTICAS)was a prospective,multicentre,real-world registry designed to assess these outcomes and the impact of centre experience.Methods 1140 severe,symptomatic intracranial arterial stenosis(ICAS)patients treated with endovascular therapy were included from 26 centres,further divided into three groups according to the annual centre volume of intracranial angioplasty and stent placement procedures over 2 years:(1)high volume for≥25 cases/year;(2)moderate volume for 10–25 cases/year and(3)low volume for<10 cases/year.Results The rate of 30-day stroke,transient ischaemic attack or death was 9.7%(111),with 5.4%,21.1%and 9.7%in high-volume,moderate-volume and low-volume centres,respectively(p<0.05).Multivariable logistic regression confirmed high-volume centres had a significantly lower primary endpoint compared with moderate-volume centres(OR=0.187,95%CI:0.056 to 0.627;p≤0.0001),while moderate-volume and low-volume centres showed no significant difference(p=0.8456).Conclusion Compared with the preceding randomised controlled trials,this real-world,prospective,multicentre registry shows a lower complication rate of endovascular treatment for symptomatic ICAS.Non-uniform utilisation in endovascular technology,institutional experience and patient selection in different volumes of centres may have an impact on overall safety of this treatment.Yabing Wang Tao Wang Adam Andrew Dmytriw Kun Yang Liqun Jiao Huaizhang Shi Jie Lu Tianxiao Li Yujie Huang Zhenwei Zhao Wei Wu Jieqing Wan Qinjian Sun Bo Hong Yongli Li Liyong Zhang Jianfeng Chu Qiong Cheng Yiling Cai Pengfei Wang Qi Luo Hua Yang Baijing Dong Yang Zhang Jun Zhao Zuoquan Chen Wei Li Xiaoxin Bai Weiwen He Xueli Cai Maimai Ti Osama O Zaidat 2022Stroke & Vascular Neurology2022,7,2:3
7PrP mutants with different numbers of octarepeat sequences are more susceptible to the oxidative stress显示文摘One of the physiological functions of cellular prion protein(PrP C )is believed to work as a cellular resistance to oxidative stress,in which the octarepeats region within PrP plays an important role.However,the detailed mechanism is less clear.In this study,the expressing plasmids of wild-type PrP (PrP-PG5)and various PrP mutants containing 0(PrP-PG0),9(PrP-PG9)and 12(PrP-PG12)octarepeats were generated and PrP proteins were expressed both in E.coli and in mammalian cells.Protein aggregation and formation of carbonyl groups were clearly seen in the recombinant PrPs expressed from E.coli after treatment of H2O2.MTT and trypan blue staining assays revealed that the cells expressing the mutated PrPs within octarepeats are less viable than the cells expressing wild-type PrP.Statistically significant high levels of intracellular free radicals and low levels of glutathione peroxidase were observed in the cells transfected with plasmids containing deleted or inserted octarepeats.Remarkably more productions of carbonyl groups were detected in the cells expressing PrPs with deleted and inserted octarepeats after exposing to H2O2.Furthermore,cells expressing wild-type PrP showed stronger resistant activity to the challenge of H2O2 at certain extent than the mutated PrPs and mock. 8The effect of collagen hydrogels on chondrocyte behaviors through restricting the contraction of cell/hydrogel constructs显示文摘Collagen is a promising material for tissue engineering,but the poor mechanical properties of collagen hydrogels,which tend to cause contraction under the action of cellular activity,make its application challengeable.In this study,the amino group of type I collagen(Col I)was modified with methacrylic anhydride(MA)and the photo-crosslinkable methacrylate anhydride modified type I collagen(CM)with three different degrees of substitution(DS)was prepared.The physical properties of CM and Col I hydrogels were tested,including micromorphology,mechanical properties and degradation properties.The results showed that the storage modulus and degradation rate of hydrogels could be adjusted by changing the DS of CM.In vitro,chondrocytes were seeded into these four groups of hydrogels and subjected to fluorescein diacetate/propidium iodide(FDA/PI)staining,cell counting kit-8(CCK-8)test,histological staining and cartilage-related gene expression analysis.In vivo,these hydrogels encapsulating chondrocytes were implanted subcutaneously into nude mice,then histological staining and sulfated glycosaminoglycan(sGAG)/DNA assays were performed.The results demonstrated that contraction of hydrogels affected behaviors of chondrocytes,and CM hydrogels with suitable DS could resist contraction of hydrogels and promote the secretion of cartilage-specific matrix in vitro and in vivo.Longpeng Dong Qingli Liu Yongli Gao Hengxing Jia Wenling Dai Likun Guo Hongsong Fan Yujiang Fan Xingdong Zhang 2021Regenerative Biomaterials2021,8,4:2
9Investigation of Micro-mechanical Response of Asphalt Mixtures by a Three-dimensional Discrete Element Model显示文摘The micro-mechanical response of asphalt mixtures was studied using the discrete element method. The discrete element sample of stone mastic asphalt was generated first and the vehicle load was applied to the sample. A user-written program was coded with the FISH language in PFC3 D to extract the contact forces within the sample and the displacements of the particles. Then, the contact forces within the whole sample, in asphalt mastic, in coarse aggregates and between asphalt mastic and coarse aggregates were investigated. Finally, the movement of the particles in the sample was analyzed. The sample was divided into 15 areas and a figure was drawn to show how the balls move in each area according to the displacements of the balls in each area. The displacements of asphalt mastic balls and coarse aggregates were also analyzed. 11Gas-liquid mass transfer in the gas-liquid-solid mini fluidized beds显示文摘The gas-liquid-solid mini fluidized bed(GLSMFB)combines the advantages of fluidized bed and micro-reactor,and meets the requirements for safety and efficiency of green development of process industry.However,there are few studies on its flow performance and no studies on its mass and heat transfer performance.In this paper,the characteristics of gas-liquid mass transfer in a GLSMFB were studied in order to provide basic guidance for the study of GLSMFB reaction performance and application.Using CO_(2)absorption by NaOH as the model process,the gas-liquid mass transfer performance of GLSMFB was investigated.The results show that the liquid volumetric mass transfer coefficient and the gas-liquid interfacial area both increase with the increase of the superficial gas velocity within the experimental parameter range under the same given superficial liquid velocity.At the same ratio of superficial gas to liquid velocity,the liquid volumetric mass transfer coefficient increases with the increase of the super-ficial liquid velocity.Fluidized solid particles strengthen the liquid mass transfer process,and the liquid volumetric mass transfer coefficient is about 13%higher than that of gas-liquid mini bubble column.Tingting Dong Yongli Ma Mingyan Liu 2022Particuology2022,20,10:1

17Excited-state dynamics in light-harvesting complex of Rhodobacter sphaeroides显示文摘Photodynamics of peripheral antenna complexes,light-harvesting complex (LH2) of Rhodobacter (Rb) Sphaeroides 601,was studied using femtosecond pump-probe technique at different laser wavelengths. The obtained results reveal dramatic dynamical evolutions within B800 and B850 absorption bands of antenna complexes LH2. At excitation wavelength around 835 nm,a sharp photobleaching signal was observed which was assigned to the contribution of the two-exciton state,which was further confirmed by the power dependence measurement. Rate equations with eight-level scheme were used to calculate the population evolution in LH2 and the transient dynamics under femtosecond pulse excitation.
