1Polymorphisms in PPARD,PPARG and APM1 associated with four types of Traditional Chinese Medicine constitutions显示文摘Based on the theory of constitution of Traditional Chinese Medicine (TCM), the human population is divided into nine constitutions including one balanced constitution (Normality) and eight unbalanced constitutions (Yang-deficiency, Yin-deficiency, Phlegm-wetness, Qi-deficiency, Wetness-heat, Blood stasis, Depressed constitution, and Inherited special constitution). Different constitutions have specific metabolic features and different susceptibility to certain diseases. However, whether a genetic basis accounts for such constitution classifica-tion is yet to be determined. Here we performed a genetic study to assess the association between genetic variations of metabolic genes in-cluding PPARD, PPARG and APM1 and the constitutions. A total of 233 individuals of the Han population in China were classified into four groups, Normality, Yang-deficiency, Yin-deficiency and Phlegm-wetness with whom 23 single nucleotide polymorphisms (SNPs) in the three genes were genotyped using polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) method. Biased distri-bution of PPARD rs2267669 and rs2076167, APM1 rs7627128 and rs1063539 in Yang-deficiency, PPARG Pro12Ala in Yin-deficiency and PPARD rs2076167, APM1 rs266729 and rs7627128 in Phlegm-wetness were observed. The frequencies of Haplotype13 (Hap13) of PPARG in Yin-deficiency, Hap25 of APM1 in Yang-deficiency and Hap2 of PPARD and Hap14 of PPARG in Phlegm-wetness, were significantly different from those in Normality, suggesting those might be group-associated haplotypes. 2Epigenetic regulation in B-cell maturation and its dysregulation in autoimmunity显示文摘B cells have a critical role in the initiation and acceleration of autoimmune diseases, especially those mediated byautoantibodies. In the peripheral lymphoid system, mature B cells are activated by self or/and foreign antigens andsignals from helper T cells for differentiating into either memory B cells or antibody-producing plasma cells.Accumulating evidence has shown that epigenetic regulations modulate somatic hypermutation and class switchDNA recombination during B-cell activation and differentiation. Any abnormalities in these complex regulatoryprocesses may contribute to aberrant antibody production, resulting in autoimmune pathogenesis such as systemiclupus erythematosus. Newly generated knowledge from advanced modern technologies such as next-generationsequencing, single-cell sequencing and DNA methylation sequencing has enabled us to better understand B-cellbiology and its role in autoimmune development. 3Gene admixture in ethnic populations in upper part of Silk Road revealed by mtDNA polymorphism显示文摘To evaluate the gene admixture on the current genetic landscape in Gansu Corridor (GC) in China, the upper part of the ancient Silk Road which connects the Eastern and Central Asia, we examined mitochondrial DNA (mtDNA) polymorphisms of five ethnic populations in this study. Using PCR-RFLP and sequencing, we analyzed mtDNA haplotypes in 242 unrelated samples in three ethnic populations from the GC region and two ethnic populations from the adjacent Xinjiang Uygur Autonomous Region of China. We analyzed the data in comparison with the previously reported data from Eastern, Central and Western Asia and Europe. We found that both European-specific haplogroups and Eastern Asian-specific haplogroups exist in the Gansu Corridor populations, while a modest matrilineal gene flow from Europeans to this region was revealed. The Gansu Corridor populations are genetically located between Eastern Asians and Central Asians, both of who contributed significantly to the maternal lineages of the GC populations. 5Rational design of novel ultra-small amorphous Fe_(2)O_(3)nanodots/graphene heterostructures for all-solid-state asymmetric supercapacitors显示文摘Constructing graphene-based heterostructures with large interfacial area is an efficient approach to enhance the electrochemical performance of supercapacitors but remains great challenges in their synthesis.Herein,a novel ultra-small amorphous Fe_(2)O_(3)nanodots/graphene heterostructure(a-Fe_(2)O_(3)NDs/RGO)aerogel was facilely synthesized via excessive metal-ion-induced self-assembly and subsequent calcination route using Prussian blue/graphene oxide(PB/GO)composite aerogel as precursors.The deliberately designed a-Fe_(2)O_(3)NDs/RGO heterostructure offers a highly interconnected porous conductive network,large heterostructure interfacial area,and plenty of accessible active sites,greatly facilitating the electron transfer,electrolyte diffusion,and pseudocapacitive reactions.The obtained a-Fe_(2)O_(3)NDs/RGO aerogel could be used as flexible free-standing electrodes after mechanical compression,which exhibited a significantly enhanced specific capacitance of 347.4 F·g^(-1)at 1 A·g^(-1),extraordinary rate capability of 184 F·g^(-1)at 10 A·g^(-1),and decent cycling stability.With the as-prepared a-Fe_(2)O_(3)NDs/RGO as negative electrodes and the Co_(3)O_(4)NDs/RGO as positive electrodes,an all-solid-state asymmetric supercapacitor(a-Fe_(2)O_(3)NDs/RGO//Co_(3)O_(4)NDs/RGO asymmetric supercapacitor(ASC))was assembled,which delivered a high specific capacitance of 69.1 F·g^(-1)at 1 A·g^(-1)and an impressive energy density of 21.6 W·h·k·g^(-1)at 750 W·k·g^(-1),as well as good cycling stability with a capacity retention of 94.3%after 5,000 cycles.This work provides a promising avenue to design high-performance graphene-based composite electrodes and profound inspiration for developing advanced flexible energy-storage devices.Chenxiao Wu Zhifang Zhang Zhonghui Chen Zuimin Jiang Huiyu Li Haijing Cao Yongsheng Liu Yanyan Zhu Zebo Fang Xiangrong Yu 2021Nano Research2021,14,4:6

7A dual-active Co-CoO heterojunction coupled with Ti_(3)C_(2)-MXene for highly-performance overall water splitting显示文摘Development of cost-effective and highly-efficient bifunctional hydrogen evolution reaction(HER)and oxygen evolution reaction(OER)catalysts is crucial for overall water splitting in practical utilization.Herein,we proposed a novel non-noble metal bifunctional,HER/OER electrocatalyst by synergistically coupling a dual-active Co-based heterojunction(Co-CoO)with high conductive and stable two-dimensional Ti_(3)C_(2)-MXene(defined as Co-CoO/Ti^(3)CrMXene).A series of characterizations and theoretical calculations'verify that the synergistic effect of metallic Co with HER activity and Coo with OER performance leads to superb bifunctional catalytic performance,and Ti_(3)C_(2)-MXene can enhance electrical conductivity and prevent the aggregation of the Co-based catalysts,thereby improving both the activity arid stability.Co-Co0/Ti_(3)C_(2)-MXene presents low onset potential(11onse1)of 8 mV and,Tafel slope of 47 mV·dec^(-1) for HER(close to that of Pt/C)and 17onset of 196 mV and Tafel slope of 47 mV·dec^(-1) for OER(superior to:that of Ru02).Assembled as an electrolyzer,Co-CoO/Ti_(3)C_(2)-MXene shows a low voltage of 1.55 V at 10 mA·cm^(-2),high Faradaic efficiency and remarkable stability.It can be driven by a solar cell of-1.55 V for consecutive production of hydrogen and oxygen gases.Dezheng Guo Xin Li Yanqing Jiao Haijing Yan Aiping Wu Ganceng Yang Yu Wang Chungui Tian Honggang Fu 2022Nano Research2022,15,1:2
8Evolutionary characteristics of exons expressed at different abundance levels in mammals显示文摘With comparative genomics approaches, we evaluated the evolutionary characteristics of conservation of exons which are expressed abundantly, moderately or lowly in mammals. Using non-coding regions and pseudogenes as controls, sequence identity, phastCons and Ka/Ks analyses were carried out and our results showed that as the exons of high abundance are highly conserved, the minor and low exons also showed conservative characteristics in evolution. 14Mn-doped Ti-based MOFs for magnetic resonance imaging-guided synergistic microwave thermal and microwave dynamic therapy of liver cancer显示文摘Currently,precise ablation of tumors without damaging the surrounding normal tissue is still an urgent problem for clinical microwave therapy of liver cancer.Herein,we synthesized Mn-doped Ti MOFs(Mn-Ti MOFs)nanosheets by in-situ doping method and applied them for microwave therapy.Infrared thermal imaging results indicate Mn-Ti MOFs can rapidly increase the temperature of normal saline,attributing to the porous structure improving microwave-induced ion collision frequency.Moreover,Mn-Ti MOFs show higher 1O2 output than Ti MOFs under 2 W of low-power microwave irradiation due to the narrower band-gap after Mn doping.At the same time,Mn endows the MOFs with a desirable T1 contrast of magnetic resonance imaging(r2/r1=2.315).Further,results on HepG2 tumor-bearing mice prove that microwave-triggered Mn-Ti MOFs nearly eradicate the tumors after 14 days of treatment.Our study offers a promising sensitizer for synergistic microwave thermal and microwave dynamic therapy of liver cancer.Qiongyu Qin Ming Yang Yu Shi Haijing Cui Chunshu Pan Wenzhi Ren Aiguo Wu Jianqing Hu 2023Bioactive Materials2023,,9:1
15Alternations in the gut microbiota and metabolome with newly diagnosed unstable angina显示文摘Gut microbiota plays an important role in coronary heart disease, but its compositional and functional changes in unstable angina(UA) remain unexplored. We performed metagenomic sequencing of 133 newly diagnosed UA patients and 133 sex-and age-matched controls, and profiled the fecal and plasma metabolomes in 30 case-control pairs. The alpha diversity of gut microbiota was increased in UA patients:the adjusted odds ratios(ORs) per standard deviation increase in Shannon and Simpson indices were 1.30(95% confidence interval, 1.01-1.70) and 1.36(1.05-1.81), respectively. Two common species(depleted Klebsiella pneumoniae and enriched Streptococcus parasanguinis;P ≤ 0.002) and three rare species(depleted Weissella confusa, enriched Granulicatella adiacens and Erysipelotrichaceae bacterium 6_1_45;P ≤ 0.005) were associated with UA. The UA-associated gut microbiota was depleted in the pathway of Lphenylalanine degradation(P = 0.001), primarily contributed by Klebsiella pneumoniae. Consistently, we found increased circulating phenylalanine in UA patients(OR = 2.76 [1.17-8.16]). Moreover, Streptococcus parasanguinis was negatively correlated with fecal citrulline(Spearman's r_(s)=-0.470, P = 0.009), a metabolite depleted in UA patients(OR = 0.26 [0.08-0.63]).
