AXL is a candidate receptor for SARS-CoV-2 that promotes infection of pulmonary and bronchial epithelial cells显示文摘The current coronavirus disease 2019(COVID-19)pandemic presents a global public health challenge. The viral pathogen responsible, severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),binds to the host receptor ACE2 through its spike(S)glycoprotein, which mediates membrane fusion and viral entry.Although the role of ACE2 as a receptor for SARS-CoV-2 is clear, studies have shown that ACE2 expression is extremely low in various human tissues, especially in the respiratory tract. Thus, other host receptors and/or co-receptors that promote the entry of SARS-CoV-2 into cells of the respiratory system may exist. In this study,we found that the tyrosine-protein kinase receptor UFO(AXL)specifically interacts with the N-terminal domain of SARS-CoV-2 S.Using both a SARS-CoV-2 virus pseudotype and authentic SARS-CoV-2,we found that overexpression of AXL in HEK293T cells promotes SARS-CoV-2 entry as efficiently as overexpression of ACE2,while knocking out AXL significantly reduces SARS-CoV-2 infection in HI 299 pulmonary cells and in human primary lung epithelial cells.Soluble human recombinant AXL blocks SARS-CoV-2 infection in cells expressing high levels of AXL.The AXL expression level is well correlated with SARS-CoV-2 S level in bronchoalveolar lavage fluid cells from COVID-19 patients.Taken together, our findings suggest that AXL is a novel candidate receptor for SARS-CoV-2 which may play an important role in promoting viral infection of the human respiratory system and indicate that it is a potential target for future clinical intervention strategies. | Shuai Wang Zongyang Qiu Yingnan Hou Xiya Deng Wei Xu Tingting Zheng Peihan Wu Shaofang Xie Weixiang Bian Chong Zhang Zewei Sun Kunpeng Liu Chao Shan Aifu Lin Shibo Jiang Youhua Xie Qiang Zhou Lu Lu Jing Huang Xu Li | 2021 | Cell Research2021,31,2: | 39
Sonic hedgehog signaling in kidney fibrosis: a master communicator显示文摘The hedgehog signaling cascade is an evolutionarily conserved pathway that regulates multiple aspects of embryonic development and plays a decisive role in tissue homeostasis. As the best studied member of three hedgehog ligands, sonic hedgehog(Shh) is known to be associated with kidney development and tissue repair after various insults. Recent studies uncover an intrinsic link between dysregulated Shh signaling and renal fibrogenesis. In various types of chronic kidney disease(CKD), Shh is upregulated specifically in renal tubular epithelium but targets interstitial fibroblasts, thereby mediating a dynamic epithelialmesenchymal communication(EMC). Tubule-derived Shh acts as a growth factor for interstitial fibroblasts and controls a hierarchy of fibrosis-related genes, which lead to the excessive deposition of extracellular matrix in renal interstitium. In this review, we recapitulate the principle of Shh signaling, its activation and regulation in a variety of kidney diseases. We also discuss the potential mechanisms by which Shh promotes renal fibrosis and assess the efficacy of blocking this signaling in preclinical settings. Continuing these lines of investigations will provide novel opportunities for designing effective therapies to improve CKD prognosis in patients. | Dong Zhou Roderick J.Tan Youhua Liu | 2016 | Science China(Life Sciences)2016,59,9: | 18
Astronomical tuning and magnetostratigraphy of Neogene biogenic reefs in Xisha Islands, South China Sea显示文摘Biogenic reefs are one of two major depositional types in the South China Sea, and are constructed by coral, algae and bryozoa. The West Pacific is a major area of biogenic reef development and plays a critical role in the global carbon cycle. However, the lack of geochronological studies in previous works inhibits our understanding of their contributions. Herein, we conduct a cyclostratigraphic and magnetostratigraphic study on Neogene biogenic reefs using the XK–1 core that was drilled at the Shidao Island,Xisha(Paracel) Islands. The main findings of this study are:(1) the establishment of reliable magentostratigraphy for Ledong, Huangliu, Meishan and Sanya Formations;(2) the magnetic susceptibility variation can be inferred as growth index and tuned to the 405–ka long eccentricity cycle;(3) the astronomical geochronology suggests that the bottom ages for Ledong, Yinggehai, Huangliu, Meishan, and Sanya Formations are 2.2 Ma, 5.7 Ma, 10.4 Ma, 16.6 Ma, and 24.3 Ma, respectively; and (4) Earth's eccentricity and obliquity played predominant roles in biogenic reef establishment on orbital to tectonic timescales. Thus, the reported geochronology offers an opportunity to test the contributions of various factors and hypothesize their roles in the global carbon cycle in future. | Liang Yi Zhimin Jian Xinyu Liu Youhua Zhu Daojun Zhang Zhenfeng Wang Chenglong Deng | 2018 | Science Bulletin2018,63,9: | 12

Low-cost,μm-thick,tape-free electronic tattoo sensors with minimized motion and sweat artifacts显示文摘Electronic tattoos(e-tattoos),also known as epidermal electronics, are ultra-thin and ultra-soft noninvasive but skin-conformable devices with capabilities including physiological sensing and transdermal stimulation and therapeutics. The fabrication of e-tattoos out of conventional inorganic electronic materials used to be tedious and expensive.Recently developed cut-and-paste method has significantly simplified the process and lowered the cost. However, existing cut-and-paste method entails a medical tape on which the electronic tattoo sensors should be pasted, which increases tattoo thickness and degrades its breathability. To address this problem, here we report a slightly modified cut-and-paste method to fabricate low-cost,open-mesh e-tattoos with a total thickness of just 1.5μm.E-tattoos of such thinness can be directly pasted on human skin and conforms to natural skin texture. We demonstrate that this ultra-thin,tape-free e-tattoo can confidently measure electrocardiogram(ECG),skin temperature, and skin hydration.Heart rate and even respiratory rate can be extracted from the ECG signals.A special advantage of such ultra-thin e-tattoo is that it is capable of high-fidelity sensing with minimized motion artifacts under various body movements.Effects of perspiration are found to be insignificant due to the breathability of such e-tattoos. | Youhua Wang Yitao Qiu Shideh Kabiri Ameri Hongwoo Jang Zhaohe Dai YongAn Huang Nanshu Lu | 2018 | npj Flexible Electronics2018,2,1: | 9
Receptome profiling identifies KREMEN1 and ASGR1 as alternative functional receptors of SARS-CoV-2显示文摘Host cellular receptors play key roles in the determination of virus tropism and pathogenesis. However, little is known about SARS-CoV-2 host receptors with the exception of ACE2.Furthermore,ACE2 alone cannot explain the multi-organ tropism of SARS-CoV-2 nor the clinical differences between SARS-CoV-2 and SARS-CoV,suggesting the involvement of other receptor(s).Here,we performed genomic receptor profiling to screen 5054 human membrane proteins individually for interaction with the SARS-CoV-2 capsid spike(S)protein.Twelve proteins, including ACE2,ASGR1,and KREMEN1,were identified with diverse S-binding affinities and patterns.ASGR1 or KREMEN1 is sufficient for the entry of SARS-CoV-2 but not SARS-CoV in vitro and in vivo.SARS-CoV-2 utilizes distinct ACE2/ASGR1/KREMEN1(ASK)receptor combinations to enter different cell types, and the expression of ASK together displays a markedly stronger correlation with virus susceptibility than that of any individual receptor at both the cell and tissue levels. The cocktail of ASK-related neutralizing antibodies provides the most substantial blockage of SARS-CoV-2 infection in human lung organoids when compared to individual antibodies. Our study revealed an interacting host receptome of SARS-CoV-2,and identified ASGR1 and KREMEN1 as alternative functional receptors that play essential roles in ACE2-independent virus entry, providing insight into SARS-CoV-2 tropism and pathogenesis,as well as a community resource and potential therapeutic strategies for further COVID-19 investigations. | Yunqing Gu Jun Cao Xinyu Zhang Hai Gao Yuyan Wang Jia Wang Juan He Xiaoyi Jiang Jinlan Zhang Guanghui Shen Jie Yang Xichen Zheng Gaowei Hu Yuanfei Zhu Shujuan Du Yunkai Zhu Rong Zhang Jianqing Xu Fei Lan Di Qu Guoliang Xu Yun Zhao Dong Gao Youhua Xie Min Luo Zhigang Lu | 2022 | Cell Research2022,32,1: | 8
Bile acids evoke placental inflammation by activating Gpbar1/NF-κB pathway in intrahepatic cholestasis of pregnancy显示文摘Intrahepatic cholestasis of pregnancy(ICP)is a cholestatic disorder with potentially deleterious consequences for fetuses.Although a clear correlation between the elevated levels of maternal serum bile acids and deficient fetal outcome has been established in clinical practice, the underlying mechanisms remain elusive.Herein,we report that bile acids induce NF-κB pathway activation via G protein-coupled bile acid receptor 1(Gpbar1),with consequent upregulation of inflammatory genes in trophoblasts, leading to aberrant leukocyte infiltration and inflammation in placenta.Ursodeoxycholic acid(UDCA),a drug used clinically to treat ICP,competes with other bile acids for binding with Gpbar1 and thus inhibits bile acid-induced inflammatory response in trophoblasts and improves fetal survival in pregnant rats with obstructive cholestasis.Notably, inhibition of NF-κB by andrographolide is more effective than UDCA in benefiting placentas and fetuses.Thus,anti-inflammation therapy targeting Gpbar1/NF-κB pathway could be effective in suppressing bile acid-induced inflammation and alleviating ICP-associated fetal disorders. | YouHua Zhang YouDong Pan ChangDong Lin YaJuan Zheng Hao Sun HaiLong Zhang JunLei Wang MengYa Yuan Tao Duan QiaoLing Du JianFeng Chen | 2016 | Journal of Molecular Cell Biology2016,8,6: | 8
Oxygen and carbon isotope records of calcareous nanno-fossils for the past 1 Ma in the southern South China Sea显示文摘Oxygen and carbon isotopic analyses have been carried out on calcareous nannofossils from ODP Site 1143 in the southern South China Sea. The results demonstrate that the δ18O values of calcareous nannofossils for the past 1 Ma vary systematically with those of planktonic and benthic foraminifera from the same site. But the average δ18O value of nannofossils is 1.869‰ higher than that of planktonic foraminifera, and is 3.855‰ lower than that of benthic foar-minifera. There are systematic differences between the δ13C values of nannofossils, planktonic foarminifera and benthic foraminifera. The average δ13C of nannofossils is 0.756‰ higher than that of benthic foraminifera, and is 0.460‰ lower than that of planktonic foraminifera. The δ13C values of nannofossils exhibit a significant positive shift beginning near isotope stage 14 and lasting until stage 8. This study also finds that there is a close ralationship between the δ13C of nannofossils and the percentage of Florisphaera profundo. | 11 | RBD-Fc-based COVID-19 vaccine candidate induces highly potent SARS-CoV-2 neutralizing antibody response显示文摘The pandemic of coronavirus disease 2019(COVID-19)caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)has posed serious threats to global health and economy, thus calling for the development of safe and effective vaccines. The receptorbinding domain(RBD)in the spike protein of SARS-CoV-2 is responsible for its binding to angiotensin-converting enzyme 2(ACE2)receptor.It contains multiple dominant neutralizing epitopes and serves as an important antigen for the development of COVID-19 vaccines. Here,we showed that immunization of mice with a candidate subunit vaccine consisting of SARS-CoV-2 RBD and Fc fragment of human IgG,as an immunopotentiator, elicited high titer of RBD-specific antibodies with robust neutralizing activity against both pseudotyped and live SARS-CoV-2 infections. The mouse antisera could also effectively neutralize infection by pseudotyped SARS-CoV-2 with several natural mutations in RBD and the IgG extracted from the mouse antisera could also show neutralization against pseudotyped SARS-CoV and SARS-related coronavirus(SARSr-CoV).Vaccination of human ACE2 transgenic mice with RBD-Fc could effectively protect mice from the SARS-CoV-2 challenge. These results suggest that SARS-CoV-2 RBD-Fc has good potential to be further developed as an effective and broad-spectrum vaccine to prevent infection of the current SARS-CoV-2 and its mutants,as well as future emerging SARSr-CoVs and re-emerging SARS-CoV. | Zezhong Liu Wei Xu Shuai Xia Chenjian Gu Xinling Wang Qian Wang Jie Zhou Yanling Wu Xia Cai Di Qu Tianlei Ying Youhua Xie Lu Lu Zhenghong Yuan Shibo Jiang | 2020 | Signal Transduction and Targeted Therapy2020,5,1: | 7

Expression of NAC1 up-stream regulatory region and its relationship to the lateral root initiation induced by gibberellins and auxins显示文摘A 1050 bp up-stream regulatory fragment of the transcription factor gene NAC1 in Arabidopsis thaliana was isolated using polymerase chain reaction(PCR)based techniques. The fragment was used to substitute the 35S promoter of the pBI121 plasmid to construct a β-glucuronidase gene(GUS)expression system. The construct was introduced into tobacco(Nico- tiana tabaccum)plants by the Agrobacterium-mediated transferring method.GUS expression pattern was studied by using the transgenic lines. The results showed that the GUS driven by the NAC1 up-stream regulatory region was specifically expressed in the root meristem region, basal areas of the lateral root primordium and the lateral roots. The GUS expression was induced by 3-indolebutyric acid (IBA)and gibberellins(GA3 and GA4+7).The results indicated that the up-stream regulatory fragment of NAC1 responded to plant hormones. The fragment might be involved in both auxins and gibberel- lins signaling in promoting the development of lateral roots. | WANG Youhua 1 ,DUAN Liusheng 1 ,LU Mengzhu 2 ,LI Zhaohu 1 ,WANG Minjie 2 &ZHAI Zhixi 1 1.State Key Laboratory of Plant Physiology and Biochemistry,Department of Agronomy,College of Agronomy and Biotech- nology,China Agricultural University,Beijing 100094,China 2.Forestry Institute,Chinese Academy of Forestry,Beijing 100091,China | 2006 | Science China(Life Sciences)2006,49,5: | 5
A novel STING agonist-adjuvanted pan-sarbecovirus vaccine elicits potent and durable neutralizing antibody and T cell responses in mice, rabbits and NHPs显示文摘The emergence of SARS-CoV-2 variants and potentially other highly pathogenic sarbecoviruses in the future highlights the need for pan-sarbecovirus vaccines. Here,we discovered a new STING agonist,CF501,and found that CF501-adjuvanted RBD-FC vaccine(CF501/RBD-FC)elicited significantly stronger neutralizing antibody(nAb)and T cell responses than Alum-and cGAMP-adjuvanted RBD-FC in mice.Vaccination of rabbits and rhesus macaques(nonhuman primates,NHPs)with CF501/RBD-FC elicited exceptionally potent nAb responses against SARS-CoV-2 and its nine variants and 41 S-mutants,SARS-CoV and bat SARSr-CoVs.CF501/RBD-FC-immunized hACE2-transgenic mice were almost completely protected against SARS-CoV-2 challenge, even 6 months after the initial immunization.NHPs immunized with a single dose of CF501/RBD-FC produced high titers of nAbs.The immunized macaques also exhibited durable humoral and cellular immune responses and showed remarkably reduced viral load in the upper and lower airways upon SARS-CoV-2 challenge even at 108 days post the final immunization.Thus,CF501/RBD-Fc can be further developed as a novel pan-sarbecovirus vaccine to combat current and future outbreaks of sarbecovirus diseases. | Zezhong Liu Jie Zhou Wei Xu Wei Deng Yanqun Wang Meiyu Wang Qian Wang Ming Hsieh Jingming Dong Xinling Wang Weijin Huang Lixiao Xing Miaoling He Chunlin Tao Youhua Xie Yilong Zhang Youchun Wang Jincun Zhao Zhenghong Yuan Chuan Qin Shibo Jiang Lu Lu | 2022 | Cell Research2022,32,3: | 5
Habitat suitability modeling of amphibian species in southern and central China:environmental correlates and potential richness mapping显示文摘Successful wildlife management must take into account suitable habitat areas.Information on the correlation between distribution ranges and environmental conditions would, therefore, improve the efficacy of in-situ conservation of wildlife. In this contribution, correlations between environmental factors and the distribution of 51 amphibians in southern and central China were investigated.Ecological niche factor analysis(ENFA) at a spatial resolution of 1° latitude×1° longitude identified a mixture of climatic and habitat factors as important predictors of the occurrence of individual species. The aims of the present work were(i) to evaluate potential distributions of amphibians based on the suitability of areas;(ii) to identify the major environmental descriptors upon which they depend;and(iii) to identify areas of potential high richness that have been overlooked in available inventories. Most of the predicted species ranges of species covered the majority of southern and central China.Six richness hotspots were predicted,of which four have been described previously, but two overlooked(SE Fujian and SE Qinghai). The prediction model was considered to be relatively accurate and it is recommended that these two new potential hotspots should be subjected to further evaluation and sampling efforts.Amphibians have high ecological preference for high humidity and precipitation, and low annual frost days.ENFA is a useful tool in wildlife conservation assessment because it is able to identify potential hotspots where studies on the correlations between environmental descriptors and the occurrence of particular species could be focused. | CHEN YouHua | 2013 | Science China(Life Sciences)2013,56,5: | 5
A highly potent and stable pan-coronavirus fusion inhibitor as a candidate prophylactic and therapeutic for COVID-19 and other coronavirus diseases显示文摘The development of broad-spectrum antivirals against human coronaviruses(HCoVs)is critical to combat the current coronavirus disease 2019(COVID-19)pandemic caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)and its variants,as well as future outbreaks of emerging CoVs.We have previously identified a polyethylene glycol-conjugated(PEGylated)lipopeptide,EK1C4,with potent pan-CoV fusion inhibitory activity. However,PEG linkers in peptide or protein drugs may reduce stability or induce anti-PEG antibodies in vivo. Therefore,we herein report the design and synthesis of a series of dePEGylated lipopeptide-based pan-CoV fusion inhibitors featuring the replacement of the PEG linker with amino acids in the heptad repeat 2 C-terminal fragment(HR2-CF)of HCoV-OC43.Among these lipopeptides,EKL1C showed the most potent inhibitory activity against infection by SARS-CoV-2 and its spike(S)mutants,as well as other HCoVs and some bat SARS-related coronaviruses(SARSr-CoVs)tested.The dePEGylated lipopeptide EKL1C exhibited significantly stronger resistance to proteolytic enzymes, better metabolic stability in mouse serum, higher thermostability than the PEGylated lipopeptide EK1C4,suggesting that EKL1C could be further developed as a candidate prophylactic and therapeutic for COVID-19 and other coronavirus diseases. | Jie Zhou Wei Xu Zezhong Liu Chao Wang Shuai Xia Qiaoshuai Lan Yanxing Cai Shan Su Jing Pu Lixiao Xing Youhua Xie Lu Lu Shibo Jiang Qian Wang | 2022 | Acta Pharmaceutica Sinica B2022,12,4: | 5

Ameliorative effects of potassium on drought-induced decreases in fiber length of cotton(Gossypium hirsutum L.) are associated with osmolyte dynamics during fiber development显示文摘Fiber length of cotton(Gossypium hirsutum L.)decreases under drought stress, potassium(K)could diminish the decreased caused by drought, but the mechanism associated with this alleviation effect is not clear. We evaluated the effect of K on fiber elongation using two cotton cultivars,Simian 3 and Siza 3,grown in well-watered and drought-stressed conditions.Potassium fertilizer(K2O)was applied 0,150,or 300 kg ha?1 in each growing condition.Drought stress reduced the final fiber length due to a decline in the maximum rate of rapid elongation(Vmax,mmday?1).The application of K alleviated the droughtinduced fiber length reduction by increasing Vmax.At 10 and 15 days post-anthesis(DPA),drought significantly reduced osmotic potential(OP)and increased K+and malate contents at all K rates, relative to well-watered conditions, which was associated with increased activities of phosphoenolpyruvate carboxylase(PEPC),V-ATPase,PPase,and PM H+-ATPase in cotton fiber. However, the relative contribution of K+and malate to OP declined under drought in comparison with well-watered condition.Compared with control without K,K application decreased OP and increased the accumulation of osmolytes(K+,malate and soluble sugar)as well as the activities of related enzymes in fiber irrespective of water treatments. Moreover,K application increased osmotic adjustment during drought, and improved the contribution of K+and malate to OP,especially under drought stress. This study showed that drought decreased fiber length by reducing Vmax, and K application ameliorates the decline in fiber elongation due to drought by enhancing osmolytes accumulation and their contribution to OP in fiber cells. | Wenqing Zhao Haoran Dong Rizwan Zahoor Zhiguo Zhou John L.Snider Yinglong Chen Kadambot H.M.Siddique Youhua Wang | 2019 | The Crop Journal2019,7,5: | 4
