Nuclear Aurora kinase A switches m^(6)A reader YTHDC1 to enhance an oncogenic RNA splicing of tumor suppressor RBM4显示文摘Aberrant RNA splicing produces alternative isoforms of genes to facilitate tumor progression, yet how this process is regulated by oncogenic signal remains largely unknown. Here,we unveil that non-canonical activation of nuclear AURKA promotes an oncogenic RNA splicing of tumor suppressor RBM4 directed by m^(6)A reader YTHDC1 in lung cancer.Nuclear translocation of AURKA is a prerequisite for RNA aberrant splicing, specifically triggering RBM4 splicing from the full isoform(RBM4-FL)to the short isoform(RBM4-S)in a kinase-independent manner. | SiSi Li YangFan Qi JiaChuan Yu YuChao Hao Bin He MengJuan Zhang ZhenWei Dai TongHui Jiang SuYi Li Fang Huang Ning Chen Jing Wang MengYing Yang DaPeng Liang Fan An JinYao Zhao WenJun Fan YuJia Pan ZiQian Deng YuanYuan Luo Tao Guo Fei Peng ZhiJie Hou ChunLi Wang FeiMeng Zheng LingZhi Xu Jie Xu QingPing Wen BiLian Jin Yang Wang Quentin Liu | 2022 | Signal Transduction and Targeted Therapy2022,7,5: | 4
Cytosolic phospholipase A2α modulates cell-matrix adhesion via the FAK/paxillin pathway in hepatocellular carcinoma显示文摘Objective: To explore the effect of cytosolic phospholipase A2α(cPLA2α) on hepatocellular carcinoma(HCC) cell adhesion and the underlying mechanisms. Methods: Cell adhesion, detachment, and hanging-drop assays were utilized to examine the effect of cPLA2α on the cell-matrix and cell-cell adhesion. Downstream substrates and effectors of cPLA2α were screened via a phospho-antibody microarray.Associated signaling pathways were identified by the functional annotation tool DAVID. Candidate proteins were verified using Western blot and colocalization was investigated via immunofluorescence. Western blot and immunohistochemistry were used to detect protein expression in HCC tissues. Prognosis evaluation was conducted using Kaplan-Meier and Cox-proportional hazards regression analyses. Results: Our findings showed that cPLA2α knockdown decreases cell-matrix adhesion but increases cell-cell adhesion in HepG2 cells. Microarray analysis revealed that phosphorylation of multiple proteins at specific sites were regulated by cPLA2α. These phosphorylated proteins were involved in various biological processes. In addition, our results indicated that the focal adhesion pathway was highly enriched in the cPLA2α-relevant signaling pathway. Furthermore, cPLA2α was found to elevate phosphorylation levels of FAK and paxillin, two crucial components of focal adhesion. Moreover, localization of p-FAK to focal adhesions in the plasma membrane was significantly reduced with the downregulation of cPLA2α. Clinically, cPLA2α expression was positively correlated with p-FAK levels. Additionally, high expression of both cPLA2α and p-FAK predicted the worst prognoses for HCC patients. Conclusions: Our study indicated that cPLA2α may promote cell-matrix adhesion via the FAK/paxillin pathway, which partly explains the malignant cPLA2α phenotype seen in HCC. | Piao Guo Yuchao He Lu Chen Lisha Qi Dongming Liu Ziye Chen Manyu Xiao Liwei Chen Yi Luo Ning Zhang Hua Guo | 2019 | Cancer Biology & Medicine2019,16,2: | 2
Polymeric Complex Nanoparticles Enable the Fabrication of Mechanically Superstrong and Recyclable Poly(aryl ether sulfone)-based Polymer Composites显示文摘It is a long-term pursuit, and also,a challenge to significantly improve the mechanical strength of thermoplastic polymers using readily dispersed polymers as nanofillers. In this study,we demonstrated that in situ formed carboxylic acid-functionalized poly(aryl ether sulfone)(PAES-COOH)/polyvinylpyrrolidone(PVPON)complex nanoparticles can significantly enhance the mechanical strength of PAES-COOH by mixing PAES-COOH with a small fraction of PVPON.The PAES-COOH/PVPON10%composite,which contained∼10 wt%PVPON,exhibited a tensile strength of∼104.8 MPa and Young’s modulus of∼932.2 MPa,which were∼2.0 and∼1.7 times higher than those of the PAES-COOH,respectively.The PAES-COOH/PVPON nanoparticles which were uniformly dispersed in PAES-COOH matrices and had strong hydrogen-bonding interactions with PAES-COOH,served as nanofillers to reinforce the mechanical strength of the PAES-COOH.The PAES-COOH/PVPON_(10%)composites possessed excellent solvent-assisted healability, and the fractured composites could be healed to restore their original mechanical strength.Meanwhile, the PAES-COOH/PVPON_(10%)composites could be recycled multiple times, and yet retained their shape integration and their original mechanical strength. | Xingyuan Lu Yuchao Luo Yixuan Li Chunyang Bao Xiaohan Wang Ni An Guibin Wang Junqi Sun | 2020 | CCS Chemistry2020,2,1: | 1
Fulgide Derivative-Based Solid-State Reversible Fluorescent Switches for Advanced Optical Memory显示文摘Solid-state fluorescent switches with reversible luminescence characteristics have attracted considerable attention because of their broad applications in advanced photonics, such as anticounterfeiting inks, optical writing and erasing, and biological imaging.Herein,we have fabricated a solid-state reversible fluorescent switch under alternating UV(365 nm)and visible light treatments based on a fulgide(FUL)-functionalized tetraphenylethylene(TPE)derivative(TPE-FUL)containing a photochromic group FUL and aggregation-induced emission(AIE)luminogen TPE.TPE-FUL exhibited excellent reversible absorption and luminescence owing to the interconversion between open TPE-FUL(O-TPE-FUL)and closed TPE-FUL(C-TPE-FUL).Photophysical and theoretical investigations revealed that the luminescence of O-TPE-FUL is based on the local excited state of the TPE moiety, whereas the fluorescence quenching of C-TPE-FUL originates from the intramolecular charge transfer from the TPE to the FUL moiety. The excellent reversible photoswitching properties of TPE-FUL in the solid state allows for its potential use in advanced optical memory applications, such as anticounterfeiting, optical writing and erasing, and information encryption. | Yang Jiao Runqing Yang Yuchao Luo Leijing Liu Bin Xu Wenjing Tian | 2022 | CCS Chemistry2022,4,1: | 1
Ideality analysis and general laws of bubble swarm microflow for large-scale gas-liquid microreaction processes显示文摘The flow ideality of bubbly microflow remains unclear even though it is vital for the design of microreactors, especially the ideality of bubble swarm microflow for large-scale gas-liquid microreaction processes. This work is the first time to report the ideality analysis of the microbubble swarm in a relatively large microchannel. The bubble swarm microflow has undergone two conditions:quasihomogeneous plug flow and liquid phase/gas-liquid qua si-homogeneous phase two-phase laminar flow. Both the deviations of void fraction and bubble velocity from the ideal plug flow can divide into two parts, and the two transition points simultaneously happen at the velocity ratio of 1.25.There exists a critical capillary number to maintain the quasi-homogeneous plug flow, which could be regarded as the general laws for the design of gas-liquid microreactors. Finally,a novel model is developed to predict the bubble velocity. This work could be very helpful for the large-scale gas-liquid microreactors design. | Lin Sheng Yuchao Chen Jian Deng Guangsheng Luo | 2022 | Chinese Journal of Chemical Engineering2022,,10: | 1

Compounded chelating agent derived from fruit residue extracts effectively enhances Cd phytoextraction by Sedum alfredii显示文摘Chelating agent is known as the enhancer for metal phytoextraction;however, there is still a lack of efficient and environmentally sustainable chelators. Here, lemon residue extraction(LRE),prepared from 11 kinds of fruit wastes, was combined with N,N-bis(carboxymethyl)glutamic acid(GLDA),and tea saponin(T.S.)for the compounded plant-derived chelator(CPC),and their influences on Cd phytoextraction by the hyperaccumulator Sedum alfredii was evaluated.Among these fruits, the lemon residue extracted the most significant amount of Cd from the soil. The most effective CPC was at the volume ratio of three agents being 15:4:1(LRE:GLDA:T.S.).Compared with the deionized water, the solubility of three Cd minerals was increased by 85–256 times, and Cd speciation was substantially altered after CPC application. In the pot experiment,CPC addition caused evident increases in plant shoot biomass,Cd phytoextraction efficiency, and organic matter content compared with EDTA and nitrilotriacetic acid(NTA)application.CPC induced fewer changes in bacterial community composition compared with EDTA and had no pronounced influence on microbial biomass carbon and bacterialα-diversity,suggesting CPC had a subtle impact on the microbiological environments. Our study provides a theoretical base for the reutilization of fruit wastes and the development of environmental-friendly chelator that assists Cd phytoextraction. | Jipeng Luo Jiabin Liang Yuchao Song Xinyu Guo Youzheng Ning Nanlin Liu Heping Zhao Tingqiang Li | 2021 | Soil Ecology Letters2021,3,3: | 1
Giant piezoresponse and promising application of environmental friendly small-ion-doped ZnO显示文摘In recent years, with the growing concerns on environmental protection and human health, new materials, such as lead-free piezoelectric materials, have received increasing attention. So far, three types of lead-free piezoelectric systems have been widely researched,i.e.,perovskites,bismuth layer-structured ferroelectrics, and tungsten-bronze type ferroelectrics. This article presents a new type of environmental friendly piezoelectric material with simple structure, the transition-metal(TM)-doped ZnO.Through substituting Zn2+ site with small size ion,we obtained a series of TM-doped ZnO with giant piezoresponse, such as Zn0.975V0.025O of 170 pC/N,Zn0.94Cr0.06O of 120 pC/N,Zn0.913Mn0.087O of 86 pC/N and Zn0.988Fe0.012O of 127 pC/N.The tremendous piezoresponses are ascribed to the introduction of switchable spontaneous polarization and high permittivity in TM-doped ZnO.The microscopic origin of giant piezoresponse is also discussed.Substitution of TM ion with small ionic size for Zn2+ results in the easier rotation of noncollinear TM-O1 bonds along the c axis under the applied field, which produces large piezoelectric displacement and corresponding piezoresponse enhancement.Furthermore,it proposes a general rule to guide the design of new wurtzite semiconductors with enhanced piezoresponses. That is,TM-dopant with ionic size smaller than Zn2+ substitutes for Zn2+ site will increase the piezoresponse of ZnO significantly. Finally,we discuss the improved performances of some TM-doped ZnO based piezoelectric devices. | PAN Feng LUO JingTing ANG YuChao WANG XuBo ZENG Fei | 2012 | Science China(Technological Sciences)2012,55,2: | 0
Precise Detection and Visualization of Cyclooxygenase-2 for Golgi Imaging by a Light-Up AggregationInduced Emission-Based Probe显示文摘Precision-targeted detection of cancer in a rapid and sensitive fashion remains a significant challenge in prevention, management, and in particular, treatment.Herein,we have designed and synthesized a unique cyclooxygenase-2(COX-2)fluorescence probe,dimethylamine-9,10-distyrylanthracene-indomethacin(NDSA-IMC),that could visualize the site of highly expressed COX-2 in the Golgi apparatus of cancer cells, using 9,10-diatyrylanthracene derivative as the luminous unit and a selective inhibitor, indomethacin(IMC),as the recognizing moiety for COX-2.In an aqueous solution, the free state NDSA-IMC showed a weak emission in the absence of COX-2 but enhanced emission in the presence of the enzyme due to the restriction of intramolecular motion of aggregation-induced emission-active NDSA-IMC when bound to COX-2.Cell imaging and flow cytometry experiments indicated that NDSA-IMC could discriminate between cancer and normal cells and visualize the Golgi apparatus of cancer cells via specific targeting of COX-2.Therefore,NDSA-IMC might potentially detect early cancer lesions and ultimately mitigate the population of cancer burden in society. | Yuchao Luo Song Zhang Hui Wang Quan Luo Zhigang Xie Bin Xu Wenjing Tian | 2022 | CCS Chemistry2022,4,2: | 0
A Stiffness-Tunable Composite with Wide Versatility and Applicability Based on Low-Melting-Point Alloys显示文摘Flexible materials are essential in bionic fields such as soft robots. However, the lack of stiffness limits the mechanical performance of soft robots and makes them difficult to develop in many extreme working conditions, such as lifting and excavation operations. To address this issue,we prepared a stiffness-tunable composite by dispersing low-melting-point alloy into thermosetting epoxy resin.A dramatic and rapid change in stiffness was achieved by changing the state of matter at lower temperatures, and accurate control of the composite modulus was achieved by controlling the temperature. When the alloy content is at 30vol%,the tensile modulus changes 41.6 times, while the compressive modulus changes 58.9 times.By applying the composite to a flexible actuator, the initial stiffness of the actuator was improved by 124 times, reaching 332 mN/mm.In addition, the use of stiffness-tunable materials in the wheel allowed for timely changes in the grounding area to improve friction. These flexible materials with manageable mechanical properties have wide applicability in fields including bionics, robotics, and sensing. Our findings provide a new approach to designing and developing flexible materials with improved stiffness and controllability. | Jiawei Xiong Bo Sun Chunbao Liu Konghua Yang Yuchao Luo Yunhong Liang Zhiwu Han Lei Ren Zhaohua Lin | 2023 | Journal of Bionic Engineering2023,20,6: | 0
Transfer Behavior of Carbendazim from Dried Yingshan Yunwu Tea Leaves into Tea Infusions显示文摘Tea infusion is the main route of human exposure to pollutants in tea,and it is vital to investigate the transfer rate of pollutants from tea leaves to infusion.Carbendazim is a commonly used systemic fungicide, the transfer behavior of which in Yingshan Yunwu tea is not clear.Hence, this study firstly established the analytical method for determination of carbendazim with great accuracy and precision via modified QuEChERS method and ultra-performance liquid chromatography-fluorescence detection(UPLC-FLD).Then,the transfer behavior of carbendazim from tea leaves and infusion was systematically investigated. Results indicated that water temperature and proportion of tea to water ration would obviously increase the transfer rate of carbendazim, but times of infusion repetition showed negative correlation with the transfer rate of carbendazim. In addition, brewing time seemed to have lesser impact on the transfer rate. This study will be helpful for the risk assessment of carbendazim residue and provide the guidance for tea brewing. | Xinyu LIU Yuchao TIAN Haijian LIU Qingling ZHU Nan LUO Zhiling HU Chang LIU | 2023 | Agricultural Biotechnology2023,12,5: | 0
Effects of Househould Cleaning Methods on Carbendazim Residue in Apples显示文摘Carbendazim, belonging to fungicides, was commonly used for controlling crop diseases, resulting in frequent residues in fruits and vegetables.Washing was the initial step of household fruit processing for cleaning and reducing pollutant contents. However, the effect of washing on the removal of carbendazim in apples was not systemically studied. Thus,in this study,an analytical method was firstly established for the determination of carbendazim with great accuracy and precision through quick, simple, cheap, effective and safe(QuEChERS)method and ultra-performance liquid chromatography-fluorescence detection(UPLC-FLD).Then,the influencing factors were systematically researched to unveil their effects on the carbendazim residue in apples. The results indicated that water temperature, washing products and washing method could obviously affect the carbendazim content in apples. In addition, the best cleaning scheme for removal of carbendazim was soaking apples for 5 min in a cleaning solution, which was prepared by dissolving 8 g of commercial fruit washing soup with 500 ml of water at 45℃.Under this condition, the removal rate of carbendazim in the washing step reached 73.91%.This study will be helpful for providing guidance for household cleaning methods for controlling pesticide residues. | Zhiling HU Yuchao TIAN Qingling ZHU Haijian LIU Nan LUO Fan WANG Ziyi ZHANG Furong LIU Chang LIU Mengrong CHENG | 2023 | Agricultural Biotechnology2023,12,6: | 0
