| 1 | Effect of methyl tert-butyl ether on adipogenesis and glucose metabolism in vitro and in vivo显示文摘Methyl tert-butyl ether (MTBE),as a widely used gasoline additive,is suspected of being environmentally toxic.MTBE accumulates mainly in adipose tissue,but its effect on obesity or obesity-related metabolic disorders has not been well understood yet.Therefore,we examined the effect of MTBE on the adipose function and the related metabolic processes with both 3T3-L1 cell line and C57BL/6J mice model.We found that exposure to MTBE at the environmental relevant concentration (100 μmol/L) could significantly induce differentiation of preadipocyte and disturb insulin-stimulated glucose uptake of mature adipocyte.The in vivo observation in male mice showed a positive correlation of visceral white adipose tissue (vWAT) expansion and cell size increase with MTBE treatment in 14 weeks.Glucose tolerance and insulin sensitivity tests demonstrated that MTBE at 1000 μg/(kg·day) disturbed the systemic glucose metabolism in a gender-specific manner,which might be partly attributed to the alterations of gut microbiota community at genus level with respect to Akkermansia,Clostridium XIVb,and Megamonas.In summary,our study characterized the effect of MTBE on adipose tissue function and glucose homeostasis in vitro and in vivo,and revealed that systemic disorders of the glucose metabolism might be modulated by the related gut microbiota. | Yue Tang Qidong Ren Qing Wen Caixia Yu Xinni Xie Qing Hu Yuguo Du | 2019 | Journal of Environmental Sciences2019,31,11: | 4 |
| 2 | A neglected GABAergic astrocyte:Calcium dynamics and involvement in seizure activity显示文摘Gamma-aminobutyric acid(GABA) contributes substantially to neurocognitive function as an important inhibitory neurotransmitter in the human cerebral cortex. However, the pathophysiology of disorders such as epilepsy are not well understood, since GABA agonists are not quite effective in treating epilepsy. Knowledge of the mechanism of action of GABA would contribute to review previously proposed anti-epileptic processes by GABA agonists. In this study based on recent experiments on GABAergic astrocytes, we developed a modified GABAergic astrocyte model, and successfully simulated a long-lasting Ca^(2+) oscillation in astrocytes after 0.5-s stimulation of GABAergic transmission. We then incorporated this GABAergic astrocyte model into a classical Ullah-Schiff seizure model and surprisingly found that this GABAergic astrocyte model functions to hinder the anti-epileptic action of GABA agonists, thereby explaining their low efficiency in previous experiments. | 4 | Acute toxicity variation of hydroxyl benzophenone UV filters during photoinduction–chlorination disinfection processes显示文摘Benzophenones(BPs), a group of widely used UV filters, exert multiple, significant toxicity effects. The 11 BPs were selected as target compounds, and the photobacterium acute toxicity test and an index for acute toxicity formation potential(ATFP) were used to evaluate the toxicity variation of BPs before and after a photoinduction–chlorination disinfection process.Orthogonal experiments were performed at different pH values and chlorine dosages. The characteristics of ATFP values for 11 BPs after a photoinduction–chlorination process can be summarized as follows:(1) The ATFPs decreased as the hydroxyl group number increased in BPs molecules.(2) For those BPs with the same hydroxyl group number, the ATFPs were higher when the hydroxyl groups were located at the 3-or 4-position than those at the 2-position; the BPs with hydroxyl groups distributed on two benzene rings had higher ATFPs than those on one ring.(3) Introducing a methoxyl group and sulfonic acid group into BP molecules increased the ATFP values.(4) The ATFPs were p H-dependent, the values of which were lowest at the neutral condition and highest at the acid condition.(5) The ATFPs increased and then decreased as the chlorine dosage increased. | 10 | Integrating structure and function:mapping the hierarchical spatial heterogeneity of urban landscapes显示文摘Background:Cities are social-ecological systems characterized by remarkably high spatial and temporal heterogeneity,which are closely related to myriad urban problems.However,the tools to map and quantify this heterogeneity are lacking.We here developed a new three-level classification scheme,by considering ecosystem types(level 1),urban function zones(level 2),and land cover elements(level 3),to map and quantify the hierarchical spatial heterogeneity of urban landscapes.Methods:We applied the scheme using an object-based approach for classification using very high spatial resolution imagery and a vector layer of building location and characteristics.We used a top-down classification procedure by conducting the classification in the order of ecosystem types,function zones,and land cover elements.The classification of the lower level was based on the results of the higher level.We used an objectbased methodology to carry out the three-level classification.Results:We found that the urban ecosystem type accounted for 45.3%of the land within the Shenzhen city administrative boundary.Within the urban ecosystem type,residential and industrial zones were the main zones,accounting for 38.4%and 33.8%,respectively.Tree canopy was the dominant element in Shenzhen city,accounting for 55.6%over all ecosystem types,which includes agricultural and forest.However,in the urban ecosystem type,the proportion of tree canopy was only 22.6%because most trees were distributed in the forest ecosystem type.The proportion of trees was 23.2% in industrial zones,2.2%higher than that in residential zones.That information“hidden”in the usual statistical summaries scaled to the entire administrative unit of Shenzhen has great potential for improving urban management.Conclusions:This paper has taken the theoretical understanding of urban spatial heterogeneity and used it to generate a classification scheme that exploits remotely sensed imagery,infrastructural data available at a municipal level,and object-based spatial analysis.For effective planning and management,the hierarchical levels of landscape classification(level 1),the analysis of use and cover by urban zones(level 2),and the fundamental elements of land cover(level 3),each exposes different respects relevant to city plans and management. | Yuguo Qian Weiqi Zhou Steward T.A.Pickett Wenjuan Yu Dingpeng Xiong Weimin Wang Chuanbao Jing | 2020 | Ecological Processes2020,9,1: | 1 |

| 17 | SUPER Score Contributes to Warning and Management in Early-Stage COVID-19显示文摘Background:Some COVID-19 patients deteriorate to severe cases with relatively higher case-fatality rates,which increases the medical burden.This necessitates identification of patients at risk of severe disease.Early assessment plays a crucial role in identifying patients at risk of severe disease.This study is to assess the effectiveness of SUPER score as a predictor of severe COVID-19 cases.Methods:We consecutively enrolled COVID-19 patients admitted to a comprehensive medical center in Wuhan,China,and recorded clinical characteristics and laboratory indexes.The SUPER score was calculated using pa-rameters including oxygen saturation,urine volume,pulse,emotional state,and respiratory rate.In addition,the area under the receiver operating characteristic curve(AUC),specificity,and sensitivity of the SUPER score for the diagnosis of severe COVID-19 were calculated and compared with the National Early Warning Score 2(NEWS2).Results:The SUPER score at admission,with a threshold of 4,exhibited good predictive performance for early identification of severe COVID-19 cases,yielding an AUC of 0.985(95%confidence interval[CI]0.897-1.000),sensitivity of 1.00(95%CI 0.715-1.000),and specificity of 0.92(95%CI 0.775-0.982),similar to NEWS2(AUC 0.984;95%CI 0.895-1.000,sensitivity 0.91;95%CI 0.587-0.998,specificity 0.97;95%CI 0.858-0.999).Com-pared with patients with a SUPER score<4,patients in the high-risk group exhibited lower lymphocyte counts,interleukin-2,interleukin-4 and higher fibrinogen,C-reactive protein,aspartate aminotransferase,and lactate dehydrogenase levels.Conclusions:In conclusion,the SUPER score demonstrated equivalent accuracy to the NEWS2 score in predicting severe COVID-19.Its application in prognostic assessment therefore offers an effective early warning system for critical management and facilitating efficient allocation of health resources. | Yuan Bian Qi Han Yue Zheng Yu Yao Xinhui Fan Ruijuan Lv Jiaojiao Pang Feng Xu Yuguo Chen | 2023 | Infectious Medicine2023,2,4: | 0 |
| 18 | Establishment of highly efficient transgenic system for black soldier fly(Hermetia illucens)显示文摘The black soldier fly(BSF),Hermetia illucens,is a promising insect for miti-gating solid waste problems as its larvae are able to bioconvert organic waste into valuable biomass.We recently reported a high-quality genome assembly of the BSF;analysis of this genome sequence will further the understanding of insect biology and identify genes that can be manipulated to improve efficiency of bioconversion.To enable genetic manip-ulation of the BSF,we have established the first transgenic methods for this economically important insect.We cloned and identified the ubiquitous actin5C promoter(Hiactin5C-p3k)and 3 endogenous U6 promoters(HiU6:1,HiU6:2,and HiU6:3).The Hiactin5C pro-moter was used to drive expression of a hyperactive variant of the piggyBac transposase,which exhibited up to 6-fold improvement in transformation rate when compared to the wild-type transposase.Furthermore,we evaluated the 3 HiU6 promoters using this trans-genic system.HiU6:1 and HiU6:2 promoters provided the highest knockdown efficiency with RNAi and are thus promising candidates for future Clustered Regularly Interspaced Short Palindromic Repeats(CRISPR)development.Overall,our findings provide valuable genetic engineering toolkits for basic research and genetic manipulation of the BSF. | Zongqing Kou Xingyu Luo Yuguo Jiang Bihui Chen Yu Song Yaohui Wang Jun Xui Jeffery K.Tomberlin Yongping Huang | 2023 | Insect Science2023,30,4: | 0 |
| 19 | A novel predictor of unsustained return of spontaneous circulation in cardiac arrest patients through a combination of capnography and pulse oximetry: a multicenter observational study显示文摘BACKGROUND:Unsustained return of spontaneous circulation(ROSC)is a critical barrier to survival in cardiac arrest patients.This study examined whether end-tidal carbon dioxide(ETCO_(2))and pulse oximetry photoplethysmogram(POP)parameters can be used to identify unsustained ROSC.METHODS:We conducted a multicenter observational prospective cohort study of consecutive patients with cardiac arrest from 2013 to 2014.Patients’general information,ETCO_(2),and POP parameters were collected and statistically analyzed.RESULTS:The included 105 ROSC episodes(from 80 cardiac arrest patients)comprised 51 sustained ROSC episodes and 54 unsustained ROSC episodes.The 24-hour survival rate was significantly higher in the sustained ROSC group than in the unsustained ROSC group(29.2%vs.9.4%,P<0.05).The logistic regression analysis showed that the difference between after and before ROSC in ETCO_(2)(ΔETCO_(2))and the difference between after and before ROCS in area under the curve of POP(ΔAUCp)were independently associated with sustained ROSC(odds ratio[OR]=0.931,95%confi dence interval[95%CI]0.881-0.984,P=0.011 and OR=0.998,95%CI 0.997-0.999,P<0.001).The area under the receiver operating characteristic curve ofΔETCO_(2),ΔAUCp,and the combination of both to predict unsustained ROSC were 0.752(95%CI 0.660-0.844),0.883(95%CI 0.818-0.948),and 0.902(95%CI 0.842-0.962),respectively.CONCLUSION:Patients with unsustained ROSC have a poor prognosis.The combination ofΔETCO_(2) andΔAUCp showed signifi cant predictive value for unsustained ROSC. | Jing Yang Hanqi Tang Shihuan Shao Feng Xu Yangyang Fu Shengyong Xu Chen Li Yan Li Yang Liu Joseph Harold Walline Huadong Zhu Yuguo Chen Xuezhong Yu Jun Xu | 2024 | World Journal of Emergency Medicine2024,15,1: | 0 |
